Evaluation of the Anti-Hypercholesterolemic and Antioxidant Activity of Mentha pulegium (L.) Aqueous Extract in Normal and Streptozotocin- Induced Diabetic Rats

Objective:Mentha pulegium L., an aromatic plant belonging to Lamiaceae family, is widely used by local population against diabetes, hypertension and cardiovascular disorders. The present study aimed to evaluate the in vivo antihypercholesterolemic effect of aerial parts aqueous extract (A.P.A.E) of Mentha pulegium (M. pulegium) in normal and streptozotocin-induced diabetic rat. Additionally, the in vitro antioxidant activity of M. pulegium A.P.A.E has been estimated.Methods:The effect of M. pulegium (L.) A.P.A.E (20 mg of lyophilized A.P.A.E per kg body weight) on plasma lipid profile was investigated in normal and streptozotocin (STZ) diabetic rats (n=6) treated for 15 days with M. pulegium (L.) A.P.A.E oral administration. The antioxidant activity was evaluated using DPPH (1-1-diphenyl 2-picryl hydrazyl) radical scavenging activity. Estimation of total polyphenol contents in A.P.A.E of M. pulegium was determined with the Folin- Ciocalteu reagent by the method using gallic acid as a standard phenolic compound. Also, flavonoids were estimated according to the method based on the formation of a complex flavonoidaluminium.Results:The results show that 15 days of M. pulegium A.P.A.E oral administration alleviated hyperlipidemia in diabetic rats by lowering significantly (p<0.01) the plasma cholesterol levels without affecting the triglycerides (TG) levels significantly. However, no significant decrease in plasma cholesterol and plasma triglycerides has been shown in diabetic control rats. In addition, A.P.A.E oral administration has exerted some increasing activity on plasma HDL-c level (54.84 %), whereas the HDL-c level, in diabetic control rats, has significantly decreased (p<0.05). On the other hand, M. pulegium A.P.A.E showed a high amount of flavonoid (83.07 ± 0.58 mg EQ / g of extract) and phenolic compounds (239.08 ± 35.40 mg EAG/g of extract). Also, according to the DPPH radical scavenging activity, this aqueous extract has demonstrated a significant antioxidant activity.Conclusion:We conclude that 15 days of M. pulegium A.P.A.E oral administration exhibited a significant antihypercholesterolemic effect and has also demonstrated a not negligent increase in HDL-c levels without affecting the triglycerides levels. Furthermore, A.P.A.E exhibited a potent antioxidant activity. Therefore, phytochemical compounds and antioxidant activity of M. pulegium A.P.A.E may be seemingly implicated in the antihypercholesterolemic effect demonstrated in this study.

Many plants possess antioxidants that exhibit additive or synergistic activities. The antioxidant activities of the root of Curcuma longa Linn extracts extracted different solvents were investigated by using several established in vitro systems: α,α-diphenyl-β-picrylhydrazyl (DPPH) radical scavenging activity, hydrogen Peroxide scavenging activity (HPSA), nitric oxide radical scavenging activity (NOSA) and ferric reducing antioxidant power (FRAP). The result showed that methanol extract exhibited greater antioxidant activity in vitro which was statistically significant compared to the other extracts. Based on the in vitro results, the methanol extract was subjected to column chromatography. Six pooled fractions (FI-FVI) were evaluated for in vivo antioxidant activity in liver and kidney of alloxan-induced diabetic rats using a total of forty-five (45) rats which were grouped into nine (9) groups of five (5) rats. The in vivo antioxidants showed a significant decrease in superoxide dismutase (SOD), catalase (CAT) and gluthatione peroxidase (GPx) levels in both liver and kidney of Alloxan-induced diabetic rats. These changes were significantly reversed after treatment with methanol fraction II and the standard drug. Thus, Curcuma longa Linn may be useful in the management of diabetes and oxidative stress.

