Bovine tuberculosis is a serious problem for agriculture all over the world and causes huge economic losses. At the same time, infections caused by Mycobacterium bovis not only affect animal husbandry and the food industry, but also pose a threat to public health, as M. bovis is often isolated as a pathogen of tuberculosis, especially in patients with various immune deficiencies. The situation is aggravated by the lack of simple and accessible methods for diagnosing tuberculosis in cattle, which significantly impedes the monitoring of infection and the application of appropriate preventive measures. Serological tests, due to their relatively cheap cost, rapidity, rather high sensitivity and specificity, are able to compete with the allergic test during mass examinations of cows. In the present study, we present the results on the study of the antigenic structure of mycobacteria and the isolation of the most specific antigenic components from the lysate of the M. Bovis cell wall destroyed on the Fast Prep 24 device. The developed method allows obtaining a serologically active antigenic fraction with a high degree of purity with a molecular weight of 28 kDa.

The problem of animal schistosomiasis is multifaceted. It intertwines veterinary, health, social and economic issues. The current state of the epizootology of schistosomiasis in the ecological conditions of the Aral Sea region is presented. Spontaneous infection of farm animals with mature Schistosoma turkestanica was 55.2% in cattle, sheep – 33.0%, horses – 21.5%, goats – 15.5% and camels – 10.2%. At individual farms of the Bozatau, Kungradsky, Muinaksky, Amudarya and other districts, cattle infection reached up to 100% with a high degree of infection intensity. We noted the cyclical nature of the epizootic of schistosomiasis in cattle in the Aral Sea region. The alternation of cyclicity is observed at specified intervals – from 15 to 20 years. The ups and downs in the infection intensity alternate with a certain accuracy, according to which epizootics of schistosomiasis can be predicted for several years in advance. Currently, science has developed the necessary prerequisites for a sharp decrease in the incidence of animals with schistosomiasis and the elimination of invasion in individual foci. This is achieved by a complex of therapeutic and preventive measures, as well as the use of progressive methods and technology of animal husbandry

Recently, large-scale screening for COVID-19 has presented a major challenge, limiting timely countermeasures. Therefore, the application of suitable rapid serological tests could provide useful information, however, little evidence regarding their robustness is currently available. In this work, we evaluated and compared the analytical performance of a rapid lateral-flow test (LFA) and a fast semiquantitative fluorescent immunoassay (FIA) for anti-nucleocapsid (anti-NC) antibodies, with the reverse transcriptase real-time PCR assay as the reference. In 222 patients, LFA showed poor sensitivity (55.9%) within two weeks from PCR, while later testing was more reliable (sensitivity of 85.7% and specificity of 93.1%). Moreover, in a subset of 100 patients, FIA showed high sensitivity (89.1%) and specificity (94.1%) after two weeks from PCR. The coupled application for the screening of 183 patients showed satisfactory concordance (K = 0.858). In conclusion, rapid serological tests were largely not useful for early diagnosis, but they showed good performance in later stages of infection. These could be useful for back-tracing and/or to identify potentially immune subjects.

Poult enteritis and mortality syndrome (PEMS) is one of the most significant problem affecting turkeys and continues to cause severe economic losses worldwide. Although the specific causes of PEMS remains unknown, this syndrome might involve an interaction between several causative agents such as enteropathogenic viruses (coronaviruses, rotavirus, astroviruses and adenoviruses) and bacteria and protozoa. Non-infectious causes such as feed and management are also interconnected factors. However, it is difficult to determine the specific cause of enteric disorders under field conditions. Additionally, similarities of clinical signs and lesions hamper the accurate diagnosis. The purpose of the present review is to discuss in detail the main viral possible causative agents of PEMS and challenges in diagnosis and control.

Precision multiple-beam interference methods are applied to the study of the topography of octahedral faces of three diamonds. A new crossed fringe technique of high sensitivity is described. Evidence is adduced to support the view that the curvature of diamond faces and the occurrence of triangular pits are to be attributed to growth and not to solution. The observations thus settle a long-standing controversy. Irregular depressions which are observed are attributed to a solution mechanism. New features associated with the triangular growth pits are described and interpreted. A crystal surface with regions showing an abnormally high degree of optical plane uniformity is described. The surface of a polished diamond is described.

The experience of implementing the food safety management system at OJSC Kuban Confectionery plant is shown. Physical, chemical and microbiological factors that reduce the safety of products produced at the Kuban Confectionery plant-flour confectionery products and products of the chocolate production workshop, have been established. Preliminary programs developed and implemented at the plant to prevent the risk of contamination of products are listed. It was found that the introduction of preventive measures allowed: to gradually reduce the number of critical control points at the plant from 88 to 4, to ensure that there are no complaints about the quality and safety of products from regulatory authorities, to reduce by 10% in 2019 the number of claims from consumers compared to 2018.

