Tinospora cordifolia (wild) Hook.f. (Thomas) is an important medicinal plant distributed throughout in Sri Lanka and commonly known as Rasakinda in Sinhala and Giloy in English. It is widely used in Sri Lankan traditional medical system and Ayurveda for the treatment of diabetes mellitus, fever, arthritis, skin diseases and for Rasayana (rejuvenating) therapies due to its anti-inflammatory, hypoglyceamic, immunomodulatory, antioxident, anti-allergy, antipyretic, antiarthritic and various other medicinal properties. Imported T. cordifolia stems and Sri Lankan grown T. cordifolia stems can be found in Sri Lankan herbal market. Moreover, different varieties of Tinospora species known as Tikthakinda, Bukinda and Gatakinda are available in the market under the name of Rasakinda which leads to adulteration. Hence, in this research an attempt was made to develop standards for genuine T. cordifolia grown in Sri Lanka. Stems of T. cordifolia evaluated for macroscopical, microscopical, physico-chemical, phytochemical constituents, TLC and HPTLC fingerprint patterns. Microscopical examinations of T. cordifolia stem exhibited the wheel shaped appearance at the transverse cut surface, which is a main characteristic feature of the family Menespermaceae. Stem also showed the mucilage cells and abundant starch granules. Percentages of total ash, water soluble ash and acid insoluble ash, were 9.1%+0.1, 2.31%+0.1, <0.1% respectively. Among the percentages of extractable matter of T. cordifolia, highest amount was shown in hot water extract (16.2 %+0.3). Heavy metals (Hg, As, Cd, Pb) were within the limits given in WHO guidelines. Phytochemical screening revealed the presence of phenols, saponins, tannins, steroids, flavonoids, terpenoids and cardiac glycosides. TLC fingerprint of T. cordifolia was developed using butanol: ethyl acetate: acetic acid: water in a ratio of 5: 8: 6:2 v/v and compared with one of its marker compound, Berberine. The HPTLC fingerprint patt ern of T. cordifolia showed a spot bearing the same Rf value corresponds to Berberine, at wavelength 254 nm. In conclusion, the results obtained from this study can be used as a standard reference for Sri Lankan grown T. cordifolia stems.

Elaeocarpus ganitrus Roxb, (E. ganitrus) known as Rudraksha belongs to family- Eleocarpaceae. It has a reflecting position in Hinduism and Ayurveda whereas traditionally it has mentioned to cure various health problems like fever, skin diseases, mental problems, wound healing etc. The present study was designed to study the microscopic and macroscopic analysis, physiochemical parameters, quantitative microscopy, phytochemical screening of E. ganitrus leaves as per WHO guidelines and evaluate the antiulcer potential of aqueous extract of E. ganitrus (AEEG) and ethanolic extract of E. ganitrus (EEEG) at the doses of 200 mg/kg and 400 mg/kg using pylorus ligation induced ulcers model, biochemical parameters. Hepatic, cardiac, hematological parameters have also done to find out the effect of different extracts on other major organs. Microscopic analysis proved the presence of covering trichomes, upper epidermis, lower epidermis, stomata, phloem, xylem etc. Ash value, water soluble ash, acid soluble ash, water soluble extract, alcohol soluble extract, loss on drying, swelling index, foaming index found to be 4.3 ± 0.52, 0.2 ± 0.33, 2.0 ± 0.2, 13.7 ± 0.25, 12.5 ± 0.55, 9.8 ± 0.23, 3.6 ± 0.04, more than 100. Different quantitative parameters were found out. Phytochemical analysis of different extracts showed the presence of various primary and secondary metabolite like alkaloids, glycosides, tannin, phenolic compounds etc. Pharmacological potential showed that extracts treated, and sucralfate treated groups showed significantly decreases in ulcer index in all above-mentioned models, biochemical studies clearly showed significant decreases in volume, pH, free acidity, total acidity of gastric content and increases in gastric mucus parameters like protein, total hexoses, hexosamine, fucose, sialic acid and DNA level. The level of antioxidant enzymes like LPO (Lipid peroxidation), SOD (Superoxide dimutase) were decreased and CAT (Catalase) level was increased. Level of PC (Plasma corticosterone) was decreased. Hematological, hepatic, cardiac parameters found to be normal during extracts treatment. Histopathological analysis clearly supports the biochemical studies at various doses and it was found to be effective in dose dependent manner. The obtained scientific data may be helpful to prepare the monograph of the plant and E. ganitrus has antiulcer potential in a dose dependent. Detailed study needed for better exposure of plant.

