Phosphate Solubilization Potentials of Rhizosphere Fungi Isolated from Insecticide Treated Soil

Abstract Insecticides are used widely to control a variety of pests and often residues of these insecticides are left in soil which may have impact on the phosphate solubilization potentials of rhizosphere fungi. Rhizosphere soils were collected from carefully uprooted cowpea seedlings with hand trowel on 50, 70 and 90th days of germination containing the lambda-cyhalothrin and dimethoate insecticides. Fungi associated with the above samples were identified by standard microbiological techniques. Screening for phosphate solubilization potential of the isolates was done by spot inoculation on Pikovskaya agar by measuring the clear zones around the colonies supplemented with tricalcium phosphates [Ca3(PO4)2]. The phosphatase produced by the fungal species was optimized using parameters such as incubation time, pH, temperature, carbon source and nitrogen source in submerged fermentation. The isolated rhizosphere fungi were identified as Rhizopus stolonifer, Aspergillus niger, Aspergillus fumigatus, Aspergillus terreus, Trichoderma viride, Arthroderma fulvum and Fusarium oxysporum. Among the isolates, Trichoderma viride showed the best ability to solubilize phosphate with solubilization index of 2.82 with dimethoate of 12.5ml/L of water. The optimization study for the enzyme production showed that the best incubation time for phosphatase production was 72 hours by Trichoderma viride. It was observed that the optimum pH for production of phosphatase by Trichoderma viride was 6.5. This study suggests that all the isolated fungi especially Trichoderma viride can adapt to these insecticidal treatments, which make them useful as biofertilizers to increase uptake of phosphorous in plants.

Pieces of art stored in museum depots and display rooms are subject to fungal colonization that leads to bio-deterioration processes. Deteriorated wooden sculptures and art photographs temporarily stored in the quarantine room of the Cultural Center of Belgrade were subject to mycological analyses. Twelve fungal species were identified on the wooden substratum and five species were detected on photograph surfaces. Trichoderma viride, Chaetomium globosum and Alternaria sp. were the fungi with proven cellulolytic activity detected on the examined cellulose substrata. Indoor air mycobiota were estimated to 210.09 ? 8.06 CFU m-3, and the conidia of fungus Aspergillus niger were the dominant fungal propagules in the air of the examined room.

Trichoderma species are known as excellent biocontrol agents against soil-borne pathogens that cause considerable crop losses. Eight strains of Trichoderma were isolated from five Egyptian regions. They identified based on translation elongation factor-1α (TEF1) sequencing as four different Trichoderma species: Trichoderma asperellum, Trichoderma harzianum, Trichoderma viride, and Trichoderma longibrachiatum. Optimal growth conditions (temperature and media), and the phosphate solubilization capability of Trichoderma strains were evaluated in vitro. Further, the ability of these strains to antagonize Fusarium solani, Macrophomina phaseolina, and Fusarium graminearum was also evaluated. The results revealed that Trichoderma harzianum (Th6) exhibited the highest antagonistic ability against F. solani, M. phaseolina and F. graminearum with inhibition rates of 71.42%, 72.97%, and 84.61%, respectively. Trichoderma viride (Tv8) exhibited the lowest antagonism against the same pathogens with inhibition rates of 50%, 64% and 69.23%, respectively. Simple-sequence repeats (SSRs) and random amplified polymorphic DNA (RAPD) markers were used to evaluate the genetic variability of the Trichoderma strains. The results revealed that of 45 RAPD amplified bands, 36 bands (80%) were polymorphic and of SSRs amplified 36 bands, 31 bands (86.11%) were polymorphic. The amplification of calmodulin and β-1,3-endoglucanase was noted at 500 bp and 230 bp, respectively. Data indicated that T. viride (Tv8) had the highest phosphate solubilization index (10.0 mm), while T. harzianum (Th6) had the lowest phosphate solubilization index (4.0 mm). In conclusion, T. harzianum (Th6) had the highest antagonistic activity in dual culture assay along with the growth rate; while T. viride (Tv8) had the highest phosphate solubilization activity. There are still gaps in obtaining new formulations, selecting potent Trichoderma strains to confirm disease control in planta. For improving Trichoderma recommendation in the organic agricultural system and sustaining the fertility of the soil, the field application of highly antagonistic biocontrol agents in different types of soil and plant species will be the first approach toward bio-pesticide treatments along with bio-fertilizer inoculation. Furthermore, secondary metabolites will be investigated for the most promising strains with the combination of different pathogens and application timing.

