This study has been carried out in 2017-2018 in order to determine seed dormancy and effective germination depth wild mustard (Sinapis arvensis L.). The in-vitro dormancy breaking experiments (tip breaking, sanding, H2SO4 application, holding in flowing and still water, GA3, KNO3 and GA3+KNO3 combination application) has been applied to wild mustard seeds collected from wheat field in Tokat province and has been applied to wild mustard seeds collected from wheat field in Tokat province and the most effective method was determined as 1000 ppm GA3+KNO3 with 98% impact on seed germination at 15°C within 72 hours. In contrast germination rate has been calculated as 5% in control plants. Furthermore 15°C was assessed as optimum temperature for seed germination was the most effective temperature and during depth studies 100% of wild mustard seeds germinated at 3-5 cm. Because of the difficulies with the work with seeds and plants that have dormancy, these data will contribute future studies.

This study aimed to determine the type of seed dormancy and to identify a suitable method of dormancy-breaking for an efficient seed viability test of Lysimachia coreana Nakai. To confirm the effect of gibberellic acid (GA3) on seed germination at different temperatures, germination tests were conducted at 5, 15, 20, 25, 20/10, and 25/15 °C (12/12 h, light/dark), using 1% agar with 100, 250, and 500 mg·L−1 GA3. Seeds were also stratified at 5 and 25/15 °C for 6 and 9 weeks, respectively, and then germinated at the same temperature. Seeds treated with GA3 demonstrated an increased germination rate (GR) at all temperatures except 5 °C. The highest GR was 82.0% at 25/15 °C and 250 mg·L−1 GA3 (4.8 times higher than the control (14.0%)). Additionally, GR increased after cold stratification, whereas seeds did not germinate after warm stratification at all temperatures. After cold stratification, the highest GR was 56.0% at 25/15 °C, which was lower than the GR observed after GA3 treatment. We hypothesized that L. coreana seeds have a non-deep physiological dormancy and concluded that 250 mg·L−1 GA3 treatment is more effective than cold stratification (9 weeks) for L. coreana seed-dormancy-breaking.

Abstract Wild mustard (Sinapis arvensis L.) is a widespread weed of the southeastern cropping region of Australia. Seed germination ecology of S. arvensis populations selected from different climatic regions may differ due to adaptative traits. Experiments were conducted to evaluate the effects of temperature, light, radiant heat, soil moisture, salt concentration, and burial depth on seed germination and seedling emergence of two [Queensland (Qld) population: tropical region; and Victoria (Vic) population: temperate region] populations of S. arvensis. Both populations germinated over a wide range of day/night (12 h/12 h) temperatures (15/5 to 35/25 C), and had the highest germination at 30/20 C. Under complete darkness, the Qld population (61%) had higher germination than the Vic population (21%); however, under the light/dark regime, both populations had similar germination (78 to 86%). At 100 C pretreatment for 5 min, the Qld population (44%) had higher germination than the Vic population (13%). Germination of both populations was nil when given pretreatment at 150 and 200 C. The Vic population was found tolerant to high osmotic and salt stress compared with the Qld population. At an osmotic potential of −0.4 MPa, germination of Qld and Vic populations was reduced by 85% and 42%, respectively, compared with their respective control. At 40, 80, and 160 mM sodium chloride, germination of the Qld population was lower than the Vic population. Averaged over the populations, seedling emergence was highest (52%) from a burial depth of 1 cm and was nil from 8 cm depth. Differential germination behaviors of both populations to temperature, light, radiant heat, water stress, and salt stress suggests that populations of S. arvensis may have undergone differential adaptation. Knowledge gained from this study will assist in developing suitable control measures for this weed species to reduce the soil seedbank.

