Abstract The addition of reproductive fluids (RF) to the culture media has shown benefits in different embryonic traits but its long-term effects on the offspring phenotype are still unknown. We aimed to describe such effects in pigs. Blood samples and growth parameters were collected from piglets derived from in vitro-produced embryos (IVP) with or without RF added in the culture media versus those artificially inseminated (AI), from day 0 to month 6 of life. An oral glucose tolerance test was performed on day 45 of life. We show here the first comparative data of the growth of animals produced through different assisted reproductive techniques, demonstrating differences between groups. Overall, there was a tendency to have a larger size at birth and faster growth in animals derived from in vitro fertilization and embryo culture versus AI, although this trend was diminished by the addition of RFs to the culture media. Similarly, small differences in hematological indices and glucose tolerance between animals derived from AI and those derived from IVP, with a sex-dependent effect, tended to fade in the presence of RF. The addition of RF to the culture media could contribute to minimizing the phenotypical differences between the in vitro-derived and AI offspring, particularly in males.

The experiment was conducted to clarify the growth and yield response of two rice cultivars, BR55 and BR43 under salt stress. Six different concentrations of NaCl viz 50, 100, 150, 200, 250 and 300 mM and distilled water (control) were applied on the rice cultivars which were grown under pot culture condition. Growth parameters like plant height, tiller number, leaf number and leaf area were negatively affected by salinity in both cultivars. Salt stress caused a significant reduction in yield in both cultivars of rice. Growth reduction was higher in BR43 than in BR55.The reduction in yield and yield parameters were found to be lower in BR55 than those in BR43. The results obtained in the present study suggest that BR55 showed higher salt tolerance than in BR43. Jahangirnagar University J. Biol. Sci. 7(2): 1-12, 2018 (December)

A field survey was conducted in 12 districts of Assam viz., Jorhat, Golaghat, Nagaon, Marigaon, Goalpara, Dibrugarh, Tinsukia, Lakhimpur, Dhemaji, Sivsagar, Kamrup and Barpeta. A total of 92 root samples were collected and 37 bacterial isolates were isolated from commercial banana cultivars. The culture filtrates extracted from 37 endophytic bacterial isolates, were screened against southern root-knot nematode, Meloidogyne incognita in vitro and under pot culture studies. The five bacterial isolates viz., EB4, EB8, BC1, BC11 and BC12 showed 100% inhibition of egg hatching and juvenile mortality of M. incognita with an exposure period of 48 and 72h. On seed bacterization, with these five promising isolates, two isolates viz.EB4, BC1 significantly enhanced germination percentage (33.33, 25.31%) and vigour index (75.5, 64.39%) of paddy, receptively. The potential bacterial isolates viz., BC1 and EB4 were identified as Lysinibacillus sp. and Pseudomonas sp., respectively , based on the morphological phenotypic and biochemical characterization. The pot culture experiment revealed that the bacterial endophytes viz., Lysinibacillus sp. (BC1) Pseudomonas sp. (EB4) significantly reduced the soil (61.64, 56.71%) and root nematode population (77.29, 68.87%), number of adult females (73.97, 69.89%), egg masses (85.63, 80.11%) and root-knot index (1.33, 1.67) of M. incognita compared to untreated control. The bacterial endophytes viz., Pseudomonas sp. (EB4), Lysinibacillus sp. (BC1) were also significantly increased the growth parameters viz., shoot length (43.33, 39.18%), and root length (78.24, 59.26%) and pesudostem girth (58.38, 52.13%).

