The genetic transformation via pollen-tube pathway in jatropha is the first alternative method that was applied in this plant. The objective of the research was to study fruit set and germination of three genotypes of jatropha following direct transformations via pollen-tube pathways. The research was conducted during April 2014 until January 2015 at jatropa’s experimental field, Pasuruan, and at green house of ICABIOGRAD, Bogor. Three genotypes of jatropha i.e., IP3A, IP3P and JcUMM18 were used. In the first experiment, split plot design was used where 3 levels of DNA plasmid concentration as a main plot and 5 levels of stigma-drip time of DNA plasmid as a subplot. In the second experiment randomized block design was used with single factor consisting of 15 combinations of concentration and stigma-drip time of DNA plasmid as treatments and control. The results demonstrated that interaction between concentration and application time of DNA plasmid did not significantly affect fruit and seeds formation of three Jatropha genotypes. Combination of DNA plasmid concentration with time of stigma-drip had significant effect on seed germination rate of IP3A genotype, but not significant on the other variables. The concentration of 0.05-0.5 µg µL-1 and application time of DNA plasmid at 1-10 hours after pollination could be applied on jatropha genetic transformation via pollen-tube pathways.

Keywords: DNA plasmid, Jatropha curcas, pCAMBIA1301, pollen-tube, stigma-drip

Long bean production in Indonesia continues to decline every year, this is due to land conversion and the lack of application of agricultural technology. One of the efforts to increase long bean production is by multiplying quality seeds using organic materials as nutrients needed by plants. This study aims to determine the response of the growth and production of long bean seeds to the interaction of strains, varieties with a concentration of liquid organic fertilizer. The research was conducted at the Vegetable Crops Research Institute, Lembang, West Bandung Regency from July 2020 to January 2021. The research method used was an experimental method with a Randomized Block Design (RBD) with a split-plot design consisting of 2 factors: the first factor (main plot) (A) was the variety and strain with 2 levels, a1 (Tavi canton variety) and a2 (pras-1 strain). The second factor (subplot) (B) is concentration of biopras-1 with 5 levels, b1 (0 cc/l-1 water), b2 (1 cc/l-1 water), b3 (2 cc/l-1 water), b4 (3 cc/l-1 water) and b5 (4 cc/l-1 water). The effect of the treatment was analyzed by means of variance and if the f test at 5% level was significantly different, then to find out the best treatment was continued with the DMRT (Duncan Multiple Range Test) at the 5% level. The results showed that there was an interaction between varieties, lines with various concentrations of biopras-1 liquid organic fertilizer on dry seed weight per plant, dry seed weight per plot, and 1000 seeds weight. Treatment of pras-1 lines with the concentration of liquid organic fertilizer biopras-1 4 cc / l of water was able to give the best results on dry seed weight per plant (35.31 gr), dry seed weight per plot (1.298.20 gr or 2.16 Ton/ha-1) and the weight of 1000 seeds (153.10 gr).

One of the diseases that often found in sorghum plants is anthracnose disease caused by Colletotrichum sp. The purposed of this study was to determine the effect of adding micro fertilizer and the use of several sorghum genotypes on the intensity of anthracnose disease. The study was conducted in Sukanegara, Tanjung Bintang, South Lampung in April 2017 - February 2018 and at the Laboratory of Plant Diseases and Pests, Faculty of Agriculture, University of Lampung. The treatments were arranged using a randomized block design in a split plot design (3 times replications), the main plot was micro nutrients (with micro addition and without micro addition) and 15 subgroups of sorghum genotypes (Numbu, Samurai 1, GH3, UPCA, GH4, P / I WHP, GH6, Super 2, GH13, P / F 51-93-C, Super 1, GH5, Mandau, GH7, and Talaga Bodas). The results showed that the addition of "ZincMicro" micro fertilizers to sorghum plants did not affect the intensity of anthracnose disease, however there were differences in the intensity of anthracnose diseases between sorghum genotypes. Numbu Genotype, GH 3, and GH 13 were relatively more resistant to anthracnose disease than the other genotypes.

