Objective: The present study was designed to identify the bioactive phytochemicals and its antibacterial and in vitro anti-inflammatory potential of aqueous and methanolic seed extract of Citrullus lanatus.Methods: The phytochemical screening of both the aqueous and methanolic seed extract was carried out qualitatively to identify the major Phyto-constituents present in the extracts. The antimicrobial activity of the extracts was evaluated against six pathogenic bacterial strains by agar well diffusion method and the Minimum inhibitory concentration (MIC) was determined by broth dilution method. In vitro anti-inflammatory activity of C. lanatus seed extracts was evaluated by using human red blood cell (HRBC) membrane stabilization and inhibition of albumin denaturation method.Results: The results of the study indicated that both the extracts of the seed having antimicrobial activity, while the methanolic extract showed more significant activity against the tested organism than aqueous extract. Methanol extract had the lowest MIC of 1.562 mg/ml against Escherichia coli, Staphylococcus aureus, Klebsiella pneumonia, Pseudomonas aeruginosa and Bacillus subtilis, whereas in aqueous extract was highly sensitive to Bacillus subtilis, E. coli and Klebsiella pneumonia with MIC of 3.125 and 6.25 mg/ml, respectively. Methanolic extracts exerted comparative higher anti-inflammatory activity than aqueous extract.Conclusion: Present study provides a firm evidence to support that the synergistic effect of C. lanatus seed extracts having potent anti-inflammatory and antimicrobial property, which might serve as an effective drug for various microbial infections and inflammatory disorders.

Background: Lawsonia inermis (L. inermis) is perennial plant commonly called henna. It is frequently cultivated in Sudan. Beside its uses cosmetics for staining hands and as hairs dyes‚ it was reported to be useful in jaundice, enlargement of spleen, calculus affliction and skin disease. Method: This descriptive study was done during the period from December 2014 to April 2015 in order to determine the invitro antimicrobial activity of L. inermis (henna) leaves extract against standard and clinical isolates from wound swabs, including Staphylococcus aureus, Escherichia coli, Proteus species, Klebsiella pneumonia, and Pseudomonas aeruginosa. These organisms were collected from different hospitals in Khartoum State including: Soba University Hospital, Military Teaching Hospital, and Laboratory Management Center. The in vitro antimicrobial susceptibly testing was performed using cup plate diffusion method. The activity of L. inermis Linn leaves extract was controlled with four reference antibiotics including gentamicin, oxacillin, ciprofloxacin, and impinim. Results: When aqueous extract of L. inermis Linn examined against standard bacteria and clinical isolates result showed that all standard bacteria were inhibited at 100%, 50%, and 25% concentration. All clinical isolates were successfully inhibited at 100%, 50%, 25%, and 12.5%. In contrary, the activity of methanolic extract of L. inermis Linn against standard bacteria showed that all standard bacteria were inhibited at 100%, 50% concentration, However, the clinical isolates showed an inhibition rate various depending on the concentration of methanolic extract of L. inermis Linn with S. aureus being most sensitive isolate. Conclusion: We conclude that aqueous and methanolic extract of henna exhibited antimicrobial activity against all types of tested organisms both clinical and standard isolates. But the aqueous extract shows superior inhibition ability than the methanolic.

