This study was designed to explore the gross and microscopic structures of the testes of Black Bengal bucks (n = 14) with special emphasis on the seminiferous tubules. A quantitative comparison of the various cell types in the seminiferous tubules of the testes were done. Biometrical values of testes were recorded. The left testes were significantly larger than the right. The mean length, width, weight and circumference of the left testis were 6.7 ± 0.1 cm, 3.9 ± 0.0 cm, 66.9 ± 0.8 gm and 13.4 ± 0.2 cm, respectively. The mean length, width, weight and circumference of the right testis were 6.3 ± 0.0 cm, 3.8 ± 0.0 cm, 66.5 ± 0.8 gm and 13.1 ± 0.1 cm, respectively. For microscopic studies tissue samples were evaluated with quantitative techniques. The testis was encapsulated by tunica toward the mediastinum testis. The testicular parenchyma was divided into convoluted seminiferous tubules and Leydig cells, which were found in the intertubular spaces. The Bangladesh Veterinarian (2018) 35(1&2): 40-46

Morphological study of the testis of adult Sudanese Chicken : gallus domesticus The adult chicken testes were two bean - shaped , large and soft , the left testis is usually higher in position and larger in size than the right one . The testis is active during cold weather with the mean diameter of the seminiferous tubules being 126^m in the chicken . it is less active during the hot season with the mean diameter of the seminiferous tubules being 135^m in the chicken. The non - breeding season seemed to be characterized by a decline in the spermatogenic activity only and not by complete spermatogenesis

SummaryThe spiny rat (Proechimys guyannensis) is a neotropical rodent that is used in biomedical research, particularly research related to chronic resistance to epilepsy and infectious diseases. To our knowledge, there are few reports concerning the reproductive biology of this species. Therefore, besides providing basic biometric and morphometric data, in the present study we investigated testis function and spermatogenesis in adult spiny rats. The mean testis weight and gonadosomatic index obtained were 1.63 ± 0.2 g and 1.15 ± 0.1% respectively. Based on the development of the acrosomic system, 12 stages of the seminiferous epithelium cycle were characterized. Stages VI and VII presented the highest frequencies (~17–19%), whilst stages II to V showed the lowest frequencies (~2–4%). The most advanced germ cell types labelled at 1 h or 20 days after BrdU injections were respectively preleptotene/leptotene spermatocytes at stage VII and elongated spermatids at stage III. The mean duration of one cycle was 7.5 ± 0.01 days and the entire spermatogenic process lasted 33.7 ± 0.06 days (~4.5 cycles). The seminiferous tubules (ST) occupied ~96 ± 1% of the testis parenchyma, whereas Leydig cells comprised only 1.5 ± 0.4%. The number of Sertoli cells (SC) per testis gram and the SC efficiency (spermatids/SC) were respectively 78 × 106 ± 11 × 106 and 7.9 ± 1. The daily sperm production per testis gram (spermatogenic efficiency; daily sperm production (DSP)/g/testis) was 78 × 106 ± 8 × 106. To our knowledge, this spermatogenic efficiency is among the highest found for mammals investigated to date and is probably related to the very short duration of spermatogenesis and the very high ST percentage and SC number obtained for this species.

The insectivorous bat Myotis nigricans is widely distributed throughout the Neotropics, including Brazil, and has a reproductive biology that is affected by climate and food availability. To evaluate the reproductive capacity of this species, morphofunctional parameters of the testes were correlated with environmental variables and the body condition of individuals captured. After bats had been killed, their testes were removed, fixed in Karnovsky’s fluid for 24 h and embedded in resin for evaluation by light microscopy. The mean annual tubulosomatic index (0.58%) and the percentage of seminiferous tubules in the testes (88.96%) were the highest ever recorded for the Order Chiroptera. The percentage of Leydig cells and volume of the cytoplasm of Leydig cells were higher in the rainy than dry season (80.62 ± 3.19% and 573.57 ± 166.95 μm, respectively; mean ± s.d.). Conversely, the percentage of nuclei of the Leydig cells in the dry season (26.17 ± 3.70%; mean ± s.d.) and the total number of Leydig cells (6.38 ± 1.84 × 109; mean ± s.d.) were higher in the dry season. The results of the present study could help in future conservation of these bats because they provide a better understanding of the bats’ reproductive strategies and how the species can adapt to changes.

