To guide the cultivation of superior Chinese fir plantlets, we designed an L16(4)4 orthogonal experiment to determine how leaf age and exogenous hormones influence key growth processes in leaf cuttings. Hormone concentration and treatment duration significantly affected leaf cuttings in all three age categories; 6-benzylaminopurine (6-BA), 1-naphthaleneacetic acid (NAA), and treatment time exerted the strongest effects on callus initiation rates. Additionally, NAA had the largest effect on the rooting rate across all cuttings, and all three hormones significantly influenced the bud germination rate. Based on our experimental results, expected optimal treatments for callus initiation were 10 mg∙L−1 indole-3-butyric acid (IBA) for 10 min, 30 mg∙L−1 NAA for 15 min, and 10 mg∙L−1 NAA plus 30 mg∙L−1 IBA for 10 min. For the rooting rate, the expected optimal treatment was 50 mg∙L−1 NAA and 40 mg∙L−1 IBA for 5–20 min. Finally, for bud germination, optimal treatments were 20 min of immersion in water, 30 mg∙L−1 6-BA plus 50 mg∙L−1 NAA for 15 min, and 30 mg∙L−1 6-BA for 5 min. Plantlet formation only occurred in the <one-year-old leaves, and at very low rates (maximum 5.8%); this outcome is likely attributable to the mother plant’s relatively old age (five years). Plantlet formation from cuttings is dependent on ensuring the rooting rate after callus initiation. Therefore, to promote rooting rates and bud germination, we recommend leaving more xylem at the base of leaf cuttings.

AbstractIn order to improve dimensional stability and durability of wood, furfurylation of poplar and Chinese fir wood using newly developed furfuryl alcohol (FA) formulation combined with a common vacuum and pressure impregnation process was studied. An orthogonal experiment was designed to optimize the furfurylation process for the two wood species. The weight percent gain (WPG), equilibrium moisture content (EMC), anti-swelling efficiency (ASE), modulus of rupture (MOR), modulus of elasticity (MOE), as well as resistance to mold, decay fungi, and termites were evaluated. The results showed that nearly all the properties of the furfurylated wood could be improved to various extents. The average ASE of the furfurylated Chinese fir and poplar could reach as high as 80, 71, 92% and 79, 90, 75% in tangential and radial directions, and by volume, respectively, higher than most previously reported wood modification processes. Furthermore, the modified wood had excellent biological durability, with nearly 100% mold resistance, strong decay and termite resistance. Finally, processing parameters with 50% FA, 105–115 °C curing temperature, and 5–8 h curing time were therefore recommended for pilot-scale production of furfurylated poplar and Chinese fir wood based on range analysis.

AbstractAlthough seed dormancy of temperate legumes is well understood, less is known about it in species that grow in subalpine/alpine areas. This study investigated dormancy and germination of four Vicia species from the Tibetan Plateau. Fresh seeds of V. sativa were permeable to water, whereas those of V. angustifolia, V. amoena and V. unijuga had physical dormancy (PY). One year of dry storage increased the proportion of impermeable seeds in V. angustifolia, but showed no effect on seed coat permeability in V. amoena or V. unijuga. Seeds of all four species also had non-deep physiological dormancy (PD), which was especially apparent in the two annuals at a high germination temperature (20°C). After 1 year of storage, PD had been lost. The hydrotime model showed that fresh seeds obtained a significantly higher median water potential [Ψb(50)] than stored seeds, implying that PD prevents germination in winter for seeds dispersed without PY when water availability is limited. After 6 months on the soil surface in the field, a high proportion of permeable seeds remained ungerminated, further suggesting that PD plays a key role in preventing germination after dispersal. Addition of fluridone, an inhibitor of abscisic acid (ABA) biosynthesis, evened-out the differences in germination between fresh and stored seeds, which points to the key role of ABA biosynthesis in maintaining dormancy. Further, fresh seeds were more sensitive to exogenous ABA than stored seeds, indicating that storage decreased embryo sensitivity to ABA. On the other hand, the gibberellic acid GA3 increased germination rate, which implies that embryo sensitivity to GA is also involved in seed dormancy regulation. This study showed that PY, PD or their combination (PY+PD) plays a key role in timing germination after dispersal, and that different intensities of dormancy occur among these four Vicia species from the Tibetan Plateau.

A retrospective study on 15 non-vital immature incisor teeth was done using Ca(OH)2 Pulpdent® paste. A success rate of 100 percent was achieved within one year. The variables influencing the time taken for apexification were also evaluated. The teeth were followed up to a period of 24 months. It was found that older children having narrow open apex had a shorter treatment time than the younger children (NS); teeth without periapical infection showed some amount of root growth and closing of apex that was faster than those with periapical infection (p&lt;0.001). The calcified bridge formed following apexification is a porous structure. This investigation provides information about the time taken and procedure required to achieve apical barrier formation in non-vital immature incisors.

To meet the requirement for high transparency of agar gel used in tissue culture, bacteriological applications, and high-quality cosmetics and food, this study on improving the transparency of agar products was carried out by using activated carbon for decolorization and perlite for aiding filtration. The results showed that the pre-coating filtration method was better than that of the mixed slurry filtration, so an orthogonal experiment optimization was conducted using the pre-coating filtration method based on the single factor experimental results. The experimental results showed that the optimal process conditions based on the transparency index were: concentration of agar of 1.30%, activated carbon dosage of 0.80%, processing temperature of 95 °C, and treatment time of 20 min. Agar products with gel transparency of 58.93% and blue-ray whiteness value of 93.82 could be obtained under these experimental conditions. The order of the factors influencing the decolorization ratio of agar was as follows: activated carbon dosage > concentration of agar > processing temperature > treatment time. The experimental results provided effective procession methods for the production enterprises based on transparency and increased economic benefits, and it was of practical significance.