This study was carried out to investigate the effects of light spectra, additional UV-A, and different growth regulators on the in vitro germination of Senecio cineraria DC. Seeds were surface-sterilized and inoculated in MS medium to evaluate the following light spectra: white, white plus UV-A, blue, green, red or darkness. The maximum germinability was obtained using MS0 medium under white light (30%) and MS + 0.3 mg L-1 GA3 in the absence of light (30.5%). S. cineraria seeds were indifferent to light. Blue and green lights inhibited germination. Different concentrations of gibberellic acid (GA3) (0.1; 0.4; 0.6; 0.8; 1.0 and 2.0 mg L-1) and indole-3-acetic acid IAA (0.1; 0.3 and 1.0 mg L-1) were evaluated under white light and darkness. No concentration of GA3 enhanced seed germination percentage under white light. However, when the seeds were maintained in darkness, GA3 improved germination responses in all tested concentrations, except at 1.0 mg L-1. Under white light, these concentrations also increased the germination time and reduced germination rate. Germination rate, under light or darkness, was lower using IAA compared with GA3.

Seeds from mature seed pods of Cypripedium calceolus var. parviflorum were germinated on 1/4 MSMO (Sigma) + 100ml/l coconut water + 1% sucrose +/- 8g/l agar (pH 6.0), and with or without prechilling at 5C for 8 weeks. Protocorm with apex (stage 3) was use as an index of germination. Seeds sown on agar medium withou chilling treatment resulted in a 40% germination rate in 120 days but the germination was very uneven. Seeds germinated on agar medium with prechilling developed more synchronously with 92% germination in 60 days (ie. about 120 days after sowing). Suspension culture of seeds without prechilling resulted in 85% germination after 90 days. The synchronization of seed germination in suspension culture was intermediate between that on agar with and without prechilling. Protocorms germinated in suspension culture appeared morphologically identical to those germinated on agar medium. All stage 3 protocorms developed further on the same agar medium in darkness. Both agar and suspension culture in media containing coconut water provided reliable seed germination methods for this orchid species.

The present work was carried out to investigate the effects of gamma radiation on regeneration of Capsicum annuum L. var Kulai via in vitro. Seeds of C. annuum were irradiated with various doses of gamma ray (0, 20, 40, 60, 80, 100, 200, 300, 400, 500, and 600 Gy) emitted from the Caesium-137 source at the rate of 4.31 Gy per minute. Irradiated seeds grown on MS medium without hormone for hypocotyl and cotyledon preparation as explant for in vitro regeneration. Seed germination rate revealed significant variation between treatments, and seeds started to germinate between 6 to 17 days. Irradiated seeds between 0-60 Gy were observed to germinate in less than 10 days. All explants including hypocotyl and cotyledon were cultured on MS medium with different concentrations of BAP in combination with AgNO3 to observe the response of these explants to different hormone concentrations. From the observation, calluses were induced in 90% of hypocotyl and cotyledon explants in all treatments. The characteristics of calluses were varied with greenish friable, greenish compact, yellowish watery, yellowish friable and yellowish compact. In other treatments, calluses were found in purple, bright yellow and yellowish orange. On the other hand, shoot regeneration was observed in treatment between 40-100 Gy. In conclusion, gamma radiation gave impact on seed germination, seedling growth performance, in vitro callus formation and shoot regeneration of Capsicum annuum var. Kulai

The experiment was carried to evaluate the efficacy of five different plant extracts i.e. Adhatida vasica, Tagetes patula, Vinca rosea, Lowsonia alba and Andrographis paniculuta on seed germination, vigor index and seed borne and seed associated mycoflora of wheat varieties viz. Sourav and Prodip in in vitro condition. Seed health test of the collected wheat seed revealed that the seed was associated with six different fungi viz. Bipolaris sorokiniana, Fusarium sp., Aspergillus spp., Trichoderma viride, Penicillium sp. and Rhizopus sp. Wheat seeds were treated by dipping separately into different plant extracts of 1:1, 1:2 and 1:3 dilutions. Among the plant extracts, Lowsonia alba leaf extract was observed most effective followed by A. vasica, T. patula, V. rosea and A. paniculuta in reducing seed borne and seed associated mycoflora and also increased the germination rate and vigor index. Hedysarum grandiflorum samples were collected from 7 natural populations of the Volgograd, Samara and Lugansk People's Republic and the features of seed reproduction were studied under ex situ and in vitro conditions. The optimal temperature for seed germination and the dependence of germination and seed vigor on the storage were revealed. We observed the maximum germination rate in the scarified seeds of H. grandiflorum of 2018 collection and it was equal to 70%.It was found that the seed germina-tion and seed vigor are low and quickly decrease with years of storage, while their scarification allows to increase these indicators. It is noted that the relatively low germination of the studied seeds is leveled by the rather high productivity of species in general. The optimal mode of sterilization of H. grandiflorum seeds was experimentally determined - 10% Lysofornin® 3000 solution for 5 minutes. The percentage of sterile seedlings was 60%.

