Background: The topical routes of administering drug have certain visible advantages as direct application of a drug to the site of action and persistence of action for prolonged duration. More stability and better application property of gel cause its use for topical application. Objectives: To determine and compare antibacterial efficacy of commercially available different dental gels. Methods: For the evaluation of antibacterial efficacy of commercially available different antiseptic gels on Streptococcus mutans and Lactobacillus acidophilus well-diffusion method was used. To assess the antibacterial efficacy after incubation in appropriate culture medium diameter of zone of inhibition was measured. Results: Maximum zone of inhibition was found with Curenext gel (40 mm,40mm) followed by Hexigel (30 mm, 21mm) Mucopain (17 mm, 17mm) and Dologel (7 mm, 16mm) against S. mutans and L. acidophilus respectively. Analysis of variance (ANOVA) and Post Hoc Test were used to analyze results statistically. The difference between commercially available different dental gels was statistically significant (P < 0.001) at 5% of level of significance. Conclusion: In the present study, it was seen that Curenext gel was more effective followed by Hexigel, Mucopain gel and Dologel on S. Mutans and L. Acidophilus.

Introduction: Fluoride toothpastes are a major reason for the decline in dental caries globally. For fluoride toothpaste to be effective, an adequate amount of Total Soluble Fluoride (TSF) must be available in the toothpastes. Aim: To determine and compare the Total Fluoride (TF), TSF and pH among various toothpaste brands. Materials and Methods: An in-vitro study was conducted during October 2019 on 20 toothpaste samples, which belonged to four groups namely, herbal toothpastes group, nonherbal toothpastes group, medicated toothpastes group and kids toothpastes group. Analysis of the samples was done at the Department of Environmental Engineering Laboratory, National Institute of Technology, Surathkal, Karnataka, India. The TF and the TSF concentration was determined using fluoride ion electrode. The pH was determined using pH meter. Statistical analysis was performed using Statistical Package for Social Sciences (SPSS version 19.0). Analysis of variance (ANOVA) was used by Tukey’s post-hoc test to determine the difference between groups. The level of significance was kept at 0.05. Results: The results showed that there was a significant difference between the toothpaste types for mean TF concentration (p=0.004). The post-hoc test showed that there was a significant difference between herbal and nonherbal toothpastes (p=0.041) where the mean TF concentration for nonherbal toothpastes was 1095.20 ppm when compared to herbal toothpastes was 704.40 ppm. The results showed that there was a siginificant difference between the toothpaste types for Mean TSF concentration (p<0.003). There was a significant difference between medicated and kids toothpastes (p=0.024) where the mean total soluble fluoride concentration for medicated toothpastes was 938.60 ppm when compared to the kids toothpastes was 521.20 ppm. Conclusion: The present study showed that pH of all the toothpastes were either neutral or alkaline and the TSF concentration was less when compared to TF concentrations.

Aim. The purpose of this in vitro study was to assess the antimicrobial efficacy of Biopure MTAD against E. faecalis in contaminated root canals. Materials and Methods. Forty-two single rooted extracted human teeth were inoculated with E. faecalis and incubated for four weeks. The samples were divided in two control and five experimental groups irrigated with 1.5% sodium hypochlorite solution (NaOCl); 3% NaOCl; BioPure MTAD; 1.5% NaOCl/17% EDTA; or 3% NaOCl/17% EDTA. After a one-week incubation, complete disinfection was confirmed by the absence of turbidity in the incubation media. Dentin shavings were taken from samples with no turbidity to verify whether E. faecalis was present in dentin tubules. Results were analyzed statistically using Fisher's exact test, with the level of significance set at . Results. Statistical analysis of the data obtained at Day 7 and after dentin shaving analysis showed that BioPure MTAD had significantly greater antibacterial activity than 1.5% NaOCl, 1.5% NaOCl/17% EDTA and 3% NaOCl/17% EDTA. No significant difference was detected between MTAD and 3% NaOCl. Conclusions. These findings suggest that BioPure MTAD possesses superior bactericidal activity compared with NaOCl and EDTA against E. faecalis.

