Astragalus hamosus contains valuable biologically active compounds, incl. flavonoids. The possibility for in vitro cultivation of the species as a source of important flavonoids was studied. Shoot and callus cultures were established and successfully cultivated on different nutrition media, complemented or not with growth regulators. An ultra-high performance liquid chromatography – high-resolution electrospray ionisation mass spectrometry (UHPLC-HRESIMS) qualitative and quantitative analysis of non-purified methanol extracts of these cultures was performed. It was found that the cultures produced rutin in comparable quantity. Interestingly, both shoots and callus cultures accumulated the rare triglycosides alcesefoliside and mauritianin. The quantity of mauritianin, biosynthesized in shoots, was significantly higher to that in callus cultures. Alcesefoliside, was in lower quantity, compared to mauritianin. In addition, callus cultures produced alcesefoliside trice as the shoots, besides their lower level of differentiation. These findings could serve as initial research to establish the value of in vitro cultures from A. hamosus as an alternative mean of production of pharmaceutically important flavonol glycosides.

Introduction. Geroprotectors are biologically active substances that inhibit the aging process. Many plant species are natural geroprotectors. For instance, Thymus vulgaris and Trifolium pratense are callus cultures with strong geroprotective properties. Study objects and methods. The present research featured T. vulgaris and T. pratense grown in vitro on various nutrient media. Their extracts were obtained by aqueous-alcoholic extraction using the following parameters: water-ethanol solvent Se = 30, 50, and 70 %; temperature Te = 30, 50, and 70°C; time τe = 2, 4, and 6 h. The quantitative and qualitative analysis was based on high-performance liquid mass spectrometry, gas mass spectrometry, and thin-layer chromatography. Results and discussion. The optimal extraction parameters for T. vulgaris were τe = 4 h, Te = 50°C, Se = 70 %, for T. pratense – τe = 6 h, Te = 70°C, Se = 70 %. The chromatography detected flavonoids, phenylpropanoids, simple phenols, higher fatty acids, mono- and sesquiterpenes, and aliphatic hydrocarbons. T. vulgaris appeared to have the highest content of thymol (23.580 ± 1.170 mg/mL); its thymol, apigenin, gallic, chlorogenic, and caffeic components demonstrated geroprotective properties. The extract of T. pratense possessed the highest rutin content (10.05 ± 0.35 mg/mL), and it owed its geroprotective characteristics to rutin, chlorogenic and p-coumaric acids. Conclusion. The callus cultures of T. vulgaris and T. pratense proved to be promising sources of geroprotective biologically active substances.

The onion (<em>Allium cepa</em>) is a vegetable used extensively all over the world both for culinary purposes as well as in medicine. Its medicinal values are due to the high levels of biologically-active compounds present within the bulb. There are various phytochemicals of therapeutic importance found in <em>A. cepa</em>. Quercetin, a flavonoid, is one of these phytochemicals and it is a potent antioxidant. <em>Allium cepa</em> is a dietary supplement and is beneficial for diverse ailments, thus justifying its status as a valuable medicinal plant. Due to its medicinal significance, elicitation of total antioxidants and quercetin levels have been attempted to enhance their production in tissue callus cultures. This study reports in vitro enhancement of total antioxidants and quercetin in <em>A. cepa</em> using methyl jasmonate as an elicitor. A reverse phase-high performance liquid chromatography (RP-HPLC) method was used with an isocratic system and a flow rate of 1.0 mL min<sup>−1</sup> and a mobile phase of acetonitrile: 1% v/v acetic acid (60%:40% v/v). The detection wavelength was 362 nm and the retention time 8.79 minutes. Total antioxidant and quercetin contents were maximal with 100 µM of methyl jasmonate in leaf tissue callus cultures at 84.61 ±6.03% and 0.81 ±0.03 mg g<sup>−1</sup> dry cell weight, respectively. They decreased with further increases of methyl jasmonate at 200 µM. The increase in total antioxidant and quercetin contents were 2.3- and 13.9-fold, respectively. The optimization of methyl jasmonate as an elicitor, as well as the determination of a suitable concentration in <em>A. cepa</em> in callus cultures, will be helpful for enhanced production of various other secondary metabolites of therapeutic significance. This could be beneficial for the pharmaceutical and neutraceutical industries for herbal drug formulations.

