The aim of the study was to determine the effect of herbicides and herbicides used with biostimulants on the content of total and true protein in potato tubers. The three-year field experiment was carried out using the random two-way split-plot arrangement in three repetitions. The examined factors were: I—three cultivars of potato: Bartek, Gawin, Honorata; II—five method applications of herbicides and herbicides with biostimulants: 1. Control—mechanical weeding, 2. Harrier 295 ZC (linuron + clomazone), 3. Harrier 295 ZC + Kelpak SL (linuron + clomazone and extract from algae Ecklonia maxima), 4. Sencor 70 WG (metribuzin), 5. Sencor 70 WG + Asahi SL (metribuzin and sodium p-nitrophenolate, sodium o-nitrophenolate, sodium 5-nitroguolacolate). On the plots sprayed with herbicides and herbicides mixed with biostimulants, a significant increase in the content of total and true proteins and the share of true protein in total protein in relation to tubers harvested from the control object was found. Genetic features of cultivars determined the content of total and true protein in potato tubers. The highest total and true protein was accumulated by tubers of the Bartek cultivar, and the least by the Honorata cultivar.

The aim of the study was to determine the interaction of herbicides and herbicides with biostimulants on the accumulation of dry matter and starch in potato tubers. In a three-year field experiment based on the method of randomised sub-blocks, two factors were taken into account. The first factor were potato cultivars: Bartek, Gawin, Honorata. The second factor were five methods of herbicides and biostimulants application: (1) the control object without chemical protection (CO); (2) herbicide linuron + clomazone (Harrier 295 ZC) (H); (3) herbicide linuron + clomazone (Harrier 295 ZC) and biostimulant Ecklonia maxima (Kelpak SL) – (H + K); (4) herbicide metribuzin (Sencor 70 WG) – (S); (5) herbicide metribuzin (Sencor 70 WG) and biostimulant sodium para-nitrophenol, sodium ortho-nitrophenol, sodium 5-nitroguaiacol (Asahi SL) – (S + A). The cultivars and weather conditions significantly affected the content and yields of dry matter and starch. The herbicides and biostimulants used determined the starch accumulation as well as dry matter and starch yields. Most starch in tubers (more by 3.7 g/kg) and the highest dry matter and starch yields (more by 2.87 and 1.79 t/ha, respectively), compared to the control object, were obtained after the application of the herbicide Sencor 70 WG and biostimulant Asahi SL.

Field experiments were carried out at the Agricultural Experimental Nuisance Station of University of Podlasie in Siedlce in the years 1999–2001. The influence of herbicides and their mixtures on total proteins content and on proper proteins in eatable potato tubers were analysed. The weeding chemical agents (pesticides) used in the potato treatment increased the total protein content by 0.09–0.18% and proper proteins content by 0.15–0.34% in the tubers in contrast with the ones of the control object. The statistical analysis also revealed the importance of the cultivar and weather conditions during the experimental years.

The aim of the study was to determine the effect of region, variety, year and ecological and conventional way of cultivation on the content of total protein and nitrates in potato tubers in three-year field trials. The effects of selected factors on nitrate and protein contents were similar, more significant for nitrates. Growing regions, varieties and years of cultivation caused statistically significant effects whereas the effect of ecological way of cultivation was less pronounced and showed a tendency without significance. Higher region showed lesser average nitrate (113.4 mg/kg) and protein content (4.27% dry matter) in comparison with lower region (143.7 mg/kg and 4.72% dry matter). The highest nitrate content was found in Impala and Karin varieties (199.2 and 167.6 mg/kg); Karin variety was also rich in total proteins (4.86% dry matter). Ecological way of cultivation showed no significant difference on nitrate content (136.9 mg/kg) and protein content (4.41%) as compared with conventional way of cultivation (153.9 mg/kg, 4.32%).

