Objective: to determine the antioxidant effects of Nucleinat sodium in patients with uncomplicated recurrent pyelonephritis. Material and methods. We have analyzed the indicators of oxidant/ antioxidant (O/ A) balance in the blood of 55 patients with uncomplicated recurrent pyelonephritis. Women were randomized into 2 groups: I (n=30) consisted of patients treated with Nucleinat sodium at a dose of 0.25 mg 4 times daily for 14 days to II (n=25) included patients who used only antibacterial therapy (according to the set sensitivity identified pathogens). The control group consisted of 20 conditional healthy donors, matched for sex and age. Results. In the blood ofpatients prior to treatment were observed reduction of total peroxidase activity of red blood cells (p<0.0001), ceruloplasmin (p=0,03), SH–groups (p=0.001) and increase in serum MDA, and oxidative stress index (p<0.0001) compared with the conditionally healthy donors. Comprehensive treatment with Nucleinat sodium statistically significantly led to normalization of these parameters and decrease of urine N–acetyl–p–D– glucosaminidase and p–galactosidase (p<0,001 and p=0.04, respectively). Conclusions. For the treatment of uncomplicated recurrent pyelonephritis appropriate to apply Nucleinat sodium at a dose of 0.25 mg 4 times / day, which helps to reduce the activity of oxidative processes and the negative impact of oxidative stress on the patient, and increases of antioxidant protection.

Objective The present study aimed to assess the global oxidant and antioxidant status in infants born to preeclamptic mothers and their correlation with cardiac functions. Study Design We compared 40 infants born to preeclamptic mothers with 40 premature infants born to normotensive mothers. We assessed the relationship between echocardiographic measurements and total antioxidant capacity (TAC) and total oxidant status (TOS) values. Results In the study group, TAC, TOS, and oxidative stress index (OSI) levels were significantly higher in the cord blood (p = 0.03, 0.04, and 0.039, respectively) than in the control group. We did not observe any correlation between echocardiographic measurements and TAC, TOS, and OSI levels in infants born to preeclamptic mothers. Conclusion Compared with the control group, despite higher TAC levels in infants born to preeclamptic mothers, concurrent elevated OSI levels reveal that the oxidant–antioxidant balance is disturbed in favor of oxidants. Furthermore, the findings of this study suggest that echocardiographic parameters are unaffected by the oxidant status.

Diabetes mellitus is one of the serious global health problems affecting a significant proportion of both developed and developing countries. Overproduction of free radicals and oxidative stress has been associated with the development of diabetic complications. In the present study, the antioxidant effects of metformin (MET), glibenclamide (GLI), and repaglinide (REP) were evaluated in alloxan-induced diabetic rats. The findings from this study may possibly help in understanding the efficacy of these standard drugs in managing the complications arising from diabetes mellitus (DM). Alloxan (130 mg/kg BW) was administered as a single dose to induce diabetes. Four (4) groups of rats (n=6) were used; group 1 served as diabetic control while groups 2, 3, and 4 were the diabetic test groups that received MET (25 mg/kg), GLI (2.5 mg/kg), and REP (0.5 mg/kg), respectively. The result of the study showed significant (p<0.05) improvement in the altered antioxidant enzymes (SOD, CAT) and GSH concentration in diabetic treated rats compared with the diabetic control group. MET and REP produced significant effect on the MDA concentration while GLI showed insignificant reduction in the MDA concentration compared with the diabetic control. Findings from this study suggest that the administration of MET, GLI, and REP exerts significant antioxidant effects in alloxan-induced diabetic rats, thus contributing to the protective effect against oxidative stress-induced damage during diabetic complications.

Abstract: Malnutrition because of protein deficiency on diet caused antioxidant deficiency state and oxidative stress on brain that marked by increased MDA level. The aim of this study was to analyze the antioxidant effects of saluang (Rasbora spp.) on brain MDA level in malnourished rats. This was an experimental study with a Posttest Only with Control Group Design, consisted of malnutrition control group (M), group that feed with saluang (S), group that given standard feeding (P), and group that given standard feeding added with DHA supplement (D). Brain MDA level measured with TBARS method in spectrophotometer. Mean brain MDA level for each group in a row is M = 210,750 μΜ, S = 194,125 μΜ, P = 202,625 μΜ, and D = 200,875 μΜ. Kruskal-Wallis and Mann Whitney test showed that there were significant differences between the group that feed with saluang and the other groups (p<0.05). Based on this research, concluded that the administration of saluang (Rasbora spp.) significantly affects the decrease of brain MDA level in malnourished rats. Keywords: antioxidant, saluang fish, MDA, malnutrition Abstrak: Malnutrisi karena defisiensi protein akan menimbulkan keadaan kekurangan antioksidan dan menimbulkan stres oksidatif pada otak yang ditandai dengan peningkatan kadar MDA. Penelitian ini merupakan penelitian eksperimental dengan Posttest Only with Control Group Design, terdiri dari kelompok kontrol malnutrisi (M), kelompok pemberian pakan saluang (S), kelompok pemberian pakan standar (P), dan kelompok pemberian pakan standar yang ditambah suplemen DHA (D). Kadar MDA otak diukur dengan menggunakan metode TBARS secara spektrofotometer. Rerata kadar MDA otak masing-masing kelompok perlakuan berturut-turut adalah M = 210,750 μΜ, S = 194,125 μΜ, P = 202,625 μΜ, dan D = 200,875 μΜ. Uji Kruskal-Wallis dan uji Mann Whitney menunjukkan bahwa terdapat perbedaan bermakna antara kelompok yang diberi pakan saluang dengan kelompok lainnya (p<0,05). Berdasarkan penelitian yang dilakukan dapat diambil kesimpulan bahwa pemberian ikan saluang (Rasbora spp.) berpengaruh secara bermakna terhadap penurunan kadar MDA otak tikus putih malnutrisi. Kata-kata kunci: antioksidan, ikan saluang, MDA, malnutrisi

