Cytotoxicity of Palladium Nanoparticles Against Aspergillus Niger

Background: Investigating the bioactivity of nanoscale materials against microorganisms give a comprehensive, proactive understanding of nanomaterial toxicity and explore their potential for applications. Methods: The aim of this study is to assess the changes in the mycelium growth and proteomics for filamentous fungi Aspergillus niger (A. niger) caused by palladium nanoparticles (Pd NPs). In this study, quantitative analysis of the mycelium growth and protein content of A. niger upon incubation with different concentrations of Pd NPs (0-1350 ng/mL) were reported. Matrix Assisted Laser Desorption/ Ionization Mass Spectrometry (MALDI-MS) was also used to analyze the changes of the proteins content of A. niger in different medium using two different matrices; α-cyano-4-hydroxycinnamic acid (CHCA), and sinapinic acid (SA). We found that Pd NPs decrease the mycelium growth of A. niger. Results: The protein contents are increased at low concentrations of Pd NPs, while it decreases with high concentration. MALDI-MS results show change of the protein contents for A. niger upon incubation with Pd NPs. Mycelia biomass decreases at high concentration of Pd NPs. Conclusion: The biological activity of Pd NPs depends on their concentration and cell culture medium. These new findings may add valuable information about the cytotoxicity of Pd NPs.

The present study was carried out in the lab. of plant tissue culture at King Abdulaziz University to test the response of four lemon cultivars to micro-propagation using BAP, Kin and 2,4- D combinations. The used explants in this study were intermodal segments and collected from the one year old lemon seedlings which obtained from the Citrus Research Center, Najran, Saudi Arabia. The experiments were laid out in a split plot design using 4 replicates. The results revealed that there were significant differences due to genotypic and growth regulators effects and their interaction for all measured parameters except no of days to buds sprouting. Explants of ‘Shehri’ registered maximum values of no. of days to buds sprouting with 0.5mg/l-1 Kin +0.5mg/l-1 2,4-D, % sprouted buds with 0.5mg/l-1 BAP, % dead shootlets with 2mg/l-1 BAP+0.5mg/l-1 2-,4- D and length of primary shoots (mm) 1mg/l-1 BAP+0.5mg/l-1 2-,4- D. Shoots of ‘AlnEurka’ formed the highest no. of leaves with 0.5mg/l-1 BAP+1mg/l-1 2-,4- D. Low responses were observed for explants from ‘Shaary’, ‘Banzahir’ and ‘Aln-Eurka’ on culture medium supplemented with high concentration of BAP alone or with combination of 2,4- D. There were observed no sprouted buds for explants of ‘Shehri’ on culture medium complemented with high concentrations of Kin in combinations with 2,4- D. Lemon explants were successfully in vitro propagated using intermodal segments and combinations of BAP, Kin and 2,4- D at low concentrations.

ABSTRACT In order to explain the increased mammotrophic activity of pregnant rat serum, some hormones known to have an in vitro effect on the mammary gland were added to non-pregnant rat serum and their effect compared with that of pregnant rat serum. The addition of insulin to the culture medium increased the mammotrophic effect of pregnant rat serum, but a difference between non-pregnant and pregnant rat serum could be demonstrated either in the presence or absence of a high concentration of insulin. While pregnant rat serum had a mammogenic (i. e. growth-promoting) effect, cortisol had no such effect and antagonized the mitotic activity induced by the pregnant serum. Pregnant rat serum was active at such low concentrations, that the progesterone content of pregnant rat serum could not explain the mammogenic activity of the latter. Moreover, the combination of progesterone with non-pregnant rat serum did not produce other changes produced by pregnant rat serum alone such as cytoplasmic opalescence and the localized development of vacuolization and secretion. Prolactin with non-pregnant rat serum produced a mammotrophic effect which was in all respects similar to that of pregnant rat serum. An effect could be demonstrated at 0.08 μg prolactin/ml medium. However, again pregnant rat serum was active at such low concentrations that the prolactin content of pregnant rat serum could not explain the mammotrophic effect of pregnant rat serum. Moreover, the effect of pregnant rat serum could not be blocked with rabbit anti-rat prolactin serum, but the anti-serum blocked the effect of rat prolactin added to non-pregnant rat serum. The results exclude the possibility that insulin, cortisol, progesterone or prolactin can account for the mammotrophic activity in pregnant rat serum. They provide indirect evidence for the importance of rat chorionic mammotrophin.