Objective: The main objective of this study was to evaluate the free radical scavenging activity of methanol (70%) and aqueous extract of G. montana leaves which is a traditionally used herb known for its hepatoprotective activity. Methods: The in vitro antioxidant activity of G. montana extract was determined using 1, 1-diphenyl-2-picrylhydrazyl radical (DPPH), 2,2′-azinobis-(3-ethylbenzothiaziline-6-sulfonate (ABTS), Hydrogen peroxide scavenging activity, Superoxide anion radical scavenging activity and Reducing Power ability at three different concentrations (1.78µg/ml, 3.57µg/ml and 7.14µg/ml). Results: The Results revealed similar observations between the methanol and aqueous extract with respect to standard and showed potent antioxidant activity. Ascorbic acid was used as a standard, which showed IC50 value 4.71µg/ml, whereas, methanol and aqueous extract showed 5.08 µg/ml and 5.69 µg/ml. Three different concentrations were used which showed a dose-dependent non-significant increase in percent inhibition. Conclusion: Findings indicate that this plant is a good source of antioxidant and can be used for the treatment of diseases as such medicinal plant extracts are natural products and they are comparatively safe, eco-friendly, less expensive and locally available. Hence, the validation of the effects of these herbal remedies will have to be undertaken for their wider acceptance.

This work was conducted to evaluate the antioxidant activity of the essential oil obtained from the aerial part of Mentha pulegium. The antioxidant power of the essential oil obtained by hydrodistillation using a Clevenger-type apparatus was evaluated by using three methods: free radical-scavenging activity, reducing power and liver lipid peroxidation assay. Results showed that Mentha pulegium oil displayed good quality according to its physicochemical characteristics, and a higher yield 5.1 ± 0.2%. The essential oil of Mentha pulegium showed a higher DPPH radical scavenging activity 90.54 ± 1.5 % at a concentration of 1000 µg/mL. This value was close to the results obtained with ascorbic acid 96.23 ± 1.2%, and catechin 94.50 ± 1.4%. This oil exhibited significant potential for reducing iron (the value observed by optical density was 1.8 ± 0.3), while ascorbic acid and catechin provided an OD of 2.069 ± 0.03 and 2.66 ± 0.016 at the same concentration of 1000 µg/mL. The tested oil protected against lipid peroxidation induced by Fe+2, and considerably increased the percentage of anti lipid peroxidation in a dose-dependent manner. The studied oil displayed a good degree of antioxidant activity and can be exploited in food and pharmaceutical industries.

Aims: The current investigation aimed to assess the antioxidant, antidiabetic and antilipidemic effects of the aqueous extract of aerial part of Cotula cinerea (C. cinerea). Background: Cotula cinerea (Del). which belongs to the Asteraceae family is commonly used traditionally for the treatment of diabetes. Objective: The objective of the study was to study the effect of the aqueous C. cinerea extract on glucose and lipid metabolism in normal and streptozotocin-induced diabetic rats using a single and repeated oral administration. Methods: A preliminary phytochemical screening and the quantification of phenolic and flavonoid contents as well as the antioxidant activity using three methods (DPPH, FRAP and ABTS) were carried out. The effect of a single and repeated (15 days of treatment) oral administration of the aqueous extract of aerial part of Cotula cinerea (AEAPCC) at a dose of 20 mg/kg on glucose and lipid profile was examined in normal and streptozotocin-induced diabetic rats. Additionally, histopathological examination of the pancreas and liver was carried out according to the Hematoxylin-Eosin method. Results: AEAPCC (20 mg/kg) showed a significant blood glucose-lowering activity in both normal and diabetic rats after a single and repeated oral administration during 15 days. The aqueous extract was also able to decrease the plasma triglycerides levels in both normal and diabetic rats after 15 days of oral treatment at a dose of 20 mg/Kg while no effect was observed on plasma cholesterol levels. In addition, the results show that AEAPCC exhibits an in vitro antioxidant activity using different tests. Histopathological analysis of the pancreas and liver of AEAPCC-treated diabetic rats has revealed that AEAPCC had a beneficial effect on the architecture of these organs while no improvement of glucose tolerance was noticed using the glucose tolerance test. Furthermore, the results showed that the extract is rich in several phytochemical compounds and exhibited an important antioxidant activity. The phytochemical screening revealed that AEAPCC contains polyphenolic compounds, flavonoids, tannins, alkaloids, saponins, quinones, sterols, terpenoids, anthroquinones and reducing sugars. Whereas, it is free from glycosides. Conclusion: In conclusion, this study demonstrates that Cotula cinerea possesses a beneficial effect on diabetes. Further investigations are required to study the mechanism of action of the antidiabetic effect of this plant.

