ABSTRACTPhaseoulus lunatus L., a variety of baby lima bean, which is produced in the coastal region of Ecuador, is a profitable crop of that country. Various cultivars of this common bean are considered a sources for nutraceutical compounds, such as bioactive peptides. To assess the potential biologic activities of protein isolates and hydrolysates of P. lunatus baby lima beans, this study evaluates the proteins antimicrobial, antioxidant and anti-inflammatory activities. Antioxidant activity was measured by the TBARS method. In-vitro anti-inflammatory activity was measured by the inhibition of denatured protein as well as a diffusion method, according with CLSI guidelines by antimicrobial activity. Both fractions (isolate and hydrolysates) showed anti-inflammatory and antioxidant activity. However, protein hydrolysates (pH 5) had a better performance than protein isolates. The same effect was observed in antimicrobial activity, when protein hydrolysates had a broad-spectrum antimicrobial activity against Gram-negative and Gram-positive bacteria. These preliminary studies suggest that P. lunatus baby lima beans could have a considerable biological activity for nutraceutical applications.

Objective: The present study was designed to identify the bioactive phytochemicals and its antibacterial and in vitro anti-inflammatory potential of aqueous and methanolic seed extract of Citrullus lanatus.Methods: The phytochemical screening of both the aqueous and methanolic seed extract was carried out qualitatively to identify the major Phyto-constituents present in the extracts. The antimicrobial activity of the extracts was evaluated against six pathogenic bacterial strains by agar well diffusion method and the Minimum inhibitory concentration (MIC) was determined by broth dilution method. In vitro anti-inflammatory activity of C. lanatus seed extracts was evaluated by using human red blood cell (HRBC) membrane stabilization and inhibition of albumin denaturation method.Results: The results of the study indicated that both the extracts of the seed having antimicrobial activity, while the methanolic extract showed more significant activity against the tested organism than aqueous extract. Methanol extract had the lowest MIC of 1.562 mg/ml against Escherichia coli, Staphylococcus aureus, Klebsiella pneumonia, Pseudomonas aeruginosa and Bacillus subtilis, whereas in aqueous extract was highly sensitive to Bacillus subtilis, E. coli and Klebsiella pneumonia with MIC of 3.125 and 6.25 mg/ml, respectively. Methanolic extracts exerted comparative higher anti-inflammatory activity than aqueous extract.Conclusion: Present study provides a firm evidence to support that the synergistic effect of C. lanatus seed extracts having potent anti-inflammatory and antimicrobial property, which might serve as an effective drug for various microbial infections and inflammatory disorders.