Rheumatoid arthritis (RA) is a chronic, autoimmune connective tissue disease of unknown etiology. RA affects about 1% of the human population, women suffer three times more often than men, with the peak incidence between the age of 40 to 50. The up-to-date criteria from 2010 for the diagnosis of RA include: occurrence and duration of clinical signs, indicators of inflammation and serological tests. Neopterin, a protein released by macrophages, is a sensitive indicator of inflammation and the severity of RA. Regarding the serological tests, anti-cyclic citrullinated peptide antibodies represent a well-known marker with the specificity for RA of about 98%. The antibodies may be present in the serum of patients even a few years before the first clinical signs of the disease, heralding erosive changes in the joints and more severe course of RA. The literature also contains reports about autoantibodies anti-CarP and anti-Sa/ anti-MCV, which may occur in people with pain and swelling of joints and precede full-blown development of RA as well as reflect disease activity. Serological diagnosis of RA may be supported by some genetic tests based on PCR for detecting mutations e.g. C1858T in the PNPN22 gene. In turn, the quantitative analysis of different classes of miRNAs seems justified in order to better classify patients showing symptoms of RA. Further studies are needed that take into account the role of different markers in the development of RA, and confirm the high sensitivity and specificity of these markers in the diagnosis of the disease.

In time the environmental conditions could damage textiles (materials/ artifacts) causing the need to develop better non-destructive or at least micro-destructive analysis techniques of the samples. There are ethnographic textile artifacts that were treated in the past with various pesticides, that have not been mentioned in any document. These are often re-treated with chemicals by museum staff as a method of preventing pest infestation. Due to the progressive use of many pesticides, this paper was focused on the detection and quantification of three pesticides: malathion, methoxychlor, and permethrin (cis- and trans- isomers). Gas chromatography is one of the most widely used analytical techniques for characterizing volatile organic compounds and therefore was the analytical method of choice for the present study. Because these analytes are found at trace levels, the detection and quantification limits of analytes are very small and it is necessary to optimize and validate a SIM method - that allows the mass spectrometer to detect specific compounds with high sensitivity. In SIM mode, the instrument is set to collect data at selected masses of interest, thus increasing the accuracy and precision of the quantitative results. The present paper is aimed to develop this type of method with specificity and selectivity, high precision (expressed in terms of repeatability and intermediate accuracy), accuracy, suitable working range and linearity, and high degree of series homogenity.

Most metabolites of aflatoxin B1 (AFB1), especially exo-AFB1-8,9-epoxide (AFBO), can induce the production of reactive oxygen species (ROS) to vary degrees, causing oxidative stress and liver damage, and ultimately induce liver cancer in humans and animals. Duck is one of the most sensitive animals to AFB1, and severe economic losses are caused by duck AFB1 poisoning every year, but the exact mechanism of this high sensitivity is still unclear. This review highlights significant advances in our understanding of the AFB1 metabolic activation, like cytochrome P450s (CYPs), and AFB1 metabolic detoxification, like glutathione S-transferases (GSTs) in poultry. In addition, AFB1 may have other metabolic pathways in poultry, such as the mutual conversion of AFB1 and aflatoxicol (AFL) and the process of AFBO to produce AFB1-8,9-dihydrodiol (AFB1-dhd) and further metabolize it into detoxification substances. This review also summarized some exogenous regulatory substances that can alleviate AFB1-induced oxidative stress.

As a test for detecting middle ear disease among preschool children, tympanometry — as opposed to audiometry — has three advantageous attributes: a high degree of sensitivity, minimal need for subject cooperation, and total objectivity. For these reasons interest has arisen in tympanometry as a method for screening, i.e., identifying children with previously undetected middle ear disease. However, uncertainty persists concerning the importance of detecting apparently asymptomatic middle ear effusions, and concerning optimal methods, or even the advisability, of treating them. Further, the sensitivity and specificity of tympanometry depend on how the pass-fail cutoff point is defined. Defining this cutoff point so as to achieve high sensitivity may result in excessively low specificity, with the production of large numbers of false-positives who then become overreferrals. Data are presented to show how the validity of the test may be increased to some extent by attention to the gradient of “negative-pressure” tympanograms. At the present time, given the various aforementioned uncertainties, and with adequate validation as to the presence or absence of disease often lacking in reported studies of impedance screening in preschool populations, the cumulative results of these studies do not warrant embarking on large-scale screening programs. What is needed instead is additional research to explore the issue further.