The research was carried out in 2018–2020 at the Agricultural Experimental Farm of the University of Natural Sciences and Humanities in Siedlce. The experiment was established on medium soil, in a split-plot design in three replications. The following factors were investigated: I – two medium-early varieties of potato were Malaga and Oberon, II – five ways of using herbicide and herbicide with biostimulators: control object – mechanical care, Avatar 293 ZC (clomazone + metribuzin), Avatar 293 ZC and PlonoStart, Avatar 293 ZC and Aminoplant as well as Avatar 293 ZC and Agro-Sorb Folium. The following factors were assessed: percentage of infected tubers, average degree of sample infection and average degree of tuber poison infection. The obtained results showed that the methods of using the herbicide and the herbicide with biostimulators significantly reduced the average degree of infection of potato tubers with rhizoctonosis compared to the tubers collected from the control object.

SummaryThe size, total protein, fibrinogen and 5-HT content were evaluated in density subpopulations of human and canine platelets fractionated in linear arabinogalactan gradients. The methodology was assessed to ascertain that platelet separation was by density and to discard artifactual changes and platelet release during the procedure. EDTA or PGEi increased the size of human PRP-platelets, but not of dog platelets. In humans, high density (HD) platelets were 1.26 times larger and contained 1.88 times more fibrinogen, 2.23 times more 5-HT and 1.37 times more protein than low density (LD) platelets; in dogs, these density cohorts did not differ in protein content, but LD platelets were 1.29 times larger and had 1.33 times more fibrinogen and 5-HT than HD platelets. These findings suggest that cell density is mostly dependent on the protein content per unit volume of platelets (and not on dense bodies). The differences in fibrinogen and 5-HT content between HD and LD cohorts in humans and dogs may be related to platelet age. The difference in volume between HD and LD platelets in dogs is of uncertain interpretation.

In our previously described Hep G2/24-hour/total protein assay, protein levels were measured by using the Lowry method. This assay was the best acute in vitro assay for the prediction of human toxicity within the Multicentre Evaluation of In Vitro Cytotoxicity (MEIC) study. In order to increase the MEIC data-base with a wider range of chemicals, we were interested in introducing the more practical 3-(4-carboxybenzoyl)-quinoline-2-carboxaldehyde (CBQCA) method for the quantification of the total protein content. Therefore, we investigated whether the same good results for the prediction of acute human toxicity would be obtained with the CBQCA method. The cells were treated for 24 hours, then cytotoxicity was determined by measuring the total protein content with CBQCA. The results were quantified by using the PI50c: the concentration (in mM) of test compound required to reduce the total protein content measured with the CBQCA-method by 50% as compared to the control cells. The results were compared with the PI50, the corresponding value when the Lowry method was used. A relatively low correlation was observed between PI50 and PI50c, reflecting the large and unexpected, differences when using the two protein assays. However, when comparing the log PI50c with the human toxicity, a correlation coefficient of r2 = 0.761 ( n = 44) was obtained for exactly the same series of MEIC chemicals. This value is clearly higher than that for the Lowry method ( r2 = 0.695). Compared to the Lowry method originally used, the Hep G2/24-hour/CBQCA total protein assay has the additional important advantage that it can be very easily adapted for large-scale analyses with robotic systems, including the on-line calculation of the results.

Hydroxyapatite (HA) has been used in orthopedic, dental, and maxillofacial surgery as a bone substitute. The aim of this investigation was to study the effect of surface topography produced by the presence of microporosity on cell response, evaluating: cell attachment, cell morphology, cell proliferation, total protein content, and alkaline phosphatase (ALP) activity. HA discs with different percentages of microporosity (< 5%, 15%, and 30%) were confected by means of the combination of uniaxial powder pressing and different sintering conditions. ROS17/2.8 cells were cultured on HA discs. For the evaluation of attachment, cells were cultured for two hours. Cell morphology was evaluated after seven days. After seven and fourteen days, cell proliferation, total protein content, and ALP activity were measured. Data were compared by means of ANOVA and Duncan’s multiple range test, when appropriate. Cell attachment (p = 0.11) and total protein content (p = 0.31) were not affected by surface topography. Proliferation after 7 and 14 days (p = 0.0007 and p = 0.003, respectively), and ALP activity (p = 0.0007) were both significantly decreased by the most irregular surface (HA30). These results suggest that initial cell events were not affected by surface topography, while surfaces with more regular topography, as those present in HA with 15% or less of microporosity, favored intermediary and final events such as cell proliferation and ALP activity.
