Biomonitoring study of deoxynivalenol exposure and association with typical cereal consumption in Swedish adults

Deoxynivalenol (DON) is a mycotoxin of the trichothecene family commonly found in cereals infested with different Fusarium species. DON acts primarily on the gastrointestinal and immune system and is suspected to be an underlying agent causing several outbreaks of gastrointestinal disorder among humans, which prompts studies of human exposure and estimations of intake among populations. However, assessing human exposure to mycotoxins is associated with several difficulties. Therefore, a study was undertaken among adults (18-80 years) in a subgroup of Riksmaten, the Swedish national survey investigating dietary habits, examining both the association between urinary DON concentration and dietary intake of cereals, and estimations of daily DON intake. The results indicate that exposure to DON is common among Swedish adults, as this mycotoxin was detected in 292 out of 326 urine samples (90%) at levels ranging from non-detectable to 65.8 ng DON/ml urine with a median level of 2.9 ng/ml. Furthermore, urinary DON (ng/mg creatinine) was associated with intake (g/day) of total cereal grain as well as whole grain. Urinary DON was also significantly associated with breakfast cereals and porridge consumption (P<0.05). Estimated DON intake in this study ranged between 2.5 and 5,443 ng/kg body weight (bw). 1% of the individuals had estimated intakes above the group provisional maximum tolerable daily intake (PMTDI; 1 μg/kg), whereas the mean and median intakes of 159 and 84 ng DON/kg bw, respectively, were considerably below the PMTDI. Along with the toxicological profile of DON, no serious health implications are to be expected for the majority of Swedish adults, although a potential health concern remains for some high cereal consumers. In conclusion, biomonitoring could prove to be a valuable tool for observing DON exposure among populations.

Toxic element pollution is a serious global health concern that has been attracting considerable research. In this study, we elucidated the major routes of exposure to three toxic elements (mercury, cadmium, and lead) and two essential elements (manganese and selenium) through diet, soil, house dust, and indoor air and assessed the potential health risks from these elements on women from the coastal area of Miyagi prefecture, Japan. Twenty-four-hour duplicate diet, house dust, soil, and indoor air samples were collected from 37 participants. Cd, Pb, Mn, and Se concentrations were measured using inductively coupled plasma mass spectrometry, and Hg concentrations using cold vapor atomic absorption spectrometry. We found that soil and house dust were the primary reservoirs of these elements. Diet contributed most strongly to the daily intake of these elements, with mean values of 0.72, 0.25, 0.054, 47, and 0.94 μg/kg/day for Hg, Cd, Pb, Mn, and Se, respectively. The mean hazard quotient of Hg was 1.53, indicating a high potential health risk from Hg exposure in daily lives. The intakes of other elements were below the tolerable limits. Future studies with a larger sample size are warranted to confirm our findings.

ABSTRACT Aflatoxin B1 (AFB1) is the most potent of the dietary aflatoxins, and its major metabolite, aflatoxin M1 (AFM1), is frequently found in the breast milk of lactating mothers. The aim of this study was to assess the occurrence and factors associated with AFM1 contamination of breast milk collected from lactating mothers in Lebanon. A total of 111 breast milk samples were collected according to the guidelines set by the World Health Organization. Samples were analyzed with a competitive enzyme-linked immunosorbent assay between December 2015 and November 2016. A survey was used to determine the demographic and anthropometric characteristics of participating lactating mothers. Dietary habits were assessed using a semiquantitative food frequency questionnaire. Mean (±standard deviation) concentration of AFM1 in the breast milk samples was 4.31 ± 1.8 ng/L, and 93.8% of samples contained AFM1 at 0.2 to 7.9 ng/L. The mean concentration of AFM1 was significantly lower (P &lt; 0.05) in fall and winter (4.1 ± 1.9 ng/L) than in spring and summer (5.0 ± 1.7 ng/L). None of the samples exceeded the European Commission regulation limit (25 ng/L) for infant milk replacement formula. AFM1 contamination was significantly associated (P &lt; 0.05) with the daily consumption of white cheeses but not with the consumption of meat or cereal products. No significant association (P &gt; 0.05) was observed between AFM1 concentrations in breast milk and anthropometric sociodemographic factors (age and level of education) or the governorate of residence of the nursing mothers. The mean AFM1 estimated daily intake was found to be 0.69 ng/day/kg of body weight. Although the incidence of AFM1 contamination was low, our first-of-its-kind study highlights the importance of conducting investigations on mycotoxin contamination in breast milk and of developing protection strategies to tackle the exposure of infants to this potent chemical hazard.