Using natural products as antioxidant agents has been beneficial to replace synthetic products. Efforts have been made to profile the antioxidant capacities of natural resources, such as medicinal plants. The polyphenol content of Himalayan rhubarb, Rheum emodi wall, was measured and the antioxidant activity was determined using DPPH and ABTS+ assay, and the oxidative stress was assessed using SOD enzymatic assay. Five different solvent fractions, n-hexane, n-butanol, ethyl acetate, dichloromethane, and water, were used for screening the antioxidant capacity in effort to determine the optimum extraction solvent. The total phenolic contents for R. emodi fractions ranged from 27.76 to 209.21 mg of gallic acid equivalents (GAE)/g of dry weight. DPPH and ABTS+ assay results are presented into IC50 values, ranged from 21.52 to 2448.79 μg/mL and 90.25 to 1718.05 μg/mL, respectively. The ethyl acetate fraction had the highest antioxidant activity among other fractions. Also, n-butanol and water fractions showed significantly lower IC50 values than the positive control in DPPH radical scavenging activity. The IC50 values of SOD assay of fractions ranged from 2.31 to 64.78 μg/mL. A similar result was observed with ethyl acetate fraction showing the highest SOD radical scavenging activity. The study suggests that the ethyl acetate fraction of R. emodi possess the strongest antioxidant activity, thus the most efficient in extracting antioxidant contents. Moreover, a highly significant correlation was shown between total polyphenol content and antioxidant activity screening assays. The compounds related to the antioxidant activity of R. emodi were identified to myricitrin, myricetin 3-galloyl rhamnoside, and myricetin, which have not been reported in studies about R. emodi before.

The present study aimed to evaluate the potential of standardized methanolic extracts from seven Ficus deltoidea varieties in inhibiting the formation of AGEs, protein oxidation, and their antioxidant effects. The antiglycation activity was analyzed based on the inhibition of AGEs, fructosamine, and thiol groups level followed by the inhibition of protein carbonyl formation. The antioxidant activity (DPPH radical scavenging activity and reducing power assay) and total phenolic contents were evaluated. After 28 days of induction, all varieties of Ficus deltoidea extracts significantly restrained the formation of fluorescence AGEs by 4.55–5.14 fold. The extracts also reduced the fructosamine levels by 47.0–86.5%, increased the thiol group levels by 64.3–83.7%, and inhibited the formation of protein carbonyl by 1.36–1.76 fold. DPPH radical scavenging activity showed an IC50 value of 66.81–288.04 μg/ml and reducing power activity depicted at 0.02–0.24 μg/ml. The extent of phenolic compounds present in the extracts ranged from 70.90 to 299.78 mg·GAE/g. Apart from that, correlation studies between the activities were observed. This study revealed that seven varieties of Ficus deltoidea have the potential to inhibit AGEs formation and possess antioxidant activity that might be attributed to the presence of phenolic compounds.

