ENTRIKIM THERAPEUTIC EFFICACY IN PIGS WITH ENZOOTIC PNEUMONIA

The number of T-lymphocytes in the subacute course decreases to 28.9±1.4% and in the chronic course it increased to 69.1±1.2%. However, against the background of lymphopenia, the absolute value of lymphocytes also decreased to 0.70±0.038·109 /l in the acute form of the disease. In chronic pneumonia, the number of lymphocytes was at the level of 0.44±0.04·109 /l. The T-x/T-c ratio also changed, depending on the form of enzootic pneumonia. With subacute – 1.7, and with chronic – 2.28. The entrikim treatment of piglets with a subacute course carried out for 7 days and a blood test 5 days after the implementation of sanitation showed that the number of Tlymphocytes increased twofold to 1.40±0.04·109 /l, while the Tx/Tc ratio was 2, 28, which corresponds to the physiological norm. After the treatment of piglets with a chronic course, all analyzed parameters characterizing the state of T-cell immunity testified to its normal functioning. A decrease in the total number of T-lymphocytes, individual EAK populations, indicate a depressive state of lymphopoiesis. A decrease in the number of T-lymphocytes to 0.44±0.04·109 /l in chronic course up to indicates a deep suppression of cellular immunity. In the subacute form of the disease, the number of B-lymphocytes decreased to 14.07±1.92%, but increased to 32.64±0.3% in the chronic course. But against the background of lymphopenia in subacute form, the absolute value of lymphocytes decreased to 0.36±0.04·109 /l, and in chronic lymphopenia to 0.20±0.06·109 /l. In the subacute form of the disease, the content of Ig A (1.49±0.03 mg/ml) and Ig G (1.39±0.02 mg/ml) does not differ from healthy animals, but a low level of Ig M (11.0±0.12 mg/ml) indicates a negative effect of mycoplasmas on humoral immunity. In chronic course, there is a sharp decrease (by 30%) Ig A to 1.49±0.03 and vice versa, a sharp increase by 30% in the Ig G content (11.8±011 mg/ml). After treatment of piglets with a subacute course of the disease, the number of B-lymphocytes increased by 2 times, and after treatment with m with a chronic course of the disease, by 3.5 times to 0.70±0.04·109 /L. The use of entrikim has a positive effect on humoral immunity, as the amount of Ig G increases by 45%. The therapeutic efficacy of entrikim increases due to the synergistic interaction of enrofloxacin, trimethoprim and tilmicosin phosphate. The therapeutic efficacy of entrikim in the subacute course was 90.7%, and in the chronic one – 74.4%.

The paper deals with researching of T- and B-lymphocytes relative quantity and functional activity in broiler chicken blood after using of animal origin immunomodulators in conditions of pre-slaughter stress. The authors determined the relative amount of T- and B-lymphocytes and their individual populations in the reaction of spontaneous rоsetting with the sheep erythrocytes in blood. Besides, the differentiated count of rоsetting lymphocytes with the various degree of functional activity was conducted. The spleen extract (70% alcohol solution in volume of 1.4 ml per chicken) was added to the diet of broiler chicken of experimental groups by aerosol method. This extract was obtained with/ without ultrasound application. 70% alcohol solution in the same volume and using the same method was added to the diet of broiler chicken of the control group five days before slaughter. The authors have not established probable increase of T-lymphocytes general quantity in broiler chicken blood in both experimental groups. It is shown that pre-slaughter stress in broiler chicken caused by weaning has immuno-suppressive effect on T- and B-lymphocytes in blood, which is accompanied by their quantity and functional activity decrease. T- and B-lymphocytes amount and functional activity of T- and B-cell immunity was stimulated after adding immunomodulators of natural origin to broiler chicken diet. Spleen extract polyamines as immunomodulators and antistressors most effectively influenced on some of cell immunity indices before slaughter – it is necessary to note the increase in T-helper lymphocytes in the broiler chickens blood caused by lymphocytes with medium (6–10) – by 18% (Р < 0.05) and high density receptors (М) – by 35% (Р < 0.05) compared to the control. It is shown that decrease of T-lymphocytes quantity in broiler chicken blood is caused by lymphocytes with law (3–5) – by 22% (Р < 0.01) and high (M) – by 11% (Р < 0.05) density receptors with increase of medium density receptors (6–10) – by 15% (Р < 0.05) compared to the control. The results obtained in the experiment can be used in the investigations of cell immunity indices of farm animals in order to increase the organism resistance and to correct their pre-slaughter stress.