Background: Desmodium velutinum (Fabaceae) is one of the wild edible plants used traditionally to treat diarrhoea, haematuria, infertility and also externally used to treat scorpion bites. Though various parts of the plant have been used for therapeutic purpose, panchanga of the plant has not been studied for its Pharmacognostical and phytochemical characteristics in detail, following scientific parameters. Hence in the present study Desmodium velutinum whole plant has been studied for its pharmacognostical and phytochemical characters and compared with commonly used species Desmodium gangeticum. Methods: Whole plant of Desmodim velutinum was collected from Paikmal, Odisha and Desmodium gangeticum was collected from Junagarh, Gujarat in the month of November 2019 following good collection practices and shade dried. Pharmacognostical study and analytical study were performed following standard pharmacopoeial procedures. Results: Whole plant (Panchanga) of former is sweetish and latter is bitter. Powder microscopy of Desmodium gangeticum showed presence of rhomboidal crystals and lignified annular and spiral vessels. Desmodium velutinum showed presence of prismatic crystals and lignified pitted vessels. Physico-chemical analysis showed higher loss on drying in Desmodium gangeticum, and higher water soluble extractive values of Desmodium velutinum. Phytochemical estimation showed the presence of Proteins, tannins, and carbohydrates in both samples. HPTLC study showed similar Rf value 0.82 and 0.94 at 254 and 0.03 and 0.41 at 366nm. Conclusion: Basing upon the analytical and microscopic similarities Desmodium velutinum may be considered as substitute of Desmodium gangeticum

This study deals with the Pharmacognostical physicochemical and phytochemical evaluation of the whole plant of Piper longum L. Pharmacognostical studies include macroscopic, microscopic and physicochemical evaluation. Physicochemical constants showed the values of total ash (10.8%), acid insoluble ash (1.3%), water soluble ash (3.2%), sulphated ash (12%), nitrated ash (1.85%), hot extraction (11.2%), water soluble extractive (4.8%), ethanol soluble extractive (12.4%), ether soluble extractive (5.6%), loss on drying (29%), swelling index (0.33) and mucilage content (0.8%) w/w. The preliminary phytochemical screening of the alcoholic extract showed presents of alkaloids, phenolic compounds, tannins, mucilage, starch, steroid and flavonoids. This study helps to development of different standard parameters of the plant which help the plant identification and authentication. Antibiotic resistance has become a global concern and hence, the search for other source of antimicrobials initiated to find a way to control infections in future. The main objective of this paper is to screen Giloy (Tinospora cordifolia) for its antibacterial activity. The stem of Tinospora cordifolia is used to prepare extract for determining it’s in vitro antibacterial activity as per the agar well diffusion method. In the agar well diffusion method 100μl of 24 hr broth culture of bacteria was aseptically and evenly swabbed on Mueller Hinton agar plates. Wells of about 8 mm diameter were aseptically cut using sterile cork-borer. 100 μl of plant extracts of different concentrations were then placed into the separate wells. The plates were incubated at 37 oC for 24hr. Antimicrobial activity of the giloy was determined by measuring the diameter of zone of inhibition. The methanolic extract of Tinospora cordifolia showed 13, 11, 9 and 5 mm zone of inhibition in S. aureus cultures by using 100, 75, 50 and 25 mg/ml concentration, respectively while hot water extract of Tinospora cordifolia showed 14, 12, 10 and 8 mm zone of inhibition for S. aureus by using 100, 75, 50 and 25 mg/ml concentration, respectively and the cold extract of Tinospora cordifolia showed 10, 8, 5 and 0 mm zone of inhibition for S. aureus by using 100, 75, 50 and 25mg/ml concentration, respectively. The methanolic extract of Tinospora cordifolia indicated 12, 10, 6 and 4 mm zone of inhibition in cultures of E.coli by using 100, 75, 50 and 25 mg/ml concentration, respectively and the hot water extract of Tinospora cordifolia showed 16, 14, 12 and 10 mm zone of inhibition in cultures of E.coli by using 100, 75, 50 and 25mg/ml concentration, respectively. The cold water extract of Tinospora cordifolia showed 13, 10, 8, and 5 mm zone of inhibition in cultures of E.coli by using 100, 75, 50 and 25 mg/ml concentration, respectively. It has been observed that Tinospora cordifolia showed very promising results as indicated by the zone of inhibition of bacterial culture through agar well diffusion method that varies from few mm to few cm. This study indicates the in-vitro antibacterial effect of Giloy which needs further validation through in-vivo studies.

