Food allergies are a very serious problem among consumers. The most common food allergies involve animal products, but they can also involve fruits such as berries. We aimed to determine whether organic farming is useful for the production of high-quality and safe fruits. Three varieties of raspberries ‘Laszka’, ‘Glen Ample’ and ‘Polka’ from organic and conventional production (neighboring farms) were collected over the two years of the experiment. Quantitative and qualitative analysis of phenolic compounds was carried out, and the content of Bet v1 and profilin was determined. The organic raspberries contained a lower level of phenolic compounds, especially anthocyanins. Conventional fruits were characterized by a higher allergenic potency than organic ones. We found a strong link between their anthocyanin content and the allergy status of conventional raspberry fruits. Therefore, organically produced raspberries are safer for consumers.

Blackberry bushes have increasingly drawn producers’ and consumers’ interest since they find their cultivation characteristics, production and nutraceutical qualities advantageous. Both chemical and mineral characteristics, bioactive compounds and antioxidant activity found in blackberries are extremely relevant factors to be considered when cultivars are chosen. Chemical analyses of the nutritional composition of fruit is important, since they can help studies of quality genotypes. Besides, fruit with high nutritional values contribute to human health when they are often consumed. This study aimed at evaluating chemical and mineral characteristics, besides bioactive compounds and antioxidant activity, of different blackberry bush genotypes grown in an organic farming system in Pelotas, Rio Grande do Sul (RS), Brazil. Production, soluble solid content, hydrogenionic potential, total titratable acidity, SS/TA ratio, minerals, phenolic compounds, anthocyanins and antioxidant activity of six blackberry bush genotypes – ‘Tupy’, ‘BRS Xingu’, Selection Black 178, Selection Black 128, Selection Black 112 and Selection Black 145 – were evaluated. It was a randomized block design with three replicates of every plant. Selection Black 178 yielded fruits with the highest value of SS/TA ratio and had the highest accumulated production. The highest anthocyanin content was found in ‘BRS Xingu’. Selection Black 112 had the highest total contents of phenolic compounds and the highest antioxidant activity, whereas ‘Tupy’ exhibited their lowest values. Selection Black 178 adapted well to the organic farming system, a fact that did not happen to Selection Black 128.

AbstractBackgroundRNAs to be used in transcriptome analysis must be of high quality and pure in order to ensure maximum representation of the expressed genes. RNA isolation is difficult in hazelnut tissues containing large amounts of secondary metabolite, phenolic compounds and the cell wall structure. Commonly used protocols for RNA isolation are those that require a lot of labor and time and also do not allow sufficient RNA isolation when applied to tissues rich in phenolic compounds. This study was aimed to develop an efficient method for isolation of total RNAs from bud of hazelnut to be used in RNA sequencing.Materials and methodsAn optimized new method was successfully applied on three different hazelnuts genotypes (Çakıldak, Palaz, Tombul) and about 25 times higher amount of total RNAs per mg fresh tissues were obtained compared to classical CTAB method. Different methods have been tried for the isolation of RNA from hazelnut tissues and the determination of the quality of the obtained RNAs.ResultsThe quality and quantity of isolalated total RNAs were determined by spectrophotometer, electrophoresis and PCR. This success has been caught without any compromise of purity since A260/A280 ratios ranged from 1.90 to 2.04 and A260/A230 ratios were >2.0 in all purified RNAs.ConclusionThe total RNAs isolated with new protocol was found to be suitable for RNA sequencing and other molecular applications.

The current study was carried out to evaluate the accumulation of phenolic compounds in distinct developmental stages of Wheatgrass (Tritium aestivum). The wheatgrass was grown in the container/tray in the standard condition in the laboratory space provided by the department. Overnight soaked seeds were then sowed in the soil containers for 25 days. The accumulation of phenolics compounds was qualitatively and quantitatively checked in the leaves at 5, 10,15,20,25 days. Phenolics compounds were sequentially extracted in methanol, n-hexane, and acetone. For qualitative analysis, various biochemical tests were carried out and phenolics compounds were quantitatively estimated by Folin-Ciocalteu reagent. Among three solvents methanol extract was found to accumulate phenolics compounds in increasing concentration at various developmental stages i.e. 5th days - 0.7135 mg/ml, 10th days 1.2614 mg/ml, 15th days- 1.1244 mg/ml; 20th days - 1.8093 mg/ml and 25th days - 4.0693 mg/ml. On the qualitative check, methanol extract was found to be positive for tannins, terpenoids, and glycosides. These phenolics could be analyzed for antidiabetic and antimicrobial activities are the future perspective of this study.

Roselle contains high phenolic compounds, mainly anthocyanins that are not stable with pH, metal ions, light exposure, temperature, oxygen, and enzymatic activity. The stability of phenolic compounds can be improved by nanoencapsulation. This research was aimed to evaluate the effect of inulin, inulin-chitosan and inulin-chitosan-maltodextrin with varying concentrations as encapsulants towards the physicochemical properties and encapsulation efficiency of nanocapsules product by spray drying. Roselle extract nanocapsules were prepared using various types and concentrations of encapsulants (inulin, inulin-chitosan and inulin-chitosan-maltodextrin). The solubility of nanocapsules ranged from 69.31 - 83.2%, while the hygroscopicity of nanocapsules was varied, approximately 17.89 - 23.79%. Nanocapsules moisture content was approximately 2.83 - 4.27%, while the total phenolic content of nanocapsules ranged from 6.74 - 13.41 mg GAE/g DW. The total anthocyanin of roselle extract nanocapsules was approximately 2.25 - 4.82 mg/g DW. The encapsulation efficiency of phenolic compounds in this study were approximately 60.31 - 77.13%. Nanocapsules with inulin-chitosan-maltodextrin (2.4%-2.4%-0.2%) had good properties of nanocapsules such as good solubility, high total phenolic content and total anthocyanin content. Nanocapsules with 5% inulin and inulinchitosan-maltodextrin (2.4%-2.4%-0.2%) had particle size of 641.4 and 411.1 nm respectively. The nanocapsules had a spherical shape, smooth surfaces but also a few had indentations.

