Goldfish (Cyprinus carpio Linn) punten breeds is a type of freshwater fish that has greatpotential in terms of quality and quantity. Seed production from generation to generation hasdecreased quality. An alternative effort was made to maintain the seed quality of geneticengineering with triploidization process using heat shock treatment. The heat shock can preventpolar body II from splitting on eggs during cleavage of meiosis II level resulting in 3Nchromosome or sterile fish. The purpose of this research is to analyze chromosome number intriploidization process of punten breeds fish and to analyze the effect of temperature shockduration on hatching rate of goldfish (Cyprinus carpio Linn) Punten Breeds. This research wasdescriptive research with 2 treatments and 3 replications in each treatment that was heat shocktreatment 40 ºC at minute 2 after fertilization with difference of heat shock duration 1,5 minutes(treatment A) and shock time temperature 2 minutes (treatment B). The parameters observedwere triploid success, hatching rate, survival rate, and growth of of goldfish (Cyprinus carpioLinn) Punten Breeds, analyzed by T-test with 95% confidence interval. The result of heat shockduration of 1.5 minutes showed the percentage of triploid fish 53,57% and shock timetemperature 2 minute 43,64%. The result of T-test of both treatments had no significant effect onall observation parameters but had varied relationship that was enough and inversely related toall observation parameters

<p><strong><em>Abstract</em></strong></p><p>Carp is a freshwater fish, body shape is long and somewhat rounded with a small head, the high back and across large scaly body.The development of aquaculture carp (Cyprinus carpio) is progressing very rapidly with cultivation system are manifold. Pisciculture technology will not form without yielding seeds followed by genetic improvement effort. To improve the quality of the parent, the parent needs to be purified race carp (Cyprinus carpio) that exist so that the resulting pure strain. The success of seed carp, especially at this stage of the enlargement is determined by the quality of the seed. Breeding programs developed at this time is the method gynogenesis. Ginogenesis method is the process of female gamete without interference from male gamete gene.</p><p>The experimental design used in this study is completely randomized design (CRD) is equipped with 3 treatments and 3 replications. Analysis using analysis of variance, with the distribution table F or F test that compares the value of F arithmetic with F table.</p><p>Based on the analysis of research data on the influence of the length of time a different heat shock against the hatchability of eggs carp (Cyprinus carpio) method gynogenesis meiosis, based on the results of research, treatment A showed optimal results with an average 17.33, treatment B showed 12. 33 and C treatment showed 7.00. Based on calculations obtained ANOVA calculated F value (47.29)&gt; F 1% (18.00) but larger than F 5% (6.49) then H1 is rejected at the level of a = 1% and H0 highly significant conclusions (hight significant).</p><p> </p><p>Key words : Heat Shock, <em>Gynogenesis Meiosis, Hatching rate</em>.</p>

The easiest, cheapest, simpliest and efficient polyploidization method was heat shock treatment. Each fish species has different tolerance either initial time, temperature or duration of heat shock. The aim of this study were to show and determine the heat shock effects on differentiation of hatching rate, abnormality, survival rate, and successful polyploidization of common carp. The method that used in this study was experiment. Treatment that used was heat shock 40° C during 1.5 minutes of common carp eggs to 3 and 29 minutes after fertilization. Ten replicates were carried out for each treatment. Parameters test were hatching rate, abnormality, survival rate and polyploidy induction by counting of nucleolus number. Data analysis that used was descriptive. The result of this study indicated that heat shock treatment influenced on hatching rate, abnormality, survival rate and polyploidy induction of the common carp polyploidization. Hatching rate of common carp, triploidy and tetraploidy were 22.63 ± 8.36 percent and 11.10 ± 8.60 percent, abnormality 13.81 ± 4.67 percent and 24.86 ± 8.37 percent, survival rate 52.64 ± 8.46 percent and 55.04 ± 8.15 percent and polyploidy induction 70 ± 7.07 percent and 60 ± 7.07 percent, respectively.

