Objective: This research aimed to examine the anti-oxidant activity, antielastase activity, and the content of total phenolic and total flavonoid of R. fraxinifolius unripe fruit. Methods: The dried unripe fruit was extracted using Soxhlet apparatus with sequence solvent: n-hexane, ethyl acetate, and methanol. Each extract was determined the anti-oxidant and antielastase activity, total phenolic, and total flavonoid content. Result: The result showed the extracts (n-hexane, ethyl acetate, and methanol) gave anti-oxidant IC50>200; 186.84; and 19.74 ppm, and the ability of elastase inhibition was 6.84+0.9%; 52.23+7.1%; and 57.81+5.5% at 100 ppm, respectively. The methanolic extract contained phenolic 202.2 mg GAE/g extract and flavonoid 43.89 mg QE/g extract. Conclusion: R. fraxinifolius unripe fruit has shown potential as a DPPH (α, α-diphenyl-β-picrylhydrazyl) radical scavenger and anti-elastase. This study provides an excellent effect to underline the importance of R. fraxinifolius unripe fruit, and it can be developed as nutricosmetics, nutraceuticals, or herbal anti-wrinkle cosmetics.

Penelitian pendahuluan tentang aktivitas antioksidan ekstrak etanol dan ekstrak etil asetat daun dewandaru (Eugenia uniflora L.) menunjukkan potensi anti radikal bebas yang tinggi dari kedua ekstrak. Oleh karena itu telah dilakukan fraksinasi ekstrak etanol dan ekstrak etil asetat daun dewandaru, serta pengujian aktivitas antiradikal dari masing-masing fraksi. Selain itu, penelitian ini juga bertujuan untuk mengetahui apakah ada korelasi antara kandungan fenol dan flavonoid total dalam berbagai fraksi tersebut dengan aktivitas antiradikalnya. Daun dewandaru diekstraksi dengan kloroform, etil asetat, dan etanol secara maserasi bertingkat. Selanjutnya dilakukan fraksinasi terhadap masing-masing ekstrak etanol dan ekstrak etil asetat menggunakan kromatografi kolom gravitasi dengan fase diam silika G60 dan fase gerak campuran perbandingan tertentu antara kloroform, etil asetat, etanol, dan air secara gradien kepolaran. Aktivitas antiradikal diukur secara spektrofotometri dengan pereaksi DPPH (2,2-diphenyl-1-picrylhidrazyl) dan sebagai pembanding digunakan vitamin E. Penentuan kadar fenol dan flavonoid total dalam fraksi secara spektrofotometri menggunakan pereaksi Folin Ciocalteu untuk penetapan kadar fenol total, sedangkan untuk penentuan kadar flavonoid total menggunakan pereaksi AlCl3. Hasil penelitian menunjukkan bahwa fraksi-fraksi dari ekstrak etil asetat dan ekstrak etanol daun dewandaru mempunyai aktivitas antiradikal yang cukup tinggi. Lima fraksi dari ekstrak etanol memiliki aktivitas antiradikal lebih tinggi dibanding vitamin E. Aktivitas antiradikal paling tinggi ditunjukkan dengan nilai ARP (antiradical power) sebesar 3496,5 mg DPPH/mg sampel, sedangkan vitamin E sebesar 1776,20 mg DPPH/mg sampel. Korelasi antara kadar fenol maupun flavonoid total dalam berbagai fraksi tersebut dengan aktivitas antiradikalnya menunjukkan korelasi positif dengan korelasi sebesar 0,55 dan 0,45. Berdasarkan hasil diatas dapat disimpulkan bahwa fraksi-fraksi polar daun dewandaru mempunyai aktivitas antiradikal yang lebih tinggi daripada vitamin E, namun aktivitas tersebut berkorelasi rendah dengan kandungan fenol dan flavonoid totalnya.   Preliminary research on the antioxidant activity of ethanol and ethyl acetate extracts of Dewandaru leaves (Eugenia uniflora L.) showed high anti-free radical potency from both extracts. Therefore fractionation of the ethanol and ethyl acetate extracts of dewandaru leaves was carried out, as well as evaluation of antiradical activity of each fraction. In addition, this study also aimed to determine whether there is a correlation between the phenol and total flavonoid content in various fractions with their anti-inflammatory activity.Dewandaru leaves were extracted by sequentially maceration with chloroform, ethyl acetate and ethanol. Furthermore, the ethanol and ethyl acetate extracts were fractionated  using gravity column chromatography with silica G60 as stationary phase and increasing polarity of mobile phase by  varying the ratio of chloroform, ethyl acetate and ethanol. Anti-free radical activity was determined using spectrophotometer and DPPH (2,2-diphenyl-1-picrylhidrazyl), vitamin E was used as a standard. Determination of total phenol and flavonoid content was determined using spectrophotometer, Folin Ciocalteu reagent was used to determine total phenolic content, meanwhile  ALCl3 reagent was used to determine total flavonoid content.  The results showed that fractions of  ethanol and ethyl acetate extracts of dewandaru leaves showed high anti-free radical activity. Of all fractions, there 5 fractions of ethanol extract showed higher anti-free radical activity as compared to vitamin E.  The highest anti-free radical activity is indicated by ARP (antiradical power) value of 3496.5 mg DPPH/mg sample, meanwhile the ARP value of vitamin E was 1776,20 mg DPPH/mg sample. Correlation between total phenol and flavonoid content of those fractions indicated positive correlation with correlation value of 0.55 and 0.45 respectively. Based on the results, it can be concluded that polar fractions of dewandaru leaves displayed higher anti-free radical activity as compared to vitamin E, however those activitiesdid not have correlation with their total phenolic and flavonoid content

