The number of consumers seeking safe and health-beneficial foods is systematically increasing. Consumers demand more environmentally friendly and safe food options coming from organic farming. Blackcurrants fruits are a perfect source of biological active compounds, such as vitamin C, polyphenols and flavonoids as well anthocyanins. Organic agricultural practices have modified the level of bioactive compounds in blackcurrant fruits. The novelty and the aim of the presented study was to compare the concentrations of selected health-promoting antioxidants in fruits of three blackcurrants (‘Ben Adler’, ‘Tiben’ and ‘Titania’) grown in organic and conventional orchards in Poland. For bioactive compound determination, the high-performance liquid chromatography (HPLC) method was used. The obtained results showed that organic blackcurrant fruits contained significantly more vitamin C (9.4 mg/100 g f.w.), total polyphenols (189.2 mg/100 g f.w.), total phenolic acids (10.3 mg/100 g f.w.), and total flavonoids (178.8 mg/100 g f.w.), as well anthocyanins (167.9 mg/100 g f.w.), compared to conventional ones. The selected cultivars and the years of cultivation system affect the concentration of the measured polyphenols, with strong interactions between these factors. The experiment confirms the hypothesis that organic blackcurrants should be a very important part of well-balanced, healthy diet.

Our research was aimed at assessing the effect of accumulation of carotenoids, polyphenols, vitamin C and ripening stage in the rosehip fruits of two species—Rosa canina, Rosa rugosa and two cultivar—Rosa rugosa ‘Rubra’ and Rosa rugosa ‘Alba’. The amounts of carotenoids, polyphenols and vitamin C were determined using the high-performance liquid chromatography (HPLC) method. The obtained results showed that the significantly highest amount (107.15 mg 100 g−1) of total carotenoid was determined in the fruits of Rosa canina at ripening Stage V. While results indicated that significant amount of total polyphenols were established at Stages I and II in the Rosa Rugosa ‘Alba’ and Rosa rugosa ‘Rubra’ cultivars (110.34 mg 100 g−1, 107.88 mg 100 g−1 and 103.20 mg 100 g−1 103.39 mg 100 g−1). At ripening Stage I, in the fruits of Rosa rugosa the greatest increases were established in the contents of vitamin C (3036.08 mg 100 g−1).

Broccoli is a source of bioactive compounds that provide an important nutritional value. The content of these compounds can vary depending on agronomic and environmental conditions, as well as on elicitation. In this study, three crop trials were carried out to evaluate the effects of the cultivation season, the application of different dosages of methyl-jasmonate (MeJA) on the overall quality and on the total content of bioactive compounds of ‘Parthenon’ broccoli cultivated under the field conditions of southeastern Spain. Color parameters, chlorophyll content, total phenolic compounds, total flavonoids and antioxidant activity were measured to evaluate the overall quality. Moreover, individual carotenoids, phenolic compounds and glucosinolates were evaluated by high performance liquid chromatography with diode array detection (HPLC-DAD) and high performance liquid chromatography equipped with diode array detector coupled to mass spectrometer using electro spray ionization (HPLC-DAD-ESI/MSn). The content of total carotenoids, phenolic compounds and glucosinolates were higher in autumn compared with spring, showing increases of 2.8-fold, 2-fold and 1.2-fold, respectively. Moreover, a double application of MeJA increased the contents of total carotenoids, phenolic compounds and glucosinolates by 22%, 32% and 39%, respectively, relative to the untreated samples. Considering our results, the controlled and timely application of 250 µM MeJA to the aerial parts of the plants four days before harvest, on two consecutive days, seems to be a valid agronomic strategy to improve the health-promoting capacity of Parthenon broccoli, without compromising its overall quality.

Crystal guava contains high vitamin C. Vitamin C is contained in different fruits, one of the factors is the altitude of the fruit plant growing areas. This study aims to determine the level of vitamin C in the slurry of crystal guava flesh with variations of origin using the HPLC method. Samples of crystal guava are obtained from the city of Bogor, Malang, and Gunung Kidul. The samples to be analyzed are prepared as a slurry. Qualitative analysis is done by identification using KMnO4 p.a, FeCl3 p.a, AgNO3 p.a as well as comparing the retention time of the sample with vitamin C. Method validation for system suitability, linearity and precision provide results that are eligible according to the applicable regulations. Quantitative analysis using HPLC with C18 stationary phase, water: methanol mobile phase (95: 5) v/v, flow rate of 1 mL/min, and run time of 7.5 minutes. Qualitative analysis by using KMnO4, FeCl3, AgNO3, and retention time (tR) shows positive results of the vitamin C existence. The average levels of vitamin C from Bogor, Malang, and Gunung Kidul, equal to (0.4139 ± 0.004) mg/mL; (0.6746 ± 0.03) mg/mL; and 0.8608 ± 0.002 mg/mL respectively.

