Purpose : to evaluate the effects of Methanolic extract of Cowpea (Vigna unguiculata) administration on vascular endothelial growthfactor (VEGF) expression in rats with corneal-alkali induced inflammation.Methods : This was an interventional experimental study. Sixty three rats were randomly selected, 3 rats as control, 60 rats were treatedwith 1 M NaOH (as inflammation model) and administered with 25 ìM,50 ìM and 100 ìM of genistein in Cowpea (Vignaungucuilata) extract four times daily. Alkali burn by 1 M NaOH infiltration using filter paper applicated on the center cornea of righteye for 60 seconds. Aquabidest as positive control and Cowpea (Vigna ungucuilata) extract treatment perfused immediately after alkali burn.VEGF observed at 6, 24, 48, 96 and 168 hours by immunohistochemical method.Results : Each dose of Cowpea (Vigna unguiculata) extract administration significantly decreased the VEGF expression ( p = 0,00). Differences of VEGF expression based on administration time were not significant (p=0,033). Interaction between time of administrationand dose of administration had influenced the VEGF expression (R Square(r2) =24,7 %). The Linear regression between VEGF expression and dose resulted in estimated effective dose (ED) with Y = 16.486–0.079 X( Y = V E G F e x p r e s s i o n , X =d o s e o f a d m i n i s t r a t i o n ) . Considering VEGF expression in normal cornea was 12, wefound the effective dose of methanolic Vigna unguiculata extract was 61.94 ìM.Conclusion :Methanolic extract of Cowpea(Vigna unguiculata) decreased the VEGF expression on alkali burn corneal inflammationin rats but no differences in VEGF expression based on time administration. Dose of 50 ìM genistein in Methanolic extract ofCowpea (Vigna unguiculata) was close to effective dose 61.94 ìM.Keywords : VEGF expression, alkali burn, Cowpea (Vigna unguiculata) extract, cornea.

Pycnarrhena cauliflora (Miers.) Diels., local name sengkubak, is one of indigenous plants from West Kalimantan that has been used as natural flavor. Pycnorrhena cauliflora is one of species of Menispermaceae family which is rich in bisbenzylisoquinoline alkaloids. This alkaloids are known to have various biological activities including antiprotozoal, antiplasmodial, antifungal and antibacterial activities. This study aimed to investigate antimicrobial activity of  the P. cauliflora (Miers.) Diels. methanolic extracts against gram-positive and gram-negative bacteria. The methanolic extract of P. cauliflora (Miers.) Diels., root, leaf and stem were prepared by maceration. The disk-diffusion method was then used to determine the antimicrobial activity of the extracts against Streptococcus pyogenes, S. mutants, Staphylococcus aureus, S. epidermidis, Salmonella typhi, Shigella flexneri, Pseudomonas aeruginosa and Escherichia coli after 18-24 h incubation at 37 oC. Amoxicillin was used as positive control for gram-positive bacteria and ciprofloxacin was used as gram-negative bacteria. The inhibition zones were then measured in mm. Analysis were conducted in duplicates. The results showed in general the methanolic extracts of P. cauliflora (Miers.) Diels. root (inhibition zone diameter= 10-23 mm) were more active than that leaf (0-15 mm) and stem (0-17 mm) extracts against gram-positive bacteria. The zone inhibition diameter of amoxicillin as positive control was 8-42 mm. In addition, the methanolic extracts of P. cauliflora (Miers.) Diels. root (12-17 mm) were also more active than that leaf (0-12 mm) and stem (0-12 mm) extracts against gram-negative bacteria. The zone inhibition diameter of ciprofloxacin as positive control was 33-36 mm. In conclusion, the methanolic extract of P. caulifloria (Miers.) Diels. root is the most extract active against both gram-positive and gram-negative bacteria. Further study will be focused to isolate active compounds in the methanolic extract of the root.

The aim of our study was to examine the involvement of renin-angiotensin system (RAS) in estrogen-induced lactotropes proliferation and vascular endothelial growth factor (VEGF) expression in rat pituitary. The study was performed on Fisher 344 rats underwent 8-day treatment with diethylstilboestrol (DES). The proliferation index (PCNA) and VEGF expression in pituitary sections were estimated using immunohistochemical methods. Treatment with DES increased the number of PCNA-positive cells, VEGF-positive cells, and VEGF-positive blood vessels in pituitary. Stimulatory effect of estrogen on cell proliferation and VEGF expression in blood vessels was attenuated by losartan, PD123319, and captopril. VEGF immunoreactivity in pituitary cells of DES-treated rats was decreased by AT1 antagonist and not changed by AT2 blocker and ACE inhibitor. Our findings suggest the involvement of RAS in DES-induced cell proliferation and VEGF expression in pituitary. Both the AT1 and AT2 receptors appear to mediate the estrogen-dependent mitogenic and proangiogenic effects in rat pituitary.