Abstract Background: Nymphaea capensis is an aquatic flowering plant which is included in the family of Nymphaeaceae. Literature review on the plants of Nymphaeaceae family exhibited significant medicinal activities. Therefore, the objective of the present study is to evaluate possible anti-oxidant activity of crude methanol extract of N. capensis leaf.Method: In antioxidant study, methanolic plant extract was evaluated for 1,1-diphenyl,2-picrylhydrazyl (DPPH) and reducing power capacity. Moreover, total phenolic and total flavonoid content of plant extracts were determined and expressed in mg of gallic acid equivalent per gram of dry sample (mg GAE/g dry weight).Result: In the DPPH free radical scavenging assay, methanol extract showed concentration dependent inhibition of the free radicals. IC50 of Ascorbic acid was 14.84 µg/ml whereas N. capensis was 130.94 µg/ml. In case of reducing capacity, at conc. 62.5, 125, 250, 500, 1000 ug/ml, the absorbances of Ascorbic acid were 0.65, 1.12, 1.45, 1.78 and 1.89 respectively. In case of N. capensis, the absorbances were 0.46, 0.75, 1.04, 1.27 and 1.50 respectively. The extract displayed a concentration dependent increase in reducing power. In the case of total phenolic content, the methanol extract of N. capensis contained a considerable amount of phenolic contents which was 215±7 of GAE mg/g. In the case of total flavonoid content, methanol extract of N. capensis contained significant amount of flavonoid contents which was 184.75±6.78 of GAE mg/g.Conclusion: These results suggested that the methanol extract of N. capensis possess considerable anti-oxidant activity.

Ziziphora is a plant used in Turkish and Iran traditional medicine for its antibacterial activity, sedative and stomach soothing properties. Although the chemical profile of the essential oil of different Ziziphora species is well documented, data regarding plant extracts are incomplete. In this study extracts from Ziziphora taurica subsp. cleonioides were obtained using ethyl acetate, methanol and water and the chemical profile of the aerial part of the plant was elucidated. Among the compounds identified, rosmarinic acid was the most abundant (3375.67 ± 38.02 μg/mL), at the extract of methanol, followed by chlorogenic acid (3225.10 ± 16.44). Enzyme inhibition activity against α-amylase and tyrosinase was also estimated. The ethyl acetate extract showed the highest α-amylase activity (1.95 ± 0.04 mg/mL), while the best anti-tyrosinase activity was calculated for the methanolic extract (1.25 ± 0.01 mg/mL). In addition, total phenolic, flavonoid content and antioxidant activity were evaluated. According to our results, bioactivity of the plant is of great interest, nonetheless, at the same time, it is strongly depended on the solvent used during the extraction process. Our data suggest that the plant under study may be an important source to consider against metabolic, skin pigmentation and oxidative stress related disorders.

Diabetes Mellitus (DM) is one of the non communicable diseases (NCDs) with the largest number of deaths in the world. The Macaranga hispida (Blume) Mull. Arg is a source of phenolic compounds. Phenolic is grouped as polyphenols group that widely functioned as antidiabetic. The α-glucosidase enzyme plays a role in the forming of glycoproteins and glycolipids. The α-glucosidase enzyme can be used for antidiabetic activity assay because of its capability on breaking down carbohydrate into glucose in the human small intestine. This study aimed to determine phenolic content, total flavonoid content, and antidiabetic activity of M. hispida leaves extract and its fraction. The extract and fraction were tested for the phenolic levels using Folin-Ciocalteau method, the total flavonoid using alumunium chloride method, and the antidiabetic activity using α-glukosidase method. The yield of methanolic extract of the Macaranga hispida was 10.23%. Phenolic content of the ethyl acetate fraction from the methanolic extract was 8.411 mg, equivalent to gallic acid/100 mg extract. Total flavonoid content was 6.14 ± 0.31b/b μg/mL. Antidiabetic activity assay of the extract and the fractions of Macaranga hispida resulted the highest antidiabetic activity (IC50) was in the ethyl acetate fraction with IC50 value 21.91 μg / mL.

