Tanglehead (Heteropogon contortus) is a native Hawaiian grass that has been used in restoration and has potential for expanded re-vegetation use. Although interest and demand for tanglehead re-vegetation has increased, the supply of tanglehead seeds has remained limited as a result of a lack of seed production protocols addressing seed dormancy. Smoke water from burning vegetation may provide an economical and practical seed treatment because aerosol smoke has been reported to stimulate tanglehead seed germination. Dose rate and side-by-side comparison studies were conducted to evaluate the germination stimulation efficacy of food-grade liquid smoke, xylose smoke-infused water, tanglehead smoke-infused water, karrikinolide (KAR1), and cyanide (i.e., mandelonitrile and potassium cyanide). Optimum smoke water dilutions were 1% v/v for food-grade liquid smoke and undiluted for xylose smoke-infused water and tanglehead smoke-infused water. KAR1 was not stimulatory at concentrations between 0.0067 and 66.7 μm. Potassium cyanide stimulated tanglehead seed germination at concentrations between 50 to 500 μm. Germination was promoted to even greater levels with the cyanohydrin, mandelonitrile, indicating a role for benzaldehyde (a byproduct of mandelonitrile decomposition) in stimulating tanglehead seed germination. Benzaldehyde was confirmed to be stimulatory at concentrations between 50 to 100 μm. The presence of cyanide at stimulatory levels was confirmed in tanglehead smoke-infused water (i.e., ≈100 μm), but not in food-grade liquid smoke or xylose smoke-infused water. Germination with non-cyanide-containing smoke waters indicates the presence of other compounds in smoke that can stimulate tanglehead germination. In the side-by-side comparison study, food-grade liquid smoke (1% v/v) and undiluted tanglehead smoke-infused water provided consistent germination stimulation comparable to 500 μm potassium cyanide. Undiluted xylose smoke-infused water did not provide significant germination stimulation in the comparison studies. This may be the result of differences in seed batch sensitivity to the germination stimulant, seed storage duration as well as subtle differences in the preparation of xylose smoke-infused water.

The purpose of the research study the viability and vigor of plants in the nursery and look for the optimum concentration of H2SO4 solution at each storage duration that gives the distance of the highest seed viability in the nursery. Experiments conductedin the seed laboratory and in the nursery Faculty of Agriculture Winaya Mukti University Tanjungsari using randomized block design (RBD) factorial, first factor the time and concentration of sulfuric acid solution: Soaking 6 minutes in H2SO4 0.25%, 0.50%, 0.75% and 1.0%. Factor II is a seed storage duration: without being stored, 1, 2, 3 months. Analysis of the data using linear quadratic regression method. The experimental results show that the interaction between seed storage time and concentration of sulfuric acid solution on seed germination, growth speed of seeds, and seed vigor. Without storage time and concentration of sulfuric acid solution 0.75% gives the best effect compared to other treatments. At the time of storage, followed without the optimum sulfuric acid concentration as much as 0.783% gives the percentage of seed germination was 93.521%.

Field experiment on evaluation of insecticidal seed treatments against white grub Holotrichia consanguinea Blachard infesting groundnut was conducted during kharif 2018 and 2019 at farmers field. The nine insecticides evaluated as seed treatment against white grub infesting groundnut, the lowest per cent plant morality and number of grub/m2 was observed in treatment of clothianidin 50 WDG @ 2 g a.i. /kg and imidacloprid 40 + fipronil 40 WG @ 3 g a.i. /kg. These treatments were produced 2329 and 2296 yield of pod, 3970 & 3780 kg/ha yield of halum, respectively. Increased in yield over control in these treatments was recorded 57.79 and 55.56 per cent in pod, 76.29 and 64.65 per cent in halum. Whereas avoidable yield loss recorded of these treatments was recorded 36.63 and 35.71 per cent in pod, 43.27 and 39.27 per cent in halum. However, the highest 1:28.32 NICRB was recorded in treatment of chlorpyriphos 20 EC @ 30 ml a.i. /kg. The NICRB of clothianidin 50 WDG @ 2 g a.i. /kg and imidacloprid 40 + fipronil 40 WG @ 3 g a.i. /kg were recorded 1:14.95 and 1:9.20, respectively. The evaluated insecticides had no adverse effect on seed germination

