Insulin secretion is impaired in type 2 diabetes (T2D). The insulin and glucose responses to central autonomic activation induced by excitation of brain medullary TRH receptors were studied in T2D Goto-Kakizaki (GK) rats. Blood glucose levels in normally fed, pentobarbital-anesthetized GK and nondiabetic Wistar rats were 193 and 119 mg/100 ml in males and 214 and 131 mg/100 ml in females. Intracisternal injection (ic) of the stable TRH analog RX 77368 (10 ng) induced significantly higher insulin response in both genders of overnight-fasted GK rats compared with Wistar rats and slightly increased blood glucose in female Wistar rats but significantly decreased it from 193 to 145 mg/100 ml in female GK rats. RX 77368 (50 ng) ic induced markedly greater glucose and relatively weaker insulin responses in male GK rats than Wistar rats. Bilateral vagotomy blocked ic RX 77368-induced insulin secretion, whereas adrenalectomy abolished its hyperglycemic effect. In adrenalectomized male GK but not Wistar rats, ic RX 77368 (50 ng) dramatically increased serum insulin levels by 6.5-fold and decreased blood glucose levels from 154 to 98 mg/100 ml; these changes were prevented by vagotomy. GK rats had higher basal pancreatic insulin II mRNA levels but a lower response to ic RX 77368 (50 ng) compared with Wistar rats. These results indicate that central-vagal activation-induced insulin secretion is susceptible in T2D GK rats. However, the dominant sympathetic-adrenal response to medullary TRH plays a suppressing role on vagal-mediated insulin secretion. This unbalanced vago-sympathetic activation by medullary TRH may contribute to the impaired insulin secretion in T2D.

This study aimed to understand Strychnosnuxvomica and Holarrhena pubescens Stem bark extract action towards M3 receptor in controlling blood glucose levels. Strychnos nux vomica  and Holarrhena pubescens are both alkaloidal drugs can help in controlling Hyperglycemic level. This will be useful in the formulation of a new herbal drug molecule for treating diabetes. Chloroform and ethanolic extracts of selected alkaloidal plants were extracted using the soxhlet apparatus and obtained quotes were tested for acute toxicity studies and carried out anti-diabetic action on Wister albino rats for 21 days. Results obtained from Blood glucose levels and histopathological study of test groups are compared with blood glucose levels of standard group, and highly significant action was identified by the chloroform extract of Strychnos nux vomica and Holarrhena pubescens group. Moderate anti-diabetic action was observed remaining two groups of ethanolic extracts. Strychnos nux vomica and Holarrhena pubescens ethanolic extract groups are acting on M3 receptors and controlling Hyperglycemic levels.

Diabetes mellitus is a metabolic disease that always increasing in prevalence. Excessive glucose intake and oxidative stress that can lead to damage to the pancreatic cell or insulin resistance, are such factors that influence the occurrence of diabetes mellitus. Graptophyllum pictum (L.) Griff leaf contains active metabolites such as flavonoid which able to decrease free radical levels. This study aimed to prove the effectiveness of Graptophyllum pictum (L.) Griff leaves extract on blood glucose levels in alloxan-induced Wistar rats. In this study, twenty-five male Wistar rats were divided into five groups. On day 0, the negative control and each treatment group injected with 125 mg/kg dose of alloxan and would be checked on 4th day. On the 4th day after the rats had diabetes, normal group (K-1) was given only standard feed, Negative control (K-2) was given only alloxan intraperitoneally on day 0, first treatment group (K-3) was given 50 mg/kg dose of Graptophyllum pictum (L.) Griff, second treatment group (K-4) was given 100 mg/kg dose of the leaf extract, and the third treatment group (K-5) was given the leaf 200 mg/kg dose of the leaf extract. On the 10th day, fasting blood glucose was taken from the rats. Post-test only group design was used to be the experimental design. There were significant differences in the treatment groups (K-3, K-4, and K-5) compared to the negative control group (K-2) after treatment. The mean of blood glucose levels decreased with the increased doses of Graptophyllum pictum (L.) Griff leaf extract treatment. The treatment groups (K-3, K-4, and K-5) were not significant compared to the normal group (K-1).

Gynura divaricata (GD) also known as Dewa leaf is one of the Indonesian medicinal plants which are also native to India, Thailand and China. GD is widely used as antidiabetic, antihypertensive and other diseases including several tumors. However, research on the effects of GD as antidiabetic is still not widely practiced, especially in Indonesia. The aim of this research is to investigate the effect of GD ethanolic extract on blood glucose levels of alloxan-induced diabetes mice. This research is an experimental research with pre- and post-test design using 24 male DDY strain mice. Animals experiment is divided into four groups, that are one normal group and three alloxan-induced diabetes mice group that give 1% CMC (control), 400[Formula: see text]mg/kg GD ethanolic extract and 0.65[Formula: see text]mg/kg glibenclamide, respectively. Treatment was given for 14 days. Blood glucose levels were measured before and after inducing alloxan, and after 14 days treatment. At the end of study, all of animals experiment were sacrificed for histological examination. Phytochemical analysis revealed that polyphenols, flavonoids, steroids, triterpenoids, terpenoids and and quinones were present in ethanol extract of GD. The results of this study showed significant ([Formula: see text]) decrease of blood glucose levels after GD ethanolic extract and glibenclamide treatment. The results of histological examination showed improvement in pancreas damage and significant increase in the number of beta cells in GD and glibenclamide groups. The results indicate that dewa leaf ethanolic extract has antidiabetic activity and the effectiveness is the same as glibenclamide. GD also could repair damage of pancreas caused by alloxan induction.

