The stem amniote Orobates pabsti has been reconstructed to be capable of relatively erect, balanced, and mechanically power-saving terrestrial locomotion. This suggested that the evolution of such advanced locomotor capabilities preceded the origin of crown-group amniotes. We here further investigate plausible body postures and locomotion of Orobates by taking soft tissues into account. Freely available animation software BLENDER is used to first reconstruct the lines of action of hindlimb adductors and retractors for Orobates and then estimate the muscle strain of these muscles. We experimentally varied different body heights in modeled hindlimb stride cycles of Orobates to find the posture that maximizes optimal strains over the course of a stride cycle. To validate our method, we used Caiman crocodilus. We replicated the identical workflow used for the analysis of Orobates and compared the locomotor posture predicted for Caiman based on muscle strain analysis with this species’ actual postural data known from a previously published X-ray motion analysis. Since this validation experiment demonstrated a close match between the modeled posture that maximizes optimal adductor and retractor muscle strain and the in vivo posture employed by Caiman, using the same method for Orobates was justified. Generally, the use of muscle strain analysis for the reconstruction of posture in quadrupedal vertebrate fossils thus appears a promising approach. Nevertheless, results for Orobates remained inconclusive as several postures resulted in similar muscle strains and none of the postures could be entirely excluded. These results are not in conflict with the previously inferred moderately erect locomotor posture of Orobates and suggest considerable variability of posture during locomotion.

Collagenous biomaterials have been used for growing cells in vitro as well as for augmentation and replacement of hard and soft tissues. The substratum used for culturing cells is implicated in the modulation of phenotypic cellular expression, cellular orientation and adhesion. Collagen may have a strong influence on these cellular parameters when used as a substrate in vitro. Clinically, collagen has many applications to wound healing including, skin and bone substitution, tendon, ligament, and nerve replacement. In this report we demonstrate two uses of collagen. First as a fiber to support fibroblast growth in vitro, and second as a demineralized bone/collagen sponge for radial bone defect repair in vivo.For the in vitro study, collagen fibers were prepared as described previously. Primary rat tendon fibroblasts (1° RTF) were isolated and cultured for 5 days on 1 X 15 mm sterile cover slips. Six to seven collagen fibers, were glued parallel to each other onto a circular cover slip (D=18mm) and the 1 X 15mm cover slip populated with 1° RTF was placed at the center perpendicular to the collagen fibers. Fibroblast migration from the 1 x 15mm cover slip onto and along the collagen fibers was measured daily using a phase contrast microscope (Olympus CK-2) with a calibrated eyepiece. Migratory rates for fibroblasts were determined from 36 fibers over 4 days.

SummaryThe distribution of 32P-polyphosphates (lineal and cross-linked) and 32Porthophosphate in normal and tumor bearing animals has been studied. Differences between the cross-linked and the lineal form are related to a different degree of susceptibility to the hydrolysis by the phosphatases. In contrast to orthophosphate, the polyphosphates showed a lower accumulation in soft tissues which gives an advantageous reduction of the total body radiation dose.

AbstractThe development of biomedical glues is an important, yet challenging task as seemingly mutually exclusive properties need to be combined in one material, i.e. strong adhesion and adaption to remodeling processes in healing tissue. Here, we report a biocompatible and biodegradable protein-based adhesive with high adhesion strengths. The maximum strength reaches 16.5 ± 2.2 MPa on hard substrates, which is comparable to that of commercial cyanoacrylate superglue and higher than other protein-based adhesives by at least one order of magnitude. Moreover, the strong adhesion on soft tissues qualifies the adhesive as biomedical glue outperforming some commercial products. Robust mechanical properties are realized without covalent bond formation during the adhesion process. A complex consisting of cationic supercharged polypeptides and anionic aromatic surfactants with lysine to surfactant molar ratio of 1:0.9 is driven by multiple supramolecular interactions enabling such strong adhesion. We demonstrate the glue’s robust performance in vitro and in vivo for cosmetic and hemostasis applications and accelerated wound healing by comparison to surgical wound closures.

Micro arc oxidation (MAO) is a prominent surface treatment to form bioceramic coating layers with beneficial physical, chemical, and biological properties on the metal substrates for biomaterial applications. In this study, MAO treatment has been performed to modify the surface characteristics of AZ31 Mg alloy to enhance the biocompatibility and corrosion resistance for implant applications by using an electrolytic mixture of Ca3(PO4)2 and C10H16N2O8 (EDTA) in the solutions. For this purpose, the calcium phosphate (Ca-P) containing thin film was successfully fabricated on the surface of the implant material. After in-vivo implantation into the rabbit bone for four weeks, the apparent growth of soft tissues and bone healing effects have been documented. The morphology, microstructure, chemical composition, and phase structures of the coating were identified by SEM, XPS, and XRD. The corrosion resistance of the coating was analyzed by polarization and salt spray test. The coatings consist of Ca-P compounds continuously have proliferation activity and show better corrosion resistance and lower roughness in comparison to mere MAO coated AZ31. The corrosion current density decreased to approximately 2.81 × 10−7 A/cm2 and roughness was reduced to 0.622 μm. Thus, based on the results, it was anticipated that the development of degradable materials and implants would be feasible using this method. This study aims to fabricate MAO coatings for orthopedic magnesium implants that can enhance bioactivity, biocompatibility, and prevent additional surgery and implant-related infections to be used in clinical applications.

