Influence of water concentration on the content of protein and nucleic acids in organs of chick embryos of different ages

The study was carried out in 10-, 14-15-, 18-20-day-old embryos and 1-2-day-old chickens of the Hy-Line breed. In chickens during embryogenesis, the peculiarities of the relationship between the main indicator of the ontogenetic growth of organs and tissues - the content of intracellular water by previously measured values: with the concentration of protein and nucleic acids in the organs of chicken embryos of different ages - were assessed. It was found that in the period of 10-19 days of embryogenesis, the growth of organs is accompanied by a significant decrease in water content to varying degrees: noticeably higher in the liver and in the cerebral hemispheres - up to 14 days; from the 19th day, these changes are less pronounced. The results of the studied indicator in the muscles indicate advanced embryonic development with a more pronounced decrease in the water content in the red oxidative muscles in comparison with white muscles, where the decrease in the indicator is less pronounced. Selection of five typical loess in Shanxi Province, the paper adopt the indoor test methods to prepares the different compaction and different moisture content specimens which were used to test CBR value and rebound module, analyzes the impact of compactness and moisture content on the CBR value and resilient modulus and establishes the relationship of CBR and moisture content. The result shows that the level of moisture content has great impact on CBR value and rebound module. The CBR value is maximal under optimum moisture content and maximum degree of compaction condition. The times of compaction have little effect on the improvement of CBR with increasing water content. The relationship of the soaking CBR,resilient modulus and the moisture content shows a certain regularity.Key words: Loess filler;the CBR;resilient modulus;compactness;moisture content;relationship

Alkalinep-nitrophenylphosphate phosphatase (pNPPase) from the halophilic archaeobacteriumHalobacterium salinarum(previouslyhalobium) was solubilized at low salt concentration in reverse micelles of hexadecyltrimethylammoniumbromide in cyclohexane with 1-butanol as cosurfactant. The enzyme maintained its catalytic properties under these conditions. The thermodynamic “solvation–stabilization hypothesis” has been used to explain the bell-shaped dependence ofpNPPase activity on the water content of reverse micelles, in terms of protein–solvent interactions. According to this model, the stability of the folded protein depends on a network of hydrated ions associated with acidic residues at the protein surface. At low salt concentration and low water content (the ratio of water concentration to surfactant concentration;w0), the network of hydrated ions within the reverse micelles may involve the cationic heads of the surfactant. The bell-shaped profile of the relationship between enzyme activity andw0varied depending on the concentrations of NaCl and Mn2+.

The electrophoresis gel mobility shift assay is a popular method for the study of protein-nucleic acid interactions. The binding of proteins to DNA is characterized by a reduction in the electrophoretic mobility of the nucleic acid. Binding affinity, stoichiometry, and kinetics can be obtained from such assays; however, it is often desirable to image the various species in the gel bands using TEM. Present methods for isolation of nucleoproteins from gel bands are inefficient and often destroy the native structure of the complexes. We have developed a technique, called “snapshot blotting,” by which nucleic acids and nucleoprotein complexes in electrophoresis gels can be electrophoretically transferred directly onto carbon-coated grids for TEM imaging.

Austempered ductile iron (ADI) is an advanced cast iron material that has a broad field of application and, among others, it is used in contact and for conveyance of fluids. However, it is noticed that in contact with some fluids, especially water, ADI material becomes brittle. The most significant decrease is established for the elongation. However, the influence of water and the cause of this phenomenon is still not fully understood. For that reason, in this paper, the influence of different water concentrations in ethyl alcohol on the mechanical properties of ADI materials was studied. The test was performed on two different types of ADI materials in 0.2, 4, 10, and 100 vol.% water concentration environments, and in dry condition. It was found that even the smallest concentration of water (0.2 vol.%) causes formation of the embrittled zone at fracture surface. However, not all mechanical properties were affected equally and not all water concentrations have been critical. The highest deterioration was established in the elongation, followed by the ultimate tensile strength, while the proof strength was affected least.

SUMMARYA rapid, simple method is described for estimating the area of rubber leaves from two measurements on the middle and one of the side leaflets. The relationship between the area of a leaflet (A) and its length × breadth (LB), described by the expression A = 0.654 LB, does not vary between the three leaflets or between leaves of different ages, and clonal differences are slight.
