Tomato (Solanum lycopersicum L.) is one of the most important vegetable crops highly appreciated for the nutritional properties and content of beneficial compounds for human health. After its domestication, this crop has been spread throughout the world and found its secondary centre of diversification in the Mediterranean basin, where numerous landraces have been developed. Among these, ‘San Marzano’ is a famous and remunerative PDO variety for processing and fresh market consumption. ‘Re Fiascone’ is an ancient cultivar for which ongoing efforts aim to promote its diffusion and commercialisation. Both find their origin in the Campania region in Southern Italy where in the past decades, several accessions have been selected and handed down by farmers. This study reports a comprehensive assessment for morphological and biochemical traits of twenty-one accessions of the two landraces. Fruit morphology and content of sugars and flavonoids were the most discriminating parameters among cultivars. Among sugars and organic acids, fructose and citric acid were the most represented compounds, respectively. We found level of trans-lycopene and β-carotene up to 112.82 and 16.29 µg g−1 of fresh product, respectively, while ascorbic acid levels reached values up to 22.48 mg 100 g−1 of fresh weight. Molecular analysis has been performed using a double approach for microsatellite genotyping based on capillary electrophoresis and high-resolution melting. Results highlighted a separation of the accessions according to ‘Re Fiascone’ and ‘San Marzano’ identifying both unique and admixed accessions between the two groups. The study approach aims at the recovery and valorisation of local genetic resources, but also at the identification of traits of interest to transfer in breeding programs.

In order to estimate, at the molecular level, the divergence of parental lines that were used in diallel crossbreeding for production of superior offspring (F1 generation hybrids) at the Institute for Vegetable Crops, the molecular analysis using five RAPD markers for five pairs of parents has been performed. It gives an insight into their genetic polymorphism and the possibility of their further use in breeding programs. Information from this research has pioneered the application of molecular markers of onion in Serbia. Analyses were performed using the RAPD primers, which in previous studies established a high degree of polymorphism. In all five cases there was a corresponding amplification of DNA segments. From totally 50 bands analyzed, the length of fragments ranged from 500 to 3000 bp. Number of polymorphic band per example was 8 to 13. In our research at the level of the analyzed primers, a high degree of polymorphism between analyzed genotypes has been found. Based on UPGMA dendogram, analyzed genotypes were divided into two main clusters and two subclusters.

This study aimed to identification of an ecological alternative for the chemical disinfection of soil in the greenhouses from Romania. Tomato (Solanum lycopersicum L.) is one of the most popular vegetable crops in the world. The carbohydrate, vitamins, salts of important mineral elements and organic acids content of tomato fruits is very important. Tomato crops are very sensitive to climatic vagaries, so fluctuation in climatic parameters at any phase of growth can affect the yield and the fruit quality. Grafting on Solanaceae is a method which has improved and spread quickly during the past years, a similar approach to crop rotation, a practice meant to increase productivity, resistance or tolerance to biotic and abiotic stress factors and at increasing fruit quality. The research was conducted in a glass greenhouse of the Horting Institute, Bucharest, Romania. The biological material used was a Romanian tomato hybrid (Siriana F1), a Dutch tomato hybrid (Abellus F1) and four rootstocks, a Dutch tomato hybrid (Emperador F1) and three Romanian tomato cultivars (L542, L543 and L544) obtained from the Research and Development Station for Vegetable Growing, Buzău, Romania. The rootstocks have had resistance to biotic stress factors (soil diseases and pests) and the chemical disinfection of soil has was eliminated. The result of this research are presented in this paper.

AbstractLegionella pneumophila infects eukaryotic cells by forming a replicative organelle – the Legionella containing vacuole. During this process, the bacterial protein DrrA/SidM is secreted and manipulates the activity and post-translational modification (PTM) states of the vesicular trafficking regulator Rab1. As a result, Rab1 is modified with an adenosine monophosphate (AMP), and this process is referred to as AMPylation. Here, we use a chemical approach to stabilise low-affinity Rab:DrrA complexes in a site-specific manner to gain insight into the molecular basis of the interaction between the Rab protein and the AMPylation domain of DrrA. The crystal structure of the Rab:DrrA complex reveals a previously unknown non-conventional Rab-binding site (NC-RBS). Biochemical characterisation demonstrates allosteric stimulation of the AMPylation activity of DrrA via Rab binding to the NC-RBS. We speculate that allosteric control of DrrA could in principle prevent random and potentially cytotoxic AMPylation in the host, thereby perhaps ensuring efficient infection by Legionella.

Despite the remarkable interest in smart cities, noticed during the last decade, a consistent comprehension of the concept is not yet fully realized. Various definitions, ranging from exclusively technology-oriented perceptions to broader views, have been introduced, establishing a definitional polyphony and causing lack of semantic interoperability. Empirical evidence witnesses the prevalence of technology-pushed smart city initiatives as well as their failure to meet expectations in several urban domains. When planning “smart,” the relevance of ICT and their applications should be in alignment with spatial and other urban peculiarities and sub-systems' interactions, implying the need for getting deep insight into the city's ontology. The paper focuses on the extension/enrichment of an existing smart city ontology, with concepts and relationships stemming from Mediterranean small and medium-sized cities, in an attempt to outline their main key drivers and their interrelationships and fully grasp the smart city concept in the particular spatial context.

