Apples are widely used by the population as a fresh and processed product at any time of the year. The relevance of the experiment is the protection of orchards from apple pests and the cultivation of high-quality products. Over the years of experiments, the main types of pests that are found in gardens and cause the greatest damage, as well as their morphological characteristics, have been studied. Laspeyresia (Carpocapsa) pomonella L. is recognized as the most harmful apple tree in our region.

Abstract At present, embryo morphology assessment based on the observation of the embryonic morphological characteristics at several specific time points has been mainly used for selecting the high-quality embryo. However, we all know that embryo development is a dynamic process. Many research results on the correlation between the embryo morphokinetic parameters and embryo quality and development potential were inconsistent. With the help of time-lapse imaging, the development processes and outcomes of a total of 365 embryos were cultured and analyzed in this study. The results showed that the mean tPNf and t2 of the high-quality embryo were significantly shorter than the low-quality embryo; the mean t2PBe and tPNa of the high-quality embryo from the implantation group were significantly shorter than those from the non-implantation group. In addition, based on the quartile grouping of each morphokinetic parameter, the embryos that had 21.15≤tPNf≤25.30 value were significantly higher in embryo quality when compared with the embryos that had the tPNf values outside the range on Days 3. Similarly, the embryos that had values of t2≤25.60 were significantly higher in embryo quality than those with outside the range values on Days 3. Thus, we demonstrated that the morphokinetic parameter evaluated using a time-lapse embryo monitoring system can predict the embryo quality, and be benefit for the selection of the high-quality embryos and improvement for the implantation success of the patient in assisted reproductive technologies.

The authors developed a method for the automated detection and calculation of quantitative parameters of dimples of ductile fracture on the digital images of fracture surfaces obtained at different scales. The processing algorithm of fractographic images was proposed, which allowed high quality recognition of the shape and size of dimples to be achieved, taking into account the morphological features of their digital images. The developed method for identifying dimples of various physical and morphological characteristics was tested on the VT23M alloy. The test results showed that the method meets the quality requirements for the automated diagnostics of fracture mechanisms of titanium alloys.

Beacon apple tree (Malus domestica Borkh.) variants derived from gamma-irradiated scionwood on Columbia Crab seedling (CC) rootstocks were compared to both standard Beacon/clonal rootstocks, standard Beacon/CC rootstocks and Columbia Crab seedling trees. Stem or root measurements of trees classed as induced spur-type dwarf variants/CC rootstocks had larger bark to xylem ratios than trees from other growth habit classes. Stem and root bark to xylem ratios have the potential to be used as juvenile selection criteria in screening for spur-type dwarf variants in irradiation-exposed apple tree populations.

Abstract We present the data on proliferation corresponding to 10 years of continuous incubation in vitro of papaya shoots, and propose a reliable method for long-term micropropagation for papaya, using two types of explants: Microshoots from somatic embryos, and from axillary buds of papaya. Three different media were assayed. The proliferation medium (PPRM) allowed to maintain papaya shoots under continuous proliferation during 20 years, maintaining a consistent behaviour. Most of the shoots developed in PPRM rooted during the incubation, and after acclimated easily, maintaining the morphological characteristics of the parental plants, flowering and setting fruits normally. The PPRM medium consist in MS medium supplemented with NAA (0.1 mg l-1), BA (0.5 mg l-1), GA3 (0.5 mg l-1) and Adenine sulphate (40 mg l-1). The average multiplication rate was higher than 20 shoots per explant along the long-term assay. The elongation medium (PELM), was designed to recover shoots with a poor growth, and allowed the development of high quality shoots ready for rooting, and consist in a MS basal medium supplemented with NAA (0.1 mg l-1), Kin (0.5 mg l-1) and GA3 (1 mg l-1). The rooting medium (PROM) was designed to induce high quality roots from non-rooted shoots and consist in a half strength MS medium plus IBA (1mg l-1). On PROM, agar can be exchanged for expanded vermiculite. Acclimation took place inside an acclimatization tunnel under progressive hydric stress. After 4 weeks, the plant recovery rate was 90% for plants maintained under continuous proliferation during ten years.