Diarrhea is a common health complaint occurring with mild, temporary conditions to a potentially life-threatening condition. <i>Cucurbita maxima</i> (Cucurbitaceae) seed is reportedly used traditionally for the treatment of diarrheal and thus this study aimed to evaluate the in vitro antioxidants, total tannins, phenolics contents, and antidiarrheal potentials of <i>n</i>-hexane and aqueous <i>C. maxima</i> seed extracts in castor oil induced diarrheal rats. The <i>n</i>-hexane extract mainly contains oils while the aqueous extract was thick, brown solid. The aqueous and <i>n</i>-hexane seed extract of <i>C. maxima</i> expressed significant 2,2, diphenyl-1-picrythdrazyl (DPPH) scavenging activities at 6.25 – 1000 mg/mL with IC<sub>50</sub> values of 104.01 mg/mL, 29.27 mg/mL and 26.78 mg/mL for <i>n</i>-hexane, aqueous and vitamin C respectively. The hydroxyl radical scavenging activities of the <i>n</i>-hexane and aqueous seed extract of <i>C. maxima</i> were significantly lower at higher concentrations compared with that of vitamin C. Furthermore, the total antioxidant capacities of <i>n</i>-hexane (22.08mg/mL) and aqueous seed extract (11.03 mg/mL) of <i>C. maxima</i> were found to be higher than that of vitamin C (134.46 mg/mL). The aqueous extracts (658.33±380.08 mg QE/g) total tannins were not significantly different from the <i>n</i>-hexane extract (468.33±102.55 mg QE/g) while the <i>n</i>-hexane extract contains significantly higher total phenolics (2.93±1.25 mg GAE/g) compared with the aqueous extract (0.19±0.04 mg GAE/g). Percentage inhibition of stooling was found to be 57±22%, 41±16%, and 46±11% for loperamide, <i>n</i>-hexane, and aqueous respectively. The study concludes that aqueous and n-hexane seed extracts of <i>C. maxima</i> (pumpkin) possess in vitro antioxidant activities and antidiarrheal properties.

Objective: The current work was aimed to prepare a topical gel containing curcumin (CUR) for the treatment of microbial infections on skin. Methods: CUR was complexed with the β-cyclodextrin (β-CD) using kneading method in 1:1, 1:2, and 1:3 molar ratios and characterized. The inclusion complex with high aqueous solubility was loaded in the topical gel containing (2% CUR) which was prepared using carbopol, sodium CMC, and guar gum and evaluated for viscosity, spreadability, extrudability, pH, drug content, and in vitro diffusion studies. The in vitro anti-inflammatory activity of the gel was performed by albumin protein denaturation technique, the statistical analysis was done using ANOVA followed Dunnett’s t-test. The antimicrobial activity of CUR was evaluated using standard strains of Candida albicans and Escherichia coli by agar well diffusion method. Results: The complexation of CUR had an increased solubility up to 103.09 times for 1:3 molar ratio with in vitro dissolution 90.64% for 60 min. The optimized formulation F9 had viscosity of 6500.3 cps and 97.5% in vitro drug diffusion for 8 h which follows zero-order release kinetics. In vitro anti-inflammatory activity studied showed that the CUR gel has a good potency for renaturation and was as effective as standard diclofenac with 76.9% inhibition (p=0.0507). CUR showed approx. 3 mm diameter of zone of inhibition against C. albicans and E. coli. Conclusion: A stable topical gel of CUR using β-CD and carbopol was successfully prepared which showed better in vitro diffusion with promising anti-inflammatory and antifungal action.

Aim: The study evaluates the in-vitro antimicrobial activity of Hunteria umbellata against Escherichia coli, Staphylococcus aureus and Streptococcus sp. Place and Duration of Study: The study was carried out for three months in 2019 in Biochemistry Laboratory, Department of Chemical Sciences (Biochemistry unit), School of Pure and Applied Sciences, Lagos State Polytechnic, Ikorodu, Lagos- Nigeria. Methodology: The qualitative and GC-MS analysis of Hunteria umbellata methanolic seed extract were determined using standard procedure. The antimicrobial activity was evaluated by the disc diffusion method and agar well diffusion method. The experimental data was resampled 1000 times to allow for higher degrees of freedom in carrying out t-test to test for the difference of the effect of in-vitro antimicrobial activity of H. umbellata against E. coli, S. aureus and Streptococcus sp using mathematical software R language (3.6.1 version). Line plots, histogram and t-test are used to explain the effect of antimicrobial activity of H. umbellate on the selected bacteria. MIC and MBC were determined using standard methods. Results: The Phytochemical analysis of methanolic seed extract of Hunteria umbellata showed the presence of secondary metabolites like saponins, tannins, flavonoids, steroids, phenol among others. GC-MS assay of the H. umbellata seed extract revealed the presence of eight different compounds. Agar well diffusion method was characterized by inhibition zones of 18.36±0.87, 19.13±1.03 and 21.62±2.53 mm for E.coli, S. aureus and Streptococcus sp respectively at 300 mg/ml-1 and 21.70± 1.60, 23.83± 2.64 and 28.57± 1.52 for E.coli, S. aureus and Streptococcus sp respectively at 500 mg/ml. The results of the analysis show that there is a significant difference between the effects of in-vitro antimicrobial activity of H. umbellate on 3001 and 500 mg/ml on each bacteria tested at 5% level of significance. E.coli, S. aureus and Streptococcus sp were tested against 12 standard antimicrobial agents, of which six was sensitive and another six was resistance to E .coli, seven was sensitive, and five was resistance to S. aureus while four was resistance and eight sensitive to Streptococcus sp. The minimum inhibitory concentration (MIC) for E.coli, S. aureus, and  Streptococcus sp were 250, 125 and 31.25 mgml-1 while their minimum bactericidal concentration (MBC) were 500, 250 and 125 respectively. MIC and MBC tests showed that H. umbellata methanolic seed extract had noticeable bactericidal effects with MBC/MIC values ranging between 2 to 4. The extract has strong potency against these microorganisms with Streptococcus sp being the most susceptible. Conclusions: Hunteria umbellata has potential as natural therapeutic agents against E. coli, S. aureus and Streptococcus sp and they may prevent pathogenic diseases.