The microscopic features of the testis were studied in gonadally inactive Khaki Campbell duck (Anas platyrhynchos domesticus) in Bangladesh. The study was conducted in the Department of Anatomy & Histology, Bangladesh Agricultural University, Mymensingh. Five adult healthy birds of one-year-old were used for this study. The testes were collected immediately after ethical killing of the birds for histological observations. The collected tissue samples were then processed and stained with Hematoxylene & Eosin (H & E) stain for histological observations. The seminiferous tubules showed considerable involution with cessation of spermatogenesis. The basal lamina of the seminiferous tubules was irregular in outline and was invaginated into the germinal epithelium in the form of finger-like plicae or folds. Most of the lumen of the seminiferous tubules was empty and all generation of germ cells were not present in most of the seminifeous tubules. The interstitium showed a relative increase in volume and interstitial tissue consisted of loose connective tissue, interstitial cells (Leydig cells), few connective cells and blood vessels. This study first time described the microscopic features of testis of Khaki Campbell ducks in Bangladesh during inactive phases of the reproductive cycle.

The embryonic gonad is undifferentiated in males and females until a critical stage when the sex chromosomes dictate its development as a testis or ovary. This binary developmental process provides a unique opportunity to delineate the molecular pathways that lead to distinctly different tissues. The testis comprises three main cell types: Sertoli cells, Leydig cells, and germ cells. The Sertoli cells and germ cells reside in seminiferous tubules where spermatogenesis occurs. The Leydig cells populate the interstitial compartment and produce testosterone. The ovary also comprises three main cell types: granulosa cells, theca cells, and oocytes. The oocytes are surrounded by granulosa and theca cells in follicles that grow and differentiate during characteristic reproductive cycles. In this review, we summarize the molecular pathways that regulate the distinct differentiation of these cell types in the developing testis and ovary. In particular, we focus on the transcription factors that initiate these cascades. Although most of the early insights into the sex determination pathway were based on human mutations, targeted mutagenesis in mouse models has revealed key roles for genes not anticipated to regulate gonadal development. Defining these molecular pathways provides the foundation for understanding this critical developmental event and provides new insight into the causes of gonadal dysgenesis.

An H1 histone variant, H1a, has been isolated and purified from the mouse testis. Biochemical and amino acid analyses indicate its similarity with the rat testis H1a. Specific antibodies against the purified H1a have been generated in rabbits and used to study its tissue and species distribution using protein blotting procedures. We have also used the immunocytochemical technique to determine in situ distribution of H1a in spermatogenic cells and somatic tissues of the mouse. A non-random distribution of H1a has been noted in the nuclei of certain somatic cell types such as Sertoli cells, Leydig cells and neurons. By contrast, hepatocyte nuclei lacked detectable levels of H1a. In adult seminiferous tubules, the early primary spermatocyte nuclei displayed a greater level of immunoreactivity relative to other cell types. Developmental studies indicate its initial expression in the 7-day-old mouse testis concomitant with the appearance of intermediate and type B spermatogonia.