<p class="Abstract">In this work, <em>Taxithelium nepalense</em> ethanolic extract and its fractions were evaluated for the antidiabetic activity in rat based on phytochemical and free radical scavenging properties. The ethanolic extract, fraction IV and V significantly attenuated the blood glucose levels at 600, 200 and 200 mg/kg with 50.0, 33.5 and 42.0% inhibition respectively. The histopathological studies were manifesting the recuperation of damaged cells in liver and pancreas tissues. The present study aims to study the hypoglycemic effect of methanol extract of Andrographisechioides (MEAE) in streptozotocin (STZ)-induced diabetic Wistar rats. Hyperglycemia was induced in rats by single intraperitoneal injection of STZ (55 mg/kg bodyweight). Three days after STZ induction, the hyperglycemic rats were treated with MEAE orally at the doses of 200, 500, and 800 mg/kg body weight daily for 21 days. Glibenclamide (1 mg/kg, orally) was used as reference drug. The fasting blood glucose levels were measured on each 7th day during the 21 days of treatment.

Background: Diabetes mellitus (DM) is metabolic disorder syndrome characterized by hyperglycemia due to deficiency of insulin secretion. Patients of diabetic patients increases each year, so it needs proper handling. Corn flour is a source of carbohydrates with a low glycemic index to help reduce the rise in blood sugar levels. In addition, supplementation with tempeh flour containing isoflavones act to protect cells from free radical thereby inhibiting damage cells. Fiber in tempeh affect blood glucose levels because it slows the absorption of glucose.Objective: To determine the effect supplementation of corn flour with tempeh flour on blood sugar levels in diabetes mellitus Wistar rats.Methods: The study was true experimental design with Pre-posttest control group design, samples were 20 male Wistar rats aged 2-3 months were then divided into 4 groups (T0, T1, T2, T3.) T0 was injected with alloxan as 30 mg / 150 grams of BB rats are subsequently treated with standard diet; T1 was dministered by standard diet 50% + 50% corn flour; T2 was administered by standard diet 50% + 30% + corn flour tempeh flour 20%; and T3 was administered by standard diet of corn flour 50% + 15% + 35% tempeh flour for 2 weeks. Blood glucose levels were measured with a glucose kit (GOD FS). Research data were analyzed by One Way ANOVA followed by Tukey's Post Hoc test p-value <0.05.Results: There were significant differences on blood sugar levels in all groups (p = 0.000): control group (only dietary standards), T1 (administered by standard diet 50% + corn flour 50%), T2 (administered by a standard diet 50% + cornstarch 30% tempeh flour + 20%) and T3 (administered by standard diet of corn flour 50% + 15% + 35% tempeh flour).Conclusion: corn flour with tempeh flour supplementation effect on decresing of blood sugar levels.

Background: Polyphenols, antioxidants, dietary fiber, and vitamin contained in the red dragon fruit peel. Red dragon fruit peel can be processed into marmalade. Red dragon fruit peel marmalade has the potential to be a functional food. Functional food is food that has a physiological function based on scientific studies.Objectives: The objective of this study is to analyze the effect of red dragon fruit peel marmalade on fasting blood glucose levels, HDL, LDL, and triglycerides levels of hypercholesterolemic Wistar rats.Methods: This study used a pre-&post-test control group design. Hypercholesterolemic male Wistar rats were randomly assigned into five groups. Hypercholesterolemia was induced by 1% cholesterol powder and 0.5% cholic acid for two weeks. All groups received standard chow. Samples were grouped into five groups: K-; K+; K1 (0.94 g/kg b.wt/day); K2 (1.41 g/kg b.wt/day); K3 (1.88 g/kg b.wt/day). The intervention was carried out for 28 days. GDP level was measured using the GOD-PAP. HDL, LDL, and triglyceride were analyzed with spectrophotometry. GDP, HDL, LDL, and triglyceride levels were measured twice before fasting. A paired t-test and one-way ANOVA were used to analyze the data.Results: The result showed that K-; K1; K2; K3 had a significant difference between groups before and after the intervention (p<0.05). Red dragon fruit peel Marmalade was able to reduce the levels of GDP, LDL, triglycerides, and increase HDL (p <0.05).Conclusion: Red dragon fruit peel marmalade reduced fasting blood glucose levels, LDL, triglyceride levels, and increased HDL levels of hypercholesterolemic Wistar rats.