In Vitro and In Vivo studies on the efficacy of fungicides and biopesticides. Among the fungicides, in Carbedazim to the most effective as they have inhibited the mycelia growth completely of the test fungus, and Benomyl, Topsin - M, Ridomil,Vitavax were found the next best in inhibiting the mycelial growth of the pathogen up to 92.11% to 83.46% respectively. Sadabahar was least effective plant extracts which causes 42 mm of radial growth and inhibited the growth of the only 19.23%. In Vivo condition the maximum seed germination (95.50% and 95.33%), minimum wilt incidence (5.16% and 3.65%) and highest grain yield (10.50 q/ha and 10.35 q/ha) was found seed treatment with Carbendazim (0.2%). Among the test plant extracts Tulsi was lested effective, which show the minimum seed germination (80.00% and 77.50%), maximum wilt incidence (15.70% and 14.10%), and lowest grain yield (3.92 q/ha and 4.17 q/ha).

The effect of some respiratory inhibitors on light-induced Paulownia tomentosa Steud. seed germination was studied. Millimolar solution of sodium azide was sufficient to completely prevent germination induced by a 5-min red light pulse. The inhibitory effect of azide was absent if seeds were rinsed before phytochrome activation by light. Sodium azide was effective only if present in the period of Pfr activity. The escape time from azide inhibition compared to the escape from far-red light action, was delayed for about 24 hours. When azide was applied after phytochrome activation, its effect depended on how long it was present in the incubation medium. The removal of azide allowed full restoration of germination by another red light pulse and the far-red escape time did not differ from the escape of untreated, i.e. water-imbibed seeds. Potassium cyanide alone did not produce any effect in light-stimulated germination of these seeds. However, it counteracted the inhibitory effect of azide in light-stimulated germination, if applied simultaneously at a concentration three times higher.

Abstract. Puspitaningtyas DM, Handini E. 2021. Seed germination evaluation of Phalaenopsis amabilis in various media for long-term conservation. Biodiversitas 22: 5231-5238. Phalaenopsis amabilis (L.) Blume (moon orchid) is one of Indonesia's national flowers, which is mentioned in Presidential Decree No. 4/1993. Study on the orchid seed of P. amabilis was conducted to observe the longevity of seeds storage. The purpose of this study was to determine the viability of P. amabilis seeds after storage for years, then to predict the life span of the seeds, and to find out the best media germination for P. amabilis seeds. After harvesting, P. amabilis seeds were stored at –20 °C and the germination tests were carried out periodically in different periods until the seeds lose their viability to determine the optimum conditions for long-term seed storage. Four trial media cultures have been used to test seed germination by means of in vitro culture. The result showed that seeds of P. amabilis were able to germinate in 4 media i.e. Knudson C, modified Knudson C, modified Vacin & Went and modified leaf fertilizer. The best germination was on leaf fertilizer medium. Based on the research showed that the viability seeds of P. amabilis decreased sharply after 6 years stored. By using regression analysis, the life span of the seeds could be predicted can reach 9 years of storage which is sowing on the best media (leaf fertilizer).

Rhizoctonia solani Kuhn. causing banded leaf and sheath blight diseases is one of the important fungi of corn world wide. The fungus is commonly controlled by using fungicide because no resistant variety available. The objective of the study was to develop a seed treatment formulation of the selected Bacillus subtilis to control R. solani in corn. The study was conducted in the Department of Plant Pathology, College of Agriculture, University of the Philippines Los Bañòs, College, Laguna from May 2004 to August 2005, using sweet corn var. IPB Supersweet as test plant. Corn seeds were surface sterilized for 10 minutes in 1% sodium hypochlorite solution and 5% ethanol, washed thrice with sterile distilled water and air-dried. The seeds were coated with formulated B. subtilis BR23 and used for several experiments, such as evaluation for their germination and growth in the laboratory, effectively on R. solani in the baked and nonbaked field soil under greenhouse condition, and in the microplots artificially infested with R. solani. The treatment was compared with other standard seed treatment of synthetic fungicides such as captan (10 g per kg seeds) and metalaxyl (10 g per kg seeds). The experiments were designed in a completely random design with three replications. Parameters observed were seed germination, plant height, disease scores, and plant yield. Laboratory formulated B. subtilis BR23 used as seed treatment had no detrimental effects on seed germination and seedling vigor. In microplots artificially infested with a selected highly virulent R. solani, seed treatment with the same formulation increased grain yield by 27% compared to that of the control captan seed treatment with 14.4%. The studies showed the potential of B. subtilis BR23 for commercialization as a seed treatment for the control of banded leaf and sheath blight disease (R. solani) in corn.