AbstrakVanda hookeriana Rchb.f. merupakan anggrek yang dilindungi dan keberlangsungan hidupnya di alam terancam punah, sehingga perbanyakan anggrek secara in vitro sangat diperlukan. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak yeast dan pisang raja terhadap pertumbuhan tunas embrio anggrek V. hookeriana Rchb.f. Desain penelitian ini menggunakan RAL faktorial, faktor pertama adalah ekstrak yeast dengan 3 taraf: 0,5 g/L, 1 g/L, dan 1,5 g/L, faktor kedua adalah buah pisang raja dengan 3 taraf: 50 g/L, 100 g/L, dan 150 g/L. Penelitian dilakukan menggunakan teknik kultur jaringan, pengamatan dilakukan setiap bulan sekali dengan mengamati aspek morfologi plantlet. Media yang digunakan adalah Vacin-Went (VW) + kombinasi ekstrak yeast dan pisang raja. Parameter pengamatan berupa tinggi tunas, berat kering tunas, jumlah daun, diameter daun, panjang daun, berat kering daun, jumlah akar, diameter akar, panjang akar, berat kering akar, dan morfologi tunas. Hasil analisis menunjukkan bahwa perlakuan pisang raja 150 g/L + ekstrak yeast 0,5 g/L merupakan konsentrasi yang paling berperan terhadap parameter pertumbuhan anggrek V. hookeriana Rchb.f. Penelitian ini berhasil membuktikan bahwa penambahan komponen organik dalam media kultur jaringan memberikan respon pertumbuhan dan perkembangan yang baik pada eksplan. Komponen organik membantu proses fisiologis eksplan, karena komponen organik mengandung senyawa potensial yang dibutuhkan selama proses pertumbuhan dan perkembangan. Abstract Vanda hookeriana Rchb.f. is one of protected orchid and its survival in the wild was threatened by extinction, so orchid propagation was needed. This research aimed to determine the effect of yeast and pisang raja extracts to embryonal shoot growth of Vanda hookeriana Rchb.f. Experimental design used factorial RAL, first factor was yeast extract with 3 treatments (0.5 g/L; 1 g/L; & 1.5 g/L). Second factor was pisang raja with 3 treatments (50 g/L; 100 g/L; & 150 g/L). The research was conducted using in vitro culture techniques, observations were made once a month by observing the plantlet morphological parameters. The medium used was Vacin-Went (VW) + combination of yeast and pisang raja extracts. Observation parameters were shoot height, shoot dry weight, leaf number, leaf diameter, leaf length, leaf dry weight, number of roots, root diameter, root length, root dry weight and shoot morphology. The analysis showed that pisang raja treatment 150 g/L + yeast extract 0.5 g/L was the best concentration for the growth parameters of Vanda hookeriana Rchbf. This research were succeeded in proving that the addition of organic components in tissue culture media gave better response to growth and development of the explants. Organic components helps the explants in physiological ways because of  the organic components contain potential compounds needed during the growth and development process.

Abstract This study aimed to evaluate the growth, respiratory activity, and biodegradation of chlorpyrifos in cultures of Azotobacter vinelandii ATCC 12837. A strategy based on the modification of culture media and aeration conditions was carried out to increase the cell concentration of A. vinelandii , in order to favor and determine its tolerance to chlorpyrifos and its degradation ability. The culture in shaken flasks, using sucrose as a carbon source, significantly improved the growth compared to media with mannitol. When the strain was cultivated under oxygen-limited (5.5, 11.25 mmol L- 1 h- 1 ) and no-oxygen-limited conditions (22 mmol L -1 h -1 ), the growth parameters were not affected. In cultures in a liquid medium with chlorpyrifos, the bacteria tolerated a high pesticide concentration (500 ppm) and the growth parameters were improved even under conditions with a reduced carbon source (sucrose 2 g L -1 ). The strain degraded 99.6 % of chlorpyrifos at 60 h of cultivation, in co-metabolism with sucrose; notably, A. vinelandii ATCC 12837 reduced by 50% the initial pesticide concentration in only 6 h (DT 50 ).