Background: Attachments are composite auxiliaries that are used during a clear aligner orthodontic therapy to achieve difficult tooth movements. Two important factors are the planned configuration and the actual position of these auxiliary elements to obtain the desired force system. The aim of the present study was to evaluate the role of different composite materials in the correct reproduction of attachment shape and position. Methods: The materials that were considered in the study were a flowable resin, a dental restorative material, and an orthodontic composite. The attachments were created on three models of extracted teeth. Once the impressions were performed, 25 attachments of different shapes were added onto each virtual model to obtain the necessary templates to make the attachments. Each tested material was used to create a set of 25 attachments that were then scanned with an accuracy of 10μm. The resultant STL (stereolithography) files were superimposed onto the ones from the initial virtual plan, through Geomagic software, and the aligned scans were then compared while using a color map. The parameters that were calculated to make a comparison between the created attachments and the ideal ones were the maximum deviation in defect and in excess, the overflow, and the volume’s difference. In addition to these measurements comparing the three above-mentioned groups, the difference in volume between all the ideal and realized samples were analyzed. To test for differences among the three groups, a one-way Analysis of variance (ANOVA) was used with a Bonferroni post-hoc test. The level of significance was set at p < 0.05. Results: No statistically significant results were found between the three groups regarding the maximum value in defect, the maximum value in excess, and the minimum value as control, while a statistically significant difference was found between the overflow of orthodontic resin when compared to the flowable composite. Conclusions: The three materials that were used in this study were appropriate for attachment fabrication. The fidelity of attachment reproduction was similar when using the three different composites. The orthodontic composite showed more overflow when compared with the flowable one.

Infection caused by microorganisms play a crucial role in the induction of inflammation of pulpal as well as the periapical tissues. Factors like improper disinfection or obturation of the root canal space contributes to the failure of the root canal treatment. The aim of the study was to modify the contents of MTA-based sealer with nanoparticles and check for its antibacterial efficacy against E. faecalis. Silver nanoparticles (10 nm) were incorporated in MTA based sealer at various concentrations to form the test product. The antibacterial efficacy of the modified sealer was tested by well diffusion test on E. faecalis. The zone of inhibition (mm) was checked for each test product. Analysis of the results showed significant diameters of zones of inhibition (mm) as compared to sealer without nanoparticles. The zone of inhibition increased with the increase in the concentration of silver nanoparticles. The silver nanoparticle incorporated sealer can be used in the clinical setup to prevent reinfection of the root canal system and ensure the success of the root canal treatment.

Introduction: Endodontic retreatment seeks reentry to the root canal system to remove all filling material, identification and correction of pathological or iatrogenic defects. Objective: This study aimed to compare the dissolving capacity of eucalyptus oil, orange oil, and distilled water versus three types of endodontic cements. Materials and methods: Two hundred and four stainless steel molds were fabricated, where each cement sample was placed to the edge and left to set in the incubator, simulating normal oral conditions for 7 days. Subsequently, the solubility of each cement was analysed by immersing the samples in eucalyptus and orange oil for ten minutes in a static and ultrasonic environment. The solubility of the cement was evidenced by the difference between the weight before and after contact with the oils, data were statistically analysed with the ANOVA test and the post hoc Tukey bootstrap means test with 1,000 repetitions. Results: The control group and eucalyptus oil having no significant effect (p>0.05), the ultrasonic environment was found to be effective (p<0.05), whereas orange oil had better dissolution effects (p=0.004). Conclusions: The most susceptible cement was Sealapex, while the least soluble was MTA-Fillapex in all the solvents studied.

The treatment of peri-implantitis implies the decontamination of the surface of the fixture. This study aims to analyze the effect of the erbium-doped yttrium aluminum garnet laser (Er: YAG) on sandblasted and acid-etched (SLA) titanium. 30 titanium SLA disks were divided into three groups. In Group 1, the disks were left intact; on the contrary, both Groups 2 and 3 were irradiated with the Er: YAG laser at different settings, with a pulse duration of 300 μs and a period of 30 s. Group 2 was irradiated at 1 W and 100 mJ/pulse and Group 3 at 4 W and 400 mJ/pulse. The superficial changes at chemical, nano, and microscopical levels were detected through the use of Fourier-transform infrared spectroscopy, atomic force microscopy, and scanning electron microscope. The Kruskal–Wallis test, followed by the Dunn–Bonferroni Post Hoc analysis, detected the presence of statistically significant differences among the groups. The level of significance was p ≤ 0.05. Results showed that Er: YAG irradiation promoted a significant (p < 0.05) increase of oxides and a decrease of microscopical roughness and porosity on SLA disks. However, the protocol tested on group 3 seemed to be too aggressive for the titanium surface, as shown by the presence of micro-cracks and signs of coagulation, melting, and microfractures. In conclusion, Group 2 showed significantly minor surface alterations with respect to Group 3, and the increase of superficial oxide level, the decrease of porosity, and micro-roughness represent a positive alteration that could protect the materials against bacterial adhesion.