Background:: Daucus gracilis Steinh belongs to the Apiaceae family. The flowers of this plant have been used by population of western Algeria for the treatment of mouth ulcers. However, very few studies exist of literature concerning the biological properties of Daucus gracilis Steinh flowers essential oil. Objectives:: The purpose of this work was to study the chemical composition of Daucus gracilis flowers essential oil and to evaluate their antimicrobial, insecticidal and anti-inflammatory properties. Methods:: The distilled essential oil was analyzed by GC and GC-MS. The antimicrobial activity of the essential oil was evaluated using two methods i) diffusion method, and ii) Micro dilution technique. The insecticidal activity of essential oil was evaluated against adults of Tribolium confusum by fumigant test. The in vitro assessment of anti-inflammatory property of essential oil was assessed by the protein denaturation method. Results:: Daucus gracilis flowers essential oil was mainly represented by Oxygenated monoterpenes such as Geranyl acetate (18.3%), Lavandulyl acetate (15.2%), Lavandulyl isobutyrate (13.6%) and Citronellyl isobutyrate (6.8%). According to the results of antimicrobial activity the essential oil of flowers presented prominent inhibitory action against Aspergillus flavus (0.06 μg/mL), followed by Staphylococcus aureus, Escherichia faecalis, Bacillus cereus and Candida albicans with MIC values of 0.125 μg/mL. The Daucus gracilis essential oil flowers proved to be very biocidal toward adults of Tribolium confusum, mortality of 100% of the population is noted with a dose of 2 μl/L air after 24 hours of exposure. Furthermore, the oil has shown has a very good inhibition of protein denaturation comparable to Diclofenac at concentration of 30 μL/mL. Conclusion:: Daucus gracilis essential oil can be used as pharmacological tools for inflammatory, antimicrobial and insecticidal properties.

ABSTRACTPhaseoulus lunatus L., a variety of baby lima bean, which is produced in the coastal region of Ecuador, is a profitable crop of that country. Various cultivars of this common bean are considered a sources for nutraceutical compounds, such as bioactive peptides. To assess the potential biologic activities of protein isolates and hydrolysates of P. lunatus baby lima beans, this study evaluates the proteins antimicrobial, antioxidant and anti-inflammatory activities. Antioxidant activity was measured by the TBARS method. In-vitro anti-inflammatory activity was measured by the inhibition of denatured protein as well as a diffusion method, according with CLSI guidelines by antimicrobial activity. Both fractions (isolate and hydrolysates) showed anti-inflammatory and antioxidant activity. However, protein hydrolysates (pH 5) had a better performance than protein isolates. The same effect was observed in antimicrobial activity, when protein hydrolysates had a broad-spectrum antimicrobial activity against Gram-negative and Gram-positive bacteria. These preliminary studies suggest that P. lunatus baby lima beans could have a considerable biological activity for nutraceutical applications.