Aim: This study evaluated the effects of metformin in combination with a herbal capsule (glucoblock) on insulin resistance and oxidative stress index in type 2 diabetic rats. Methodology: A total of 35 male Wistar albino rats weighing between 120-220 g were used for this study. The rats were placed on high fat diet, and diabetes was induced by a single intraperitoneal injection of freshly prepared streptozotocin (STZ) (45 mg/kg body wt). Fasting plasma glucose (FPG) was determined using the glucose oxidase method. Fasting plasma insulin (FPI), total oxidant status (TOS), total antioxidant status (TAS) and superoxide dismutase (SOD) levels were quantitatively determined by a rat-specific sandwich-enzyme linked immunosorbent assay (ELISA) method. Insulin resistance (IR) was determined using the homeostatic model assessment for insulin resistance (HOMA-IR) method. Oxidative stress index (OSI) was determined by the ratio of TOS to TAS. Phytochemical analysis on the herbal capsule was done using classical methods. Results: The results revealed the presence of alkaloids (100.31μg/mg), flavonoids (131.45μg/mg), cardiac glycosides (55.93μg/mg) and saponins (61.47μg/mg) in the herbal drug glucoblock. The results showed significantly lower FPG levels in the treatment groups when compared to the diabetic control. Group 3 administered metformin had significantly higher FPG levels compared to the negative control. Group 4 administered the herbal drug glucoblock and group 5 administered a combination of metformin and glucoblock, showed no significant differences in FPG levels when compared to the negative control. The diabetic control had significantly higher FPI levels compared to the negative control and treatment groups. The treatment groups showed no significant differences in FPI levels when compared to the negative control. HOMA-IR was significantly higher in the diabetic control compared to the negative control and treatment groups. Also, HOMA-IR values in the treatment groups showed no significant difference compared to the negative control except for group 3 (metformin), that was significantly higher than the negative control. SOD was significantly lower in the diabetic control, compared to the negative control and treatment groups. There were no significant differences in SOD levels in the treatment groups compared to the negative control. TOS levels in the negative control group and treatment groups were significantly lower, compared to the diabetic control. TAS was significantly lower in the diabetic control and treatment groups compared to the negative control. OSI in the diabetic control was significantly higher, compared to the negative control and treatment groups. Also, the treatment groups had significantly higher OSI compared to the negative control. Conclusion: High fat diet and streptozotocin induction produced significant insulin resistance and oxidative stress in the diabetic rats. Glucoblock was more effective in reducing insulin resistance compared to metformin. The combination showed synergistic drug-herb reaction as glucoblock potentiated the actions of metformin. Both showed antioxidant potential but were not effective in lowering oxidative stress to normal levels. There is need to incorporate antioxidant therapy in the treatment protocol for diabetes mellitus.

Background Dental caries can be affected or altered by the body’s oxidant / antioxidant balance and oxidative stress. Aim/Hypothesis To examine the relationship between dental caries and Total Oxidant Status (TOS), Total Antioxidant Status (TAS), and Oxidative Stress Index (OSI). Design The study group consisted of 52 children with dental caries (DMFS/dfs ≥ 5), and the control group consisted of 42 healthy children (DMFS/dfs = 0). The saliva samples, unstimulated and mid-morning, were collected from all children. Participants’ saliva samples were acquired by spitting for 5 minutes, after which the samples were frozen and stored at 4 ° C and kept at -80 ° C until analysis. NCSS (Number Cruncher Statistical System) 2007 (Kaysville, Utah, USA) was used for the statistical analysis. Results TOS of the patient group cases were statistically significantly higher than those of the control group (p=0,004; p<0,01). OSI values of the patient group cases were statistically significantly higher than those of the control group (p=0,008; p<0,01). Conclusion OSI and TOS as markers of oxidative stress levels were found to be significantly higher in the group of patients with dental caries than in the healthy group.