Objective: This study was designed to evaluate the antioxidative potential of tubers of Habenaria pectinata. Methods: The tubers of H. pectinata were extracted using hexane, ethyl acetate, methanol, and water as solvents. The anti-oxidant potential of extracts was evaluated using free radical scavenging and reducing power assays. The most active methanolic extract was then fractionated into four fractions using the above-mentioned solvents. Results: The phenol and flavonoid content was found to be maximum in the methanol extracts. All the extracts and fractions showed significant levels of antioxidant activity except hexane extract. Conclusion: The tubers of H. pectinata were found to possess a significant antioxidant potential and can be explored further for isolation and preclinical investigation for the ailment of various diseased states and disorders.

<p class="Abstract">The article discusses anti-oxidant oral glucose tolerance and anti-hyperglycemic potentials of <em>Syzygium calophyllifolium</em>. Extracts from successive solvent extraction were tested for the total phenolic, tannin, flavonoid content and free radical scavenging property using DPPH, ABTS<sup>+</sup>, phosphomolyb-denum, FRAP, superoxide and metal chelating assays. Ethyl acetate extract of bark responded well against DPPH (IC<sub>50</sub> 4.13), ABTS<sup>+</sup> (36832.29 µM TE/ g extract),phosphomolybdenum (100.4 g AAE/100g extract), superoxide and metal ion radicals. The methanol extract of bark was also found as an effective radical scavenger. The leaf methanol extract also showed significant anti-oxidant ability. The bark methanol extract could potentially reduce the blood glucose level in glucose loaded and diabetic rats. The immense anti-oxidant potential of <em>S. calophyllifolium</em> leaf and bark extracts could be taken as a good source of natural anti-oxidant supplement in food to defend oxidative stress related disorders like diabetes.</p><p> </p>

Oxidative stress may lead to a number of diseases such as atherosclerosis, nephrotoxicity, liver cirrhosis, cancers, diabetes, and Alzheimer disease. Medicinal plants are an important source of antioxidants. Therefore, the antioxidant potential of Cetraria islandica was evaluated in this work. The coarse powder of leaf of Cetraria islandica was extracted in Soxhlet apparatus, with ethanol (90%) and ethanolic extract of Cetraria islandica (EECI) was further processed for phytochemical screening, total phenol content, total flavonoid content, and various in vitro antioxidant assays. The phytochemicals present in EECI were glycosides, carbohydrates, triterpenoids, proteins and amino acids, gums and mucilages, and flavonoids. The content of extractives was 8.3 mg/ml for Cetraria islandica еxtract 70%. The content of polyphenolic compounds in terms of per head acid was the highest for the sample of 70% and amounted to 0.586 mg/ml, and the content of flavonoids per standard solution quercetin - 0,012 mg/ml - also for 70% extract. By DPPH, the percentage of radicals of absorbing activity for 70% of the extract was 86%, and the antiradical activity of 0,417, indicating a sufficiently high rate of Cetraria islandica antioxidant activity at such a concentration of ethanol. The tested extracts showed next results by FRAP assay: 486 μmol/L (EE96) 135 μmol/L (EE70) and 158 μmol/L (EE40). ABTS method showed the highest result of the extract of Cetraria islandica 40%. The obtained results confirmed the high potential of the extracts as a source of phenolic compounds, in particular flavonoids.

Objective: Tagetes erecta, marigold, has various ethnomedicinal uses. It has a wide variety of bioactive components such as polyphenols and flavonoids which show different bioactivities. The aim of the present study is to quantitatively estimate total phenolic content (TPC) and total flavonoid content (TFC) of different extracts of T. erecta flowers.Methods: Extraction was done by maceration process, sequentially from non-polar to polar. Chloroform, ethyl acetate, and methanol extracts of T. erecta flowers were subjected to preliminary phytochemical screening. The extracts were analyzed for TPC and TFC using gallic acid and rutin as standard, respectively.Result: Phytochemical screening of different extracts showed the presence of carbohydrates, flavonoids, phenolics, fats, and oils. TPC and TFC in extracts of T. erecta varies with solvents. The study revealed that methanolic extract possesses the highest phenolic content, 49.76±0.29 mg gallic acid equivalents/g extract, and also maximum flavonoid content, 13.43±0.43 mg RE/g extract, among the three extracts.Conclusion: Higher value of phenolics and flavonoid indicates higher antioxidant activity. The present study revealed that methanolic extract has the highest phenolic and flavonoid content. This indicates that the flowers may possess a good antioxidant property and further research could be carried out.