The research was aimed at investingating acid detergent  fibre   neutral detergent Fiber content in corn straw fermented using Trichoderma Viride in different Incubation Time. It applied compeletey Randomizend Design consisting Of  4 treatments  and  4 repetitions,  hence, it resulted in 16 units  of  an experiment. The Treatments consisted of corn straw  withhout inoculation (control), 7%  corn straw  Fermented  with Trichoderma  viride in  1 week incubation time, 7%  corn  straw  Fermented  with Trichoderma Viride in  2 weeks incubation time, and 7% corn  straw fermented with Trichoderma Viride in 3 weeks incubation time.Analysis of variance result shows that corn straw fermented using Trichoderma  Viride in different Incubation Time gave significant inflluance toward ADF content (p0,05).  Besides ,  corn straw fermentd using Trichoderma Viride in different incubation Time gave significant influence toward NDF content(p0,05).

Two types of symptoms, blight and anthracnose, were recorded on two species of Gerbera viz. Gerbera aurantiaca L. and Gerbera viridifolia L. The most common fungal species isolated from Gerbera spp. were Alternaria citrii, A. tennuisssima, Aspergillus flavus, A. fumigatus, A. niger, A. terreus, Bipolaris hawaiiensis, Chaetomella raphigera, Cladosporium cladosporoides, Colletotrichum capsici, C. coffeanum, C. dematium, C. gleosporioides, C. lindemuthianum, C. musae, C. orbiculare, Colletorichum sp.1, Colletotrichum sp.2, Colletotrichum sp.3, Colletotrichum sp.4, Curvularia clavata, C. lunata, Fusarium nivale, F. semitectum, Phomopsis sp., Penicillium sp., Pestalotia sp., Rhizopus stolonifer, Trichoderma viride, one cleistothecial ascomycetes and one sterile fungus. The frequency percentage of association of Colletotrichum spp. with Gerbera spp. was higher than any other fungi. Bipolaris hawaiiensis is a new record for Bangladesh. Among the isolated fungi Alternaria citrii, A. tennuissima, Colletotrichum capcisi, C. dematium, C. coffeanum and Curvularia clavata were found to be pathogenic to Gerbera plant. In this optimization study, Pseudomonas aeruginosa SU-1 was producing keratinase at optimal condition of 4 days, pH – 7 and temperature 37 0C, where it was producing 23.7 U/mL After the one factor at a time, RSM was performed to understand the combination of the physical parameter that ends up for the maximum production of keratinase enzyme and the degradation percentage. The study involved in three variables (pH(A), temperature(B) and Incubation Design (C)) in three ranges (-1,0,+1) using Box-Behnken Design (BBD). The results of the analysis of variance and regression analysis of the second order model showed that the factorial effect if the degradation. The optima of the variables pH - 7, temperature - 30 and incubation time – 4 days. The isolated Pseudomonas species was subjected to feather degradation for 4 days and it was degrading 55.26 %. Keratinase was to be size of 56KDa.

Fruit wastes were incubated with the mixture of cellulolytic fungi Penicillium citrinum, Aspergillus oryzae, and Trichoderma viride to hydrolyze the cellulosic components and to increase the degree of degradation. . The batch experiments are statistically designed and performed using Box-Benhken method of Response Surface Methodology to investigate the influence of major parameters viz., incubation time, temperature, pH, moisture content and substrate concentration on cellulase enzyme production. Maximum cellulase production of 2.03 Units/ml (U/ml) was detected by the RSM method in a mixed culture containing fungi at a ratio of 1: 1: 1 under optimal conditions at an incubation time of 5.27 days, a temperature of 34.09 °C, pH 4.85, moisture content of 63.83% and a substrate concentration of 5.03%.