As a result of studying the antifungal activity of biological drugs against fungi of the genus Fusarium, causative agents of rot of roots and fruits of the apple tree, both weak and very strong mycoparasitism were noted, but in most variants of the experiment, competition for area of nutrition prevailed. Under the influence of some drugs, the shape, edge and color of the fungus colony changed. In general, for the entire sample of pathogens, the best bioagents were antagonists of the drug Trichocin, WP, which suppressed all 5 strains with Biological efficiency (BE) 50-90 % and showed hyperparasitism in 1 strain; and the drug Alirin B, WP, which inhibited the growth of 3 strains with BE 56-85 %, showing antibiosis or fungistatic antibiotic antagonism with the formation of a “sterile” zone.

Wilting of branches and leaves was observed on 4-5 year old apple trees of the varieties Delicious and Fuji in orchards located in Wushan, Gansu Province, China in April 2018. Subsequently, the stem vascular tissue and woody xylem became discolored and necrotic. The stem dieback expanded rapidly to the entire vasculature of the branches. Finally, the epidermis of the stem bases split and was covered with light pink mold. For the pathogen isolation, 25 symptomatic stems were collected from 25 symptomatic trees in 3 individual orchards. Fragments (approximately 0.5 cm in length × 0.5 cm in width) of symptomatic stems were surface sterilized and individually transferred to Petri dishes containing potato dextrose agar (PDA), and incubated for 4 days at 25°C. Five types of isolates with distinct morphological characteristics (PJ1 to PJ5) were obtained from the 25 symptomatic stems after the single spore inoculation and sub-culture. The isolation frequency of PJ1, PJ2, PJ3, PJ4 and PJ5 types was 11%, 8%, 100%, 4% and 13%, respectively, in the 25 symptomatic stems. A spore suspension of PJ1, PJ2, PJ3, PJ4 and PJ5 types was prepared by adding 5 ml of sterile distilled water in the 14-day old culture colonies and filtered through 0.22 mm Millipore membranes, and the final concentration was adjusted to 108 per ml for inoculation. Detached healthy apple stems (15 cm in length) were surface-disinfested and used to evaluate the pathogenicity of PJ1 (7 isolates), PJ2 (5 isolates), PJ3 (32 isolates), PJ4 (2 isolates) and PJ5 (9 isolates) by dipping the stems into sterilised tubs containing the spore suspension (108 per ml) of each isolate. Apple stems dipped in sterile distilled water served as the control. Each control and treatment were repeated 3 times. At day 15 and 35, the stems infected with the spore suspension of PJ3 isolates developed symptoms that were similar to those observed in the apple orchards. However, the other four types (PJ1, PJ2, PJ4 and PJ5) exhibited either no symptoms or different symptoms from those observed in the apple orchards. There were no symptoms on the control stems. After the colony of the pathogen (PJ3 type) was re-isolated from the infected stem bases 35 days inoculation. The PJ3 type isolates with same morphological characteristics as the original PJ3 type isolates were used for further examination and identification. After 4 days of incubation on PDA, the colonies of PJ3 type isolates developed velvety aerial mycelia with white or light pink color when they were viewed from the front/top side of the PDA and orange-red color when they were viewed from the reverse/bottom side. After 14 days of incubation, the color in the centre of the colonies changed to yellow green in the front view and carmine red in the reverse view of the plates. Three types of spores (microconidia, macroconidia and chlamydospores) were observed after incubation of PJ3 type isolates for 14 days. The size (width and length) of 30 conidia in each of PJ3 type isolates was measured and averaged. The microconidia were abundant on aerial mycelia and limoniform, oval or pyriform with 0-1 septa. Their size ranged from 1.94 μm to 8.05 μm in length and 1.48 μm to 3.62 μm in width. The macroconidia