Variation in cultivars can influence plant biological activities. This study aimed to identify superior cultivars while determining the variability in the phytochemical content, antioxidant, alpha-glucosidase inhibitory and antibacterial activities of cladode extracts from selected spineless Burbank cactus pear (Opuntia ficus-indica and Opuntia robusta) cultivars. Total phenolic and flavonoid contents were determined using the Folin-Ciocalteu and aluminum chloride spectrophotometric methods, respectively. Antioxidant activity was investigated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging and β-carotene linoleic acid assays. Alpha-glucosidase inhibition was determined using a spectrophotometric method and antibacterial activity using a non-polar (petroleum ether) and polar (50% methanol) extracts against two Gram-positive and two Gram-negative bacteria. Significant variation in phytochemical content, antioxidant, antidiabetic and antibacterial activities was observed amongst the cultivars. Alpha-glucosidase inhibitory activity varied widely with IC50 values ranging from 0.06 to 1.85 mg/mL. Radical scavenging activity of Polypoly cultivar was about seven fold higher than that recorded in other cultivars with low activity. Turpin and Berg x Mexican cultivars had the highest total phenolic and flavonoid contents, whilst the non-polar extract of Turpin also exhibited higher antibacterial activity against Bacillus subtilis and Escherichia coli. Sicilian Indian Fig was amongst the cultivars with a higher antioxidant activity, whilst also showing a strong inhibition against B. subtilis and E. coli. Polypoly cultivar demonstrated strong antioxidant and antidiabetic activities while its polar extract showed the highest total antibacterial activity against B. subtilis. The cultivar Malta was superior in terms of its antibacterial potency and efficacy against B. subtilis, Staphylococcus aureus and E. coli. The potential of using spineless cactus pear cladodes as a functional food with antioxidant, antidiabetic and antibacterial properties against pathogenic food spoilage bacteria in place of synthetic compounds was established. The significance of cultivar selection to increase this potential was highlighted.

Tujuan penelitian ini adalah untuk mempelajari karakteristik kimia tepung sagu baruk yang mengandung senyawa fitokimia dan mempelajari aktivitas antioksidan dari ekstrak tepung sagu baruk. Tepung sagu baruk diekstrak dengan pelarut etanol 50% dengan cara maserasi selama 24 jam. Setelah itu, tepung sagu diuji  proksimat, ekstrak dianalisis kandungan fitokimia fenolik, flavonoid dan tanin terkondensasi. Pengujian aktivitas antioksidan menggunakan radikal bebas DPPH dan daya reduksi dengan spektrofotometer UV/Vis. Hasil penelitian menunjukkan pada pengujian proksimat bahwa tepung tepung sagu baruk memenuhi syarat Standar Nasional Indonesia, tetapi berbeda dengan kadar serat kasar. Total kandungan fenolik dan flavonoid  menunjukkan bahwa tepung sagu akuades (SA) lebih tinggi daripada tepung sagu komersial (TSBK). Hasil radikal bebas sagu tepung sagu SA lebih tinggi dari TSBK sama halnya dengan kemampuan daya reduksi SA jauh lebih tinggi dari TSBK. Hasil penelitian ini menyimpulkan bahwa tepung sagu baruk memiliki kandungan fitokimia fenolik dan flavonoid yang dapat berpotensi sebagai antioksidan.Objectives of this research were to study the chemical characteristics of sago baruk flour containing phytochemicals and antioxidant activity of sago baruk flour extract. Sago baruk flour was extracted with 50% ethanol by maceration for 24 hours. Afterward, sago flour was tested for proximate and the extract was analyzed for phenolic, flavonoids, and condensed tanin. DPPH free radical was used to examine antioxidant activity and spectrophotometry method was used to determine the reduction ability. The result for proximate test showed that sago flour extract meets Indonesian Nasional Standard requirements. Total phenolic and flavonoid contents in the flour extracted with aquadest (SA) were higher than that in commercial flour (TSBK). Free radical scavenging activity of sago flour SA was higher than that of TSBK, as well as the reduction ability of SA that was much higher than of TSBK. It was concluded that sago baruk flour possess phytochemical content of phenolic and flavonoid which were potential as antioxidant.

Sorghum is a major cereal food worldwide, and is considered a potential source of minerals and bioactive compounds. Its wide adaptive range may cause variations in its agronomic traits, antioxidant properties, and phytochemical content. This extensive study investigated variations in seed characteristics, antioxidant properties, and total phenolic (TPC) and flavonoid contents (TFC) of sorghum collected from different ecological regions of 15 countries. The antioxidant potential of the seed extracts of various sorghum accessions was determined using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2′-azinobis 3-ethylbenzothiazoline-6-sulfonate (ABTS) radical scavenging assays. Significant variations in TPC were observed among the sorghum accessions. All 78 sorghum accessions used in this study exhibited significant variations in TFC, with the lowest and highest amount observed in accessions C465 and J542, respectively. DPPH scavenging potential of the seed extracts for all the accessions ranged from 11.91 ± 4.83 to 1343.90 ± 81.02 µg mL−1. The ABTS assay results were similar to those of DPPH but showed some differences in the accessions. Pearson’s correlation analysis revealed a wide variation range in the correlation between antioxidant activity and TPC, as well as TFC, among the sorghum accessions. A wide diversity range was also recorded for the seed characteristics (1000-seed weight and seed germination rate). A dendrogram generated from UPGMA clustering, based on seed traits, antioxidant activity, TPC, and TFC was highly dispersed for these accessions. Variations among the accessions may provide useful information regarding the phytoconstituents, antioxidant properties, and phytochemical contents of sorghum and aid in designing breeding programs to obtain sorghum with improved agronomic traits and bioactive properties.

