Breast cancer is the most dominant type of cancer in Indonesia. Traditional medicine using tapak liman (Elephantopus scaber L.) as an anticancer becomes an option because it contains compounds such as sesquiterpenes. The purpose of this study was to know the cytotoxic activity of ethanolic extract of tapak liman on T47D cells through induction of apoptosis. The extraction method uses maceration with 96%. Total flavonoids were tested using quercetin as a spectrophotometric standard. ethanol solvent, cytotoxic test using microtetrazoliom (MTT assay), and apoptosis test using double staining test (ethidium bromide-acrydine orange) under a fluorescence microscope. The results showed the linear regression equation y = 0.0934x-0.0292 with the resulting flavonoid levels of 0.979% w / w calculated against quercetin (QE) The concentration of the compound used was 1000 µg/ml; 500 µg/ml; 250µg/ml; 125 ?g/ml, 62.5 ?g/ml; 31.25 µg/ml. The MTT assay results are the values of IC50 which are analyzed using probit analysis and apoptosis tests which are analyzed descriptively. The results of the research show that the ethanol extract of tapak liman using MTT method has IC50 value of 59 µg/ml, while the testing of double staining method shows the color of bright green which indicates early apoptosis and the color orange in the cells which undergoing final apoptosis. DNA fragmentation results shows that Tapak Liman extract induces DNA fragments. The conclusion of this study is that tapak liman textract has the potential for cytotoxic activity against T47D breast cancer cells through the induction of apoptosis

Breast cancer is one of cancer with high mortality. This cancer not only attacks women, but also men. Indonesia has many plants which potential as anticancer, such as orchids. Spathoglottis plicata is one of the orchid species that abundant in Indonesia and has a lot of antioxidant compounds which is guessed have anticancer properties. The objectives of this study were to study the cytotoxic activity and IC50 value of aquadest, ethanolic, and chloroform extracts of S. plicata’s pseudobulbs, leaves, and whole plants on T47D cells (breast cancer cells line) as well as cytotoxic activity of the specific fraction of the most toxic crude extract. S. plicata used in this study was obtained from Bungarinte nursery. Extractions were done by maceration method using aquadest, ethanol, and chloroform as the solvent. Cytotoxic test on T47D cells were done by MTT assay. The cytotoxic data were analyzed using one-way ANOVA followed by Tukey’s HSD test. The IC50 of each extracts were calculate by probit analysis. The lowest IC50 value among all extracts was fractionated and isolated by preparative TLC. The cytotoxic activity and IC50 of this fractions were analyzed. The results showed that only 2 from 9 crude extracts that able to calculate its IC50 because those two extracts have concentration dependent pattern of inhibition concentration. Chloroform extract have the lowest IC50 value (369,837 μg/mL). Then, this extract fractionated by eluen n-hexane : ethyl acetate 4:1. Four fractions were collected. The lowest IC50 value is fraction IV (144,41 μg/mL). Based on the results it could be concluded that S. plicata leaves have moderate potency to develop as anticancet agents, especially on breast cancer. ABSTRAK Pengobatan kanker menggunakan bahan alam terus dikembangkan. Salah satu tanaman yang memiliki efek sitotoksik Ocimum basilicum, L. tujuan penelitian ini adalah mengetahui aktivitas sitotoksik dari ekstrak etanol Ocimum basilicum (EEOB) terhadap sel kanker payudara T47D dan MCF7. Ocimum basilicum diekstraksi menggunakan metode maserasi dengan pelarut etanol 70%. Pengujian aktivitas sitotoksik menggunakan metode MTT assay dengan  seri konsentrasi EEOB 1000; 500; 250; 125; 62.5; 31,25 µg/mL. Hasil uji aktivitas sitotoksik EEOB memperlihatkan nilai IC50 pada sel T47D dan MCF-7 sebesar 399.86 µg/ml dan 387.76 µg/ml. Kata Kunci—Sitotoksik, Ocinum basilicum L., T47D, MCF-7  ABSTRACT             Cancer treatment using natural ingredients continues to be developed. One of the plants that is proven to have cytotoxic activity is basil leaves (Ocimum basilicum, L.). This study aims to determine the cytotoxic activity of ethanol extract of basil leaves (EEBL) on T47D and MCF-7 breast cancer cells. Basil leaves were extracted using maceration  with ethanol 70%. The cytotoxic test was perfomed using MTT assay with various EEBL concentrations: 1000; 500; 250; 125; 62.5; 31,25 µg/mL. The use of natural products has been widely used as a resource of new bioactive chemical compounds. One of them is the Paku Atai Merah (Angiopteris ferox Copel) tuber which has long been used empirically by the Dayak tribe of East Kalimantan as an anti-cancer. The purpose of this study was to determine the anticancer cytotoxic activity of the extract and fractions of Paku Atai Merah tuber against T47D breast cancer cells in vitro. Extract of Paku Atai Merah tubers was obtained by maceration method using ethanol solvent until obtained the ethanolic extract then fractionated using various solvents to obtain n-hexane, ethyl acetate, and aqueous-ethanol fractions. The cytotoxic effect was carried out based on the MTT assay. Phytochemical screening tests showed positive results for the presence of flavonoid, phenolic, tannin, saponin and steroid compounds. The results of the cytotoxic activity study showed that the ethyl acetate fraction had moderate cytotoxic activity in T47D cancer cells with an IC50 value  of 84.8 µg/ml. Ethanol extract (513.06 µg/ml) and n-Hexane frsction (881.97 µg/ ml) were also included in the weak category. This study indicates that ethyl acetate fraction can be developed as a supportive therapy for breast cancer treatment.