A new infertility syndrome has recently been described in Finnish Yorkshire boars. Typical for the syndrome is total akinesia and severe tail malformation of the spermatozoa. Morphometric analysis was performed on semen smears from 20 affected and 18 control boars and on testicular tissue sections from 5 affected and 4 control boars. Semen morphometry revealed that, in affected boars, the length of the sperm tails was only 33% of that of the controls (15.4 μm vs. 47.0 μm, P < 0.0001). Typical for the spermatozoa of affected boars was also an abundant frequency of proximal cytoplasmic droplets (72.4% vs. 6.9%, P < 0.0001), whereas no major sperm-head abnormalities were recorded. In the testicular tissue samples, viewed at light microscopic level, the volume densities of seminiferous tubules or interstitium did not differ. The most characteristic change in the seminiferous epithelium of the affected boars was a reduced number of elongated spermatids. Densities of Sertoli cells and Leydig cells between affected and control boars did not differ. The ultrastructure of testicular tissue from affected boars showed severe alterations in the assembly of the midpiece and tail of the spermatozoa. As well, a typical finding in the seminiferous epithelium of affected boars was conspicuous deposition of lipid droplets. The pathogenesis of this syndrome severely affects spermiogenesis and motility. Spermatozoa have malformed, short tails, which never become motile. This syndrome is not manifested in the structure or function of other ciliated cells in the affected animals.

A 4-year-old boy with an undescending left testis, penoscrotal hypospadia and bilateral microphthalmia was admitted to our hospital. Chromosome analysis revealed a karyotype of 46, XX del(x)(p2 2,31) and the sex-determining region of the Y chromosome (SRY) was negative. The right testis was located in the scrotum and a left cystic ovary-like gonad, a salpinx and a unicorn uterus were found in the left inguinal canal. Histologically the gonad was an ovotestis in which primordial follicles covered infantile seminiferous tubules. Microphthalmia is observed in some congenital syndromes caused by interstitial deletion of the X chromosome. This case suggested that the short arm of the X chromosome was involved in the differentiation of the gonad. Very closely located follicles and infantile seminiferous tubules indicated that induction of meiosis in the fetus was controlled by the local microenvironment in follicles and seminiferous tubules, and not by the systemic hormonal condition.

Abstract Background Medico-legal or forensic autopsy is a special type of autopsy performed under the orders of a legal authority in circumstances involving unnatural, suspicious or criminal deaths. Gross and histopathological studies of viscera in medico-legal autopsies have been observed to serve as crucial tool in the detection of undiagnosed disease conditions and the resulting findings may closely represent the general population. Methods A prospective cross sectional study was carried out at the Muhimbili National Hospital (MNH) department of pathology, to medico-legal autopsies received at the MNH mortuary. The study included 103 cases in an eight month period. Both lungs were weighed for each case and tissue samples were collected from each lobe, in both lungs. Other gross findings were observed and recorded. Tissue sections were made and stained with hematoxylin and eosin and evaluated under a microscope. Results Out of 103 cases studied, 88.3% were male with mean age of 35.8 + 16.6 years. In females, the mean age was 31.6 + 10.4 years. The mean weight of lungs was found to be 627.3 + 138.5g and 591.7 + 129.7g for the right and left lung, respectively, in males. In females, the mean weight of lungs was 616.7 + 166.1g and 583.3 + 155.8g for the right and left lung, respectively. More than half (53.4%) of lung samples had some pathological disease, and the leading diagnosis was pulmonary congestion/oedema (22.3%) followed by lobar pneumonia (18.5%). Age was found to relate with the observed histological diagnosis of the lungs. Conclusion Males, especially the youth and adults are the main victims of medico-legal deaths. The study also revealed high prevalence of undiagnosed lung diseases in victims of medico-legal deaths. These findings may reflect heavy burden of lung diseases in the general population.

Pituitary glands were grafted under the capsule of the left testis of rats to induce high levels of prolactin in this organ. One hundred days after implantation, significantly increased levels of prolactin were found in the tissue and the venous plasma of the left testis. Although the levels of testosterone in testicular venous plasma were not raised, the testicular content of testosterone was increased when compared to the right testis. The ratio of testosterone and dihydrotestosterone was not affected in the pituitary-grafted testis. Since the seminiferous tubules adjacent to the pituitary graft appeared to be completely normal, it is concluded that in the rat high levels of prolactin have no direct inhibitory effect on testicular functions.