According to the World Health Organization, about 2 million new couples experience infertility annually, and their number is growing. An effective way to overcome infertility is assisted reproductive technology (ART). Cryopreservation will rationally solve the issue of preservation and further use of embryos: to delay pregnancy for some time considering womans desire and to prevent ovarian hyperstimulation syndrome. Embryo freezing allows to reduce the rate of repeated ovarian stimulation and perform preimplantation genetic diagnosis. Over the past decades, various cryotransfer options have been proposed to increase ART treatment efficacy, including the use of a culture medium with a high concentration of hyaluronic acid, but there are conflicting data on the use of such a medium in ART programs. Aim. Evaluation of thawed embryo transfers efficacy using the hyaluronic acid-containing culture medium. To achieve the goal, the following tasks were set: to evaluate clinical and medical history data of patients with tubal infertility in cryoprotocols, to analyze the previous cycle of in vitro fertilization and embryological stage, to evaluate the effectiveness of the culture medium with a high content of hyaluronic acid. Materials and methods. A detailed description of the patient sample, inclusion and exclusion criteria, embryological stage, embryo grading, devitrified embryo transfer technique. The article includes 3 tables which present the groups general clinical characteristics, the embryological stage, the rate of pregnancy, depending on the cultivation day. Results. The authors established that in patients with a history of pelvic surgery and sexually transmitted infections, it is advisable to use the culture medium with a high content of hyaluronic acid to transfer the thawed embryo. It was shown that pregnancy rate is 1.5 times higher when transferring devitrified embryos on the 5th day of development with the use of hyaluronic acid-containing culture medium. The conclusion about the pregnancy rate in obese patients is not indisputable, which requires further study. The authors also provide practical recommendations on the use of the culture medium with hyaluronic acid in cryoprotocols. Conclusion. The study allows to optimize the devitrified embryo transfer in patients with tubal infertility using a culture medium with a high content of hyaluronan. This work has undoubted scientific and practical significance.

Abstract Background This work aims to study the optimal conditions of the fermentation culture medium used for the production of extracellular enzymes (amylase, cellulase, lipase, and protease) from previously isolated Aspergillus niger strains in date by-products. Results The five most powerful isolates selected based on the zone of degradation formed on Petri plates by the substrate were subjected to the quantitative evaluation of their enzymatic production. All five strains showed almost similar API-ZYM profiles, with minor variations observed at the level of some specific enzyme expression. The production of cellulase and amylase was depending on pH and incubation temperatures. ASP2 strain demonstrated the high production rate of amylase (at pH 5 and 30 °C) and cellulase (at pH 6 and 30 °C) for 96 h of incubation. Conclusion The A. niger showed the ability to produce several extracellular enzymes and can be used in the valorization of different agroindustrial residues.

Abstract To cryopreserve cells, it is essential to avoid intracellular ice formation during cooling and warming. One way to achieve this is to convert the water inside the cells into a non-crystalline glass. It is currently believed that to accomplish this vitrification, the cells must be suspended in a very high concentration (20–40%) of a glass-inducing solute, and subsequently cooled very rapidly. Herein, we report that this belief is erroneous with respect to the vitrification of one-cell rat embryos. In the present study, one-cell rat embryos were vitrified with 5 μL of EFS10 (a mixture of 10% ethylene glycol, 27% Ficoll, and 0.45 M sucrose) in cryotubes at a moderate cooling rate, and warmed at various rates. Survival was assessed according to the ability of the cells to develop into blastocysts and to develop to term. When embryos were vitrified at a 2,613 °C/min cooling rate and thawed by adding 1 mL of sucrose solution (0.3 M, 50 °C) at a warming rate of 18,467 °C/min, 58.1 ± 3.5% of the EFS10-vitrified embryos developed into blastocysts, and 50.0 ± 4.7% developed to term. These rates were similar to those of non-treated intact embryos. Using a conventional cryotube, we achieved developmental capabilities in one-cell rat embryos by rapid warming that were comparable to those of intact embryos, even using low concentrations (10%) of cell-permeating cryoprotectant and at low cooling rates.

The effects of wheat germ agglutinin on Drosophila embryonic cell lines growing on cover-glasses was examined by scanning electron microscopy. At low concentrations of the lectin (5-10 mug/ml), cells spread against the glass surface and fused to form syncytia. At high concentration, damage to the cell surface was evidenced as extensive membrane shrivelling and loss of surface microfilaments. Fusion also occurred under these conditions. There was some indication that the morphology of cells in division remains undisturbed by wheat germ agglutinin. The coalescence of cells and morphologic disotrtion induced by wheat germ agglutinin were not inhibited by N-acetylglucosamine, the hapten inhibitor of the lectin, under the conditions utilized in this study.

Abstract The present study aimed to identify biomarkers to assess the quality of in vitro produced (IVP) bovine embryos in the culture media. IVP embryos on Day (D) 5 of development were transferred to individual drops, where they were maintained for the last 48 h of culture. Thereafter, the medium was collected and the embryos were transferred to the recipients. After pregnancy diagnosis, the media were grouped into the pregnant and nonpregnant groups. The metabolic profiles of the media were analyzed via electrospray ionization mass spectrometry, and the concentrations of pyruvate, lactate, and glutamate were assessed using fluorimetry. The spectrometric profile revealed that the media from embryos from the pregnant group presented a higher signal intensity compared to that of the nonpregnant group; the ions 156.13 Da [M + H]+, 444.33 Da [M + H]+, and 305.97 Da [M + H]+ were identified as biomarkers. Spent culture medium from expanded blastocysts (Bx) that established pregnancy had a greater concentration of pyruvate (p = 0.0174) and lesser concentration of lactate (p = 0.042) than spent culture medium from Bx that did not establish pregnancy. Moreover, pyruvate in the culture media of Bx can predict pregnancy with 90.9% sensitivity and 75% specificity. In conclusion, we identified markers in the culture media that helped in assessing the most viable IVP embryos with a greater potential to establish pregnancy.

