This study aims to investigate the effects of probiotics and Chinese medicine polysaccharides (CMPs) on growth performance, blood indices, rumen fermentation, and bacteria composition in lambs. Forty female lambs were randomly divided into four groups as follows: control, probiotics, CMP, and compound (probiotics + CMP) groups. The results showed that probiotics treatment increased the concentrations of blood glucose (GLU) and immunoglobulin G (IgG) and enhanced rumen microbial protein contents but declined the value of pH in rumen fluid compared with the control (P < 0.05). Furthermore, supplementation with CMP enhanced the average daily gain (ADG) and the contents of IgA, IgG, and IgM in the serum but decreased the F:G ratio compared with the control (P < 0.05). Besides, both CMP and compound (probiotics + CMP) treatments decreased the ratio of acetic acid and propionic acid compared with the control (P < 0.05). High-throughput sequencing data showed that at the genus level, the relative abundance of Veillonellaceae_UCG-001 in the probiotics group was increased, the relative abundance of Succiniclasticum and norank_f__Muribaculaceae in the CMP group were enhanced, and the relative abundance of Ruminococcaceae_UCG-002 in the compound group was raised compared with the control (P < 0.05). In summary, supplementation with probiotics can promote rumen protein fermentation but decrease the diversity of bacteria in rumen fluid; however, CMP treatment increased the relative abundance of Fibrobacteria, changed rumen microbial fermentation mode, increased the immune function, and ultimately improved the growth performance.

Abstract BackgroundThis study was conducted to evaluate the effects of three rearing systems (FL: flooring litter rearing, MC: multilayer cage rearing, PN: plastic net rearing) and narasin inclusion or not on growth performance, gastro-intestine development and health of broilers. A total of 2400 one-day-old Ross 308 mixed-sex broilers (1:1 ratio of males and females) were used in a completely randomized design utilizing a 3 × 2 factorial arrangement of treatments, with 12 replicates per treatment. In each replicate for FL, MC and PN consisted of: 34 birds per pen, 30 birds per cage, and 36 birds per pen, respectively, ensuring that the density of each rearing system was the same (12 birds/m2).ResultsLower ADG (average daily gain), ADFI (average daily feed intake) and FCR (feed conversation ratio) observed in MC group than those of the other two systems from 1 to 36 days of age (P < 0.05). Narasin inclusion in diets decreased ADFI and FCR significantly (P < 0.05). MC and PN rearing systems reduced the relative weight of the gizzard significantly (P < 0.05). Compared with FL rearing, MC reduced the relative weight of the duodenum, jejunum and ileum (P < 0.05). The mRNA expression levels of the ileal IL-1β and IFN-γ in FL was higher than those in PN and MC groups (P < 0.05). Narasin decreased the mRNA expressions of TNF-α in the ileum (P < 0.05). Different rearing systems changed the ileal microflora structure of broilers. The FL system increased ileal microbial diversity of broilers and relative abundance of Actinobacteria. Narasin combined with MC increased the relative abundance Proteobacteria of broilers. Conclusionbirds reared in PN had higher body weight. MC birds had poorer intestinal development and health condition, higher abundance of Proteobacteria, but better FCR. FL rearing appeared to be propitious for gastro-intestinal development and health. Narasin inclusion in diets improved FCR and changed the relative abundance Proteobacteria of broilers.

Oxidative stress commonly occurs in pig production, which can severely damage the intestinal function of weaned piglets. This study was conducted to investigate the effects of D-galactose with different levels used to induce chronic oxidative stress on growth performance, intestinal morphology and gut microbiota in weaned piglets. The results showed that addition of 10 and 20 g/kg BW D-galactose reduced average daily gain and average daily feed intake from the first to the third week. 10 g/kg BW D-galactose increased the concentration of serum MDA at the second and third week. 10 g/kg BW D-galactose significantly influenced the jejunal and ileal expressions of GPx1, CAT1, and MnSOD. The results of 16S rRNA sequencing showed that compared with the control, 10 and 20 g/kg BW D-galactose significantly decreased the relative abundance of Tenericutes, Erysipelotrichia, Erysipelotrichales, and Erysipelotrichaceae, while increased the relative abundance of Negativicutes, Selenomonnadales, and Veillonellaceae. The results indicated that treatment with 10 g/kg BW/day D-galactose for 3 weeks could induce chronic oxidative stress, reduce the growth performance and alter gut microbiota in weaned piglets.