Calcium hydroxide (CH) is an intracanal medicament that has been widely used in endodontics, which can eliminate bacteria because of its high alkalinity. However, E. faecalis is resistant to CH. Triple antibiotic paste (TAP) is a mixture of ciprofloxacin, minocycline, and metronidazole, and is highly effective against E. faecalis. Hence the main aim of this study was to find the antimicrobial efficacy of CH and TAP combination against E. faecalis. The study was done by agar diffusion method, three wells were punched in Tryptone soya agar and filled with CH, TAP, and the combination of both. The zone of inhibition values was recorded and subjected to statistical analysis using SPSS. One way ANOVA and Post Hoc tests were used to compare the means. The combination of CH with TAP was found to be significantly better than CH and TAP used alone (p-value <0.05).Within the limits of this study, it can be concluded that TAP is more efficient when compared with CH,The combination of CH and TAP proved to be more effective when compared to the two when used alone.

Aim and Objectives: To compare and evaluate the antibacterial efficacy of various nanoparticles incorporated in orthodontic primer with that of conventional antimicrobial agents at different concentrations on Streptococcus mutans ( S. mutans) strain. Materials and Methods: Transbond XT Primer was mixed with 2.5% and 5% benzalkonium chloride (BAC), 0.2% and 2.5% chlorhexidine, 1% and 3% titanium dioxide (TiO2) nanoparticles, 0.2% and 0.5% nanohydroxyapatite, and 0.2% and 0.5% silica-doped nanohydroxyapatite powders. Antibacterial activity against S. mutans for all the materials was evaluated by the disk diffusion method for periods of 48 (T1) and 72 (T2) hours. Results: There was a significant increase in the antimicrobial activity of the orthodontic primer modified by the addition of antibacterial agents. The highest zone of inhibition against S. mutans was observed for silica-doped nanohydroxyapatite of 0.5% (11.03 mm) among all the nanoparticles, which was similar to the conventional antibacterial agents used in our study. Conclusions: • Among all the groups, BAC at 5% concentration showed the highest antimicrobial activity, and the least activity was exhibited by 1% TiO2 nanoparticles. • Silica-doped nanohydroxyapatite at 0.5% expressed the greatest antibacterial activity among all the nanoparticles. • All the materials showed sustained antibacterial activity even after 72 hours.

The purpose of this study was to evaluate the antibacterial efficacy of silver Nanoparticles, Nanoparticle calcium hydroxide and calcium hydroxide as intracanal medicament against Enterococcus faecalis bio film formed on root dentinThe Minimal inhibitory concentration test and Anti microbial test were conducted for silver nanoparticle, Nanoparticle calcium hydroxide. Teeth were inoculated with E.faecalis strains for 1month to establish a standard mono species bio film model. Biofilm formation is confirmed using SEM. The intracanal medicaments were placed inside the canal and colony forming units (CFU) were counted before and 7 days later the placement of the medicament.MIC for AGNP is 25µg/200µl and for NCH is 12.5µg/200µl. The Antimicrobial test conducted shows zone of inhibition about 11mm and 14mm for AGNP and NCH respectively at 500µg. The calcium hydroxides do not show any antibacterial activity at this concentration. In antibacterial efficacy test using CFU count, the AGNP and NCH shows 75% and 61% of antibacterial efficacy respectively whereas calcium hydroxide has exhibited just 11% of antibacterial efficacy. Silver Nanoparticles and nanoparticle calcium hydroxide had statistically significant difference in the reduction in the number of colonies when compared to calcium hydroxide by post hoc turkey test and p-value of <0.01 was considered as significant in this study.

Aim: To evaluate the effect of Probiotic used in PMMA Temporary restorations on gingival inflammatory response by its action on the marginal gingival epithelial cells. Materials and Methods:  This is an in vitro Interventional pilot study using discs of  PMMA ( Group A ) and PMMA  incorporated with Probiotics (Group B . The effect of probiotics incorporated PMMA on strains of Porphyromonos gingivalis  grown on culture plates will  be assessed using zone of inhibition test and minimum inhibitory concentration was assessed . Data was recorded, tabulated and statistically evaluated using SPSS software. Results: The zone of Inhibition in the Group B was (Mean = 16.30mm) is comparatively higher than that of Group A with mean value 12.92mm. Results states that among the various concentration of Probiotic Lozenges (2.5, 5, 10, 15 and 20 µg/ml) used to determine the antibacterial activity , Highest mean zone of inhibition was observed with 20 µg/ml with 16 mm, followed by 15 µg/ml  with 14mm , 10 µg/ml  with 9mm, 5 µg/ml  with 11 mm & with the lowest inhibition observed in 2.5 µg/ml  with 8mm. There is significant correlation between concentration of probiotics and its antibacterial efficacy. The increase in concentration of probiotics in the PMMA discs is directly proportional to its antibacterial efficacy against P. gingivalis. Conclusion: Thus the study proved a significant correlation between Probiotic used in PMMA Temporary crowns and reduction in gingival Inflammation at the margins of the restoration which will be beneficial when used in crowns used in patients undergoing Orthodontic treatment or Immediate Implant Loading with acrylic crowns.
