Background: Major changes in the metabolic functions in high-yielding dairy cows occur during the transitional period and during lactation. Parturition and lactogenesis are accompanied by many physiological changes that facilitate the maintenance of homeostasis Consequently, physiological situations leading to a negative energy balance are coupled to an increased uncontrolled rate of body fat mobilisation and the increased fatty acids accumulation in hepatocytes and blood ketone bodies, resulting in disturbances of the morphological and physiological liver integrity. The objective of the present study was to estimate metabolic status in late pregnant, early lactation and full lactation Holstein dairy cows on the basis changes of blood concentrations of selected biochemical markers.Materials, Methods & Results: The experiment included 36 Holstein cows. Three groups of clinically healthy cows were chosen from the herd. Group 1 consisted of late pregnant cows (n = 12) from 30 to 1 day (20 ± 15) to partus; Group 2 comprised early lactation cows (n = 12) in the first month of lactation (15 ± 12 days), and Group 3 included full lactation cows (n = 12) between 60 to 90 days of lactation (81 ± 30 days). Blood samples were collected from all cows, by  punction of the jugular vein. Biochemical testing for markers in the blood serum showed significantly lower values (P < 0.05) of glucose, TG, Tchol. and urea in early cows than in full lactation and late-pregnant cows. The blood concentration of NEFA and BHB  was significantly increased (P < 0.05) in the group of cows in early lactation compared to the other groups of cows. The mean tBIL. concentration and the serum AST, GGT and ALT  activities were markedly increased (P < 0.01) in the lactation cows compared to the late pregnant cows. Furthermore, the intensity of lipomobilisation (NEFA or BHB concentrations) correlated positively (P < 0.05) with the markers of cell damage or liver function impairment (tBIL., serum AST, ALT and GGT activities), but negatively (P < 0.05) with the circulating concentrations of compounds synthesised in liver (glucose, TG and urea).Discussion: In dairy cows, it was observed that up to 50 % of females exhibited some lipid accumulation in liver in the first 3 weeks after calving and that fatty liver occurs primarily in this period.  Liver can be categorized into mild, moderate and severe fatty liver as dependent on the degree of pathology and a mild fatty infiltration of liver in dairy cows during lactation is considered to be almost physiological. The blood BHB and NEFA concentrations are markers of lipomobilisation and positively associated with the ketosis and liver steatosis intensity. The simultaneous and parallel variations observed between the extent of the fat infiltration in liver and the serum BHB and NEFA concentrations in puerperal cows clearly indicated that the intense lipomobilisation in the post-partum period has induced lipid overloading and ketogenesis in the liver. On the other hand, it was observed significant decreases in the serum biochemical markers, at least partially synthesised in the liver, such as glucose, TG, Tchol., urea, albumin and TP during the postpartum period. However, the liver steatosis has induced some cellular lesions as evidenced by significant increases in the serum tBIL. concentrations and in the AST, ALT, GGT and LDH enzyme activities in puerperal cows. All these biochemical metabolites may be used as important biochemical markers in the determination of the metabolic status in high-yielding dairy cows during the transition period and during lactation.

The objective of the present study was to determine metabolic status in late pregnant (n = 15) and puerperal (n = 15) Simmental dairy cows. The various blood metabolites and serum enzyme activities were determined by photometric methods. The early lactation cows had the indicative values of the beta-hydroxybutyrate (BHB) (> 1.20 mmol/l) but did not display any clinical signs, which means that they had a typical subclinical condition. The lipomobilization markers, serum BHB and non-esterified fatty acids (NEFA) concentrations, were markedly enhanced (P<0.05) in early lactation cows. Liver steatosis compromised hepatocyte metabolism, leading to significantly weaker (P<0.05) circulating concentrations of glucose, triglyceride (TG) and urea, and induced some cellular lesions as evidenced by significant increases (P<0.05) in the serum bilirubin concentrations and theaspartate transaminase (AST) enzyme activities in early lactation cows. On the basis biochemistry estimation, early lactation cows had metabolic disturbances which were associated with ketosis, and some degree of hepatic lesions, probably due to fat infiltration.

Introduction: Hormonal and metabolic changes, as well as energy imbalance, can affect health, production and reproductive performance of dairy cows. In the present study, we evaluated phagocytosis and respiratory burst neutrophil activity during the transition period and early lactation and compared it with biochemical and hematological parameters in dairy cows. Methodology: Simmental cows (n = 21) were enrolled in the study. Whole blood samples were collected weekly from 3 weeks pre- calving until 6 weeks post calving. Basic metabolic and blood parameters were assessed by routine laboratory analyses, while neutrophil functions were analyzed by commercial test kits. Results: Optimal neutrophil response was observed pre and post calving. The highest value was recorded in the 6th week after calving (89.54 ± 7.61%) and being significantly higher (p < 0.01) as compared to values recorded at two and one week before and one week after calving. The percentage of activated neutrophils was high during the entire study period: from 70.80 ± 5.22% at the beginning of the study to 89.54 ± 7.61% at the end of the study. During the study period, production of Reactive Oxidative Species by neutrophils was positively correlated with β-hydroxybutyrat and non-esterified fatty acids values (0.454** and 0.423**, respectively) and calcium levels (0.164* and 0.212**, respectively). Conclusions: The most prominent changes in all parameters had no influence on phagocytic and respiratory burst activity of neutrophils. Neutrophil function is preserved at the optimal level during the transition period and early lactation in Simmental cows.