AbstrakSuatu senyawa obat dapat menjadi kemoterapi kanker adalah dengan cara menskrining terlebih dahulu tumbuhan obat yang berpotensi sebagai obat antikanker. Salah satunya adalah tanaman obat daun nimba (Azadirachta indica L.Juss) yang terbukti secara significant menyebabkan apoptosis pada beberapa jenis sel line kanker. Dalam penelitian ini, ekstrak ethanol dari A. indica dipelajari untuk melihat efeknya pada pertumbuhan sel kanker payudara manusia jenis MDA-MB-231 dengan menggunakan tes untuk proliferasi yaitu MTT assai dan untuk mengetahui perubahan morphologi dari apoptosis selnya dengan menggunakan TUNEL assay Ekstrak daun nimba (A. indica) dapat menurunkan keberadaan jumlah sel kanker dengan cara menghambat perkembangan daripada sel tersebut dan menginduksi proses apoptosis pada sel kanker tersebut. Hasil pemeriksaan MTT assai didapatkan nilai IC50 nya adalah 55 ug / mL. Kematian MDA-MB231 sel yang disebabkan oleh ekstrak daun nimba (A.indica) ditemukan melalui mekanisme apoptosis yang secara morfologinya menunjukan ciri ciri dari kematian secara apoptosis seperti kondensasi dari nucleus, membrane nukleus yang melebur dan akhirnya terjadinya fragmentasi dari DNA. Analisis struktur dalaman sel juga mengungkapkan karakteristik apoptosis yaitu marginasi dari kromosom yang disertai dengan fragmentasi DNA dan selanjutnya akan terbentuk badan apoptotik pada sel kanker yang diinkubasi dengan ekstrak tersebut. Pada penelitian ini juga dijumpai peningkatan jumlah sel apoptosis dari hari 1 sampai hari 3 inkubasi oleh ekstrak nimba. Ekstrak ethanol A.indica mungkin mengandung senyawa bioaktif(s) yang menyebabkan kanker payudara MDA-MNB 231 mengalami kematian sel secara apoptosis. Penelitian lebih lanjut masih diperlukan untuk mengetahui mekanisme tumbuhan ini membunuh sel kanker MDA-MB 231.Kata kunci: Studi In vitro, Azadirachta indica, apoptosis, TUNEL assayAbstractA screening is conducted on plants that have potential as anticancer is a promising way for discovering novel chemotherapeutic compound. A medicinal plant neem leaf (Azadirachta indica L.Juss) intake has been shown to induce significant levels of apoptosis in various cancer cells. In this present study, ethanol extract of Azadirachta indica was studied for its effects on growth in MDA-MB 231 human breast cancer cells using assays for proliferation (MTT assay) and mechanisme of cell apoptosis using TUNEL assay. Neem leaf extract decreased cell viability, inhibited cell proliferation, and induced cell apoptosis. Result of MTT assay was 55 μg/mL of neem remarkably reduced cell viability of MDA-MB 231 cells. MDA-MB231 cell death elicited by the extract was found to be apoptotic in nature based the indication of nucleus condensation, shrinkage of nucleus membrane and also DNA fragmentation which are a hallmark of apoptosis. In addition, ultrastructural analysis also revealed apoptotic characteristics which are the presence of chromatin margination and apoptotic bodies in the extract-treated cells. There was an increase in the number of apoptotic cells from day 1 to day 3 post incubation with neem extract. Thus, the results from this study strongly suggest that the ethanol extract of A.indica may contain bioactive compound(s) that caused breast carcinoma, MDA-MNB 231 cell death by apoptosis. It’s needed to do advance research to know more deeply the mechanism this plant on breast cancer cell line MDA-MB.Keywords:In vitro study, Azadirachta indica, apoptosis, TUNEL assay