Pisang Ambon (Musa paradisiaca L.) is one type of bananas usually consumed by Indonesian people. Besides its flesh which has high nutrition, its peels also has antioxidant activity. Antioxidants has the ability to reduce oxidative damage in people’s body with diabetes mellitus. Therefore, this study aimed to determine the antioxidant activity of the aqueous  extract of Pisang Ambon peels and to determine it’s effective dose as an antidiabetic agent in hyperglycemic mice. This study used male mice which all have been intravenously induced with alloxan at a dose of 50 mg/kgBW. They were then divided into five groups. The first two groups got Na CMC 0.5% (negative control) and glibenclamide 0.65 mg/kgBW (positive control), while the other three got  the aqueous  extract of Pisang Ambon peels successively at doses of 400, 800, and 1200 mg/kgBW. The data were statistically analyzed using ANOVA (Analysis of Variance) at 95% confidence interval with parameter of blood glucose levels difference between before and after treatment. The results showed that the aqueous extract of Pisang Ambon peels had antidiabetic activity at an effective dose of 400 mg/kgBW in hyperglycemic mice which was comparable to glibenclamide

In the present study, we investigated the antinociceptive effects of the plant Vigna unguiculata spp dekindtiana using chemical and thermal tests in mice. The peripheral and the central analgesic activities of the methanol extract and its fractions were investigated in-vivo in albino mice using acetic acid induced-writhing test and hot plate models respectively. The result of the central analgesic effect showed that the methanol extract (VUME) at 400 mg/kg produced a significant (p<0.05) delay in reaction time in mice on hot plate compared to the control. Various fractions of the extract showed more potency compared to the crude extract. In acetic writhing model, the extract and the fractions demonstrated dose dependent reduction in writhing reaction induced by acetic acid in mice. The reduction was significant when compared to control which was suggestive of the analgesic effect of the plant. It was also seen that the extract and fractions showed an improved analgesic effect compared to diclofenac used as positive control in this model. Yohimbine (alpha adrenergic receptor antagonist) and cyproheptadine (serotonergic receptor antagonist) reversed the antinociceptic effect of the fractions in the hot plate model demonstrating the possibility of adrenergic and serotonergic involvement in eliciting the analgesic effect. Naloxone on the other hand, caused a reversal only in the butanol fraction meaning that this fraction may contain active principles that may mediate their analgesic effect through the opioid mechanism. In the writhing test, yohimbine abolished the analgesic effect of both hexane and butanol fractions. This may therefore, suggest that the analgesic effect of these fractions may be mediated through adrenergic pathway. In conclusion, the plant V. unguiculata subspecies dekindtiana possesses active principles with potential analgesic activity, establishing the folkloric use of the plant in managing pain.

Introduction. Increased angiogenesis in bone marrow is one of the characteristics of chronic myeloid leukaemia (CML), a clonal myeloproliferative disorder that expresses a chimeric bcr/abl protein. Vascular endothelial growth factor (VEGF) is one of the most potent and a specific regulator of angiogenesis which principally targets endothelial cells and regulates several of their functions, including mitogenesis, permeability and migration. The impact of elevated VEGF expression on the course of chronic myeloid leukaemia is unknown. Objective. The aim of this study was the follow-up of VEGF expression during the course of CML. Methods. We studied VEGF expression of 85 CML patients (median age 50 years, range 16-75 years). At the commencement of the study, 29 patients were in chronic phase (CP), 25 in an accelerated phase (AP), and 31 in the blast crisis (BC). The temporal expression (percentage positivity per 1000 analysed cells) VEGF proteins over the course of CML were studied using the immunohistochemical technique utilizing relevant monoclonal antibodies. It was correlated with the laboratory (Hb, WBC and platelet counts, and the percentage of blasts) and clinical parameters (organomegaly, duration of CP, AP, and BC) of disease progression. Results. The expression of VEGF protein was most pronounced in AP (ANOVA, p=0.033). The level of VEGF expression correlated inversely with the degree of splenomegaly (Pearson, r=-0.400, p=0.011). High expression of VEGF correlated with a shorter overall survival (log rank, p=0.042). Conclusion. Immunohistochemically confirmed significance of the expression of VEGF in dependence of the CML stage could be of clinical importance in deciding on the timing therapy. These data suggest that VEGF plays a role in the biology of CML and that VEGF inhibitors should be investigated in CML.

The present study was designed to authenticate the anti-anxiety activity (by using elevated plus maze model) of methanolic extract of the leaves of Gentiana diffusa. by authors Swiss Albino mice were treated with different doses of the leaf extracts (200 mg / kg p.o.) and Diazepam (2mg/ kg, p.o) was used as a positive control. Results of study show that methanolic extract in higher doses (200 mg/kg) possesses marked anti-anxiety activity and was comparable to the effect produced by diazepam. The plant can be developed as a commercial source of anxiolytic agent. Further studies are in process to isolate the active constituent responsible for this activity and mechanism of action.