Objective: The objective of the present study was to determine oxidant and antioxidant status in infants with hyperbilirubinemia and/or kernicterus and to find whether there is a relationship between bilirubin level and oxidant/antioxidant status. Patients: The study includes 69 full-term newborns (neonates with hyperbilirubinemia needing phototherapy [Group 1, n = 36] and neonates with kernicterus [Group 2, n = 33]) and 25 age-matched healthy newborn. Results: Plasma total antioxidant capacity (TAC) and serum total oxidant status (TOS) were significantly higher in Groups 1 and 2 than the control group. There was a significant difference between Group 1 and control cases for malondialdehyde (MDA; p < 0.001). Total free sulfhydryl group (TTHI) values were significantly elevated in Group 1 compared to Group 2 and control cases. Correlation analysis showed that the correlation between total bilirubin (TB) and TAC, TOS, MDA and oxidative stress index may be expressed by a quadratic curve. After phototherapy, a statistically significant increase in nitrite level was observed. Conclusion: We demonstrated that the relationship between serum TB and antioxidants and oxidative stress could be expressed by a quadratic correlation curve.

This research studied the interrelationship between the immune and oxidative-antioxidant systems in a group of individuals who had lived for a long time in areas contaminated with radionuclides after the Chernobyl catastrophe and as a result experienced prolonged exposure to small doses of ionizing radiation. We have examined a group of 100 students aged 18–24, where 50 of them formed the control group and the remaining 50 belonged to the experimental group as they arrived from the territories of enhanced radioecological control (IV radiation zone, density of soil contamination by isotope 137Cs 3.7 x 104 – 18.5 x 104 Bq/m2). Here we determined the level of cortisol, leukocytes and their populations, the levels of lymphocyte subpopulations with phenotypes CD3+, CD5+, CD4+, CD8+, CD16+, CD72+, immunoregulatory index CD4+/CD8+, indicators of phagocytic activity of neutrophils and monocytes, IgG (H), IgM (H), IgA (H), malondialdehyde (MDA), ceruloplasmin (CP), transferrin (Tr), sulfhydryl (SH); and also calculated the oxidative stress index (OSI). We performed the analysis twice: in the absence/presence of additional emotional stress such as an examination session. The studies showed an increase in the oxidative stress index in the group examined from the experimental cluster, especially in terms of emotional stress. At the same time, the neutrophil level increased, but phagocytic activity of neutrophils and monocytes, the relative and absolute number of lymphocytes with phenotypes CD3+, CD5+, CD4+, CD4+/CD8+, and IgG levels decreased. Consequently we revealed the negative correlation between the indexes of oxidative stress in the group of examined (the oxidative stress index (ISO)/the level of malonic dialdehyde (MDA)) and the parameters of phagocytic activity of monocytes, the immunoregulatory index CD4+/CD8+, and the number of lymphocytes with the CD16+ phenotype. In this study we demonstrated the decrease in the participation of ceruloplasmin (CP) as an important antioxidant factor in maintaining the immune homeostasis of the group examined from radiation-contaminated areas compared with control group. The evidence of this is the lack of reliable correlation between ceruloplasmin level and immune system parameters. Moreover we found that radiation-induced intensification of oxidative processes in the experimental group grew in conditions of additional stresses of an emotional nature. Besides, it was accompanied by a significant correlation in the level of oxidative stress and phagocytic activity parameters. Reducing phagocytic activity and the CD4+/CD8+ index on the background of oxidative stress increase can be considered as a sign of immune system ageing, while a decrease in the number of lymphocytes with the CD16+ phenotype is a sign of antitumor defense inhibition. Thus, we draw the conclusion that the inhabitants of the territories of strengthened radioecological control, undergoing exposure to small doses of ionizing radiation from birth, show a significant imbalance of redox homeostasis, which creates the preconditions for immunoreactivity pathology development at the level of both innate and acquired immunity.

Background and Objectives. The aim of this study was to investigate inflammatory indicators and oxidative status in patients with asbestos exposure with and without mesothelioma and to compare results with data from healthy subjects.Methods. Eighty people with exposure to environmental asbestos and without any disease, 46 mesothelioma patients, and a control group of 50 people without exposure to environmental asbestos were enrolled in this prospective study. Serum total oxidant level (TOL), total antioxidant capacity (TAC), and oxidative stress index (OSI), CRP, transferrin, ceruloplasmin,α-1 antitrypsin, ferritin, and copper levels were measured.Results. Mesothelioma group exhibited higher TOL, OSI,α1-antitrypsin, ferritin and copper levels as compared to the other groups (P<0.001,P=0.007,P<0.0001,P<0.001, andP<0.001, resp.). Transferrin was lower in the mesothelioma group than in the other two groups (P<0.001). The asbestos group had higher TOL, TAC,α1-antitrypsin, and transferrin levels (P<0.001,P<0.001,P<0.001, andP<0.001, resp.), as well as lower OSI and ferritin levels as compared to the control group (P<0.001andP<0.001).Conclusions. We believe that elevated acute phase reactants and oxidative stress markers (TOL and OSI) in the mesothelioma group can be used as predictive markers for the development of asbestos-related malignancy.

