Characterization, ACE inhibitory and Antioxidative Properties of Peptide Fractions Obtained from White Shrimp (Litopenaeus vannamei)

Background: Aquatic organisms are considered to be an important source of bioactive peptides with a high antioxidant and antihypertensive capacity. Therefore, the objective of this study was to hydrolyse peptide fractions from white shrimp (Litopenaeus vannamei) muscle by Alcalase and Protamex and to evaluate the angiotensin I-converting enzyme (ACE) inhibitory and the antioxidant activities. Methods: Protein hydrolysates of White shrimp were obtained by enzymatic hydrolysis using Alcalase and Protamex until the degree of hydrolysis reached 10% and 20%. Peptide fractions were obtained from White shrimp protein hydrolysates by ultrafiltration using membranes with sizes of 10 and 3 kDa. The antioxidant activity was evaluated for the three peptide fractions (F1: >10 kDa, F2: 3-10 kDa and F3: <3 kDa). To measure the antihypertensive activity, fractions with molecular sizes of less than 3 kDa were used. Results: The fractions obtained with Alcalase showed greater inhibitory effects on the ACE. In general, the molecular weight of the fractions influenced the antioxidant activity, with fractions smaller than 3 kDa having a high capacity for sequestering the DPPH radical, while peptide fractions with a size greater than 10 kDa presented higher reducing power. However, in capturing the ABTS radical, a high antioxidant capacity was observed for both fractions. Conclusion: The results suggest white shrimp would be an attractive raw material for the manufacture of antioxidant and anti-hypertensive nutraceutical ingredients.

Background: There has been some evidence that proteins are potentially excellent source of antioxidants, antihypertensive and antimicrobial peptides and enzymatic hydrolysis is an effective method to release these peptides from protein molecules. The functional properties of protein hydrolysates depends on the protein substrate, the specificity of the enzymes, the conditions used during proteolysis, degree of hydrolysis, and the nature of peptides released including molecular weight, amino acid composition, and hydrophobicity.Context and purpose of this study: The biomass of Kluyveromyces marxianus was considered as a source of ACE inhibitory, antioxidant and antimicrobial peptides. Results: Autolysis and enzymatic hydrolysis were completed respectively, after 96 h and 5 h. Overall, trypsin (18.52% DH) and chymotrypsin (21.59% DH) treatments were successful in releasing antioxidant and ACE inhibitory peptides. Autolysate sample (39.51% DH) demonstrated a poor antioxidant and ACE inhibitory activity compared to trypsin and chymotrypsin hydrolysates. The chymotrypsin 3-5 kDa (301.6±22.81 μM TEAC/mg protein) and trypsin< 3 kDa (280.16±39.16) permeate peptide fractions showed the highest DPPH radical scavenging activity. The trypsin <3 kDa permeate peptide fraction showed the highest ABTS radical scavenging (1691.1±48.68 μMTE/mg protein) and ACE inhibitory (IC50=0.03±0.001 mg/ml) activities. The fraction (MW=5-10 kD) obtained after autolysis treatment showed antibacterial activity against St. aureus and Lis. monocytogenes in well diffusion screening. The minimum inhibitory concentration (MIC) value was 13.3 mg/ml against St. aureus and Lis. monocytogenes calculated by turbidimetric assay and it showed bactericidal activity against St. aureus at 21.3 mg/ml protein concentration. Conclusions: Taken together, the results of this study reveal that K. marxianus proteins contain specific peptides in their sequences which can be released by enzymatic hydrolysis and autolysis. It was reported that 8-hydroxygenistein (8-OHG) was synthesized by methylation, bromination, methoxylation, and demethylation using cheap and readily available biochanin A as raw material. All synthesized products were structurally confirmed by ultra-high-performance liquid chromatography (UHPLC), infrared spectroscopy, mass spectrometry, 1H-nuclear magnetic resonance (NMR), and 13C-NMR. In addition, we examined the antioxidant capacity of 8-OHG using 6 different methods such as 1,1-diphenyl-2-picrylhydrazyl radical scavenging, 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonate) radical (ABTS) scavenging, nitric oxide radical (NO) scavenging, superoxide radical (O2 −•) scavenging, reducing power assay, and total antioxidant activity using ascorbic acid (VC) as a positive control. Compared with VC, 8-OHG exhibited higher total antioxidant activity and stronger scavenging activity on ABTS, NO, and O2 −•. These results indicate that 8-OHG is an excellent antioxidant agent and may be effective in preventing damage induced by free radical.

Microalgae protein hydrolysates (MPH) were obtained by enzymatic hydrolysis of defatted microalgae meal using neutral protease. The protein recovery, degree of hydrolysis, and the antioxidant activities of the hydrolysates were investigated. The results demonstrated that hydrolysates prepared by neutral protease at 50 °C for 4 h exhibited the strongest antioxidant activity. Under these conditions, the 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical scavenging activity and the reducing power of the hydrolysates were 68.3%, 50.8% and 1.303, respectively.

