Blackberry fruits are recognized as functional foods while blackberry leaves are outside this classification and they also contain active compounds with health-promoting potential. Therefore, the aim of this study was the phytochemical analysis of blackberry leaves of varieties (Chester, Loch Ness, Loch Tay and Ruczaj) and screening of their biological activity (antioxidant potential, possibility of inhibition of enzymes, anti-inflammatory and microbial activity). The following compounds from selected groups: phenolic acids (caffeic acid, ellagic acid, gallic acid, syringic acid), flavonols (quercetin, kaempferol) and their glycosides (rutin, isoquercetin, hyperoside) and flavon-3-ols (catechin, epicatechin) were chromatographically determined in the aqueous and hydroalcoholic leaves extracts. All tested blackberry leaves extracts showed antioxidant effects, but the highest compounds content (TPC = 101.31 mg GAE/g) and antioxidant activity (e.g., DPPH IC50 = 57.37 μg/mL; ABTS IC50 = 24.83 μg/mL; CUPRAC IC50 = 62.73 μg/mL; FRAP IC50 = 39.99 μg/mL for hydroalcoholic extracts) was indicated for the Loch Tay variety. Blackberry leaf extracts’ anti-inflammatory effect was also exceptionally high for the Loch Tay variety (IC50 = 129.30 μg/mL), while leaves extracts of the Loch Ness variety showed a significant potential for microbial activity against Lactobacillus spp. and Candida spp. Summarizing, the best multidirectional pro-health effect was noted for leaves extracts of Loch Tay variety.

Oxidative stress and inflammation are interlinked processes. The aim of the study was to perform a phytochemical analysis and to evaluate the antioxidant and anti-inflammatory activities of ethanolic Mahonia aquifolium flower (MF), green fruit (MGF), and ripe fruit (MRF) extracts. Plant extract chemical composition was evaluated by HLPC. A DPPH test was used for the in vitro antioxidant activity. The in vivo antioxidant effects and the anti-inflammatory potential were tested on a rat turpentine oil-induced inflammation, by measuring serum nitric oxide (NOx) and TNF-alpha, total oxidative status (TOS), total antioxidant reactivity (TAR), oxidative stress index (OSI), 3-nitrothyrosine (3NT), malondialdehyde (MDA), and total thiols (SH). Extracts were administrated orally in three dilutions (100%, 50%, and 25%) for seven days prior to inflammation. The effects were compared to diclofenac. The HPLC polyphenol and alkaloid analysis revealed chlorogenic acid as the most abundant compound. All extracts had a good in vitro antioxidant activity, decreased NOx, TOS, and 3NT, and increased SH. TNF-alpha was reduced, and TAR increased only by MF and MGF. MDA was not influenced. Our findings suggest that M. aquifolium has anti-inflammatory and antioxidant effects that support the use in primary prevention of the inflammatory processes.

The herb <em>Acalypha indica </em>which belongs to Euphorbiaceae family has multiple medicinal properties which include anti-oxidant, anti-bacterial, anti-fungal, anti-inflammatory, antiulcer, anti-helmintic, anti-cancerous, anti-venom, and neuro-protective activity. The present study was designed to evaluate the phytochemical, antimicrobial and anti-oxidant activity of <em>Acalypha indica </em>leaves extracts in different solvent extractions like methanol, hexane, ethyl acetate and petroleum ether. Fresh leaves of the plant were collected and shade dried. Dried leaves were milled to obtain powder. Powder was subjected to soxhlet extraction using solvents and extracts were successively obtained. Phytochemical analysis was conducted following standard methods.  Phytochemical analysis showed the presence of Alkaloids, Phenols, Saponins, Flavanoids and Amino acids. Leaf extract of methonal have shown the highest anti-oxidation capacity than hexane, ethyl acetate and petroleum ether. Anti-microbial activity has been performed on microbes like <em>Bacillus sps</em>, <em>E.coli,</em> <em>Psuedomonas sps</em> and <em>Streptococcus sps.</em> A highest value of zone of inhibition was found in methanol extract against <em>E.coli.</em> These results provide evidence that <em>Acalypha indica </em>leaf extract possesses vital phytochemicals, antimicrobial and antioxidant properties. Hence this plant can be studied further for drug analysis for finding potent medicines for diseases.