Apple pomace is a by-product of apple processing industries with low value and thus frequent disposal, although with valuable compounds. Acidified hot water extraction has been suggested as a clean, feasible, and easy approach for the recovery of polyphenols. This type of extraction allowed us to obtain 296 g of extract per kg of dry apple pomace, including 3.3 g of polyphenols and 281 g of carbohydrates. Ultrafiltration and solid-phase extraction using C18 cartridges of the hot water extract suggested that, in addition to the apple native polyphenols detected by ultra-high-pressure liquid chromatography coupled to a diode-array detector and mass spectrometry UHPLC-DAD-ESI-MSn, polyphenols could also be present as complexes with carbohydrates. For the water-soluble polyphenols, antioxidant and anti-inflammatory effects were observed by inhibiting chemically generated hydroxyl radicals (OH•) and nitrogen monoxide radicals (NO•) produced in lipopolysaccharide-stimulated macrophages. The water-soluble polyphenols, when incorporated into yogurt formulations, were not affected by fermentation and improved the antioxidant properties of the final product. This in vitro research paves the way for agro-food industries to achieve more diversified and sustainable solutions towards their main by-products.

This study investigated the anti-obesity effect of a polysaccharide-rich red algae Gelidium amansii hot-water extract (GHE) in high-fat (HF) diet-induced obese hamsters. GHE contained 68.54% water-soluble indigestible carbohydrate polymers. Hamsters were fed with a HF diet for 5 weeks to induce obesity, and then randomly divided into: HF group, HF with 3% guar gum diet group, HF with 3% GHE diet group, and HF with orlistat (200 mg/kg diet) group for 9 weeks. The increased weights of body, liver, and adipose in the HF group were significantly reversed by GHE supplementation. Lower plasma leptin, tumor necrosis factor-α, and interleukin-6 levels were observed in the GHE+HF group compared to the HF group. GHE also increased the lipolysis rate and decreased the lipoprotein lipase activity in adipose tissues. GHE induced an increase in the phosphorylation of AMP-activated protein kinase (AMPK) and the protein expressions of peroxisome proliferator-activated receptor alpha (PPARα) and uncoupling protein (UCP)-2 in the livers. The decreased triglyceride and total cholesterol in the plasma and liver were also observed in obese hamsters fed a diet with GHE. These results suggest that GHE exerts a down-regulation effect on hepatic lipid metabolism through AMPK phosphorylation and up-regulation of PPARα and UCP-2 in HF-induced obese hamsters.