Semen Allii Fistulosi (PSAF) is the seed of Allium fistulosum L. of the Liliaceae family. The purpose of this study was to extract, characterize, and evaluate the antioxidant activity in vitro of proteins. Using single factor and orthogonal design, the optimum conditions of extraction were determined to be as follows: extraction time 150 min, pH 8.5, temperature 60 °C, and ratio (v/w, mL/g) of extraction solvent to raw material 35. The isoelectric point of the pH was determined to be about 4.4 and 10.2, by measuring the protein content of PSAF solutions at different pH values. The amino acid composition of PSAF was determined by high performance liquid chromatography (HPLC), and the results suggested that the species of amino acids contained in the PSAF was complete. Sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS–PAGE) analysis showed the molecular weight was mainly between 40 and 55 kDa, and Fourier-transform infrared spectroscopy (FTIR) characterized prevalent protein absorption peaks. PSAF exhibited potent scavenging activities against DPPH assays, via targeting of hydroxyl and superoxide radicals, while chelating Fe2+ activity and demonstrating weak reducing power. This work revealed that PSAF possessed potential antioxidant activity in vitro, suggesting potential for use of PSAF as a natural antioxidant.

Protein hydrolysates from Tuna frame (TFPH) and Pollock frame (PFPH) were prepared by papain, respectively.The yield, the basic composition content, the antioxidant activity and functional properties (solubility, emulsifying and foaming ability) and the degree of hydrolysis of the protein hydrolysates were evaluated. Results suggest that solubility, antioxidant activity of protein hydrolysate from Pollock frame are better than that of tuna frame, but the yield is lower than that of tuna frame.

Deer blood was hydrolyzed using Alcalase with hydrolysis time ranged form 0 to 6 h, and the degree of hydrolysis (DH) of protein hydrolysates increased with increasing hydrolysis time (P < 0.05). The reducing power, radicals scavenging activities and Cu2+-chelation ability of deer blood hydrolysate (DBH) significantly enhanced with increasing hydrolysis time (P < 0.05). The antioxidant activity of DBH, indicated by thiobarbituric acid-reactive substance (TBARS) values in a liposome-oxidizing system, increased with increasing DH (P < 0.05). The results indicated that antioxidant activity of DBH depended on hydrolysis time, and the hydrolyzed deer blood could be a potent food antioxidant.

ABSTRACTThis study was conducted to evaluate the digestibility of biofloc meal collected from catfish culture as a feed raw material for Pacific white shrimp Litopenaeus vannamei diet. A basal feed with 43% crude protein content was used as a control diet and mixed with 2% of binders and 0.5 % of Cr2O3 as a marker for digestibility. The experimental diets were made by mixing 67.5% of the basal diet with 30% of biofloc meal, 2% of binders and 0.5 % of Cr2O3. Nine units of glass tanks (90 cm ×40 cm× 35 cm) filled with 100 L seawater were used as the experimental culture units. White shrimp with an average body weight of 5.61 ± 0.09 g was randomly distributed to each experimental tank at a density of 20 shrimp/tank. The feed was offered at a level of 5% shrimp biomass per day at a frequency of four times a day. The results showed that the dry matter digestibility of feed with 30% biofloc meal in shrimp were similar to that of the reference diet. However, protein and fat digestibility of feed containing biofloc meal were considerably higher than those of the reference diet. Feeding shrimp with 30% biofloc meal diet resulted in higher survival and specific growth rate and lower feed conversion ratio than those of the control. The digestibility of bioflocs dry matter, protein and lipid in Pacific white shrimp obtained in this study were 54.9%, 76.3% and 79.3%, respectively.Keywords: biofloc, digestibility, catfish, shrimp  ABSTRAKPenelitian ini dilakukan untuk mengevaluasi kecernaan tepung bioflok yang diambil dari media pemeliharaan ikan lele sebagai bahan pakan udang vaname Litopenaeus vannamei. Pakan yang mengandung kadar protein 43% digunakan sebagai pakan control dan dicampur dengan 2% binder dan 0.5% Cr2O3 sebagai marker untuk kecernaan. Pakan perlakuan dibuat dengan mencampurkan 67.5% pakan control dengan 30% tepung bioflok, 2% binder dan 0.5% Cr2O3. Penelitian menggunakan sembilan unit akuarium (90 cm ×40 cm ×35 cm) yang diisi 100 L air laut. Udang vaname dengan bobot rata-rata 5.61 ± 0.09 g ditebar secara acak pada setiap akuarium perlakuan pada kepadatan 20 ekor/akuarium. Pakan diberikan dengan tingkat pemberian pakan 5% biomassa per hari sebanyak empat kali sehari. Hasil penelitian ini menunjukkan bahwa kecernaan pakan dengan 30% tepung bioflok tidak berbeda nyata dengan pakan acuan. Namun kecernaan protein dan lemak pakan yang mengandung tepung bioflok terlihat lebih tinggi daripada pakan kontrol. Pemberian pakan dengan tepung bioflok sebanyak 30% juga menghasilkan tingkat kelangsungan hidup, laju pertumbuhan spesifik udang dan rasio konversi pakan yang lebih baik daripada udang yang diberi pakan kontrol. Kecernaan bahan, protein dan lemak tepung bioflok pada udang yang didapat dalam penelitian masing-masing adalah 54.9%, 76.3% dan 79.3%.Kata-kata kunci: bioflok, ikan lele, kecernaan, udang