Despite a huge number of studies, the uniqueness of antiparasitic immunity is so great that there is still insufficient knowledge of the factors contributing to the manifestation of the characteristics of immunity in mixed parasitic diseases of rabbits. Therefore, the question of the influence of the association of pathogens Treponema cuniculi and Eimeria sp. on indicators of cellular immunity of rabbits is relevant. The study was conducted on 59 male rabbits age 3–5 months of the Californian breed, selected by analogy. Animal were separated into two groups: healthy animals (control group) and sick animals (research group). Intensity of invasion was determined by the method of the Mac-Master. It has been established that the level of damage of rabbits by spirochetosis and eimeriosis was, on average, 1155.17 ± 184.87 and 6668.97 ± 284.16 pathogens in 1 g of feces. The count of T- and B-lymphocytes was determined by the method of spontaneous rosette-formation with sheep erythrocytes. Parasitizing the association of pathogens Treponema cuniculi and Eimeria sp. was revealed a high number of leukocytes (1.22 times, P < 0.001), which increased mainly due to lymphocytes, which were 1.45 times higher (P < 0.001), as well as neutrophilic metamyelocytes – 1.48 times (P < 0.05), eosinophils – 1.68 times (P < 0.001) and basophils – 1.57 times (P < 0.001) compared with similar blood parameters of healthy animals. In the blood of sick rabbits, the absolute number of T-lymphocytes (1.56 times, P < 0.001) and B-lymphocytes (3.02 times, P < 0.001) was significantly higher in comparison with a low number of O-lymphocytes (3.46 times, P < 0.001) compared with the control. This indicates the redistribution of lymphocytes to cells that carry T and B lymphocyte receptors on the plasma membrane. The absolute number of T-lymphocytes became high due to T-helpers, which in these animals were higher both in absolute (1.87 times, P < 0.001) and percentage (by 9.18%, P < 0.001) compared to control. Moreover, the percentage of T-suppressors in the blood of rabbits of the experimental group was significantly lower on 5.46% (P < 0.05) compared with the same blood count of healthy animals. Such a redistribution of the T-cell population in the peripheral blood of this group of rabbits led to an increase in the immunoregulatory index by 1.64 times (P < 0.01) than in healthy ones. High IRI and the number of T-active lymphocytes (by 28.23%, P < 0.05) in the blood of rabbits with parasitism of the association of pathogens Treponema cuniculi and Eimeria sp. indicate increased immune system tension.