Introduction. Recently, mass media published a number of articles doubting health promoting effect of manuka honey (MH). On the other hand, many researchers emphasize beneficial effect of honey on human digestive tract, respiratory system and skin. However, the honeys tested for their health promoting effects came from certified sources and met the applicable harvesting, storage and packing standards between the production and testing. Aim. The purpose of the study was to compare quality features of manuka honey (MH) imported to and sold in, Poland: methylglyoxal (MGO), 5-hydroxymethylfurfural (HMF) and diastase number (DN) declared for product lots by producers and distributors vs. the actual figures. Material and methods. The study tested 45 samples of MH coming from 9 producers, acquired from different local suppliers (importers and distributors), marketed in Poland in 2014-2015. The contents of MGO were measured using the high-performance liquid chromatography (HPLC) method. The contents of HMF were measured using high-performance liquid chromatography with spectrophotometric detection (HPLC-UV) in accordance with the regulation by the Polish Minister of Agriculture. DN were determined using the photometric method acc. To the regulation by the Polish Minister of Agriculture. Results. A review of the test results revealed a relationship between the declared and the actual contents of MGO. For the MH with MGO > 500, the actual values were different more frequently. For the MH with higher declared MGO contents (< 500), commercial quality of the products was worse (too high HMF and too low DN). Conclusions. The MH with MGO under 500 mg/kg can be recognized as satisfactory in terms of MGO contents and commercial quality.

We studied 19 different tropical fruits traditionally consumed in the coastal lowlands of Ecuador to determine their chemical composition and antioxidant activity. Carambola (Averrhoa carambola L.) had the highest total phenolic, flavonoid, and total antioxidant capacity values, whereas guava fruits (Psidium guajava L.) had the highest vitamin C values. The main organic acids identified were lactic, citric, and acetic acids, and the highest amount of lactic acid was found in soursop fruits (Annona muricata L.), whereas Ecuadorian ivory palm (Phytelephas aequatorialis Spruce) and guava fruits had the highest acetic acid content. Guava also had the highest citric acid content; the highest concentration of oxalic acid was found in carambola. In terms of sugar content, giant granadilla (Passiflora quadrangularis L.) had the highest values of glucose, and red mombin (Spondias mombin L.) had the largest values for fructose and guava for sucrose. Chili pepper (Capsicum chinense Jacq) proved to be the main source of carotenoids, lutein, and β-carotene, anthocyanins, and vitamin C. The results here increase our knowledge regarding the composition of the main fruits consumed on the west coast of Ecuador to facilitate recommendations as potential sources of health-promoting compounds.

Abstract The present investigation was to estimate the total phenolic content and composition of flavonoids and hydroxycinnamic acids in herbal infusions obtained from aerial parts of three Lamiaceae species (Dracocephalum moldavica, Ocimum americanum and Satureja hortensis). The total phenolic content of herbal infusions was determined using a spectrophotometric method, whereas the individual phenolics were assessed by high-performance liquid chromatography (HPLC). The HPLC method was developed and validated. The total phenolic content was measured by applying the Folin-Ciocalteu method with reference to gallic acid. Results were in the range from 29.39 to 65.38 mg estimated as gallic acid equivalents per gram of dry herb. The phenolic profile was, in turn, analysed by HPLC and consisted of gallic acid, hydroxycinnamic acids (caffeic, chlorogenic, ferulic, and rosmarinic) and flavonoids (rutin, hyperoside, quercitrin, quercetin, apigenin, apigenin-7-glucoside and catechin) in different concentrations. Rosmarinic acid was the predominant component among the hydroxycinnamic acids in herbal infusions of all three plants. This was found to be in the range of 3.64 to 5.28 mg per gram of dry herb. Apigenin-7-glucoside, quercitrin and hyperoside were the prevailing flavonoid components of the infusions.