A protocol for efficient and rapid regeneration of medicinal plant (Valeriana officinalis L.) was developed by in vitro culture of crown division. The effect of three different culture media, MS, B5 and MB (a new combined medium) containing 12 combinations of BAP and NAA was evaluated on regeneration and growth parameters. Results revealed a significant response of media used for shoot regeneration. The best frequency of regenerated plantlets (80%) and number of regenerated shoots (9.43) were obtained on MB containing BAP (0.5 mg/l) + (0.25 mg/l). The maximum length of shoots was found in the MB with BAP (1 mg/l) + (0.5 mg/l). A positive influence of time was noticed on rooting of regenerated shoots. All acclimatized plantletswere successfully established in a greenhouse and showed normal growth. This protocol provides a technique for mass propagation of valerian and helpful for future genetic improvement studies.Plant Tissue Cult. & Biotech. 24(2): 263-271, 2014 (December)

This work dealt with the study of growth parameters, pigments production, and bioenergetic aspects of the microalga Dunaliella tertiolecta in different culture media. For this purpose, cultures were carried out in Erlenmeyer flasks containing F/2 medium, Bold’s Basal medium, or an alternative medium made up of the same constituents of the Bold’s Basal medium dissolved in natural seawater instead of distilled water. D. tertiolecta reached the highest dry cell concentration (Xmax = 1223 mgDM·L−1), specific growth rate (µmax = 0.535 d−1), cell productivity (PX = 102 mgDM·L−1·d−1), and photosynthetic efficiency (PE = 14.54%) in the alternative medium, while the highest contents of carotenoids (52.0 mg·g−1) and chlorophyll (108.0 mg·g−1) in the biomass were obtained in Bold’s Basal medium. As for the bioenergetic parameters, the biomass yield on Gibbs energy dissipation was higher and comparable in both seawater-based media. However, the F/2 medium led to the highest values of moles of photons absorbed to produce 1 C-mol of biomass (nPh), total Gibbs energy absorbed by the photosynthesis (ΔGa) and released heat (Q), as well as the lowest cell concentration, thus proving to be the least suitable medium for D. tertiolecta growth. On the other hand, the highest values of molar development of O2 and consumption of H+ and H2O were obtained in the alternative medium, which also ensured the best kinetic parameters, thereby allowing for the best energy exploitation for cell growth. These results demonstrate that composition of culture medium for microalgae cultivation has different effects on pigments production, growth kinetics, and bioenergetics parameters, which should be taken into consideration for any use of biomass, including as raw material for biofuels production.

Skeletal muscle-tissue engineering can be applied to produce cell-based meat for human consumption, but growth parameters need to be optimized for efficient production and similarity to traditional meat. The addition of heme proteins to plant-based meat alternatives was recently shown to increase meat-like flavor and natural color. To evaluate whether heme proteins also have a positive effect on cell-based meat production, bovine muscle satellite cells (BSCs) were grown in the presence of hemoglobin (Hb) or myoglobin (Mb) for up to nine days in a fibrin hydrogel along 3D-printed anchor-point constructs to generate bioartificial muscles (BAMs). The influence of heme proteins on cell proliferation, tissue development, and tissue color was analyzed. We found that the proliferation and metabolic activity of BSCs was significantly increased when Mb was added, while Hb had no, or a slightly negative, effect. Hb and, in particular, Mb application led to a very similar color of BAMs compared to cooked beef, which was not noticeable in groups without added heme proteins. Taken together, these results indicate a potential benefit of adding Mb to cell culture media for increased proliferation and adding Mb or Hb for the coloration of cell-based meat.

This study determined the effect of growth media and culture concentration on the growth, proximate, and microelement composition of Ankistrodesmus falcatus. The culture of A. falcatus was done using three media, namely Modified COMBO Medium (COMBO), Bold’s Basal Medium (BBM), and Bristol, at two concentrations (50% and 100%). The results obtained show that the cell density (>3.5 × 107 cells/mL), optical density (>0.24), and specific growth rate (>0.429%/day) were significantly higher (p ≤ 0.05) in BBM and COMBO than in Bristol (<3.1 × 107 cells/mL; <0.23; <0.416%/day, respectively) at both concentrations. However, biomass was higher in BBM (>2.20 g/L) than in COMBO (1.87–2.13 g/L), while Bristol had the lowest value observed (1.70–1.73 g/L). Biochemical and microelement composition showed variations between media and at the different concentrations, with higher values observed in BBM and COMBO. Based on the growth parameters and nutritional composition, it was concluded that BBM and COMBO were better media for the propagation of A. falcatus growth than Bristol. The study also demonstrated that the microalgae can be cultured using half of the media’s concentration to lower production costs.