Objective. Street-vended foods offer numerous advantages to food security; nevertheless, the safety of street food should be considered. This study has investigated the level of potential toxic metal (Pb, Cd, Hg, Sb, Mn, and Al) contamination among street-vended foods in Benin City and Umunede. Methods. Twenty street food samples were purchased from vendors at bus stops. Metals were analyzed with atomic absorption spectrophotometry. The methods developed by the US EPA were employed to evaluate the potential health risk of toxic metals. Results. The concentrations of the toxic metals in mg/kg were in the range of Pb (0.014–1.37), Cd (0.00–0.00017), Hg (0.00–0.00014), Sb (0.00–0.021), Mn (0.00–0.012), and Al (0.00–0.22). All the toxic metals except Pb were below permissible limit set by WHO, EU, and USEPA. The daily intake, hazard quotient, and hazard index of all toxic metals except for Pb in some street foods were below the tolerable daily intake and threshold value of 1, indicating an insignificant health risk. Total cancer risk was within the priority risk level of 1.0E-04 but higher than the acceptable risk level of 1E-06. Conclusion. Consumption of some of these street foods is of public health concern.

Introduction: Exposure to mercury (Hg) by consumption of fish is a recent health concern. So, it is important to evaluate the health risks related to canned fish consumption. The purpose of this study was to investigate the potential health risk based on Hg concentration in people who consumed canned fish with a probabilistic approach in Isfahan City, the central province in Iran. Materials and Methods: In this study, 20 popular brands of canned fish prepared in Iran and other countries were selected and analyzed for Hg concentration with atomic absorption spectrometer. The results were compared with the European Communities and JECFA guidelines. Then, a probabilistic method with Monte-Carlo simulation was used to assessment the Provisional Tolerable Daily Intake (PTDI) and the Hazard Quotient (HQ) for consumers in Isfahan City. Results: The average Hg concentrations in samples were 0.251 ± 0.204 and 0.189 ± 0.152 µg/g in canned fish of Iran and other countries, respectively. The Hg level was found below the guideline limit for European Communities and JECFA. The estimated PTDI was 0.037 µg person/day and HQ was 0.074. Conclusion: The results indicated that canned fish available in the markets of Iran did not have a health risk for adults. Moreover, canned fish consumption has a possible influence on the risk estimate and its risk should be assessed  for vulnerable groups.

Background. Cereal grains and their products are among the basic components of the diet all over the world. Their consumption varies depending on the dietary habits in each country. Apart from ingredients valuable from a health point of view, these products can also be a source of contaminants. The European Food Safety Authority (EFSA) included these foodstuffs in the group of main contributors to the dietary exposure to nickel by different groups of the population. Objective. The aim of the studies was to determine the nickel content in cereal grains and selected cereal products commercially available in Poland and to assess the exposure of consumers to nickel intake with these foodstuffs. Material and methods. A total of 56 samples of cereal grain and cereal products from trade were tested. Nickel content was determined after microwave mineralization of the samples by atomic absorption spectrometry with a graphite furnace atomization (GFAAS). The estimated exposure for the different groups of the population was assessed and compared with the Tolerable Daily Intake (TDI) value established by the European Food Safety Authority based on an updated risk assessment at 13 μg/kg body weight (b.w.) per day. Results. The mean (MB) and 95th percentile (MB) nickel content in investigated samples of cereal grains and their products were 0.66 mg/kg and 1.93 mg/kg. The mean content of Ni in the analyzed samples of grains was 1.16 mg/kg. Obtained results in this group of products ranged from 0.10 mg/kg for rye to 4.80 mg/kg for millet. In the group of grain products, the mean (MB) concentration of nickel was 0.61 mg/kg (95th percentile (MB) 1.84 mg/kg). The highest nickel level was determined in the samples of bran, groats, and flakes compared to other grain-based products. The highest content of Ni in the group of cereal products was found in the samples of roasted buckwheat 1.81 mg/kg and oat flakes 2.53 mg/kg. Significantly lower nickel content was observed in barley groats as well as for pasta and flour. The estimated average exposure of adults and children to nickel intake with grains and grain-based products ranged from 1.1% to 13.4% of TDI and it does not pose a health risk for consumers. Conclusions. Based on the obtained results, it was found that the nickel content in the tested samples of cereals and their products does not pose a health risk, even though the contamination of individual samples was significant.