Investigations into the germination behaviour of seed of the widespread Australian arid zone leguminous tree Acacia aneura Benth. are described. Hardseededness has been overcome in routine laboratory studies, but attempts to understand the mechanism by which it is overcome in nature were unsuccessful. The seed germinates equally well in the dark and in the light within the temperature range 15-36°C. Temperatures between 20 and 30° are optimal for germination, and carbon dioxide concentrations up to 5 % increase the germination rate above that displayed in ambient air. A simple model has been developed which suggests that seed of this species should have been available and should have germinated in c. one year in every nine, since records began in parts of western New South Wales where mulga regeneration is absent.

The effect of four whole-tree sprays at monthly intervals on the fruit mineral element content and on the incidence of storage disorders in Merton apples was investigated for three years. Calcium nitrate, boric acid, zinc nitrate and naphthaleneacetic acid (NAA) were applied singly and in all possible combinations. There were no significant treatment interactions, which implied an absence of synergistic or antagonistic effects between spray components. Calcium nitrate was the only compound whose application consistently reduced the incidence of bitter pit and breakdown following cool storage for 4 months. In one year NAA reduced the incidence of these disorders, but it also reduced mean fruit size and increased the concentration of most of the mineral elements, including calcium, in the fruit at harvest. Zinc nitrate sprays had no effect. Boron application promoted the development of core flush in one year and of breakdown in two years, but did not affect the incidence of bitter pit. No relationship was observed between the levels of calcium and boron in the fruit.

Callus cultures of Juniperus virginiana L. (varieties ‘Hetzii’, ‘Glauca’, ‘Grey Owl’) were derived from fresh leaves of garden-grown trees on Schenk and Hildebrandt medium supplemented with 3.0 mg/L of α-naphthaleneacetic acid, 0.2 mg/L of kinetin and 15 mg/L of ascorbic acid. The growth characteristics of one-year-old and two-years-old cultures were determined. The maximum biomass in all varieties was achieved on the 35th day of the cultivation period. The increase in fresh weights of two-years-old callus cultures, when compared with one-year-old callus cultures, was as follows: variety ‘Hetzii’ by 25%, variety ‘Glauca’ by 29% and variety ‘Grey Owl’ by 49%. J. virginiana suspension cultures (varieties ‘Hetzii’, ‘Glauca’, ‘Grey Owl’) were derived from two-years-old callus cultures on Schenk and Hildebrandt medium supplemented with 3.0 mg/L of α-naphthaleneacetic acid, 0.2 mg/L of kinetin and 15 mg/L of ascorbic acid. The maximum biomass of all varieties was found on the 21st day of the cultivation period. These results indicate that a sub-cultivation interval of 35 days for callus cultures and of 21 days for suspension cultures can be recommended. The callus and suspension cultures of J. virginiana of the variety ‘Glauca’ have the best survivability and thus provide the most biomass.

To enhance decay resistance, the effect of heat treatment and the variation of chemical composition on Chinese Fir and Pine were investigated in this study — heat treatment temperature was 170°C, 190°C and 210°C, respectively, heat treatment time was 2, 3 and 4hours, respectively. Both of them were subsequently exposed to white-rot fungus and brown-rot fungus. The results showed that:(1) With the increasing of the heat treatment, decay resistance of Chinese Fir and Pine were improved, anti-corrosion of Pine after being heat treated at 190°C which were exposed to write-rot fungus can reach I, anti-corrosion of Chinese Fir after being heat treated at 170°C treated which were exposed to brown-rot fungus could reach I yet, After being heat treated at 210°C for 3 hours , the Chinese fir samples had no measurable weight loss when exposed to the write-rot fungus.(2) There was no remarkable influence on both Chinese Fir and Pine by heat treatment time.(3) The moisture content of Chinese Fir and Pine were lower than the moisture content that the rot fungus need, macromolecule chains such as cellulose and hemicellulose broke down, their contents decreased, and the hemicellulose decomposed into acetic acid, they prevented the growth of rot fungus.

Abstract The high valuable endemic commodities in Papua, Masoyi’s (Cryptocarya massoy) population facing great threat due to unsustainable harvest system. Generative propagation faces significant challenges due to seed characteristics and habitat conditions. Controlled conditions and the role of hormones have an important effect on generative growth. This study aimed to determine the influence of axenic culture with sterilization treatments Isothiazolone Biocide (IB) and 1-Naphtaleaneacetic Acid (NAA) in Murashige and Skoog (MS) medium on seed regeneration and to observe the development of seedlings at the acclimatization stage. The tissue culture method was used. The highest percentage of axenic cultures (57%) was obtained with 5% of BI. The germination rate of masoyi seeds was achieved by 100%. Furthermore, it showed varied responses depending upon concentrations of NAA, the addition of 1 ml l−1 NAA in MS medium is recommended. Acclimatization has been successfully carried out in the greenhouse (67% survival rate) and excellent seedlings growth at nursery (52.35 + 0.6 cm in height after one year transferred). The impact of the controlled conditions and the addition of NAA to axenic cultures in vitro increased the germination of masoyi seeds. Axenic culture and hormones were also important requirements for mass propagation of masoyi by tissue culture.