Rosmarinus eriocalyx Jord. & Fourr is a well-known aromatic and medicinal plant whose consumption serves to remedy a number of disorders, evergreen bush endemic to Algeria. The present study aimed it investigating the in-vitro anti-inflammatory and antimicrobial for two extracts of Rosmarinus eriocalyx. The antioxidant activity was evaluated by DPPH and the reducing power essay, anti-inflammatory activity with the Human Red Blood Cell (HRBC) membrane stabilization method. However, the antimicrobial activity was tested with three bacterial strain Pseudomonas aeruginosa ATCC 27853, Staphylococcus aureus ATCC25923 and Bacillus subtilis ATCC6633. The results show an excellent antioxidant and an interesting inflammatory activities but a weak power against the used strains therefore. © 2020 International Formulae Group. All rights reserved. The aim of present study was to estimate the in vitro antioxidant potential and antimicrobial activity of hydroalcoholic extract of Vernonia cinerea. Antioxidant activity was assessed by using 2, 2- diphenyl-1-picryl-hydrazyl (DPPH) assay using ascorbic acid as standard antioxidant. The extract was found to scavenge effectively the free radicals. The total flavonoid contents were determined by established methods and were found to be 0.547 mg/100mg in quercitin equivalents. Antimicrobial activity was performed against 2 stains of human pathogenic bacteria by well diffusion method. Hydroalcoholic extract of Vernonia cinerea showed good antimicrobial activity against gram positive bacteria. The antioxidant activities may be attributed to the presence of significant amounts of flavonoid compounds. Results indicated that hydroalcoholic extract of Vernonia cinerea possess significant antioxidant effect in dose dependent manner, followed by the hydroalcoholic extract of Vernonia cinerea possessed good antimicrobial activity. Background: The Algerian medicinal plant Thymus pallidus Coss. is traditionally known to be used as an anti-infective, expectorant, healer of wounds and against inflammatory and gastrointestinal diseases. Objective: The aim of the present study is to investigate the antioxidants and the antibacterial activities of the aerial part extracts of this plant. Methods: Polyphenols extraction was performed with methanol 85%, followed by preliminary fractionation based on the polarity of the solvent. The extraction and fractionation process gave the crude extract (CrE), chloroform extract (ChE), ethyl acetate extract (EtAE) and aqueous extract (AqE). The antioxidant activity of extracts was evaluated using in vitro assays. While the antimicrobial activity was determined using disk diffusion method. Results: Quantitative analysis of polyphenol and flavonoids showed that EtAE includes the highest amount (P≤0.001) with 495.12±1.91 μg GAE/mg of extract and 97.25±2.84 μg QE/mg of extract, for both polyphenols and flavonoids respectively. The results of the antioxidant activity of extracts showed that the EtAE exhibited the highest inhibition activity on XO (IC50 =51.6±0.2 μg/ml) compared to other extracts (P≤0.001). Moreover, the EtAE was the most effective fraction in free radical scavenging assay against ABTS radical and hydrogen peroxide (EC50= 3.21±0.03 μg/ml and EC50 =12.12±0.05 μg/ml, respectively) compared to other extracts (P≤0.01). Using β-carotene/ linoleic acid bleaching assay, the AqE and ChE recorded a considerable inhibition effect on linoleic acid oxidation with a ratio of 81.81% and 80.86%, respectively. In the antimicrobial activity evaluation, the extracts exhibited moderate antibacterial activity against bacterial strain, with inhibition zone ranging from 07 to 14 mm. In addition, the EtAE showed a good action spectrum against six bacterial strains. In conclusion, the EtAE of Thymus pallidus showed good antioxidant activity and antibacterial effect. Conclusion: The obtained results revealed the presence of a strong correlation between the antioxidant and antibacterial activities of the extract and its total phenolic content. Furthermore, they reported that the EtAE has a considerable antioxidant capacity. This can be considered as an alternative natural source of antioxidants used generally as additives in food and pharmaceutical preparation.

Background:: Daucus gracilis Steinh belongs to the Apiaceae family. The flowers of this plant have been used by population of western Algeria for the treatment of mouth ulcers. However, very few studies exist of literature concerning the biological properties of Daucus gracilis Steinh flowers essential oil. Objectives:: The purpose of this work was to study the chemical composition of Daucus gracilis flowers essential oil and to evaluate their antimicrobial, insecticidal and anti-inflammatory properties. Methods:: The distilled essential oil was analyzed by GC and GC-MS. The antimicrobial activity of the essential oil was evaluated using two methods i) diffusion method, and ii) Micro dilution technique. The insecticidal activity of essential oil was evaluated against adults of Tribolium confusum by fumigant test. The in vitro assessment of anti-inflammatory property of essential oil was assessed by the protein denaturation method. Results:: Daucus gracilis flowers essential oil was mainly represented by Oxygenated monoterpenes such as Geranyl acetate (18.3%), Lavandulyl acetate (15.2%), Lavandulyl isobutyrate (13.6%) and Citronellyl isobutyrate (6.8%). According to the results of antimicrobial activity the essential oil of flowers presented prominent inhibitory action against Aspergillus flavus (0.06 μg/mL), followed by Staphylococcus aureus, Escherichia faecalis, Bacillus cereus and Candida albicans with MIC values of 0.125 μg/mL. The Daucus gracilis essential oil flowers proved to be very biocidal toward adults of Tribolium confusum, mortality of 100% of the population is noted with a dose of 2 μl/L air after 24 hours of exposure. Furthermore, the oil has shown has a very good inhibition of protein denaturation comparable to Diclofenac at concentration of 30 μL/mL. Conclusion:: Daucus gracilis essential oil can be used as pharmacological tools for inflammatory, antimicrobial and insecticidal properties.