The exact cause of breast cancer is unknown; it is a multifactorial disease. It is the most diagnosed and the second killer cancer among women. Breast cancer can be originated from tissues of breast or secondary from other organs via metastasis. Generally, cancer cells show aberrant metabolism and oxidative stress when compared to noncancerous tissues of breast cancer patients. The current study aims at evaluating glutamate and glucose metabolism through GDH and LDH enzyme activities, oxidant, and antioxidative status among breast cancer patients attending referral hospitals of Addis Ababa, Ethiopia. Result. Catalytic activities of glutamate dehydrogenase, lactate dehydrogenase, and oxidative stress index were significantly increased in both serum (4.2 mU/ml, 78.6 mU/ml, and 3.3 : 1, resp.) and cancerous tissues (1.4 mU/ml, 111.7 mU/ml, and 2.15 : 1, resp.) of breast cancer patients as compared to those in serum of control group (3.15 mU/ml, 30.4 mU/ml, and 2.05 : 1, resp.) and noncancerous tissues of breast cancer patients (0.92 mU/ml, 70.5 mU/ml, and 1.1 : 1, resp.) (P≤0.05). Correspondingly, ratios of reduced to oxidized glutathione were significantly decreased in both serum (20 : 1) and cancerous tissues (23.5 : 1) of breast cancer patients when compared to those in serum of control group (104.5 : 1) and noncancerous tissues of breast cancer patients (70.9 : 1) (P≤0.05). Conclusion. Catalytic activities of GDH and LDH, ratios of GSH to GSSG, and concentration of TOS among breast cancer patients were significantly higher than were those among control group and noncancerous tissues of breast cancer patients, while TAC of breast cancer patients is significantly lower than that of control group and normal tissues of breast cancer patients.

Introduction: Anesthesia induced during a surgical intervention, the duration of the surgical intervention, and the surgical intervention itself tend to affect immune functions, resulting in the formation of free radicals in the metabolism. Free radicals can cause postoperative disorders by targeting biomolecules in the cell, such as lipids, carbohydrates, proteins, and DNA. In the present study, we used remifentanil or esmolol to induce a controlled hypotension in patients who were undergoing septorhinoplasty under general anesthesia, and we planned to compare the effect of these agents on hemodynamics and oxidative stress relative to the control group. Methodology: A total of 75 patients aged between 18 and 65 y, ASA I-II, planned to undergo elective septorhinoplasty, were included in this study. Patients were randomly divided into the following three groups: Group R (remifentanil group, n = 25); Group E (esmolol group, n = 25); and Group C (control, n = 25). Anesthesia was induced with 2 mg/kg propofol 2 mg/kg, fentanyl 1 µg/kg, and rocuronium 0.6 mg/kg. Immediately after induction, Group R was started loading dose of remifentanil 1 µg/kg/min, followed by infusion at 0.25–0.50 µg/kg/min. In Group E, a loading dose of esmolol 500 µg/kg was given for 1 min, then infusion was continued @ 150–300 µg/kg. A targeted mean arterial pressure (MAP) of 55–65 mmHg was aimed. In Group C, remifentanil was infused at 0.1–0.2 µg/kg/min until a MAP of 70–100 mmHg was reached. During operation; systolic (SAP), diastolic (DAP) and mean arterial pressure (MAP), heart rate (HR), peripheral oxygen saturation (SpO2), EtCO2 (end tidal CO2) were recorded before induction, after induction, after intubation, at 5-min intervals during the first 30 min, and then at 10-min intervals during the intervention. The amounts of remifentanil and esmolol consumed by the patients during the operation were calculated and recorded. Blood samples that were taken twice, preoperatively and postoperatively, for malondialdehyde (MDA), superoxide dismutase (SOD), total oxidant level (TOL), total antioxidant level (TAL), and oxidative stress index (OSI). Results: MAP showed a greater decrease starting from the 25th min and 40th min after intubation in remifentanil group and esmolol group respectively, compared to the control group. In the remifentanil and control groups, there was a statistically significant decrease in the postoperative OSI levels compared to the preoperative levels. One the other hand, in the esmolol group, there was no statistically significant difference between the preoperative and postoperative median OSI levels. There was a significant increase in the postoperative TAL of the remifentanil group compared to the preoperative level.
