In this study, fish feed supplemented by probiotic, prebiotic and synbiotic were applied to enhance the immune response and survival rate of the fish against Streptococcus agalactiae infection. Treatments used in this study, were as follows: feed containing 1% (v/v) of probiotic (C), feed containing 2% (v/v) of prebiotic (D) and feed containing mixed of 1% (v/v) of probiotic + 2% (v/v) of prebiotic (E, defined as synbiotic). Control fish (both positive, A; and negative, B; treatments) were fed by fish feed without containing neither probiotic nor prebiotic. In this study, test fish used monosex (all male) red tilapia with the average of body weight of 13,43±2,97 gram. The fish were fed by supplemented feed within first 30 days pre injection. After that, the treated fish were challenged by 109 CFU/ml of S. agalactiae. Nile tilapia fed by synbiotic (treatment E) had higher immune response (haemoglobin, neutrofil, and phagocyte indices) than control, but were not significantly different than those of treatment C, and D. After challenged test by S. agalactiae, treatment C, D and E resulted significantly higher resistance than that of control. The survival rate of fish fed by supplemented feed containing probiotic (C), prebiotic (D) and synbiotic (E) were 76%, 76% and 80%, respectively; higher than than positive control (50%). The results showed that addition of probiotic, prebiotic and synbiotic in fish feed could increase immune response and survival rate against S. agalactiae.Keywords : tilapia, probiotic, prebiotic, synbiotic, Streptococcus agalactiae

Streptococcus agalactiae (Sta) of Lancefield group B is the primary etiological agent of bacterial meningitis in Nile tilapia and newborn humans. Thus, the study of this disease is of fundamental importance for aquaculture and human medicine. Additionally, elucidation of the mechanisms involved in the host–pathogenic response is important for the success of new therapies. In the present study, we elucidated important aspects of the innate immune response in the brain tissue of Nile tilapia (Oreochromis niloticus) infected by Sta. The neuroinflammatory process in the meninges started with the migration of MHC class II and CD68 + cells, production of TNF-alpha, and the effective immune response to Sta was mediated by the increased iNOs+. In conclusion, the present study brings a partial understanding of the pathophysiological and neuroinflammatory mechanisms in meningitis in Sta infected tilapia, enabling important advances in the therapy of this disease as well as the possibility of using this biological model to understand human meningitis.

This study aimed to evaluate the effect of probiotic on growth and survival of tilapia (Oreochromis niloticus) against Streptococcus agalactiae infection. The test materials used were commercial probiotic bacteria and Streptococcus agalactiae. The probiotic bacteria were administered by mixing into the feed with a dose of 0 mL (without probiotics), 10 mL / kg of feed, 15 mL / kg of feed and 20 mL / kg of feed.  Feed was given 3 times a day at 08.00, 12.00 and 16.00 for 21 days. After that, the challenge test was done with Streptococcus  agalactiae bacteria by cohabitation. Bacteria with a density of 107 cells / mL were mixed into the water and allowed for 2 hours. The result showed that B treatment (10 mL / kg of feed) gave the best result with 93,33 percent of  survival rate. Statistical analysis showed that the survival of fish in treatments B, C and D was significantly different from treatment A (P <0.1). However, there was no significant difference between treatments B, C, and D. It was also found that the addition of probiotic in feed did not affect the growth of Nile tilapia (Orechromis niloticus). Keywords : Probiotic bacteria,  nile tilapia,  Streptococcus  agalactiae, growth,  infection

Nile Tilapia or Tilapia Fish (Oreochromis niloticus) is one of commodity with a high number of consumers. But, Streptococcus agalactiae attack caused a decline in production. One way to reduce these diseases by using natural materials of songga wood (Strychnos lucida R.Br) where the songga wood contains many compounds like alkaloid, phenols, flavonoids, triterpenoids, which can inhibit bacterial growth by damaged cell membrane. The results showed that the inhibitory songga wood extract against Streptococcus agalactiae was highly significant, and shows with the highest survival rate of 90% and proved by theincreasingantibodytitersafteraddingsonggawood extract.