Thirty five fungal species, representing 20 genera were found to be associated with Bipolaris leaf blight infected leaves of 21 wheat varieties, collected from eight districts (Dhaka, Gazipur, Dinajpur, Joypurhat, Pabna, Sirajgonj, Kushtia and Chuadanga) of Bangladesh. Fungi were isolated following tissue planting method. The isolated fungi were Alternaria alternata, A. triticina, Arthirinium sp., Aspergillus flavus, A. fumigatus, A. niger, A. terreus, Aspergillus sp., Bipolaris cynodontis, B. oryzae, B. sorokiniana, B. tetramera, B. victoriae, Bispora antenata, Chaetomium globosum, Chaetophoma sp., Cladosporium cladosporioides, Coniothyrium sp., Curvularia affinis, C. lunata, C. pallescens, Drechslera dematioidea, D. hawaiiensis, Epicoccum purpurascens, Eurotium sp., Fusarium moniliforme, F. nivale, F. semitectum, Nigrospora oryzae, N. sacchari, Penicillium digitatum, Pestalotiopsis guepinii, Rhizopus stolonifer, Syncephalastrum racemosum and Trichoderma viride. The present study investigated the development of high sugar production by optimization of an enzymatic hydrolysis process using both conventional and statistical methods, as well as the production of ethanol by the selected wastepaper source. Among four sources of pretreated wastepaper including office paper, newspaper, handbills and cardboard, office paper gave the highest values of cellulose (87.12%) and holocelluloses (89.07%). The effects of the amount of wastepaper, the pretreatment method and the type of enzyme on reducing sugar production from office paper were studied using conventional methods. The highest reducing sugar production (1851.28 µg L−1; 37.03% conversion of glucose) was obtained from the optimal condition containing 40 mg of office paper, pretreated with stream explosion and hydrolysed with the combination of cellulase from Aspergillus niger and Trichoderma viride at the fixed loading rate of 20 FPU g−1 sample. The effects of interaction of wastepaper amount and enzyme concentration as well as incubation time were studied by a statistical method using central composite design. The optimal medium composition consisted of 43.97 µg L−1, 28.14 FPU g−1 sample and 53.73 h of wastepaper, enzyme concentration and incubation time, respectively, and gave the highest amount of sugar production (2184.22 µg L−1) and percentage conversion of glucose (43.68%). The ethanol production from pretreated office paper using Saccharomyces cerevisiae in a simultaneous saccharification and fermentation process was 21.02 g L−1 after 36 h of cultivation, corresponding to an ethanol volumetric production rate of 0.58 g ethanol L−1 h−1.

The objective of this paper is to establish the significance of the mycoremediation of contaminants such as Cd2+ to achieve sustainable and eco-friendly remediation methods. Industries such as electroplating, paint, leather tanning, etc. release an enormous amount of Cd2+ in wastewater, which can drastically affect our flora and fauna. Herein, we report on the in vitro bioadsorption of Cd2+ ions using fungal isolates obtained from different contaminated industrial sites. The detailed studies revealed that two fungal species, i.e., Trichoderma fasciculatum and Trichoderma longibrachiatum, were found to be most effective against the removal of Cd2+ when screened for Cd2+ tolerance on potato dextrose agar (PDA) in different concentrations. Detailed adsorption studies were conducted by exploring various experimental factors such as incubation time, temperature, pH, inoculum size, and Cd2+ salt concentrations. Based on optimum experimental conditions, T. fasciculatum exhibited approximately 67.10% removal, while T. longibrachiatum shows 76.25% removal of Cd2+ ions at pH 5.0, 120 h incubation time, at 30°C. The inoculum sizes for T. fasciculatum and T. longibrachiatum were 2.5% and 2.0%, respectively. Finally, the morphological changes due to Cd2+ accumulation were examined using scanning electron microscopy (SEM). Further, Fourier transform infrared spectroscopy (FTIR) spectroscopy reveals the presence of various functional groups (-CH, –C=O, NH and –OH), which seem to be responsible for the efficient binding of Cd2+ ions over the fungal surfaces.
