This research was conducted to isolate, identify, and characterize pathogenic Vibrio spp., causative agents of vibriosis in grouper at Brackishwater Aquaculture Development Center (BADC), Situbondo. Twenty-nine isolates were isolated from wound, gills, eyes, liver, and ren of grouper with vibriosis on Thiosulphate Citrate Bile Sucrose Agar (TCBSA) medium. Koch Postulate Test was conducted to determine pathogenic Vibrio spp., by intraperitoneal injection to humpback grouper (Cromileptes altivelis) (9-10 cm of total length) at 106 cells/fish in triplicates. Results indicated that the pathogenic Vibrio spp., causative agents of vibriosis in grouper at BADC, Situbondo were identified to be V. alginolyticus, V. anguillarum, V. metchnikovii, V. vulnificus, V. fluvialis, V. furnisii, and V. parahaemolyticus. Infection of the pathogenic Vibrio spp. caused acute mortality within 17-46 hours with specific disease signs like haemorhagic on fins (pinnae pectorales, pinnae abdominales, pinna analis) and also on the body. These results suggested that vibriosis should be monitored and controlled properly and quickly.

ABSTRAKKetersediaan induk dalam suatu usaha pembenihan memiliki peran yang sangat penting. Namun demikian, ketersediaan induk yang berasal dari alam sangat terbatas jumlahnya, sehingga perlu dilakukan penyediaan calon induk yang berasal dari hasil budidaya. Ikan kerapu bebek Cromileptes altivelis Valenciennes, 1828 turunan pertama (F-1) dan turunan kedua (F-2) sudah dapat diproduksi dari hasil budidaya, namun benih turunan ketiga (F-3) belum dapat diperoleh. Penelitian ini bertujuan untuk mendapatkan karakteristik morfologis dan pertumbuhan larva ikan kerapu bebek F-3 sebagai calon induk F-3, serta tingkat keberhasilan produksi benihnya. Pemeliharaan larva dilakukan dalam hatchery hingga larva menjadi benih. Parameter yang diamati meliputi panjang total dan panjang duri sirip larva, berat tubuh larva serta sintasan dan jumlah produksi benih. Hasil penelitian menunjukkan bahwa panjang total larva umur 5, 15, 25 dan 35 hari, masing-masing adalah 3,20±0,07; 4,42±1,11; 8,35±1,12 dan 12,51±3,23 mm. Duri sirip mulai terukur pada larva umur 15 hari. Berat larva umur 30 hari adalah 0,11±0,04 g. Pola pertumbuhan panjang total dan berat tubuh larva adalah eksponensial, sedangkan pola pertumbuhan duri siripnya adalah linier. Masa pemeliharaan larva hingga menjadi benih adalah ± 40 hari. Jumlah benih ikan kerapu bebek F-3 yang diproduksi dalam satu kali siklus pemeliharaan berkisar 440 hingga 2.300 ekor dari 50 ekor induk dan 3 kali siklus pemijahan dengan tingkat kelangsungan hidup 1,30% hingga 8,80%. Hasil penelitian ini mengindikasikan bahwa ikan kerapu bebek F-3 dapat diproduksi dari hasil budidaya seperti halnya pada F-1 dan F-2. ABSTRACTBroodstocks are the most important part of humpback grouper culture, but their availability in nature are limited. Therefore, it is necessary to produce broodstock candidates from culture. The first (F-1) and the second (F-2) generation of humpback grouper have already been produced but the third generation (F-3) production is still on the way. This study was conducted to find out morphological characteristic of the third generation (F-3) of humpback grouper larvae as the candidate of the third generation of broodstock, larval growth and the success rate of seed production. Larvae rearing was done in hatchery until larvae metamorphosed to be seeds. Observed variables including larval total length and spine length, larval body weight, survival rate and the juvenile productions. The study result showed the total length of 5, 15, 25 and 35 days old larvae were 3.20±0.07; 4.42±1.11; 8.35±1.12 and 12.51±3.23 mm, respectively. The spine began measured on 15 days old larvae. The body weight of 30 days old larvae was 0.11±0.04 g. The growth pattern of larval total length and body weight were exponential, while the growth of spine was linear. Rearing period from larvae to juveniles was 40 days. Number of F-3 seed production of humpback grouper produced from once rearing cycle range between 440 and 2,300 fish and the survival rate range from 1.30% up to 8.80%. Therefore, this study could indicate that seed of F-3 humpback grouper can be produced as those of F-1 and F-2.