Abstract Background Mahanintangtong is listed in the Thailand’s National List of Essential Medicines (NLEM). It is used to treat non-specific fevers and illnesses such as pharyngitis and chickenpox. In this study, we investigated the biological activities of the different medicinal plants used in the Mahanintangtong formula. Methods The plant materials were extracted by maceration and decoction. Antimicrobial activity, assessed by disc diffusion method, the minimum inhibitory concentration (MIC), and minimum bactericidal concentration (MBC) were compared with commercially available standard antibiotics. To elucidate the anti-inflammatory mechanisms, inhibition of nitric oxide (NO), tumor necrosis factor α (TNF-α), and interleukin 6 (IL-6) production was tested by Griess and ELISA techniques. Antioxidant activity was measured by ABTS and DPPH scavenging assays. Results The extracts with the best antimicrobial activities were carbonized Tectona grandis showing against Streptococcus pyogenes, Staphylococcus aureus, Methicillin-resistant Staphylococcus aureus (MRSA) and Pseudomonas aeruginosa. The ethanol extract of Dracaena loureiroi wood exhibited the highest NO and IL-6 inhibitory activity with IC50 values of 9.42 ± 1.81 and 12.02 ± 0.30 μg/mL, respectively. The ethanol extract of Pogostemon cablin had the highest TNF-α inhibitory with IC50 values of 10.68 ± 0.02 μg/mL. In anti-free radical testing, the ethanol extract of D. loureiroi displayed high antioxidant activity by both ABTS and DPPH assays. Conclusion The ethanol extracts from carbonized T. grandis and Mahanintangtong showed good antimicrobial activity, especially against S. pyogenes, and good anti-inflammatory activity. These findings are relevant to the pathogenesis of pharyngitis and justify additional studies to see if Mahanintangtong could have clinical utility.

A Gas Chromatography and Maas Spectroscopy method was developed and validated for determination and identification of α-humulene in a traditional medicinal herb Didymocarpous pedicellatus R. Br. (DP) (Gesneriaceae) and its poly herbal formulation, Safoof-e-pathar phori (SPP) including their essential oil. Hydrodistillation method were used for the isolation of essential oils from the leaves of DP and SPP herbal formulation. The proposed analysis method was found to be linear r2 = 0.999 in a wide concentration range (0.1-1000 µgmL-1), with good precision (RSD < 2.0%). Antimicrobial activity of α-humulene, DP and SPP oil was assessed by using agar well diffusion method against clinically important Gram-positive, Gram-negative bacteria and fungi. α-Humulene was found to be more active in contrast to Gram-negative bacteria while DP oil and SPP oil were exhibited maximum inhibition against fungal strains. Antioxidant activity of α-humulene, DP oil and SPP oil were determined using 2, 2-diphenyl-1-picryl hydrazyl radical (DPPH) 96 well plate method, which showed antioxidant activity in the following order: standard ascorbic acid (DPPH FRS50 = 2.40 µg) > DP oil (DPPH FRS50 = 3.68 µg) > SPP oil (DPPH FRS50 = 12.54 µg) > and α-humulene (DPPH FRS50 = 36.19 µg).