The aim of this study was to evaluate the effect of the length of oat hay on the performance, health, behavior, and rumen fermentation of dairy calves. For this purpose, two hundred and ten healthy two-day-old Holstein dairy calves were randomly allocated into three groups: basic diet (calf starter) without hay (CON), or a basic diet with oat hay at either long (OL: 10–12 cm) or short (OS: 3–5 cm) length cut. The basic diet was fed from day 4, while the hay was offered from day 14. All calves were weaned at day 56 and remained in their individual hutches till the end of the trial (day 70). Calf starter intake and fecal scores were recorded daily. Bodyweight, body size, and rumen fluid samples were collected biweekly before weaning and weekly after weaning. Overall, providing oat hay (OS and OL) in the diet increased the body weight, starter intake, and average daily gain compared to the CON group. Similarly, feeding oat hay improved rumen fermentation. More specifically, hay enhanced the rumen pH and changed the rumen fermentation type. Hay fed calves spent more time on rumination but less time performing abnormal behaviors compared to control. As it can be concluded, feeding oat hay to calves enhances the growth performance, rumen fermentation, and normal calf behaviors, implying improved animal welfare irrespective of the hay length.

Fumonisin contamination in corn is an emerging issue in animal feed production. Fumonisin disrupts the metabolism of sphingolipids and reduces growth performance. This experiment was conducted to determine the effect of feeding fumonisin-contaminated corn on growth performance and sphinganine (SA) to sphingosine (SO) ratios of 9 to 28 kg pigs. A total of 350 pigs, were used with 5 pigs/pen and 14 pens/treatment. Dietary treatments contained fumonisin-contaminated corn (50 mg/kg of fumonisin B1 + B2) blended with low fumonisin corn (10 mg/kg of fumonisin B1 + B2) to provide dietary fumonisin concentrations of 7.2, 14.7, 21.9, 32.7, and 35.1 mg/kg. From day 0 to 28, increasing fumonisin concentration decreased (linear, p < 0.001) average daily gain, average daily feed intake (linear, p = 0.055), and gain:feed ratio (linear, p = 0.016). Although these response criteria tested linear, the greatest reduction in performance was in pigs fed with 32.7 and 35.1 mg/kg of fumonisin (B1 + B2). Increasing fumonisin concentration increased the serum SA:SO ratio (linear, p < 0.001) on day 14 and 28. In summary, for 9 to 28 kg nursery pigs, increasing fumonisin linearly decreased average daily gain and gain:feed ratio. However, despite the linear response, diets containing up to 21.9 mg/kg of fumonisin did not have as dramatic a decrease in growth performance as those fed more than 32.7 mg/kg. Further research is warranted to determine the effect of fumonisin concentrations between 21.9 and 32.7 mg/kg.

Abstract Despite the broad variety of available microRNA (miRNA) research tools and methods, their application to the identification, annotation, and target prediction of miRNAs in nonmodel organisms is still limited. In this study, we collected nearly all public sRNA-seq data to improve the annotation for known miRNAs and identify novel miRNAs that have not been annotated in pigs (Sus scrofa). We newly annotated 210 mature sequences in known miRNAs and found that 43 of the known miRNA precursors were problematic due to redundant/missing annotations or incorrect sequences. We also predicted 811 novel miRNAs with high confidence, which was twice the current number of known miRNAs for pigs in miRBase. In addition, we proposed a correlation-based strategy to predict target genes for miRNAs by using a large amount of sRNA-seq and RNA-seq data. We found that the correlation-based strategy provided additional evidence of expression compared with traditional target prediction methods. The correlation-based strategy also identified the regulatory pairs that were controlled by nonbinding sites with a particular pattern, which provided abundant complementarity for studying the mechanism of miRNAs that regulate gene expression. In summary, our study improved the annotation of known miRNAs, identified a large number of novel miRNAs, and predicted target genes for all pig miRNAs by using massive public data. This large data-based strategy is also applicable for other nonmodel organisms with incomplete annotation information.