This study aims to determine the total length and scar condition of the body of whale sharks (Rhincodon typus) in Cendrawasih Bay National Park (TNTC), Papua-Indonesia. Photo-identification was used to identify individuals of the whale shark R. typus based on spot patterns behind the last gill slit of each individual. Photo-identification was also used to determine the scar of the whale shark. The total length of whale sharks were estimated based on the length of a snorkeller (assumed to be 1.6 m) swimming alongside the whale shark. We identified 21 individuals of R. typus. Of these 21 individuals, 14 were new sightings and seven were re-sightings that have been recorded in the previous photo collection database. R. typus ranged in size from 2 to 5 m total length (average 3.78 m, ±0,86, N= 21). Based on their size, all individuals of whale shark were categorized as juvenile. 52% of R. typus identified had scars and 38 % were not and 10% were unknown. The majority of whale sharks had amputation (12 individuals) and abrasion (7 individuals) scars. Scars occurred most often on the caudal fin and dorsal fin, five and four individuals respectively. This information is useful for understanding potential threats and designing better management programmes for R. typus conservation in TNTC.

A total of 65 specimens of Rhinomugil corsula (Hamilton, 1822), measuring from 8.1 cm to 28.9 cm in total length, collected from the Sitakunda coast of the Bay of Bengal, were used for the morphometric analysis during the period between March 2016 and February 2018. Twenty seven morphometric characters were selected and studied during the investigation period. The regression equations – both arithmetic and logarithmic – between the total length (TL) and 21 morphometric characters, and head length (HL) with five morphometric characters related to the head, were determined. The value of coefficient of correlation ‘r’ for each relationship was calculated and t-test for each ‘r’ value was also done. The relationships of the various measurements of the body with the total length – and head length with the five relevant characters -of R. corsula from the Sitakunda coast of the Bay of Bengal showed linear relationships, which were highly significant (P<0.01). The ranges of ‘b’ values 0.967 to 1.346 in case of the relationships between TL and 21 relevant characters, whereas 0.906 to 1.236 in case of the relationships between HL and 5 relevant characters. Introduction. Gram-positive enterococci are an integral part of the normal intestinal microflora. However, if they get into wounds or sterile cavities of the body, they may induce various diseases including wound infections, urethritis, peritonitis, sepsis, pyelonephritis, or damage other organs. These microorganisms also induce nosocomial infections and are among the causative agents of health care-associated infections (HAIs); thus, it is critical to control the spread of enterococci. Our purpose was to monitor circulation of vancomycin-resistant enterococci (VRE) in health facilities of the Karachay-Cherkess Republic (KCR) in 2016-2018. Materials and methods. We studied VRE strains isolated in the Microbiological Laboratory of the Center for Hygiene and Epidemiology in the Karachay-Cherkess Republic. During three years, the total of 281 strains of vancomycin-resistant enterococci were isolated from various biological materials received from obstetric, surgical, pediatric, and therapeutic hospitals and from outpatients undergoing treatment in polyclinics of the Karachay-Cherkess Republic. Results. According to our data, VRE rates per 100 examined patients were 2.34 in 2016, 3.4 in 2017, and 3.03 in 2018, and were comparable to the rates registered in the Russian Federation in the range of 3% to 22.7%. Discussion. When analyzing circulation of vancomycin-resistant enterococci, we established that the highest VRE rate (4.24 per 100 examined patients) was observed in maternity hospitals, while the lowest rate of 0.67% was observed in therapeutic departments. When studying sensitivity of 281 isolated VRE strains to eight antibacterial drugs, no strain resistant to all antibiotics was revealed. Sensitivity to linezolid, ampicillin, and penicillin was observed in 92.6%, 79.0%, and 66.2% of strains. Sensitivity to ciprofloxacin, gentamicin, tetracycline, and nitrofurantoin was 50% or less. VRE were mainly isolated from urine and discharge from the cervical canal and pharynx.

In this study, 45 (10 whole specimens and 35 frozen claws) frozen samples of Portunus pelagicus imported into Sicily (Italy) from the west coast of Africa were examined to assess their bacteriological characteristics and suitability for consumption. Bacteriological examination was performed on two subsamples for each whole crab. The first was the body and claw muscle; the second was a pool of viscera and gills. In the case of frozen claws, each muscle claw was a sample. An aerobic plate count at 30°C (mesophilic aerobic plate count [MAPC]) and 18°C (psychrotrophic aerobic plate count [PAPC]) for 3 days, sulfite-reducing anaerobes, total coliforms, Escherichia coli, enterococci, and Aeromonas spp. were enumerated. Detection of halophilic Vibrio spp. was also performed using salt polymixin broth as an enrichment medium and thiosulfate citrate bile salts sucrose agar as a selective medium; a further morphological and biochemical identification of suspected colonies was performed. The bacterial load of muscle and viscera and gills was low. The MAPC ranged from 0.78 to 3.26 log CFU/g, and the PAPC ranged from 0.48 to 2.41 log CFU/g. Vibrio spp., Aeromonas spp., sulfite-reducing anaerobes, and E. coli were never isolated from muscles or viscera and gills. In contrast to the findings of others, this study showed good bacteriological quality of crabs imported into Sicily from the west coast of Africa. This study also demonstrated the positive influence of the characteristics of environment of origin and postharvest handling hygiene; these parameters could be useful in the context of the application of the hazard analysis critical control point system to this production.