The aim. To study the cell immunity status under influence of 3-[5-(1H-indol-3-ylmethylene)-4-oxo-2-thioxo-thiazolidin-3-yl]-propionic acid, as a prominent 4-thiazolidinone derivative and a class of biologically active compounds with polypharmacological properties. Materials and methods. Experimental method on the model of laboratory animals (guinea pigs); intradermal allergy tests; relative and absolute content in the peripheral blood of T- and B-lymphocytes subpopulations; hematological indexies: index of the ratio of lymphocytes and monocytes, index of the ratio of neutrophils and monocytes, index of the ratio of neutrophils and eosinophils, phagocytic index, phagocytic number; ELISA; organic synthesis; pharmacological screening. Results. The effect of 3-[5-(1H-indol-3-ylmethylene)-4-oxo-2-thioxo-thiazolidin-3-yl]-propionic acid has antifungal properties and affect cellular component of immunity in vivo in the guinea pigs model. There are no changes in the skin of guinea pigs during and after chemical applications of the skin and after intradermal tests. The compound stimulate the immune cells, in particular the lymphocyte (increase in the absolute number of CD3 T-lymphocytes by 21.46 % and the absolute number of CD8 T-suppressors by 27.15 %), but with a selective inhibitory effect on certain units (decrease the relative number of NK cells CD16 by 11.57 % and B-lymphocytes CD22 by 23.08 %). There was an increase in the activity of the macrophage phagocytic system (increase in PN by 439.87 % and PI by 62.73 % at 120 minutes), which indicates the reliability of the absorbing function of phagocytes, but with a decrease in their ability to endocytosis (PCI decreased significantly by 78,72 %). Conclusions. Synthesized 3-[5-(1H-indol-3-ylmethylene)-4-oxo-2-thioxo-thiazolidin-3-yl]-propionic acid has a selective activating effect on certain parts of cellular immunity and on phagocytic activity. Derivate influence on the phagocytic activity of neutrophils is ambiguous, and the effect of the compound directed to the cellular part of the immune system does not cause cellular immunodeficiency. The studied derivative is promising for further study of the drug-like molecule with antifungal and antitumor effects

Abstract The purpose of these studies was to determine the indicators of cellular and humoral immunity of calves. Compared to calves obtained from untreated cows, the number of T-lymphocytes in young calves born from cows treated with Timolin increased the level of T-cells to 32.9%, while using Timosplenin 45%. As a result of the use of immunomodulators in treating cows with ketosis in mothers, the relative content of T-lymphocytes in the blood of calves born from them increased when using the drug Leucogen by 1.7 times, Timosplenin by 2.3 times, Timalin in 2.2 times. The use of Leucogen for cows with ketosis contributed to an increase in the percentage of B cells in the blood of calves born by 23%, Timosplenin 83%, and Timalin 42%. At the same time, the absolute number of B-lymphocytes in the treatment of “Leucogen” increased 1.9 times and reached the level of healthy calves, in the treatment with the preparations “Timosplenin” and “Timalin” - the increase was, respectively, 3.0 and 2.3 times and was higher this indicator in young animals born from healthy calves. Thus, the inclusion of dry immunomodulators in the treatment regimen of patients with ketosis leads not only to normalization of metabolism, but is also accompanied by an increase in the level of cellular immunity factors in calves born from them. Under the influence of “Leucogen,” the percentage of lymphocytes was normalized, whereas with the introduction of “Timalin” and “Timosplenin,” the content of this type of cells in newborn calves even exceeded those in healthy young.

We studied sex differences lymphocytes subpopulations of peripheral blood in adult C57Bl/6 mice during acute and chronic colitis, induced with 1% DSS. We measured subpopulations of lymphocytes with flow cytometry. We showed that in the control group the female mice had statistically significantly higher values of the relative number of regulatory and cytotoxic T lymphocytes comparing to the males. During acute colitis the females showed an increase in the relative number of Thelpers and a decrease of cytotoxic Tlymphocytes, which reflects the activation of immune response. The males had a decrease in the absolute number of leukocytes, lymphocytes and cytotoxic and regulatory T lymphocytes, probably because of an increase in migration of these cells to the inflammation locus and local lymph nodes. In chronic colitis the females had a decrease in the absolute number of leukocytes, lymphocytes, T helpers, cytotoxic T lymphocytes and B lymphocytes when comparing with acute colitis. During chronic colitis the males had a decrease in the absolute number of T helpers and B lymphocytes but an increase of regulatory T cells in comparison with the control group; in comparison with acute colitis the males with chronic colitis had higher relative and absolute number of regulatory T cells. The increase of T regulatory lymphocytes is due to an increase in their proliferation rate in the thymus and increase of their migration to the inflammatory locus – the colon. Future clinical studies may be based on these results, which show that the treatment of colitis, especially with immunotropic agents, must take sex differences into account.