  Objective: The objective of this investigation was to assess the biological activity of fresh fruit juices and freeze dried fruit juices including antioxidant and antibacterial activity.Methods: The fruits used in this study were Schleichera oleosa (Lour.) Oken, Carissa carandas L., and Sandoricum koetjape (Burm.f.) Merr. The fresh juice sample (FJS) was extracted and also prepared for freeze drying sample (FDS) by freeze dryer. The antioxidant capacity of FJS and FDS was assessed using the 2,2-diphenyl-1-picryl-hydrazyl (DPPH) assays and determination for total phenolic contents (TPCs) by Folin–Ciocalteu’s reagent. Antimicrobial tests were carried out by agar diffusion methods and evaluated by measuring the zone of inhibition against 10 isolates of pathogenic bacteria.Results: The highest antioxidant activity of FJS and FDS was found in S. oleosa and related to the scavenging effect on DPPH radical of FJS (100% concentration) and FDS (200 mg/ml) was found in the value of 93.05±0.31% and 82.18±0.33%, respectively. TPC of FJS and FDS from S. oleosa at the same concentration as in the DPPH assay was 1,003.53±1.96 ug GAE/300 g of matter and 827.77±23.15 ug GAE/100 ml of matter, respectively. Both FJS and FDS revealed antibacterial activity. The zone of inhibition of FJS was ranging from 8.3±0.6 to 24.0±1.0 mm and FDS values of 7.3±0.5-21.0±0.9 mm. The most potent sample against bacteria was FJS and FDS from S. oleosa which FJSs against Escherichia coli ESBL and FDS against Pseudomonas aeruginosa MDR2.Conclusion: Fruit juices of S. oleosa, C. carandas, and S. koetjape associated with antioxidant activity in the form of both fresh fruit juices and FDS. The result of this study showed that the value of DPPH and TPCs were slightly decreased in FDS compare with fresh juice. Screening of FJS and FDS of C. carandas revealed broad spectrum antibacterial activity. However, FJS of S. oleosa and S. koetjape showed inhibition of growth of bacteria, but few of FDS from these fruits could inhibit limited bacterial isolates. For antibacterial activity against pathogenic bacteria, the samples of fresh and freeze dried exhibited a different degree of activity.

The present study examined the effect of Lactobacillus plantarum ATCC14917 fermentation on the chemical composition and antioxidant activity of apple juice. Apple juice was fermented and examined of its antioxidant activity using chemical models and cellular antioxidant assay. Furthermore, the chemical composition of fermented apple juice was characterized by LC-MS/MS. Lactobacillus plantarum ATCC14917 fermentation showed an increase in DPPH and ABTS radical scavenging activity as well as cellular antioxidant activity of apple juice. However, fermentation decreased the total phenolic and flavonoid content. Subsequent LC-MS/MS analysis of the phenolic profile indicated that the content of 5-O-caffeoylquinic acid (5-CQA), quercetin, and phloretin with strong antioxidant activity was increased significantly after fermentation. The modified phenolic composition may contribute to the increased antioxidant activity of fermented apple juice. Our findings showed that Lactobacillus plantarum ATCC14917 fermentation may be an efficient way to enhance the bioavailability of phenolic compounds and to protect cells from oxidative stress.

The roots of Salvia miltiorrhiza are known to exhibit antioxidant and antibacterial activities. To improve the antioxidant and antibacterial activities of S. miltiorrhiza roots, the roots were fermented with Aspergillus oryzae at 25 °C for 3 weeks. The non-fermented (SME) and fermented (SMBE) roots of S. miltiorrhiza were extracted with 70% ethanol, respectively, and then fractionated with organic solvents. By fermentation, total phenolic and flavonoid contents, as well as antioxidant activity of SMBE, were increased by about 1.2 to 1.3 times compared with those of SME. The antibacterial activity of SMBE was also twice as high as that of SME. The antibacterial activity of SMBE against Bacillus cereus was lower in the n-hexane and chloroform fractions, but higher in the ethyl acetate and n-butanol fractions, compared with those of SME. These results indicate that the bioactive components of S. miltiorrhiza roots exhibiting antibacterial activity were converted to more polar compounds by fermentation of A. oryzae. Gas chromatography and mass spectrometry (GC-MS) and LC-MS analyses of SME and SMBE demonstrate that these changes are due to the acylation of dihydrofuran-2(3H)-one, dealkylation of 4-methylbenzene-1,2-diol and 4-ethylbenzene-1,2-diol, and esterification of hexadecanoic acid and (9Z, 12Z)-octadec-9,12-dienoic acid during fermentation.