Background: Inflammation is an immune response characterized by swelling, redness, pain and heat. Inflammation is main- ly managed using conventional medicines that are associated with many side effects. Plant-based remedies are considerably better alternative therapies for they have fewer side effects. Objective: This study aimed at determining the anti-inflammatory potential of dichloromethane (DCM) leaf extracts of Eucalyptus globulus and Senna didymobotrya in mice. Methods: Fresh leaves of these plants were harvested from Embu County, Kenya. Quantitative phytochemical analysis was done using Gas Chromatography-Mass Spectrometry (GC-MS). Anti-inflammatory test comprised nine groups of five animals each: normal, negative, positive controls and 6 experimental groups. Inflammation was induced with Carrageenan. One hour post-treatment, the different groups were intraperitoneally administered with the reference drug, diclofenac, 3% DMSO and six DCM leaf extracts at doses of 25, 50, 100, 150, 200 and 250mg/kgbw. Results: GC-MS results revealed α-phellandrene, camphene, terpinolene, and limonene among others. Anti-inflammatory effects showed that extract doses of 100,150,200 and 250mg/kg bw significantly reduced the inflamed paw. Doses of 200 and 250mg/kgbw in both plants were more potent and compared with diclofenac. E. globulus extract dose of 250mg kg bw reduced inflamed paw in the 1st , 2nd, 3rd and 4th hours, by 2.27,6.52,9.09 and 10.90% respectively while S.didymobotrya at similar dose ranges, inflamed paw reduced by 2.41, 5.43, 8.31 and 9.05% respectively. Conclusion: E. globulus and S. didymobotrya have potent anti-inflammatory activities, attributed to their constituent phyto- chemicals. Prostatitis is an inflammatory condition that is related to multiple infectious agents, including bacteria and fungi. Traditional herbal extracts proved efficacious in controlling clinical symptoms associated with prostatitis. In this context, the aim of the present study was to explore the efficacy of extracts from Solidago virga-aurea, Ononis spinosa, Peumus boldus, Epilobium angustifolium, and Phyllanthus niruri against bacterial (Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus cereus) and fungi strains (Candida albicans; C. tropicalis) involved in prostatitis. Additionally, anti-mycotic effects were tested against multiple species of dermatophytes (Trichophyton rubrum, T. tonsurans, T. erinacei, Arthroderma crocatum, A. quadrifidum, A. gypseum, A. currey, and A. insingulare). Antioxidant effects were also evaluated in isolated rat prostates challenged with lipopolysaccharide (LPS), and phytochemical analyses were conducted to identify and quantify selected phenolic compounds, in the extracts. Finally, a bioinformatics analysis was conducted to predict putative human and microbial enzymes targeted by extracts’ phytocompounds and underlying the observed bio-pharmacological effects. The phytochemical analysis highlighted that rutin levels could be crucial for explaining the highest antibacterial activity of P. boldus extract, especially against E. coli and B. cereus. On the other hand, in the E. angustifolium extract, catechin concentration could partially explain the highest efficacy of this extract in reducing lipid peroxidation, in isolated rat prostates stimulated with LPS. Concluding, the results of the present study showed moderate antimicrobial and anti-inflammatory effects induced by water extracts of S. virga-aurea, P. boldus, E. angustifolium, P. niruri, and O. spinosa that could be related, at least partially, to the phenolic composition of the phytocomplex.

Background: Tapinanthus globiferus (A. Rich) Teigh. (Loranthaceae) is an excellent medicinal plant in terms of efficacy and also traditionally used for the treatment of various diseases including inflammations, cancer, diabetes and strokes. This study was designed to assess the anti-inflammatory activity of the leaf extracts of T. globiferus in wistar rats and identify phytochemical constituents of the extracts responsible for the observed activity. Methods: Tapinanthus globiferus leaves was extracted with hexane, ethyl acetate and methanol in a soxhlet apparatus. The extracts were subjected to qualitative phytochemical analysis, toxicity and anti-inflammatory activity using carrageenan-induced paw oedema in wistar rats. Piroxicam (20 mg/kg) was used as reference standard. The data were analyzed by one-way analysis of variance with significant level set at p≤0.05. Results: The percentage yield from the gradient extraction of T. globiferus leaves showed methanol to be the highest and the chromatographic analysis visualized with specific reagents confirmed the presence of steroids/triterpenes, phenolic compounds and flavonoids in the leaf of T. globiferus. LD50 was above 2,000 mg/kg and no death was recorded. The hexane, ethyl acetate and methanol leaf extracts of T. globiferus at 250, 500 and 1,000 mg/kg produced a significant decrease in paw oedema (p≤0.05) with percentage inhibition at the first and third hour for hexane, ethyl acetate and methanol extract respectively. The methanol extracts recorded the highest inflammatory inhibition percentage. Conclusion: These finding revealed that the leaf of T. globiferus has anti-inflammatory activity and this justified its traditional use in the treatment of inflammation.

Objective: Plant-derived bioactive molecules are providing infinite opportunities for new drug development as they possess a wide range of actions against diseases with lesser side effects. The present study is made to analyze the qualitative phytochemicals and to evaluate in vitro antiproliferative and anti-inflammatory potentials of leaf extracts of Thespesia populnea. Methods: T. populnea leaf extracts were prepared sequentially according to the increasing polarity of the solvents, i.e., petroleum ether, chloroform, ethyl acetate, and methanol. Qualitative phytochemical analysis was performed to identify the chemical constituents of the extracts, and antiproliferative properties were evaluated against different cell lines using the 3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyltetrazolium bromide assay. Bovine serum albumin anti-denaturation assay was done to identify the anti-inflammatory activity.Results: Phytochemical analysis of the extracts revealed the presence of alkaloids, terpenoids, glycosides, tannins, flavonoids, and phenolics. The chloroform extract (CHFE) of T. populnea has a dose-dependent antiproliferative effect against acute T-cell leukemia (Jurkat E6-1), prostate Grade IV adenocarcinoma (PC-3), mouse fibroblast (L-929), and monkey kidney normal (Vero) cells. Their inhibitory concentration 50% (IC50) values were found to be 35.73±0.94 μg/ml for Jurkat E6-1, 60.79±1.84 μg/ml for PC-3, 60.88±1.45 μg/ml for L-929, and 83.482±2.05 μg/ml for Vero, respectively. CHFE also displayed the anti-inflammatory potential.Conclusion: The chloroform leaf extract of T. populnea possesses potent antiproliferative and anti-inflammatory activity. These properties present in leaf extract may be explained by the presence of biologically active constituents.