Our study is intended to prepare and evaluate the polyherbal formulation (PHF) using Nyctanthes arbor-tristis, Piper nigrum, and lemon juice. Poly Herbal Formulation F1 and F2 were synthesised and subjected to qualitative, quantitative phytochemical analysis and antioxidant activity was measured using a standard protocol. Antimicrobial activity of PHFs was observed by the agar well diffusion method. The anti-inflammatory and anti-diabetic activities (In-vitro) were studied in F1 by using inhibition of albumin denaturation method. TLC and FTIR are done for the structural elucidation in F1. The F1 and F2 exhibited the presence of flavonoids, alkaloids, tannins, phenols, terpenoids, carbohydrates, saponins, Glycosides and proteins. The F1 and F2 show significant activity against the standard in the antimicrobial and antioxidant activity. The presence of flavonoids and alkaloids may contribute to the anti-microbial activity. The Nitric oxide radical scavenging activity and a metal chelating assay of the F1 possess the highest antioxidant activity of 73.1% and 69% respectively. TLC confirms the presence of flavonoids in F1. FTIR spectrum shows the functional group of amines, alcohols, nitrites, aromatic and aliphatic compounds. These FTIR bands denote stretching and vibrational bands responsible for the compounds like flavonoids, terpenoids, phenols and amino acids. This investigation shows the excellent result in antimicrobial, antioxidant and in-vitro anti-diabetic and anti-inflammatory activities. In future, the PHF of F1 can be used as potent therapeutical agents and can be further exploited for their other beneficial effect in the pharmacological studies.

Objectives: The objectives of this study were to investigate the in vitro biological effect of the methanolic crude extract received from edible mushrooms; Flammulina velutipes on anti-inflammatory, antioxidant activity, anti-lipid peroxidation (LPO), antimicrobial activity, and anticancer activity. The scientific result obtained here was possibility of developing this variety as an ingredient in health/medical food purposes as well as a functional food.Methods: In this study, the methanol crude extract of F. velutipes has been performed and in vitro pharmacological properties such as anti-inflammatory, antioxidant activity, anti-LPO, antimicrobial activity, and anticancer activity were evaluated.Results: The results revealed that anti-inflammatory activity of crude extract was very significant with 79.81±0.87% of the dose-dependent manner. Antioxidant and anti-low-density lipoprotein (LDL) peroxidation activity of the extract showed its capacity of free radical scavenging with high-level antioxidant compounds and lipid preventive effects (2, 2-diphenyl-1-picrylhydrazyl: 79.81±0.01%, total phenol: 991.10±0.01 μg gallic acid equivalents/0.1 g dry matter, H2O2: 93.27±0.05%, LPO: 33.33±0.00%, and LDL: 57±0.03%, respectively). Analysis has also demonstrated that the methanolic extract of F. velutipes provided an antimicrobial activity to ESBL-producing Klebsiella pneumoniae. Moreover, F. velutipes extract showed a growth inhibitory effect in HepG2 cells with the 50% inhibitory concentration (IC50) value IC50=8.25±7.59 μg/ml.Conclusion: This indicated that crude extract from F. velutipes was scientifically proved as a potential source of therapeutic agent and be able to utilize as a functional ingredient for healthy or supplement food.

Ethanolic extracts of Pergularia tomentosa L. were used for the relative analysis of antioxidant and antimicrobial activity against two types of Gram-negative bacteria. (Escherichia coli G (-) and Pseudomonas aeruginosa G (-)) and a gram positive (Staphylococus aureus G (+)). Maximum activity was recorded against Escherichia coli, with a maximum inhibition diameter of 23mm using the disk diffusion method. In addition, the results revealed a higher bacterial activity against Escherichia coli with an inhibition diameter of 23mm compared to standard antibiotics Ampicillin/Nystatin with an inhibition diameter of 11.75mm. The study of the antioxidant activity by the DPPH and FRAP methods showed that vitamin C is the most active with an IC50 equal to 0.010 g/ml, followed by the crude extract, with a value of 0.012 g/ml and finally Tocopherol (VE) which seems to be the least effective with an IC50 of 0.015 g/ml. The results obtained in the present study suggest that Pergularia tomentosa L. can be used in the treatment of diseases caused by the organisms tested. Further chemical and pharmacological investigations can be carried out to isolate and identify the chemical constituents in selected plants responsible for antimicrobial activity.