AbstractGeophagic clay consumption, which is an age-long cultural practice by humans and animals in many parts of the world, and particularly in Nigeria, may have long time health effects on the consumers. This is particularly so because of the relatively high concentration of harmful minerals and toxic elements. This study sought to determine the mineralogical and trace element compositions of geophagic clay in Uteh-Uzalla area, which is underlain by the Benin Formation of Oligocene to Miocene age, in order to evaluate the potential health risk associated with the consumption of the clay. Sixteen clay samples were collected from mine face profiles of an open pit, analysed for mineral and trace element compositions, using x-ray diffraction technique and ultra-trace inductively coupled plasma mass spectrometry (ICP-MS) methods, respectively. The mean mineral concentration in % includes kaolinite, quartz and smectite (64.88, 19.98, and 9.54), respectively, among other minerals. And the mean concentrations in mg/kg for Cu (15.0), Pb (14.4), Zn (30.9), Co (8.9), Mn (39.4) and Th (10.5) among other elements were found in the clay. From the trace elements results when compared with health risk indices by Agency for Toxic Substances and Diseases Registry (ATSDR): Minimum Risk Level, recommended daily intake and estimated daily intake, it was found that the elements are far above the daily oral intake requirement. Also, considering the relatively low pH (acidic) values that were exhibited by the clays, harmful minerals and elements contained in the clay may be bioavailable in the internal system among those who are frequently involved in the consumption of the clay.

Mining is one of the major sources of heavy metals in most developing countries and this has become a major health concern to the populace due to the possibility of food contamination with these heavy metals. We investigated the levels of titanium (Ti), manganese (Mn), nickel (Ni), copper (Cu), zinc (Zn), lead (Pb), rubidium (Rb), strontium (Sr), selenium (Se), niobium (Nb) and vanadium (V) in 64 soil and  320 samples of regularly consumed staple food crops and vegetables (cassava tubers, white yam, groundnut seed, fluted pumpkin leaf and Z. zanthoxyloides from farms around Enyigba mining sites in Ebonyi State and the possible health risks due to consumption of these staple foods. Energy dispersive X-ray fluorescence (EDXRF) was used in determination of the metals in the samples respectively. The mean levels of the metals except Mn, Pb and Zn were below the threshold limits for agricultural soils. Our data also showed that food crops cultivated around this sites bio-accumulated toxic levels of these metals above the recommended legal limits set by WHO, NAFDAC and CODEX especially Pb, Zn, Cu, Ni, Se, and Rb. This perhaps may have potential health implication on the local dwellers. The estimated daily intakes (EDI) of the toxic metals through the consumption of the crop samples were below the maximum tolerable daily intake (MTDI) except lead and nickel by eating of groundnut, cassava and yam. Furthermore, the target hazard quotient (THQ), computed based on EDI of the heavy metals were above unity for the metals due to cassava, yam and groundnut consumption, suggesting significant health risk. Raised amounts of heavy metal in cassava and groundnut samples as revealed by their metal extraction coefficient computed in this study hint that they may be used as bio-indicators of ecological, health hazards and phytoremediators of Mn, Zn, Cu, and Pb.

Introduction: An instance of fish deaths in marine waters surrounding some communities in Bonny and Andoni Local Government Areas in Rivers State was reported in March-April 2020. This study investigated trace metals hazard in muscle of Tilapia (Sarotherodon melanotheron) and Silver Catfish (Chrysichthys nigrodigitatus) from Okulu River, Rivers State, Nigeria. Materials and Methods: Tissues of 24 samples of Sarotherodon melanotheron and Chrysichthys nigrodigitatus obtained from the river were analyzed using atomic adsorption spectrophotometer, and the health risk was estimated based on estimated daily intake (EDI), target hazard quotient (THQ), and total target hazard quotient (TTHQ). Results: The concentration (mg/kg) of iron, zinc, manganese, copper, cadmium, lead, and chromium in both species ranged 4.00 – 197.30, 9.20 – 35.30, 0.20 – 5.00, 0.00 – 73.10, 0.00 – 1.30, 0.00 – 54.70, and 0.00 – 0.50, respectively. The EDI of trace metals resulting from the consumption of both fish species was higher than the permissible tolerance intake (PTI) mg/day/60kg body weight. The THQ and TTHQ were < 1, indicating that the consumption of this food fish portends no known health concern. However, the carcinogenic risks exceeded the threshold level of 10-6 - 10-4, thus, reflecting carcinogenic tendency. Based on the results obtained, it must be ensured that the effluents discharged into open water bodies meet the recommended limits. Conclusion: There is a need to create awareness among consumers of food fish in the study area. There should be periodic monitoring of trace metals in surface waters and its food fish population to forestall potential health impact on humans.