A solid-liquid extraction and an HPLC method for determination of rebaudioside A from the leave parts of Stevia rebaudiana were developed. Separation method consisted of solvent extraction of leaf powder using various solvents like petroleum ether, methanol, diethyl ether and butanol followed by its purification using high performance liquid chromatography in order to obtain bioactive compound rebaudioside A. This solvent selection is very important prior to alternative extraction methods since it can be used as a pre-extraction solvents, main solvents, or co-solvents. The problem of hydrolysable components and solvent removal difficulties in the conventional extraction led us to study further the effects of solvent properties on the conventional extraction using Soxhlet method in order to determine the best solvent or solvent mixture for high extraction yield of S. rebaudiana. The chromatographic separation was realized using a C18 column, mobile phase consisting of methanol: water with UV detection at 210 nm. Based on the yield of extraction and glycosides content, methanol was found to be best solvent.

Accumulating scientific evidence suggests that medicinal plant-associated microbiota has a significant role in biosynthesis of pharmacologically relevant compounds. Suppression of S. nigra microbiota by an antibiotic treatment leads to a decreased emission of volatile terpenoids from S. nigra flowers and an altered metabolome of a host plant. However, it is not known if suppression of microbiota also affects an abundance of health promoting compounds and pharmacologically relevant properties of S. nigra. The present study reveals, that suppression of phyllospheric microbiota by an antibiotic treatment affects total phenolic content (TPC), anti-radical activity (ARA) and total flavonol content (Flav) of wild S. nigra flowers. Significance analysis of metabolites (SAM) performed using ultra high performance liquid chromatography-time of flight-masspectrometry (UHPLC-TOF-MS) based metabolomics data identifies pharmacologically relevant flavonols as metabolites discriminating between antibiotic treated and control flowers of S. nigra. Reported results substantiate need for a research investigating an effect of microbiota suppression on a biological activity of S. nigra flowers.

The apple (Malus domestica Borkh.) is one of the most widely cultivated temperate fruits globally, gaining scientific interest as a rich source of antioxidants with a demonstrated beneficial human health impact. Since a growing number of consumers are increasingly seeking safe and healthy food options, alternative fruit production systems such as organic farming, and their potential to provide safe and nutritious foods, have been gaining increasing attention in the last decades. The aim of the presented study was, therefore, to analyse and to compare the concentrations of selected health-promoting antioxidants, such as phenolic acids, flavonols, and vitamin C, in fruits of three apple cultivars (Champion, Gala, and Idared) grown in conventional and certified organic orchards in Poland. All analyses were performed using the high-performance liquid chromatography (HPLC) method. Organic apples tested within the study, compared to the conventionally grown ones, were characterised by significantly higher concentrations of phenolic acids (av. >31%) and flavonols (av. >66%) with the identified differences being consistent in all three cultivars and two seasons. The greatest production system effect was observed in the case of Idared. Significant cultivar and production season effects on the concentration of the measured fruit phenolics, with strong interactions between the two factors, were also identified. Vitamin C content in the fruits was strongly dependent on the year-to-year differences in the fruit growing conditions. The study suggests that the organic production system has a potential to provide apple fruits rich in selected health-promoting phenolic antioxidants.

Objective: To determine total phenolic and flavonoids contents and also quantify vindoline and rutin in different morphotypes of Catharanthus roseus using High-performance liquid chromatography (HPLC) method. Methods: Total flavonoids content (TFC) was determined by Aluminium chloride colorimetric and total phenolic content (TPC) was estimated by Folin-Ciocalteu reagent assay. The chromatographic separation was done by using a C18 column at room temperature and eluted with a mobile phase consisting of a mixture of phosphate buffer (pH=5.8) and acetonitrile at a flow rate of 1.0 ml/minute and detection was carried out at 254 nm. Results: TPC and TFC content was found highest in Cr00DP and lowest in Cr00WFSRE. Results also showed that the purple morphotypes Cr00DP gives more vindoline (0.3 mg/g) and rutin (18.57 mg/g) concentration compared to the pink morphotype Cr00PFRE contained 18.3 mg/g rutin and 0.2 mg/g vindoline. White morphotypes contained 0.383 mg/g rutin and 0.004 mg/g vindoline which was significantly less as compared to purple and pink morphotypes. Conclusion: The plant has significant number of alkaloids and flavonoids. The obtained outcomes from different morphotypes are thus significant for the purpose of vindoline and rutin isolation from Catharanthus roseus plant. These isolated bioactive phytoconstituents are a good candidate for further pharmacological and clinical study.