were falciform and curved in shape, mostly with 3-5 septa and a size ranging from 13.52 μm to 22.43 μm in length and 2.31 μm to 3.87 μm in width. The chlamydospores were spherical, intercalary and formed in chains on PDA plates. These morphological characteristics indicate that the PJ3 type isolates could be Fusarium tricinctum (Chen et al. 2019; Aktaruzzaman et al. 2018). To confirm the morphological identification, the sequences of internal transcribed spacer (ITS), transcriptional enhancer factor-1 (TEF-lα) and ribosomal RNA large subunit gene (LSU) of the representative isolate PJ3-3 selected from the PJ3 type isolates with same morphological characteristics were sequenced and used for molecular identification (Laurence et al. 2011; Abd-Elsalam et al. 2003; Miller et al. 1996). The sequences of ITS, TEF-lα and LSU of the PJ3-3 isolate were deposited in NCBI database with the accession numbers of MZ799356, MZ820045 and MZ820044, respectively. In BLAST analyses, the obtained sequences of the PJ3-3 isolate showed 99.47%, 100% and 99.01% identity to the corresponding region of F. tricinctum ITS (JX179207.1: 3-566 Fusarium tricinctum isolate Fyx 1), TEF-lα (MK032320.1 F. tricinctum isolate ZD3) and LSU (KC311496.1 Fusarium tricinctum isolate L12), respectively. The phylogenetic analysis clustered the PJ3-3 isolate sequences within the same clade with ITS, TEF-lα and LSU sequences of F. tricinctum isolates. Thus, the PJ3-3 isolate was identified as F. tricinctum based on the pathogenicity tests, morphological characteristics and molecular analyses. Previously, the symptoms of xylem browning and dieback were observed in the twigs of wild apple trees that were collected from wild apple forests, and the species F. avenaceum, F. solani, F. tricinctum, F. proliferatum, and F. sporotrichioides were isolated from diseased wild apple trees (Chen et al. 2019). Only F. avenaceum, F. solani, F. proliferatum, and F. sporotrichioides were reported as the pathogens causing the disease symptoms of xylem browning and dieback in wild apple trees in Xinjiang, China (Chen et al. 2019). In our present study, we found that F. tricinctum can cause stem vascular and woody xylem browning, wilting, and dieback in the apple tree varieties Delicious and Fuji. These are new symptoms discovered in our present research and different from the previous paper (Chen et al. 2019). Therefore, to our knowledge, this study is the first report of F. tricinctum causing a new disease on apple trees in China following Koch’s postulates. Our findings are important for the management of apple disease and protect apple trees in the future.

ABSTRACT The aim of this study was to evaluate the physical and morphological characteristics of the sperm-rich fraction of jackass semen. To this end, 130 ejaculates from five Pêga jackasses were collected using an open model artificial vagina. The sperm-rich fraction was collected using the split-ejaculate method and assessed for the number of mounts/ejaculate, for physical and morphological characteristics of the semen and number of doses produced/ejaculate. It was observed that all characteristics evaluated differed among the five jackasses, except for the head defect rates. The mean values obtained for the jackass sperm-rich fraction collected were: number of mounts/ejaculate - 1.27; semen volume - 20.21mL; motility - 84.53%; vigor - 4.46; motility after dilution - 80.10%; sperm concentration/mL - 894.38 x 106; total sperm/ejaculate - 16.14 x 109; number of insemination doses/ejaculate (400x106 motile sptz) - 33.39; number of insemination doses/ejaculate (800 x 106 motile sptz) - 16.69; and percentage of normal sperm - 90.46%. Thus, in the present experiment the split-ejaculate method using an open artificial vagina worked well with the jackasses, and the sperm-rich fraction of the ejaculate of Pêga jackasses had high quality and sperm concentration, allowing its use for semen processing without reducing the number of insemination doses produced per ejaculate.