AbstractThe chemical composition, antimicrobial and antioxidant activity of Eryngium palmatum, an endemic plant species from the Balkan Peninsula, were investigated. The flavonoids apigenin (9.5±0.3 mg g−1) and apigenin 7-O-glucoside (2.4±0.1 mg g−1) were determined in a methanol extract of aerial parts using HPLC analysis. The methanol extract of roots contained catechin (5.0±0.1 mg g−1), epicatechin (2.9±0.1 mg g−1), chlorogenic acid (1.6±0.0 mg g−1), gallic acid (0.9±0.0 mg g−1) and rosmarinic acid (0.9±0.2 mg g−1). GC-FID and GCMS analysis of a chloroform extract of aerial parts showed that the main volatile constituents were falcarinol, linoleic acid, hexadecanoic acid and methyl linoleate (comprising 32.6%; 24.4%; 19.9; 13.2% of the volatile fraction, respectively), while octanoic acid, tetradecanol and dodecanol dominated in the chloroform extract of the roots (34.9%; 25.8%; 22.2% of the volatile fraction, respectively). Investigation of antimicrobial activity by broth microdilution showed that the methanol and chloroform extracts of aerial parts and roots exerted a significant effect (MIC 3.5–15.6 μg mL−1) against tested Gram-positive and Gram-negative bacteria. The methanol extracts of aerial parts or roots exerted moderate ferric reducing antioxidant power, DPPH radical scavenging activity and hydroxyl radical scavenging activity.

Antioxidant activities of wampee peel extracts using five different solvents (ethanol, hexane, ethyl acetate, butanol and water) were determined by using in-vitro antioxidant models including total antioxidant capability, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, reducing power, and superoxide scavenging activity. Ethyl acetate fraction (EAF) exhibited the highest antioxidant activity compared to other fractions, even higher than synthetic antioxidant butylated hydroxyl toluene (BHT). In addition, the EAF exhibited strong anticancer activities against human gastric carcinoma (SGC-7901), human hepatocellular liver carcinoma (HepG-2) and human lung adenocarcinoma (A-549) cancer cell lines, higher than cisplatin, a conventional anticancer drug. The total phenolic content of wampee fraction was positively correlated with the antioxidant activity. This is the first report on the antioxidant and anticancer activities of the wampee peel extract. Thus, wampee peel can be used potentially as a readily accessible source of natural antioxidants and a possible pharmaceutical supplement.

ABSTRACTObjective: This study was performed to identify the phytochemicals and comparatively evaluate the antioxidant activity of Calotropis procera andAzadirachta indica by detection of total phenolics, hydrogen peroxide radical scavenging activity, and estimation of condensed tannins in differentsolvent systems and at different temperatures.Methods: Leaves of C. procera and A. indica were extracted in water, methanol by soaking dried leaf powder at room temperature and also by boilingthe leaf powder in water for 30 minutes. Phytochemical tests were performed in all of the extracts. The antioxidant activity was determined byhydrogen peroxide radical scavenging activity. Quantitative estimation of total phenolics and hydrolysable tannins was also performed.Results: The total phenolics in both leaf extracts was obtained maximum in boiled extract (40.7±1.20 mg gallic acid equivalent [GAE]/g dry extractin C. procera and 33.66±1.45 mg GAE/g dry extract in A. indica). The amount of hydrolysable tannins in both leaf extracts was found to be highest inmethanol (150±1.88 mg catechin equivalent/g dry extract in C. procera and 144.8±2.63 mg catechin equivalent/g dry extract in A. indica).Conclusion: The study showed promising results indicating that these plants are a good source of antioxidants. The majority of phytochemicals wereextracted in distilled water and methanol acts as a good solvent for extraction of tannins, whereas an increase in temperature leads to poor extractionof tannins.Keywords: Antioxidant, Phytochemicals, Phenolics, Radical, Tannins.© 2017 The Authors. The UV-Vis absorption spectrum of kaempferol and DPPH was investigated, the optimum determination wavelength and reaction time for determining the DPPH radical scavenging activity of kaempferol was 517 nm and 30 min, respectively. Kaempferol exhibited strong DPPH radical scavenging activity with a IC50 value of 0.004349 mg·mL-1, which is smaller than that of rutin, indicating that kaempferol has a stronger antioxidant activity than rutin.