Mortalities of bivalve larvae and spat linked with Vibrio spp. infection have been described in hatcheries since 1959, causing potential development of resistant bacteria. A reliable and sustainable solution to this problem is yet to be developed. Potential treatment of bacterial infection with bacteriophages is gaining interest in aquaculture as a more sustainable option for managing Vibrio spp. infection. This study assessed the effectiveness of bacteriophages (Φ-5, Φ-6, and Φ-7) against pathogenic Vibrio isolates (USC-26004 and USC-26005). These phage isolates were found to belong to the Myoviridae viral family. A total of 212 ORFs of Φ-5 were identified and annotated. The genome of this phage contained putative thymidine kinase and lysin enzyme. During infections with phages, the OD values of the isolates USC-26005 and USC-26004 remained stable at a much lower reading compared to the control after 9 h of incubation. Mortality rate of oyster (Saccostrea glomerata) larvae was 28.2 ± 3.5% in the bacteriophage treatment group, compared to 77.9 ± 9.1% in the bacterial treatment group after 24 h incubation. Findings of this study indicate that lytic phages might be utilized as potential bio-control agents of luminescent bacterial disease in oyster hatcheries.

The estuary is the ecological niche of pathogenic Vibrio spp. as it provides abundant organic and inorganic nutrients from seawater and rivers. However, little is known about the ecology of these Vibrio species in the inland brackish water area. In this study, their co-occurrence and relationships to key environmental constraints (salinity and temperature) in the Hun-Tai River of China were examined using the most probable number polymerase chain reaction (MPN-PCR) approach. We hereby report 2-year continuous surveillance based on six water indices of the Hun-Tai River. The results showed that seawater intrusion maximally reached inland as far as 26.5 km for the Hun-Tai River. Pathogenic Vibrio spp. were detected in 21.9% of the water samples. In particular, V. cholerae, V. parahaemolyticus, and V. vulnificus were isolated in 10 (10.4%), 20 (20.8.5%), and 2 (2.08%) samples, respectively. All V. parahaemolyticus strains were tdh gene negative, 10% were positive for the trh gene. Multi-locus sequence typing (MLST) divided V. parahaemolyticus strains into 12 sequence types (STs) for the Hun-Tai River. Five STs were respectively present in various locations along the Hun-Tai River. The PCR assay for detecting six virulence genes and Vibrio seventh pandemic island I and II revealed three genotypes in 12 V. cholerae isolates. The results of our study showed that seawater intrusion and salinity have profound effects on the distribution of pathogenic Vibrio spp. in the inland river, suggesting a potential health risk associated with the waters of the Hun-Tai River used for irrigation and drinking.

The present study describes Camallanus maculatus n. sp. (Nematoda: Camallanidae) from Xiphophorus maculatus (Osteichthyes: Poecilidae) collected in an ornamental fish farm in São Paulo State, Brazil. Thirty-three out of 40 examined fish were parasitized (82% prevalence) with mean intensity of 2.8 (1-20). Male specimens were 3,500µm total length and females 7,100µm total length, provided with orange-brown buccal capsule and two developed tridents. Males showed two unequal spicules, gubernaculum absent and females with vulva situated in the middle of the body. The main characteristics were the sleekly longitudinal ridges in the buccal capsule in both sexes, enlargement at the anterior end of the smaller spicule, the number of genital papillae, longer muscular and glandular portion of esophagus and round tail in female.

ABSTRACTWe investigated the abundance, distribution, and virulence gene content ofVibrio cholerae,V. parahaemolyticus, andV. vulnificusin the waters of southern Lake Pontchartrain in Louisiana on four occasions from October 2005 to September 2006, using selective cultivation and molecular assays. The three targeted pathogenic vibrios were generally below the detection level in January 2006, when the water was cold (13°C), and most abundant in September 2006, when the lake water was warmest (30°C). The maximum values for these species were higher than reported previously for the lake by severalfold to orders of magnitude. The only variable consistently correlated with total vibrio abundance within a single sampling was distance from shore (P= 0.000). Multiple linear regression of the entire data set revealed that distance from shore, temperature, and turbidity together explained 82.1% of the variability in total vibrio CFU. The log-transformed mean abundance ofV. vulnificusCFU in the lake was significantly correlated with temperature (P= 0.014), but not salinity (P= 0.625). Virulence-associated genes ofV. cholerae(ctx) andV. parahaemolyticus(trhandtdh) were not detected in any isolates of these species (n= 128 andn= 20, respectively). In contrast, 16S rRNA typing ofV. vulnificus(n= 298) revealed the presence of both environmental (type A) and clinical (type B) strains. The percentage of the B-typeV. vulnificuswas significantly higher in the lake in October 2005 (35.8% of the total) than at other sampling times (P≤ 0.004), consistent with the view that these strains represent distinct ecotypes.