Dendrobium spectabile (Blume) Miq. is one of Papuan orchids that is of economic value and threatened. At present, the optimal protocol for asymbiotic seed germination and planlet development of D. spectabile is not yet available. This research aimed to develop an optimal and comprehensive protocol for in vitro culture of D. spectabile to support the conservation and reintroduction of this species. The experiment was conducted using a completely random design, including seed sowing, protocorm subculture (subculture 1), plantlet subculture (subculture 2), and plantlet acclimatization. The highest germination rate at 3 MAS (months after sowing) was found in modified Knudson's C (92.59%). The first subculture at 4 MAP (months after planting) showed that the highest leaf and root growths were found in modified Vacin and Went (4.12 and 2.13, respectively). The second subculture at 6 MAP showed that the highest leaf growth was found in full strength of Murashige and Skoog supplemented with 100 g/l banana homogenate (5.49), while the highest number of roots and average root extension were found in half strength of Vacin and Went supplemented with 100 g/l banana homogenate (7.05 and 0.47 cm, respectively). The plantlets were best acclimatized in media consisting of tree fern fibre and sphagnum moss (2:1).

Abstract Background: In Ethiopia, species used for timber production are limited to few species. So, it is becoming necessary to find innovative ways of maintaining and improving the genetic quality of species such as Milicia excelsa by domesticating them into a wider scale of ecological and social environment. Hence, the main objective of this study was to investigate the seed germination biology and behavior of Milicia excelsa under in-vitro laboratory conditions so as to promote the domestication and establishment of seed production areas (SPA) in-situ. Methods: Seeds collected from Benchi district of Benchi-Maji zone of southwestern Ethiopia were subjected to germination biology in-vitro to determine the germination responses of M. excelsa seeds under laboratory conditions. So, a total of 1200 seeds were randomly selected, treated and prepared using four treatments procedures with six replications for paper method and 3 replications for sand method, and tested using paper methods in CRD design. So, 600 seeds were tested using paper and sand methods each where 25 seeds were tested for each replication in paper and 50 seeds for each replication in sand method. The germination process was lasted between 19 March 2020 and 15 May 2020. Result: The mean germination percentage of seeds of M. excelsa was noticed to be lower than 20%. This is below the expected (45%) germination rate of mature and healthy seed lot. Overall, seeds that were not treated using any pre-sowing treatments has shown relatively better germination percentage (19%) followed by seeds rubbed by hand (15%). Analysis of one-way t-test shows that the effects untreated (control –T1) and rubbing by hand treatments (T4) is significant at p < 0.05 compared to the effects of the other pre-sowing treatments (T2- seeds only washed with normal tap water, and T3- seeds washed and soaked in hot water for 15 minutes) on the germination capability of seeds of M. excelsa . However, no significance difference was observed between control (T1) and hand scarification (T2). Conclusions: The average germination of M. excelsa seeds combined for all treatments is lower than 20% which is lower germination when comparing with other similar research reported in other countries. Therefore, it required to include and test other pre-sowing treatments including tissue culture micro-propagation of seeds to maximize the germination of M. excelsa seeds helping to get enough seedlings required for the domestication project.

The effects of ethephon (2-chloroethylphosphoric acid) and temperature on seed germination were studied in two cultivars of Medicago truncatula (medic) and one cultivar of Trifolium subterraneum (clover) grown under different controlled conditions. Ethephon promoted germination of immature and fresh mature seeds and increased the germination rate of old mature seeds. Ethephon thiourea and carbon dioxide had different effects on the germination of seeds of the medic cultivars. At 30°C, untreated seeds germinated poorly while the presence of 10 ppm ethephon increased germination significantly.