Pterygota alata is a large deciduous tree of Malvaceae family. The present study was designed to evaluate in vitro antioxidant activity of stem bark of the plant. Ferric reducing power (FRP) test, 1, 1- diphenyl-2-picryl hydrazyl (DPPH) free radical scavenging test and total antioxidant capacity (TAC) assay were used to detect the antioxidant activity. Total phenolic and flavonoid contents were also determined as they are well known phytochemicals with antioxidant property. The bark of the plant showed efficient reducing power as well as free radical scavenging property (IC50 values 52.25 - 172.05). The bark also found rich in total phenolic and flavonoid content. The highest amount of total phenolic content was found in chloroform soluble fraction (29.898 µg/mL) followed by ethyl acetate soluble fraction (15.88 µg/mL). The highest content of total flavonoid also detected in chloroform soluble fraction (107.56 µg/mL) but followed by crude ethanol extract (98.66 µg/mL). Overall, the bark of the plant possesses significant antioxidant activity, therefore can be used as a good natural source of antioxidant. © 2014 JSR Publications. In vitro antibacterial activity of 21 plant methanolic extracts were investigated by disc diffusion method against gram-negative bacteria Escherichia coli. Amongst the extracts tested, the Datura stramonium L. and Ocimum basilicum L. showed significant antibacterial activity against the bacterial pathogen. D. stramonium showed the highest antibacterial activity followed by O. basilicum extract. Phytochemical analysis of extracts showed the presence of alkaloids, tannins, saponins, flavonoids, and phenols. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and ABTS analysis of potent extract D. stramonium showed appreciable antioxidant activity. For DPPH and ABTS assay the highest antioxidant activity was observed at 60 mg ml-1 concentration with a percent inhibition of 71.66 and 77.19 with IC50 value 9.71 and 22.16 μg ml-1 respectively. Moreover, the radical scavenging activity of the extract was lower than that observed for the synthetic antioxidants butylated hydroxyanisole (BHA )and butylated hydroxytoluene (BHT). The total phenolic content of D. stramonium extract was found to be 0.592 mg GAE/gm which was assayed using Folin ciocalteau reagent. The results provide evidence that the extract of D. stramonium and O. basilicum can be further recommended in the treatment of the infections caused by E. coli; further D. stramonium is also a potential source of natural antioxidants.

Purpose: Milk thistle (Silybum marianum (L.) Gaertn) has been used for centuries as a natural remedy for liver and gall bladder / biliary tract diseases. Silymarin is the purified and standardized extract from milk thistle seeds. The purpose of the present study is to present a comparative analysis of the total phenolic substances content and the antioxidant effect of silymarin and three different types of Silybum marianum extracts (i.e. ethanol extract, water extract and lyophilized extract). Methods: Total phenolic and flavonoid content of extracts was analysed and the antioxidant activity was assessed by determining their free radical scavenging ability by DPPH method. Results: Total phenolic content is the highest in silymarin– 134.12±3.22 mg GAE/g, followed by lyophilized water extract – 51.74±2.10 mg GAE/g. Silymarin has also been found to have the highest antioxidant activity among all extract samples (338.03 VCEAC µmol /g), followed by S.marianum lyophilised extract (123.05 VCEAC µmol/g). Conclusions: A strong correlation between total phenolic and flavonoid content in the extracts and their antioxidant activity has been found.

Dryopteris marginalis L. is a local plant in Bangladesh which is widely used as folk medicine. In this study, the chemical composition of essential oil of D. marginalis L. as well as the antioxidant activity, antibacterial activity and total phenolic content of various organic extracts were analyzed. The gas chromatography–mass spectrometry analysis was used to identify the chemical compositions. The antibacterial activity of different extracts of Dryopteris marginalis was examined against three gram-negative bacteria and two gram-positive bacteria. The antibacterial activity of different extracts was investigated using disc diffusion method and minimum inhibitory concentration showing important antibacterial activity and was measured by using broth dilution method. The antioxidant activity was determined by using 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and superoxide radical scavenging assay. The total phenolic portion of the extracts were ascertained by Folin-Ciocalteu in alkaline medium. Twenty-two volatile compounds were identified from the essential oil. The methanol extract exhibited significant inhibitory effect which was close to the value of standard antibiotic (Streptomycin). The zones of inhibition of different organic extracts against the tested bacteria were found in the range of (7-15) mm and the minimum inhibitory concentrations (MIC) were recorded (15.62-1000) µg/ml. The antioxidant activity of D. marginalis was (82.13+1.2) % and (59.21+0.45) % highest activity with DPPH and superoxide radical scavenging assay respectively. The methanol extract showed highest (147.43+3.1) mg/g GAE of total phenolic content. The extracts and essential oil of D. marginalis showing strong antibacterial activity can be considered as a potential source of natural medicine and the ingredient of modern therapeutic agents.