Holoptelea integrifolia Roxb. Planch (HI) has been used to treat various ailments including obesity, osteoarthritis, arthritis, inflammation, anemia, diabetes etc. To review the major phytochemicals and medicinal properties of HI, exhaustive bibliographic research was designed by means of various scientific search engines and databases. Only 12 phytochemicals have been reported including biologically active compounds like betulin, betulinic acid, epifriedlin, octacosanol, Friedlin, Holoptelin-A and Holoptelin-B. Analytical methods including the Thin Layer Chromatography (TLC), High-Performance Thin Layer Chromatography (HPTLC), High-Performance Liquid Chromatography (HPLC) and Liquid Chromatography With Mass Spectral (LC-MS) analysis have been used to analyze the HI. From medicinal potency point of view, these phytochemicals have a wide range of pharmacological activities such as antioxidant, antibacterial, anti-inflammatory, and anti-tumor. In the current review, it has been noticed that the mechanism of action of HI with biomolecules has not been fully explored. Pharmacology and toxicological studies are very few. This seems a huge literature gap to be fulfilled through the detailed in-vivo and in-vitro studies.

Abstract Unorganized callus and leaf/root-differentiating callus cultures of Pimpinella major have been established in liquid nutrient medium. Their capacity to accumulate rare phenylpropanoids such as epoxy-pseudoisoeugenol tiglate, epoxy-anol tiglate and anol tiglate was compared with that of seedlings and whole plants. The unorganized callus cultures were not able to accumulate any phenylpropanoids. In comparison, the leaf/root-differentiating callus culture promoted the accumulation of epoxy-pseudoisoeugenol tiglate (up to 90 mg/100 g fr.wt.) but not that of anol-derivatives. The accumulated amount of EPT in PMD-SH was comparable with that in plant seedlings.

The aim of this study was to evaluate the effect of 6-benzylaminopurine (BAP) separately and in interaction with auxins on the change in the qualitative and quantitative composition of flavonoids in the raw materials of regenerating plants Iris sibirica L. Cambridge grade in comparison with aeroponic and intact raw materials using the method of high-performance liquid chromatography. Raw materials of I. sibirica Cambridge variety obtained in vitro culture had a richer qualitative composition of flavonoids than intact plants. The dependence of the accumulation of flavonoids on the concentration of 6-benzylaminopurine in nutrient media was noted. The presence of 13 compounds was observed in extracts of 70% ethyl alcohol from regenerating plants grown at the lowest concentration of BAP (1.0 µM) within the experiment. In quantitative terms, the flavonoid apigenin was maximally determined on a medium with BAP 1 µM, and kaempferol - on media with BAP 5.0 µM, supplemented with auxins. For a medium with 7.5 µM BAP, the lowest variety of compounds was observed (9) and the lowest kaempferol content. Auxins influenced the synthesis of flavonoids. The amount of flavonoids in all variants of the experiment increased by an average of 13% in the presence of auxins. The stages of the technological process of obtaining raw materials I. sibirica Cambridge variety on the basis of clonal micropropagation and cultivation in aeroponics conditions allowed to obtain raw materials that do not contain heavy and toxic metals, are not infected with pathogens and pests. With 1 m2 of useful area of aeroponics for 1 year, it is possible to collect 5 times more raw materials than with field cultivation. According to the qualitative composition of phenolic compounds, aeroponic raw materials are identical to intact plants.

AbstractNew treatment strategies for schistosomiasis should be evaluated, since resistant strains to the only available drug, Praziquantel, have already been described. Thus, we demonstrated antiparasitic effects of ethanolic extracts of Jatropha gossypiifolia and Piper arboreum on cercariae and adult worms of Schistosoma mansoni. The bioassays were performed at 0–10 000 μg mL−1 concentration for 0–72 h. Adult worms were stained with carmine to assess external and internal damage. The chemical screening was performed using high-performance liquid chromatography. P. arboreum displayed the best cercaricidal effect, with a 100% reduction in viability in just 60 min. The extract of J. gossypiifolia was more effective against adult worms, with 100% viability reduction of male and female worms after 12 and 24 h, respectively. P. arboreum and J. gossypiifolia were equally effective in inhibiting the oviposition of S. mansoni (93% reduction) and causing damage to internal and external structures in adult worms. Flavonoids were identified in both the extracts and phenolic compounds and amides only in P. arboreum. Thus, for the first time, it was proven that ethanolic extracts of P. arboreum and J. gossypiifolia leaves are biologically active against cercariae and adult worms of S. mansoni in vitro.