The factors that influence rubber production are planted clones, plant care and tapping techniques. With the right tapping technique, both tapping time, tapping method and mature tapping age, the production potential of a superior clone of rubber plants that has been well maintained can be maintained. The technique of rubber tapping is closely related to the expected level of production, even very determining the age of the tree economy. The research objective was to determine the effect of stimulant better and tapping time on latex production. The method used was a randomized block design with two-factor split plot design. The first factor (main plot) is the time of tapping consists of 3 levels, namely; w1: 05 in the morning, w2: 06 in the morning, w3: 07 in the morning. The second factor (sub-plot), namely the stimulant dosage better consists of 4 levels, namely; b0: control, b1; 0.5 g, b2; : 1 g, b3; 1.5 g. The results showed that stimulate better treatment had a significant effect on latex volume, wet weight and latex dry weight, but was not significant for latex dryness levels. Tapping time was not significant for latex yield, latex volume, wet weight, latex dry weight and highest dry rubber content at 7am. Stimulant Better treatment has a significant effect on latex volume, wet weight, and latex dry weight, but if given at high doses it will reduce the dry content of rubber. The interaction of treatment when tapping and stimulant is better not significant for latex production. The stimulant application better does not exceed the dose of 0.5 g / principal. Doses> 0.5 g / staple will have an impact on the dry tapping field.

The research aimed to determine the optimal population ratio at each frequency of weeding for the growth and yield of maize. It was carried out from February to May 2020 in the field. There are 2 treatment factors tested arranged in a splite plot design, which was repeated 3 times using a completely randomized block design (CRBD). The first factor is the frequency of weeding as the main plot (Main Plot) consisting of no weeding, 1 x weeding (21 DAS), 2 x weeding (21 DAS and 42 DAS). The second factor is the ratio of the population as sub-plots, consisting of 100% corn + 0% peanuts, 80% corn + 20% peanuts, 60% corn + 40% peanuts, 40 % corn + 60% peanuts, 20% corn + 80% peanuts. The results showed that the growth and yield of maize in the intercropping system with peanuts gave the same response to the population ratio at all weeding frequencies. The planting density of 40% maize + 60% peanuts produced the highest response on the variables of stem diameter (24.675 mm), plant fresh weight (667.51 grams) and biomass weight (79.924 grams). While the highest response to weeding frequency was 2 times (21 DAS and 42 DAS) only on plant fresh weight (588.08 grams).

The objective of this study was to evaluate the productivity and quality of melon fruits as a function of the dose and time of application of biostimulant in the conditions of the semi-arid region of Paraíba. The experiment was carried out at the Federal University of Campina Grande, campus of Pombal - PB, Brazil, in a randomized block design in a 4 x 5 split plot scheme, with doses of biostimulant (0; 0.5; 1.0; 1, 5 and 2.0 L ha-1) and in the subset of the biostimulant application times (15; 20; 25 and 30 days before harvest - DAC), in four replications. Characteristics related to fruit production and quality were evaluated. There was no interaction between the factors of dose and application time of the biostimulant in any of the evaluated characteristics. Thus, the highest estimated values of number of fruits per plant, fruit mass and total melon production were obtained with the application of doses ranging from 0.9 to 1.5 L ha-1 and at the time of application it varied from 22,5 to 23.6 DAC. The content of soluble solids increased 5.5% when the biostimulant dose of 2.0 L ha-1 was used and 4.4% when the product was applied 15 days before harvest.

Background: Time of pre-emergence herbicide application is an important factor that decides the efficiency of herbicide. If the pre-emergence herbicide is not applied at appropriate time, it may cause ineffective weed control or toxicity to crops. With this background, a field experiment was conducted at TNAU, Madurai to optimize the time of pre-emergence herbicides namely pendimethalin and oxyfluorfen application for effective weed management in irrigated blackgram. Methods: The experiment was laid out in randomized block design with three replication. Treatments were application of pendimethalin at 0.75 kg ha-1 and oxyfluorfen at 0.2 kg ha-1 on 1, 2, 3, 4 days after sowing, weed free check and unweeded check. Result: Results of the study revealed that oxyfluorfen gave maximum weed control efficiency as compared to pendimethalin, but toxicity to the crop was observed when oxyfluorfen was sprayed after 2 days of sowing. Yield and economics were achieved higher with application of 0.75 kg ha-1 of pendimethalin at 2 DAS, which was on par with application of 0.2 kg ha-1 of oxyfluorfen at 1 DAS due to lesser toxicity. Application of 0.75 kg ha-1 of pendimethalin and 0.2 kg ha-1 of oxyfluorfen didn’t leave any herbicide residue after harvest. Hence, application of 0.75 kg ha-1 of pendimethalin at 2 DAS and 0.2 kg ha-1 of oxyfluorfen at 1 DAS can be recommended for effective weed management in irrigated balckgram.