T and B lymphocyte subpopulations were examined in the peripheral blood of children using the E and EAC rosette assays. Children under 18 months of age were found to have a decreased percentage of E-binding (T) lymphocytes and an increased percentage of EAC-binding (B) lymphocytes compared to older children (18 months to 10 years) and adults. The absolute number of E-binding and EAC-binding lymphocytes was increased in children under 18 months of age.

In the use of colostral protective factors and insufficient activity of piglets immune system, against the background of immunodeficiency after weaning from sows developing diseases, often caused by conditionally-pathogenic or pathogenic microorganisms, including non-contagious gastroenteritis. The article presents analyzes of T- and B-cell immunity parameters in weaned piglets with non-contagious gastroenteritis and after their treatment with probiotic TOYOCERIN 10⁹ and phytobiotic EXTRACT™ 6930 as part of the basic diet in combination with antibiotic (10 % solution of enrofloxacin hydrochloride) in the modern pig farm. It was established that in piglets after weaning with non-contagious gastroenteritis decreased of T- and B-cell immunity parameters. In particular, in blood decreased number of total and active T-lymphocytes, T-helpers and T-suppressors also decreased number of B-lymphocytes, which may indicate the development in these immunodeficient condition. It was established positive effect of probiotic and phytobiotic in combination with antibiotic on T-cell immunity of pigs, as indicated by the increased number of total T-lymphocytes in blood to the clinical healthy animals level. A similar tendency was observed in the number of T-helper cells, their number was higher in groups of weaned piglets which added probiotic TOYOCERIN 10⁹ and phytobiotic EXTRACT™ 6930. As to the immunoregulatory index, which is a quantitative ratio of T-helper to T-suppressor, there was no significant alteration of values in control and experimental groups of pigs. It was established positive effect of probiotic and phytobiotic in humoral immunity of piglets by increasing number of B-lymphocytes to the level of clinical healthy animals which indicate increased body ability to the active synthesis of protective antibodies. The use of probiotic TOYOCERIN 10⁹ and phytobiotic EXTRACT™ 6930 for weaned piglets with non-contagious gastroenteritis normalized number of T- and B-lymphocytes, which are the leading immunocompetent blood cells that characterize the level of animal body defense and condition of specific immunity. Perspectives for further research will be directed at determining the therapeutic effectiveness of probiotic TOYOCERIN 10⁹ and phytobiotic EXTRACT™ 6930 for weaned piglets with non-contagious gastroenteritis.

This study aims to analyze the sensitivity testing using measurements of realization of regional own-source revenues and operating expenditure and to analyze the extent of the effect of sample differences between Java and non-Java provinces by using samples outside of Java. By using sensitivity analysis, the results found the influence of audit opinion on the performance of the provincial government mediated by the realization of regional operating expenditure. More specifically, when using the measurement of the absolute value of the realization of regional operating expenditure it was found that there was a direct positive and significant influence of audit opinion on the performance of the Provincial Government. However, no significant effect of audit opinion was found on the realization value of regional operating expenditure and the effect of the realization value of regional operating expenditure on the performance of the Provincial Government. This result implies that an increase in audit opinion will be more likely to be used as an incentive for the Provincial Government to increase the realization of regional operating expenditure.

Abstract The influence of five different methods of homogenisation (1. The method according to Potter and Elvehjem, 2. A modification of this method called Potter S, 3. The method of Dounce, 4. Homogenisation by hypersonic waves and 5. Coarce-grained homogenisation with the “Mikro-fleischwolf”) on the absolute value and stability of oxygen uptake of guinea pig liver homogenates has been investigated in simultaneous measurements. All homogenates showed a characteristic fall of oxygen uptake during measuring time (3 hours). The modified method according to Potter and Elvehjem called Potter S showed reproducible results without any influence by homogenisation intensity.