Given the recent proliferation in sales programs, business colleges face a new set of challenges. Sales competencies are changing rapidly, and firms struggle with identifying and attracting sales candidates on campus. Therefore, it is important that we understand needed competencies and how the content of job advertisements may differentially appeal to various student populations. To do so, we develop a conceptual model, based on signaling theory, that focuses on how students formulate their intention to pursue a given sales position. Our research utilizes a two-study approach. First, we explore the desired entry-level sales skills communicated by employers through job advertisements. Next, we examine both student and advertisement characteristics and their distinct relationships with the satisfaction with the job ad and the intention to apply for a sales position. Our study is unique, as we examine distinct undergraduate groups’ (sales, marketing, other business, and nonbusiness students) responses to sales job advertisements. Our findings demonstrate that differences in job ad clusters and student group characteristics influence the intention to pursue a sales position. Specifically, different student majors perceive job characteristics communicated within job ads differently. As such, our research provides insight into academic programs as well as corporate sales recruiters.

Corynespora cassiicola is a highly diverse fungal pathogen that can infect more than 500 species of plants, including many economically important crops such as cotton, soybean, tomato, and cucumber. In Florida, the number one vegetable crop by market value are fresh-market tomatoes, which generate nearly half a billion dollars annually. Florida’s subtropical to tropical climate is conducive to infection and development of the target spot pathogen on tomato caused by C. cassiicola. There is no varietal resistance available for target spot of tomato, and preventative fungicide treatments are the primary method for control. In the last decade, C. cassiicola has been more frequently reported by Florida tomato growers, appearing not only more aggressive but also increasingly insensitive to various fungicides. This review brings together the most recent C. cassiicola literature, providing a history and understanding of the immense pathogen diversity and its relevance to tomato. It also provides insight into fungicide resistance development and pathogen survivability, which are important factors in providing effective control recommendations and in understanding the epidemiology of this disease, respectively.

Olive (Olea europaea L.) is a species of great economic importance in the Mediterranean basin. Italy is very important for the olive industry; in fact, olive's genetic patrimony is very rich and characterized by an abundance of cultivars. At present, the majority of ancient landraces are vegetatively propagated by farm. It is likely that the number of cultivars is underestimated because of inadequate information on minor local cultivars that are widespread in different olive-growing areas. The existence of many cultivars reinforces the need for a reliable identification method. It is important to improve the ex situ plant germplasm collection and fairly to characterize all cultivars for future breeding programs. In the present report, we used 11 loci microsatellites to characterize 211 olive cultivars of an olive collection cultivated in six regions of southern Italy. These regions represent the major area for olive cultivation in Italy and have a strategic geographical location in the Mediterranean basin. The dendrogram obtained, using the unweighted pair group method with arithmetic mean clustering algorithm, depicts the pattern of relationships between the studied cultivars. There is a clear structuring of the variability relative to the geographic origin of olive cultivars. This work, for the very high number of the Italian olive cultivars analyzed, highlights the degree and distribution of genetic diversity of this species for better exploitation of olive resources and for the design of plant breeding programs. Besides, the use of molecular markers, like simple sequence repeats, is imperative to build a database for cultivar analysis, for traceability of processed food, and for appropriate management of olive germplasm collections.

In this article, the author reflects on the process of interviewing professional footballers about the sensitive issue of money and the lessons learned from this process. The article discusses a case study approach using in-depth qualitative interviews, which generates an innovative insight into a closed social world. The focus is on the difficulties of obtaining a sample where challenges faced are discussed. The article also focuses on the interview style employed when dealing with individuals who are experienced at being interviewed regularly. It also examines the issues of being an “insider” when conducting research and ways in which this can benefit the research process. The issue of research being a messy process is also discussed as when conducting this research, it was imperative that the author could deal with unpredictability and had to be flexible to conduct the research. Finally, the article discusses the insecurities of the participants and the issues of trust, as the current position of the author led him to be seen as an “outsider” by some participants.

Increased vegetable production for the fresh market often has been suggested as an agricultural alternative that will improve the income situation of small-scale farmers. Because vegetable production is an intensive activity and high incomes per acre are possible, it has political appeal as a quick solution to the low income levels generally associated with small farm operations. This study was developed from the small farm program of the University of Florida, which has concentrated on the northern and panhandle areas of Florida. Trials conducted by the Vegetable Crops Department of the University of Florida in 1974 and 1975 indicate that by variation of the planting season and control of insects and diseases, vegetables can be produced during periods of the year when the north Florida area has not historically competed in the fresh vegetable market.