Fusarium solani H915 is a fungus derived from mangrove sediments. From its ethyl acetate extract, a new alkenoic acid, fusaridioic acid A (1), three new bis-alkenoic acid esters, namely, fusariumester A1 (2), A2 (3) and B (4), together with three known compounds (5–7), were isolated. The structures of the new compounds were comprehensively characterized by high resolution electrospray ionization-mass spectrometry (HR-ESI-MS), 1D and 2D nuclear magnetic resonance (NMR). Additionally, the antifungal activities against tea pathogenic fungi Pestalotiopsis theae and Colletotrichum gloeosporioides were studied. The new compound, 4, containing a β-lactone ring, exhibited moderate inhibitory activity against P. theae, with an MIC of 50 μg/disc. Hymeglusin (6), a typical β-lactone antibiotic and a terpenoid alkaloid, equisetin (7), exhibited potent inhibitory activities against both fungal species. The isolated compounds were evaluated for their effects on zebrafish embryo development. Equisetin clearly imparted toxic effect on zebrafish even at low concentrations. However, none of the alkenoic acid derivatives exhibited significant toxicity to zebrafish eggs, embryos, or larvae. Thus, the β-lactone containing alkenoic acid derivatives from F. solani H915 are low in toxicity and are potent antifungal agents against tea pathogenic fungi.

Abstract Balneário Camboriu (SC - Brazil) is a touristic city where the disordered growth of the urban population and the implementation of coastal works without proper evaluation generated environmental impacts and affected the sanitary quality of water and sediment of Camboriu River and marine adjacent area. One of the most recent and alarming phenomena observed are the blooms of invasive bryozoans (Arboscuspis bellula and Membraniporopsis tubigera) associated with epibenthic diatoms (Amphitetras antediluviana and Biddulphia biddulphiana). Several clues associate these phenomena, started in 2003, with the excess of nutrients and organic matter in the Camboriú cove and large coastal works such as dredging, landfills and construction of jetties, leading to changes in benthic ecological structure. Being an aesthetic and environmental health problem, the concern of the scientific community and government agencies intensified as the occurrences become more frequent and persistent. This research addresses this issue through environmental and experimental studies. Samplings of the benthic material collected by boat and diving, and blooms monitoring were the environmental approach. The laboratory work included the algal isolation and culture, in addition to growth conditions assessment and chemical biomass analysis. Monitoring data showed a seasonal trend in the blooms, with more conspicuous events in warmer months. Diatoms increase in abundance in colder months and bryozoans in the warmer ones. The diatom A. antediluviana, predominant in the blooms, grew satisfactorily in laboratory cultivation, showing better growth in media with higher concentrations of silicate and phosphate. Bryozoans showed slow growth in laboratory conditions. The deposited material collected in the environment showed low concentrations of saturated fatty acids, but the high biomass suggest a possible use for biofuels production. Biomass samples dominated by bryozoans showed moderate antimicrobial activity against Klebsiella pneumoniae. The explanation for the occurrence of these blooms are still inconclusive, but there is considerable evidence that it is a synergistic effect between the high concentration of bacteria and organic debris in the water related to local pollution and the elimination of natural competitors by coastal works.

✓ The effects of several calmodulin antagonists, such as N-(6-aminohexyl)-5-chloro-1-naphthalene-sulfonamide hydrochloride (W-7) and its dechlorinated structural analogue (W-5), on the growth and proliferation of cultured and transplanted glioma (GA-1, chemically induced from rat glioblasts) were evaluated. Under culture conditions, the concentration of W-7 necessary to exert 50% inhibition of GA-1 glioma cell growth was 50 µM. However, W-5, with a lower binding affinity to calmodulin than W-7, caused no definite inhibition of the proliferation of GA-1 cells in culture. When a low concentration of W-7 (12.5 µM) was added to the culture medium, deoxyribonucleic acid (DNA) synthesis in the GA-1 glioma cells was not markedly affected, whereas both ribonucleic acid (RNA) and protein syntheses were strongly suppressed on incubation for 24 hours. When a high concentration of W-7 (25.0 to 75.0 µM) was applied to the medium, synthesis of DNA, RNA, and protein was distinctly inhibited. When W-7 (50.0 µM) was added to the incubation medium, the calmodulin concentration in the cultured GA-1 was reduced to as much as half the control level within 2 hours, and thereafter remained at this level. Whereas control rats intraperitoneally transplanted with GA-1 cells could survive for 14 to 21 days, daily intraperitoneal injections of W-7 at concentrations of 1.0, 3.0, and 10.0 mg/kg body weight prolonged the survival span to between 21 and 26 days; this corresponded to an increased life span of about 40% compared to the controls.