AbstractThe leaves of Piliostigma thonningii are used in traditional medicine in Benin to treat inflammatory skin diseases and infections. So far, pharmacological studies of the anti-inflammatory and anti-infective effects of phytochemically characterized extracts of P. thonningii have been very limited. Therefore, we investigated the in vitro anti-inflammatory and antimicrobial effect of P. thonningii leaf extracts and analyzed the phytochemical composition of extracts of different polarities (water, 50% ethanol, and n-hexane). Quercetin-3-O-rhamnoside was confirmed as the main flavonoid in the polar extracts. GC-MS analysis identified 20 constituents of the aqueous extract and 28 lipophilic compounds of the n-hexane extract by comparison with authentic standards and spectral library data. The aqueous P. thonningii leaf extract inhibited the IL-8 and IL-6 secretion in TNF-α-stimulated HaCaT cells in a concentration-dependent manner with IC50 values of 74 µg/mL for IL-8 and 89 µg/mL for IL-6. However, an inhibitory effect of the identified quercetin-3-O-rhamnoside and its aglycone, quercetin, on the release of IL-8 and IL-6 could not be demonstrated. In the antimicrobial screening, inhibition zones for a 50% EtOH leaf extract of P. thonningii were found for Staphylococcus epidermidis, Staphylococcus aureus, methicillin-resistant Staphylococcus aureus, and Escherichia coli. For none of the microbial strains, however, the MIC was below 500 µg/mL, so that the antibacterial activity must be classified as low. As a result, our investigations primarily support the ethnomedical use of P. thonningii leaf extracts in topical inflammatory conditions. Further studies are required to identify the compounds responsible for the in vitro anti-inflammatory effects.

Abstract Background Himalayan Columbine (Aquilegia pubiflora Wall. Ex Royle) is a medicinal plant and have been used as traditional treatments for various human diseases including skin burns, jaundice, hepatitis, wound healing, cardiovascular and circulatory diseases. Till now there is no report available on phytochemical investigation of Himalayan Columbine and to the best of our knowledge, through present study we have reported for the first time, the phytochemical analysis and pharmacological potentials of different leaf extracts of Aquilegia pubiflora. Methods Four types of extracts were prepared using solvent of different polarities (Distilled water APDW, Methanol APM, Ethanol APE and Ethyl acetate APEA), and were evaluated to determine the best candidate for potent bioactivity. Phytochemical constituents in prepared extracts were quantified through HPLC analysis. Subsequently, all four types of leaf extracts were then evaluated for their potential bioactivities including antimicrobial, protein kinase inhibition, anti-inflammatory, anti-diabetic, antioxidant, anti-Alzheimer, anti-aging and cytotoxic effect. Results HPLC analysis demonstrated the presence of dvitexin, isovitexin, orientin, isoorientin, ferulic acid, sinapic acid and chlorogenic acid in varied proportions in all plant extracts. Antimicrobial studies showed that, K. pneumonia was found to be most susceptible to inhibition zones of 11.2 ± 0.47, 13.9 ± 0.33, 12.7 ± 0.41, and 13.5 ± 0.62 measured at 5 mg/mL for APDW, APM, APE and APEA respectively. A. niger was the most susceptible strain in case of APDW with the highest zone of inhibition 14.3 ± 0.32, 13.2 ± 0.41 in case of APM, 13.7 ± 0.39 for APE while 15.4 ± 0.43 zone of inhibition was recorded in case of APEA at 5 mg/mL. The highest antioxidant activity of 92.6 ± 1.8 μgAAE/mg, 89.2 ± 2.4 μgAAE/mg, 277.5 ± 2.9 μM, 289.9 ± 1.74 μM for TAC, TRP, ABTS and FRAP, respectively, was shown by APE. APM, APE and APEA extracts showed a significant % cell inhibition (above 40%) against HepG2 cells. The highest anti-inflammatory of the samples was shown by APE (52.5 ± 1.1) against sPLA2, (41.2 ± 0.8) against 15-LOX, followed by (38.5 ± 1.5) and (32.4 ± 0.8) against COX-1 and COX-2, respectively. Conclusions Strong antimicrobial, Protein Kinase potency and considerable α-glucosidase, α-amylase, and cytotoxic potential were exhibited by plant samples. Significant anti-Alzheimer, anti-inflammatory, anti-aging, and kinase inhibitory potential of each plant sample thus aware us for further detailed research to determine novel drugs.