Cervical cancer is one type of cancer that attacks women in both developed and developing countries. Cancer can arise from cell division errors or DNA damage from certain environmental exposures that cause free radicals as a factor of cancer cells. The agarwood plant (Aquilaria malaccensis  Lam) has been known by society as a health tea ingredient. The content of compounds contained in tea is believed to have anti-cancer and antioxidant activity. In previous studies found some of the active compound ingredients contained in the leaves of agarwood, such as alkaloids, flavonoids, triterpenoid, saponins and tannins. Tests that have been done previously that agarwood plants have good cytotoxic activity against cancer cells MCF-7 from one of the fractions, the n-hexane fraction. In this study, another cytotoxic potential test was carried out from aloe plants on other cancer cells, namely HeLa cells. Extraction was carried out by the Soxhletation method using 96% ethanol solvent, then fractionated with n-hexane solvent, ethyl asetate, and methanol: water. The MTT assay method is used to test the cytotoxic activity of sample research on cell cultures used. Measurements were performed using an ELISA reader at 550 nm wavelength. The results showed that the IC50 values of the four samples tested were ethanol extract (299,506 µg / ml), n-hexane fraction (120,913 µg / ml), ethyl acetate fraction (3490,476 µg / ml) and methanol fraction: water (294,060 µg) / ml). from extracts and fractions it is known that the highest value is shown by the n-hexane fraction in the moderately active/moderate category.

Treatment of cancer such as surgery, radiotherapy and chemotherapy has many side effects. Chemopreventive agent is needed to reduce the side effect and increase the effectivity of therapy. The discovery of cochemopreventive agent should consider on its selectivity to reduce side effects. The selective cochemopreventive agents work effectively in cancer cells and safe for normal cells. Buah Makassar (Brucea javanica) is a natural product that is empirically used for anti-inflammatory and antitumor. The purpose of this study is to determine the cytotoxic effect of ethanolic extract of buah Makassar against 4T1, MCF7, HeLa, and Vero cell lines. The cytotoxic test is performed by MTT assay. The parameter obtained from the cytotoxic test was IC50. Selectivity index is determined from IC50 ratio of cancer cells to normal cells. The results showed that ethanolic extract of buah Makassar has a cytotoxic activity on 4T1, MCF7, HeLa, and Vero cells with IC50 were 49,9±0,83 μg/mL; 107,6±8,14 μg/mL; 228,9±4,16 μg/mL and 395,5± 4,21 μg/mL respectively. It also has high selectivity on 4T1 metastatic breast cancer cell with selectivity index of 7,93. It can be concluded that the ethanolic extract of buah Makassar has potential to be delevoped as cochemopreventive agent especially on metastatic breast cancer. Many studies have shown the anti-cancer activities of the chemical compounds extracted from the leaves of Annona muricata or soursop plant. Cianjur and Sukabumi are quite large soursop producing area in Indonesia. This study was carried out to determine the difference of cytotoxic activity of soursop leaves ethanolic extract which were harvested from three different areas of Cianjur (I, II, III) and Sukabumi (I, II, III). The Soursop leaves were macerated with 70% ethanol using microwave assisted extraction (MAE) method. The extract was tested in vitro on breast cancer cell line MCF7 and its constituent was identified using GC-MS apparatus. The results showed that the highest cytotoxic activity with IC50 values of 9.12 µg ml-1 was determined on the extract of soursop leaves harvested from Cianjur III area. Qualitative identification of chemical constituent shows that the soursop leaves contain alkaloid, flavonoid, triterpenoid, tannin and saponin compounds. No steroid compound was detected in the extract. It can be concluded that the geographical regions affected the biochemical properties of soursop leaves.