The aim of this study was to evaluate the effect of zinc (Zn) supplementation in different commercial forms on the growth performance, health status, and Zn balance of weaners in field conditions. The animals were fed pre-starter (from the 28th to 47th day of life) and starter (from the 48th to 74th day of life) mixtures differing in Zn form and concentration. Group I was given ZnSO4 at 150 mg kg−1; Group II received pre-starter zinc oxide (ZnO) at 3000 mg kg−1 and starter at 150 mg kg−1; and Group III was given 150 mg kg−1 of zinc oxide nanoparticles (nZnO). We found that the average daily gain in Group I was significantly lower, compared to Groups II and III. A commonly accepted level of Zn (150 mg kg−1) as nZnO can be recommended, instead of therapeutic doses of Zn preparations with the same efficiency. Moreover, a lower level of Zn in the diet can prevent the excessive accumulation of this element in waste and, thus, reduce environmental damage.

Abstract Eukaryotic genomes encode thousands of small and large non-coding RNAs (ncRNAs). However, the expression, functions and evolution of these ncRNAs are still largely unknown. In this study, we have updated deepBase to version 3.0 (deepBase v3.0, http://rna.sysu.edu.cn/deepbase3/index.html), an increasingly popular and openly licensed resource that facilitates integrative and interactive display and analysis of the expression, evolution, and functions of various ncRNAs by deeply mining thousands of high-throughput sequencing data from tissue, tumor and exosome samples. We updated deepBase v3.0 to provide the most comprehensive expression atlas of small RNAs and lncRNAs by integrating ∼67 620 data from 80 normal tissues and ∼50 cancer tissues. The extracellular patterns of various ncRNAs were profiled to explore their applications for discovery of noninvasive biomarkers. Moreover, we constructed survival maps of tRNA-derived RNA Fragments (tRFs), miRNAs, snoRNAs and lncRNAs by analyzing &gt;45 000 cancer sample data and corresponding clinical information. We also developed interactive webs to analyze the differential expression and biological functions of various ncRNAs in ∼50 types of cancers. This update is expected to provide a variety of new modules and graphic visualizations to facilitate analyses and explorations of the functions and mechanisms of various types of ncRNAs.

SUMMARYA total of 70 Nellore bulls (18 ± 3 months of age) were used to determine the effects of crude glycerine (CG) replacing starch- v. fibre-based energy ingredients in low (LC; 0·40 concentrate) or high concentrate (HC; 0·60 concentrate) – on a dry matter (DM) basis – on DM intake (DMI), methane emissions and growth. Ten bulls were slaughtered (reference group) to obtain the carcass gain (CrG). The 60 remaining bulls (374 ± 24·5 kg) were allocated to a 2 × 3 factorial arrangement (two concentrate levels, LC or HC; and three feeding regimes, FR). The FR were: CO – without CG and maize as an ingredient of concentrate; CGM – inclusion of CG (0·10 of DM) replacing maize in the concentrate; and CGSH – inclusion of CG (0·10 of DM) replacing soybean hulls (SH) in the concentrate. Bulls fed LC or HC had similar DMI (kg/d) and growth. The DMI and average daily gain (ADG) were similar among FR. Concentrate level and FR tended to interact for methane emissions (g) per kg DMI. Bulls fed CGM had a greater G : F (g CrG/kg DMI) than those fed CO or CGSH diets. Increasing dietary concentrate (0·40–0·60) did not affect intake, methane emissions, or growth. Inclusion of CG in diets to replace SH in LC diets tended to decrease methane emissions from animals. When CG replaces SH in the diets, CrG and G:F (g CrG/kg DMI) are decreased compared with bulls fed CGM.