The study evaluated the concentrations of some chemical elements namely Copper (Cu), M anganese (Mn), Zinc (Zn) , Chromium (Cr), Nickel (Ni) and Lead (Pb) in the mangrove oyster ( Crossostrea gasar ) and water from Golubo creek in the Niger Delta region of Nigeria, to ascertain the impact of these metals on the investigated ecosystem. T he evaluation was carried out using Atomic Absorption spectroscopy technique. The mean concentrations of the aforementioned elements in C.gasar were 1.06 mg/kg, 0.38 mg/kg, 50.13 mg/kg, 0.14 mg/kg, 0.39 mg/kg and 1.55 mg/kg , respectively while the m ean concentrations of the elements in water were 0.65 mg/l, 0.04 mg/l, 16.47 mg/l, 0.86 mg/l, 0.85 mg/ l and 1.50 mg/l , respectively. Copper , Manganese, Zinc and Lead were bioaccumulated by C.gasar while the estimated daily intake of these elements ranged from 0.0056 mg/person/day for Chromium to 2.0 1 mg/person/day for Zinc . The M aximum Acceptable Risk values for Copper , Manganese, Zinc , Chromium, Nickel and Lead in C. gasar were 0.79, 0.05, 24.67, 0.94, 1.08 and 1.94, respectively. T he Toxicity Quotient values for the chemical elements in C.gasar ranged from 0.66 for Zinc to 0.93 for Chromium while in the case of water , the range was from 0.65 for Copper to 1.5 for Lead. With regard to health risk to man, the mean concentrations of the chemical elements in the oysters, did not exceed the Federal Environmental Protection Agency ( FEPA ) maximum allowable limit for chemical elements in food. The mean concentrations of these metals in the bivalve were , therefore , within health limits and therefore do not present an immediate health threat to consumers . It w as observed that the mean concentrations of the chemical elements in water were generally below the FEPA maximum allowable limits for chemical elements in drinking water with the exception of Lead . The study , thus , revealed that there is a potential health risk to man as a result of consuming Pb- contaminated water . Based on the findings from this study, it is advocated that the creek be closely monitored for toxic metals in order to avert the occurrence of episodic ecological accidents.

Abstract Background The prevention of metabolic syndrome (MetS) is a major public health concern in Japan. The effects of the relationship between eating behavior and nutritional intake on MetS remained unclear. To evaluate nutrition’s role in preventing or exacerbating MetS, we examined the associations among eating behavior, nutritional intake, and MetS for the baseline study in the cohort subjects undergone health checkups. Methods Four thousand and four hundred forty-seven Japanese men and women were enrolled at the Saku Central Hospital. They received an anthropometric and clinical examination and were assessed for present illness, lifestyle factors such as physical activity, smoking, drinking, and dietary habits at the enrollment. Eating behavior was analyzed by the Sakata’s Eating Behavior Questionnaire. Dietary assessment was made using a brief self-administered diet history questionnaire. Two thousand and six hundred two men and 1844 women aged more than 20 were analyzed. Results The mean age in men and women were 59.2 and 58.4 years old and the mean body mass index (BMI) were 23.7 and 22.3 kg/m2, respectively. The percentages of MetS were 20.6 in men and 6.1 in women. In some nutrients, significantly higher energy-adjusted intakes in subjects without MetS than with Mets appeared both in men and women after age adjustment. After adjusting by age, energy-adjusted intake beverages in men and cereals in women were significantly higher in subjects with MetS than those without MetS. The scores of all the categories in eating behavior were significantly worse in subjects with MetS than those without MetS. Conclusions The differences in dietary intake between subjects with Mets and without Mets were relatively small. The scores of all the categories in eating behavior were worse in subjects with MetS than without MetS. It was suggested that the problem lay in the quality of diet, not in the quantity, caused by bad eating habits. The potential influence of eating behavior and nutritional intake on MetS was presented in men and women.