The aim of this study was to characterise the bulk modulus properties of the upper arm under relaxed and controlled contraction which is defined as 25% of the maximum voluntary contraction. A new testing machine was designed to generate constant load on the upper arm and measure the deformation over time. The machine consists of a device which is effectively a cuff that applies controllable pressure on a 47 mm wide band of the upper arm. Six different loads (10, 20, 30, 40, 50 and 60 kgf) were applied over a period of time of up to a maximum of 120 seconds. The deflection-time curves obtained show strongly non-linear response of the bulk tissue. The non-linearity manifested by these deflection-time curves is in terms of both time- and load-dependency. For each load, the creep behaviour follows an exponential law typical of viscoelastic materials. At low loads (below 30kgf), the creep response increases fairly linearly as the load is increased from 10 kgf to 30 kgf. But at high loads (above 30 kgf), the creep response increases only slightly as the load is increased from 30 kgf to 60 kgf. Beyond a load of 60 kgf, the deflection or creep becomes negligible. This implies that the upper arm has reached the state of incompressibility. The creep behaviour of the upper arm was simulated using four Voigt viscoelastic models in series. The three obvious soft tissues of the upper arm, namely skin, fat and muscle, were modelled in series. The effects of blood vessels and connective tissue were also modelled in series with the other tissues.

Computerized tomography (CT), a technique conceptualized by Oldendorf in 19611 and developed by Hounsfield2 of EMI-Tronics Inc. (EMI) Central Research Laboratories, has proven to be a successful innovation in neuroradiology. Reviews by Ambrose3 in England and by Baker et al.4 and by New et al.5 in the United States have clearly demonstrated the value of this new modality in neuroradiological diagnosis. In 1975 Houser et al.6 and Harwood-Nash et al.7 provided the initial clinical and radiological data about CT in infants and children. More recently this technique has been extended to the study of tissues and organs in the body other than those in the head. This has been accomplished by modification of the original machine into a whole-body CT system. Early reviews by Ledley et al.8 and by Alfidi et al.9 suggest a significant potential for diagnosis of lesions in the abdomen, pelvis, and thorax. The advantages of CT are that it is less invasive than standard special diagnostic radiological procedures and that for the first time it provides in vivo information regarding the content and the characteristics of tissue composing organs and masses. DESCRIPTION OF EQUIPMENT In conventional radiography an image is made on radiographic film by an attenuated X-ray beam. In passing through a core of tissue, each ray of the beam is attenuated as it is absorbed and scattered by the tissue in its path. The intensity of the transmitted ray depends on the sum total of X-ray attenuation by all the different soft tissues in its path.

SUMMARYChameleons capture prey items using a ballistic tongue projection mechanism that is unique among lizards. During prey capture, the tongue can be projected up to two full body lengths and may extend up to 600 % of its resting length. Being ambush predators, chameleons eat infrequently and take relatively large prey. The extreme tongue elongation (sixfold) and the need to be able to retract fairly heavy prey at any given distance from the mouth are likely to place constraints on the tongue retractor muscles. The data examined here show that in vivo retractor force production is almost constant for a wide range of projection distances. An examination of muscle physiology and of the ultrastructure of the tongue retractor muscle shows that this is the result (i) of active hyoid retraction, (ii) of large muscle filament overlap at maximal tongue extension and (iii) of the supercontractile properties of the tongue retractor muscles. We suggest that the chameleon tongue retractor muscles may have evolved supercontractile properties to enable a substantial force to be produced over a wide range of tongue projection distances. This enables chameleons successfully to retract even large prey from a variety of distances in their complex three-dimensional habitat.

The success of current total knee replacement (TKR) devices is contingent on the kinematics and contact mechanics during in vivo activity. Indicators of potential clinical performance of total joint replacement devices include contact stress and area due to articulations, and tibio-femoral and patello-femoral kinematics. An effective way of evaluating these parameters during the design phase or before clinical use is via computationally efficient computer models. Previous finite element (FE) knee models have generally been used to determine contact stresses and/or areas during static or quasi-static loading conditions. The majority of knee models intended to predict relative kinematics have not been able to determine contact mechanics simultaneously. Recently, however, explicit dynamic finite element methods have been used to develop dynamic models of TKR able to efficiently determine joint and contact mechanics during dynamic loading conditions [1,2]. The objective of this research was to develop and validate an explicit FE model of a TKR which includes tibio-femoral and patello-femoral articulations and surrounding soft tissues. The six degree-of-freedom kinematics, kinetics and polyethylene contact mechanics during dynamic loading conditions were then predicted during gait simulation.