Commercial blueberry production has been a viable industry throughout the world for 95 years; because of blueberry is a good source of antioxidant. Blueberries are especially rich in anthocyanin, a flavonoid with potent antioxidant capacity. The aim of this study was to compare the phenolic quantities, antioxidant activities, anthocyanin, sugar and phenolic compounds of blueberries produced in Turkey with those of similar blueberry varieties produced around the world. As a result of the conducted analysis, the total phenolic content (TPC) amount found in the berries was 77.26-215.12 mg GAE/100 g, the total flavonoid content (TFC) was 30.44-91.69 mg QE/100 g and the total anthocyanin content (TAC) was 43.03-295.06 mg c3-GE/100 g. Examining the antioxidant activities of the berries, DPPH between 1.10-5.65 mg/ml, FRAP between 454.93-36832.96 µmol troloks/100 g, β-Carotene between 40.66-86.48%. It was determined that the natural berries contained much more phenolic compounds and higher antioxidant activity than that of the cultivars The result of HPLC analysis, chlorogenic acid is determined to be the dominant compound in all berries. Furthermore, fructose and glucose are found in all fruits in different quantities while sucrose is found in certain varieties of berries as well. At the end of the performed study the data indicate that wild and cultivars of blueberries are rich sources of antioxidants for local as well international industries importing this fruit for food processing and enormous products.

Aqueous extracts (20% w/w) of dried berry fruits and skins were used as sources of phenolic compounds to fortify yogurt beverages. The total phenol and anthocyanin content of the reformulated yogurts were determined, and the antioxidant properties were compared to plain yogurt (C) during storage at 4 °C for a total period of four weeks. Yogurt beverages fortified with salal berry (SB) extracts contained higher amounts of phenolic compounds (>69.9 μg GAE/mL) and anthocyanins (>19.12 mg C3G/L) compared to drinks supplemented with blackcurrant pomace (BC) extract (>50.13 μg GAE/mL and >10.80 mg C3G/L respectively). Storage affected the stability of anthocyanins, whereas total phenol content remained unaffected. Yogurts with SB displayed the highest antioxidant capacity followed by samples with BC, which is attributed to the radical scavenging effect of the bioactive compounds present with antioxidant properties. The antioxidant capacity of the yogurt beverages fortified with fruit extracts was maintained during cold storage. Findings of this study indicate that SB and BC pomace can be used as functional ingredients to increase the antioxidant potential of yogurt beverages.

Arbutus unedo L., commonly known as the strawberry-tree fruit, is an endemic species of the Mediterranean flora. Microwave extraction technology has been considered as a fast and “green” method for the production of extracts rich in bioactive compounds, although the energy consumption is high. To overcome this bottleneck, microwave was used as a pretreatment procedure in short time periods. This technique promotes the burst of intracellular vacuoles leading to an increase in the lixiviation of phenolic compounds. Different approaches were tested, namely a solvent-free irradiation (SFI), a solvent-assisted irradiation (SAI) and a pressurized solvent-assisted irradiation (PSAI). After irradiation, a solid–liquid extraction procedure was performed using a mixture of water and ethanol. A kinetic evaluation of the total phenolic content (TPC) was performed using the Folin–Ciocalteu method. For the total anthocyanin content, a UV-spectrophotometric method was used. HPLC-UV and LC-MS were used for TPC and identification of present compounds. Microwave irradiation led to an increase in TPC of extracts after SAI (52%) and PSAI (66%) along with a reduction in time of extraction from 30 min to less than 2 min. The anthocyanin content also increased by 66% for the SAI and PSAI extractions.

The present study was carried out to establish the antioxidant capacity of plant material of Cornus stolonifera (syn. Cornus sericea, Red Osier Dogwood) grown in Manitoba, by measuring total phenolic content and oxygen radical absorbance capacity (ORAC) over a 3-yr period. The presence and concentrations of flavonoids that might account for antioxidant abilities were investigated using ultra-performance liquid chromatography and a total anthocyanins assay. Both ORAC levels and phenolic content increased during the spring, peaked during the summer months (1631.65±166.30 µmol trolox equivalents g−1DW in July 2011, 220.38±2.29 mg gallic acid equivalents g−1DW in August 2012), and then declined during the fall in all 3 yr. Analysis of individual flavonoids in 2010 samples revealed that high levels of rutin, a glycoside of quercetin, were present ranging from 7.46±0.09 (July 2010) to 18.77±0.23 mg g−1dried sample (October 2010). Anthocyanin content was high in the spring, very low in the summer months, and increased in the fall over all 3 yr. Although polyphenolic and antioxidant content in Cornus spp. plants have been largely attributed to their berries, results from this study demonstrate that other plant material may also be an abundant source of these compounds.