The objective of the present study was to evaluate the effects of an antioxidant and anti-inflammatory compound found in rosemary plants (Salvia rosmarinus) named carnosic acid during the transition period of dairy cows. From day 1 to 3 after calving, 16 multiparous Holstein cows received a daily intravenous infusion of either 500 mL of saline (NaCl 0.9%; Saline; n = 8) or carnosic acid at a rate of 0.3 mg/kg of BW supplied in 500 mL of saline (CA; n = 8). Blood samples were taken at –7, 2, 5, 7, 14, and 21 d relative to parturition, then analyzed for metabolites related to energy metabolism, muscle mass catabolism, liver function, inflammation, and oxidative stress. CA infusion tended to improve milk performance; however, DMI was unaffected by treatment. At 2 d relative to parturition, CA cows had lower blood concentrations of haptoglobin, paraoxonase, FRAP, and NO2– than saline cows. After treatment infusions, haptoglobin remained lower in CA cows than saline at 5 d relative to parturition. Our results demonstrate that carnosic acid promoted positive responses on inflammation and oxidative stress biomarkers and may promote beneficial effects on lactation performance in peripartal dairy cows.

The objective of the present study was to investigate nutritional and metabolic status in Simmental cows during early and mid-lactation. Fifteen early lactating cows and 15 mid lactating cows were chosen for the investigation. Blood samples were collected to measure beta-hydroxybutyrate (BHB), nonesterified fatty acids (NEFA), triglycerides (TG), glucose and the activity of aspartate transaminase (AST). Early lactation as compared to mid lactating cows were found to have significantly higher (P<0.05) blood serum concentrations of NEFA, BHB and AST and lower blood serum concentrations of glucose (P<0.05) and TG (P>0.05). Significantly negative correlations were observed between BHB and glucose (P<0.01), BHB and TG (P<0.05), NEFA and glucose (P<0.05). Significantly positive correlations were observed between NEFA and BHB (P<0.05), NEFA and AST (P<0.05), glucose and TG (P<0.01). The results suggest that these parameters can serve as useful indicators of the nutritional and metabolic status of dairy cows during lactation.

The aim of the present study was to evaluate the potential for diagnosis of fatty liver by means of ultrasonographic measurement of liver and perivisceral adipose tissue as an alternative to blood indicators of lipomobilization and liver biopsy in periparturient high-yielding dairy cows. Thirty cows were enrolled and divided into two groups. The evaluation of body condition score (BCS), non-esterified fatty acids (NEFA), β-hydroxybutyrate (BHB), liver and perivisceral adipose tissue ultrasonographic measurement and histological liver lipid content (GdL) was performed at 15 ± 5 d prepartum (T0), 10 ± 2 d postpartum (T1), 30 ± 2 d postpartum (T2) and 50 ± 2 d postpartum (T3). Mesenteric fat thickness (the thickness of the perivascular adipose tissue) measured on ultrasound was shown to be an independent determinant of fatty liver. The cut-off of the ultrasonographic evaluation of the liver may be useful as a first and practical approach in identifying fatty liver. In conclusion, a non-invasive and reliable diagnostic method for predicting the risk of fatty liver in high yielding dairy cows has been demonstrated.

Dairy cows mobilise fat during early lactation to meet the energy deficit produced by increasing milk yield and reduced appetite. This can result in the deposition of fat in liver, kidney, muscle and other tissues. This “fatty liver syndrome” is associated with reduced fertility and an increased susceptibility to metabolic and infectious disease. Attempts to prevent the development of fatty liver are aimed at reducing fat mobilisation by: 1) calving cows in lean condition, thus increasing appetite and 2) by encouraging high feed intakes immediately after calving by the provision of feed of high energy density on a little and often basis. A useful system in this context might be out-of-parlour concentrate dispensers

The effects of precalving fat sources on fatty acid (FA) profile of colostrum and milk and on metabolic parameters in early lactation were determined. Two fat supplements (target: 21% fat, DM basis) fed at 1.8 kg DM d-1 were compared: Control (C; containing fractionated palm oil rich in 16:0) and Linseed (L; containing extruded linseed, rich in 18:3n-3). Sixteen dry Holstein (n = 7) and Brown Swiss (n = 9) cows were assigned to four groups receiving the supplements from 5 wk pre- to 4 wk post-calving in different sequences: CC, CL, LC and LL. Treatments did not affect metabolic parameters, milk yield or composition. Linseed supplementation increased proportions of 18:0, 18:1, 18:2 and 18:3 in colostrum and milk (except 18:1 for colostrum). The highest n-3 FA proportion was observed in milk of LL cows. For most 16 and 18 FA, a fast response occurred within the first week of lactation for cows that switched treatments over calving. Carry-over to milk of 18:3 and its biohydrogenation products from linseed fed precalving was low. Pre-calving feeding strategies to favour incorporation of these FA into early lactation milk were not successful, but the reasons for this failure remain unclear.Key words: Linseed, omega-3 fatty acids, transition period, milk, colostrum, dairy cows

Earlier studies have reported a relationship between degree of fatty infiltration of the liver after calving and calving interval (Reid, Roberts & Manston, 1979; Reid, 1983). The aimof the present investigation was to relate the severity of fatty liver with cow reproductive physiology and performance.