Campomanesia xanthocarpa is a plant species traditionally used in the treatment of diabetes, fever, hypercholesterolemia, obesity, and urinary tract diseases. The anti-inflammatory effects of C. xanthocarpa leaves in mice were already known. Nevertheless, studies on the anti-inflammatory activity of its seeds are still lacking. The aim of this study was to investigate the anti-inflammatory activity and acute toxicity of C. xanthocarpa seed extract, obtained from supercritical CO2 extraction (SCCO2) at 40°C and 250 bar, in mice. GC/MS analysis revealed that β-caryophyllene is the major compound present in the C. xanthocarpa SCCO2 extract. The extract (60 mg/kg, p.o.) significantly reduced the nociceptive behavior in the second phase of the formalin test and prevented the paw oedema induced by carrageenan up to 6 h after carrageenan injection. The extract (0.1–1 μg/mL) inhibited neutrophils migration induced by LPS from E. coli in vitro. This antichemostatic effect was comparable to the effect of indomethacin. Acute administration (2000 mg/kg, p.o.) of C. xanthocarpa SCCO2 extract caused no mice mortality, demonstrating that the extract is devoid of acute toxicity. These data suggest that C. xanthocarpa seeds present anti-inflammatory activity and represent a source of anti-inflammatory compounds.

AbstractBackgroundDasapatrachurnam (DPC), a multicurative powder prepared from the leaves of 10 green leafy vegetables, was developed recently with known ethnobotanical and ethnopharmacological significance. However, its functional role in curing a disease is not yet scientifically proven. The present study aims at performing the phytochemical screening of DPC and exploring its possible activity as bacteriostatic, antineoplastic and anti-inflammatory.MethodsWe performed qualitative and Fourier transform infrared spectroscopy (FTIR) to find out the presence of active compounds and tested the bacteriostatic activity in four bacterial strains namely Bacillus subtilis, Escherichia coli, Streptococcus pyogenes and Staphylococcus aureus by agar well diffusion method. We further explored the antineoplastic activity in vitro in C6 and HEK293 cell lines by cell viability assay and the anti-inflammatory activity in the ovalbumin-induced inflammation in male Wistar rats.ResultsDPC showed 60% solubility in PBS and showed the presence of flavonoids and glycosides. FTIR results indicated the presence of alkyl, ketone and aldehyde groups. The bacteriostatic activity of DPC was higher (60%) in E.coli and lower (8%) in S.aureus, when compared to streptomycin. The anti-cancerous activity of DPC in C6 and HEK293 cancer cells was similar to their respective positive controls, curcumin and camptothecin. The anti-inflammatory activity of DPC was more evident with local administration in all the parameters studied in brain hippocampus, kidney, liver and spleen in ovalbumin-induced rats.ConclusionOur results, for the first time, suggest the potentiality of the DPC in treating bacterial diseases, cancer and also inflammation. Our results also suggest the possible therapeutic role of DPC in treating chronic kidney disease.

  Aqueous extract bark of Anthocleista schweinfurthii (ASE) was investigated for anti-arthritic, anti-inflammatory activities and phytochemicals screening. Phytochemical screening was carried out according, to LC-MS procedures, the anti-inflammatory effect of ASE was evaluated in vitro inhibition of 15-lipoxygenase (15-LOX) enzyme and  proteinase inhibiting action assays. The anti-arthritic activity was performed according to the Bovine Serum Albumin (BSA) test. The chemical composition of ASE showed that it was mainly composed of isocyanate, biphenyltriol, methoxyflavanone, soscoparin and glucopyroside. In the anti-inflammatory assays, ASE showed selective inhibition of 15-lipooxygenase with IC50 value of and moderate activity against the inhibition of proteinase action with IC50 value of L. Both the methods  showed that the ASE posses good anti-inflammatory activity. ASE had strong in vitro anti-arthritic activity with IC50 of , the positive control Diclofenac had IC50 value of  The result of this study justified the use of this plant in traditional Cameroonian medicine in the treatment of inflammation.