The study investigated and compared the antimicrobial potency of aqueous (cold and hot) and ethanolic extracts of six plant parts used as herbal mixture against clinical isolate of Salmonella typhi in Okitipupa town. Extracts from six plant parts used as herbal mixture were tested for antibacterial activity against Salmonella typhi isolated from patients suffering from typhoid fever. The antibacterial activities of each of the extracts (ethanolic, cold and hot waters) were determined using agar diffusion method and the activities produced zone of inhibition ranging from 20mm-34mm. Ethanolic extract was found to be more potent being capable of exerting significant inhibitory activity on the test organism investigated with an inhibition zone of 34mm followed by the cold-water extract (28mm) while hot water extract gave the least (20mm). The phytochemical analysis showed the presence of saponins, tannins, flavonoids, phenols, glucosinolates and triterpenes in the extracts but total absence of proteins. The study confirms the medicinal value of herbal mixtures and justifies the claim of its local use in health remedies for typhoid fever caused by Salmonella typhi. There is the need therefore to develop effective combination of antimicrobial agents in purified form from higher plants and their parts for clinical trials. A galactomannan and a branched (1 → 3)-β-D-glucan were isolated from the water hyacinth plant. The galactomannan, purified from the cold water extract, is composed of D-galactose and D-mannose in a ratio of 1.0:2.8. It has a (1 → 4)-linked D-mannose backbone, one out of three D-mannose residues being substituted with a single α-D-galactosyl unit. The branched (1 → 3)-β-D-glucan isolated from the hot water extract has a main chain composed of β-(1 → 3)-linked D-glucopyranosyl residues, and two single β(1 → 6)-D-glucopyranosyl groups attached as side chains to, on average, every 5 sugar units of the main chain. In addition, the branching of the β-glucan occurs regularly at O-6 of the β-(1 → 3)-linked backbone.

Summary This paper presents results of a study on the chemical nature of kiln brown stain (KBS) that develops in kiln dried radiata pine (Pinus radiata D. Don) sapwood. KBS is a chocolate brown discolouration which develops approximately 0.5 mm under the timber surface. Stain free radiata pine was achieved during drying trials on “green” radiata pine sapwood which had been treated to extensive cold and hot water extraction. Differences in chemical composition between the sap and hot water extract were observed. Chemical analyses (of the water-soluble extracts) by a combination of chromatographic and spectroscopic techniques, demonstrated that carbohydrates, cyclitols, amino acids, protein, and phenolics were the main compound classes present. The presence of reducing sugars (glucose and fructose) and amino acids (glutamic acid) in the sap and hot water extracts support the theory that Amadori-Maillard type reactions significantly contribute to the formation of colour in KBS. Furthermore, lignin was also detected and is suspected to contribute to KBS formation. The relative contributions of colour formation to KBS intensity from either phenolics or Maillard-Amadori mechanisms is unknown.

Background: Citraka or Sveta Citraka (Plumbago zeylanica L.) is an important medicinal plant mentioned in Ayurvedic classics, belongs to the family Plumbaginaceae. The root of the plant exhibits medicinal properties. It has a broad range of pharmacological activities, including anti-inflammatory, antibacterial, antifungal, anticancer, antidiabetic, antioxidant, hepatoprotective, wound healing and cytotoxic. Objective: The aim of the study was to evaluate phyto-pharmacognostical standardization with HPTLC fingerprinting of root of P. zeylanica, one of the most accepted and beneficial medicinal plant in Ayurveda. Materials and Methods: P. zeylanica was collected from Konni, Kerala, India during the month of May 2018 and its root was studied for macroscopic, microscopic, physico-chemical, preliminary phytochemical screening and HPTLC fingerprinting following the standard protocol recommended by The Ayurvedic Pharmacopoeia of India and Quality Standards of Indian Medicinal Plants. Results: The root is long, stout, cylindrical and reddish to dark brown in colour. The transverse section of root showed 5-7 rows of small cubical or rectangular celled cork with light yellow walls, polygonal or slightly tangentially elongated cortex cells containing starch grains and coloured contents, phloem containing small groups of fibres, radially arranged xylem vessels and narrow, 1-4 seriate medullary rays. Physico-chemical analysis showed water soluble extractive value as 26.11±0.01 w/w and alcohol soluble extractive value as 15.85±0.00 w/w. Preliminary phytochemical analysis of root decoction showed the presence of alkaloids, carbohydrates, tannins, saponins, resin and quinones. Conclusion: The information obtained from this research work may be useful to establish the botanical as well as analytical standards for the root of P. zeylanica.