The objective of this research was to examine the effect of garlic (Allium sativum) at different doses on nonspecific immune response and growth of nile tilapia (Oreochromis niloticus). Experimental fish was nile tilapia measuring 8-15 cm with an average weight of 10.4 g per individual. Before used in the experiment, fish were reared in fiber tank for two weeks for acclimatization. After acclimatization, the fish were moved into glass aquarium (50 cm x 30 cm x 30 cm) at the density of 15 fish/aquarium. Each aquarium was equipped with aerator. Five doses of garlic as treatments used in this research consisted of A= 0 g/kg diet, B= 10 g/kg, C= 20 g/kg, D= 30 g/kg and E= 40 g/kg. Fish was fed experimental diet at a dose of 5% of body weight per day, twice a day at 08.00 and 17.00. Data collected at the end of the experiment included immune parameters (TLC and phagocytosis activity) and growth of fish. ANOVA was used to evaluate the effect of treatments on immune response and growth of fish while Duncan Test was used to evaluate the different effect between treatments. Research result showed that after four weeks of feeding, TLC of fish fed diet supplemented with garlic significantly different (p<0,01) as compared to that of control fish. The highest number of TLC was observed at treatment C (15.413x107 cell.ml-1) followed by treatment B (15.39x107 cell.ml-1), D (13,920 x 107 cell.ml-1), E (13,582 x 107 cell.ml-1) and A (12,195 x 107 cell.ml-1). TLC of fish in treatment C was different significantly compared to control as well as to other treatments. Phagocytosis activity of fish in treatment C also increased significantly (p<0,01) compared to control. The highest PA was achieved in treatment C and then treatment B, D, E and A. Growth of fish in treatment C was different significantly compared to control fish as well as other treatments. As conclusion, oral administration of garlic at 20 g/kg diet could enhance nonspecific immune response and growth of nile tilapia. The purpose of this research was to determine the effect of yeast (Saccharomyces cerevisiae) with different composition of 5%, 10%, 15%, 20%, and without yeast in feed formulation on growth of tilapia (Oreochromis niloticus). The experimental fish used in this study had an average lengths of 2-3 cm  and weight of 1.0 g.  This research used cage measuring 100 x 100 x 100 cm3 that was placed in outdoor concrete pond of 3.5 x 5 m2.  Each cage filled with 10 fishes with three replication.  The dose given to the fish was 10% of the weight of biomass.  The fish was fed for three weeks, 3 times a day at 08.00 am, 12.00 pm, and 16.00 pm.. Research results showed that the greatest growth was achieved in fish treated 20% yeast and the lowest was observed in control fish (without yeast). Based on statistical analysis, all treatment displayed similar effect on all growth parameters.Keywords: Fish feed, Saccharomyces cerevisiae, yeast, Nile tilapia, fish growth

This study aims to examine the effect of sponge crude extract addition to the fish feed on growth and non-specific immune response of nile tilapia (O. niloticus) and determine the most effective dose of the extract in raising the growth and the non-specific immune response of nile tilapia (O. niloticus). The sponge sample was macerated and then evaporated using a rotary vacuum evaporator until the sample became a paste and mixed with feed. The fish were fed for 14 days respectively as much as 5% of body weight per day with feeding frequency of two times a day. The results showed that addition of sponge crude extract to the fish feed gave good effect on the growth and the non-specific immune response of nile tilapia (O. niloticus) with best dose of  40 g / kg feed.Keywords: Sponge extract, fish immune response, fish growth, immune response, immunostimulant. ABSTRAKPenelitian ini bertujuan untuk menguji pengaruh penambahan ekstrak kasar spons pada pakan ikan terhadap pertumbuhan dan respon imun  non spesifik ikan nila (Oreochromis niloticus) serta menentukan dosis ekstrak kasar spons yang paling efektif dalam meningkatkan pertumbuhan dan respon imun non spesifik ikan nila (Oreochromis niloticus). Sampel spons dimaserasi kemudian dievaporasi menggunakan rotary vacuum evaporator sehingga diperoleh ekstrak kasar berbentuk pasta yang kemudian dicampurkan pada pakan. Ikan uji diberi pakan perlakuan 5% dari bobot biomassa selama 14 hari dengan frekuensi pemberian pakan dua kali sehari. Hasil penelitian menunjukan bahwa penambahan ekstrak kasar spons pada pakan ikan memberikan pengaruh yang baik terhadap pertumbuhan dan respon imun non spesifik ikan nila (Oreochromis niloticus), yang mana dosis yang terbaik adalah 40 gr / Kg pakan.Kata Kunci : Ekstrak spons, respon imun ikan, pertumbuhan ikan, respon imun, imunostimulan.