Summary Triploidization plays an important role in aquaculture and surrogate technologies. In this study, we induced triploidy in the matrinxã fish (Brycon amazonicus) using a heat-shock technique. Embryos at 2 min post fertilization (mpf) were heat shocked at 38°C, 40°C, or 42°C for 2 min. Untreated, intact embryos were used as a control. Survival rates during early development were monitored and ploidy status was confirmed using flow cytometry and nuclear diameter analysis of erythrocytes. The hatching rate reduced with heat-shock treatment, and heat-shock treatments at 42°C resulted in no hatching events. Optimal results were obtained at 40°C with 95% of larvae exhibiting triploidy. Therefore, we report that heat-shock treatments of embryos (2 mpf) at 40°C for 2 min is an effective way to induce triploid individuals in B. amazonicus.

<p>Seurukan fish (<em>Osteochilus vittatus</em>) has many advantages, besides the fish also has disadvantages which are the slow growth, so the temperature shock of triploidization technique was expected to solve the problem.   The objective of the present study was to obtain an effective temperature to increase of triploidization success of <em>seurukan</em> fish (<em>Osteochilus vittatus</em>). The experimental method and completely randomized design model were used in this study. Five levels of temperature shocks at three replicates were tasted: 4℃ (cold), 6℃ (cold), 28℃ (normal), 35℃ (heat) and 37℃ (heat). The sperms and eggs were fertilized in the plastic jar then a total 100 of fertilized eggs (zygotes) were taken randomly 3 minutes after fertilization and soaked in respective temperature for 90 seconds, and then incubated in incubation jars at the water temperature of 28-29℃. The results showed that the temperature shock gave the significant effect on the hatching and the success of triploidization success (P˂0.05), but did not give the significant effect the fertility and survival rates (P&gt;0.05). The triploid fish can be achieved using cold and heat shock, but the higher triploid fish was recorded at 37℃ was the best temperature recommended for triploidization of <em>Seurukan</em> fish.</p>

<p><em>Species, temperature shock duration, fertilization temperature from different climates can cause different hatching results, so it is necessary to do research on the treatment of variations in heat shock at the same temperature, to obtain information about the hatching rate (HR) of African catfish (Clarias gariepinus). The method used is completely randomized design (3x3). The results of the egg hatching rate (HR), with variations in the heat shock at the same temperature (35⁰C), </em><em>showed the highest rate of hatching dumbo catfish eggs at heat shock one minute 233.00%, then heat shock two minutes 212.00%, and heat shock three minutes 200.00% respectively.</em><em></em></p>

Knowledge on the quality of male and female gametes which may affect fertilisation success and larval survival is necessary to enhance fish production to meet global demand. Gamete quality is affected by factors like sex ratio, stocking density, age, size, nutrition and feeding regime. In the present study, the spermatological and fertilisation parameters as influenced by the age of brooders of Cyprinus carpio Linnaeus 1758 developed under captive condition from a single population were studied with special reference to dilution ratios. Milt was collected from 6, 12 and 18 months old brooders of C. carpio and diluted at three ratios viz., 1:40, 1:80 and 1:120 using freshwater fish saline. Sperm density, spermatocrit value, seminal plasma composition, motility duration, fecundity, fertilisation and hatching rates were studied for brooders of all age groups. The results showed that milt from 12 months old brooder produced good quality milt in terms of motility duration(69.66±4.72 sec), fertilisation rate (91.3±3.05%) and hatching rate (76.5±2.64%) at 1:40 dilution ratio.

In the present study, the ultrastructural changes of BPK cells, a fibroblast-like cell line, derived from the kidney of juvenile black porgy Acanthopagrus schlegeli, under heat shock treatment are described.The BPK cells were maintained in L-15 medium supplemented with 10% fetal calf serum and 0.15 M NaCl at 28|C2. The heating was carried out in precalibrated water baths. Monolayers of cells, grown on coverslips in parafilm-sealed petri dishes were submerged under water for 30 min at 40|C treatments. Cells were fixed in 2.5% glutaraldehyde in 0.1 M cacodylate buffer supplemented with 6.6% sucrose, postfixed in 1% OsO4 and flat embedded in Spurr’s resin. Silver section were cut parallel to the substratum, stained with uranyl acetate and Reynold’s lead citrate, and examined in a Hitachi H-600 electron microscope at 75 KV.
