A search new antibacterial compounds should continue to be done as a raw material of antibiotics. The purpose of this study, which is to get kind of marine fungi antibacterial activity and get the value of MIC and MBC. The study was conducted from the sampling sponge, marine fungi isolation, extraction, preparation of test bacteria, antibacterial activity testing and analysis of MIC and MBC. Data was analyzed by measuring the diameter of inhibition zone are formed. The results obtained, that debris ethanol fraction from marine fungi J3 can inhibit the growth of E. coli and S. aureus at a concentration of 100 mg/L to 100.000 mg/L. At the debris ethyl acetate fraction able to inhibit concentration of 1.000 mg/L to 100.000 mg/L. At the filtrate ethyl acetate fraction was able to inhibit 10.000 mg/L to 100.000 mg/L. For both types of test bacteria, MIC and MBC values are indicated by the highest debris ethanol fraction and lowest by the fraction of ethyl acetate filtrate.

Peel of red dragon fruit (Hylocereus polyrhizus) is one of the plants used as an antibacterial agent as it contains saponin triterpenoid compounds, flavonoid compounds, and alkaloid compounds which can have antibacterial activity. This research aims to determine the antibacterial effect of n-hexane, ethyl acetate, and ethanol fraction of red dragon fruit’s peel against Escherichia coli and Staphylococcus aureus by the concentration of 10mg/ml, 20mg/ml, 40mg/ml, 80mg/ml dan 160mg/ml. This research was conducted by using laboratory experiments. The simplicia was macerated with 96% ethanol and fractionated by n-hexane and ethyl acetate. The phytochemical screening of the fraction was n-hexane fraction containing saponin and alkaloid, while the ethyl acetate fraction contained saponin and flavonoid. Kanamycin was used as a positive control, while DMSO was used as a negative control. According to this research, the MIC value of ethanol fraction, n-hexane fraction, and ethyl acetate fraction were 80mg/ml, 20mg/ml, and 80mg/ml, respectively, for E. coli and all fractions were 10mg/ml for S. aureus. Based on the average diameter of the inhibition zone, the largest diameter zone in E. coli was ethyl acetate fraction with 160mg/ml concentration  that was  10,33mm. Meanwhile, in S. aureus n-hexane fraction, it was 160mg/ml, which was 11,20mm. This result showed that the n-hexane fraction has good gram-positive activity while the ethyl acetate fraction has good activity on gram-negative.

Abstract Endosymbiont fungi from marine sponges are a rich source of medicinally active compounds. Indonesia has a huge number of marine sponges. This research was conducted to determine the antibacterial potential of endosymbiont fungi isolated from marine sponges collected from Kotok Kecil Island, Seribu Islands. The screening was conducted with two methods, the modified GIBEX and disc diffusion. Species of marine sponges were isolated, namely Petrosia sp., Stylissa carteri, Cinachyrella australiensis, Callyspongia sp., Petrosia nigrians, and Stylissa massa, and obtained 9 isolates of endosymbiont fungi. The GIBEX test against Escherichia coli and Streptococcus mutans showed that ethyl acetate extract had strongest antibacterial activity. Disc diffusion test of ethyl acetate extract, five isolates had antibacterial activity against S. mutans and four against E. coli. The isolate that showed the strongest antibacterial activity was from Petrosia sp. Which has an inhibition zone of 8.4 mm against E. coli and 7.45 mm against S. mutans. The main active compounds from the isolate of Petrosia sp. are butylhydroxytoluene and phthalic acid di-(2-propylpentyl) esters. Based on this study, we concluded that the endosymbiont fungi of marine sponges are potential to be developed for further development as source of antibacterial agents.

Objective: This research aims to analyze the ability of robusta coffee leaves fraction extract to inhibit the growth of Staphylococcus aureus and Escherichia coli and also determine the minimum inhibitory concentration (MIC). Methods: Antibacterial activity evaluated by the disc diffusion method observed in four types of fraction of extract robusta coffee leaves (n-hexane, ethyl acetate, ethanol, and water). Each extract divided into three various concentrations, 5%, 10%, and 15%. Determination of antimicrobial activity in vitro by the disk diffusion method. Results: Ethyl acetate fraction of coffee leaves extract produced the largest diameter zone of inhibition of bacterial growth compared to other extraction fractions of 17.28 mm in E. coli and 18.58 mm in S. aureus. The MIC of coffee leaves extract fraction water, ethyl acetate, and n-hexane on E. coli and S. aureus is 5%, while the fraction ethanol MIC is 10%. Conclusion: The antibacterial effect of ethyl acetate fraction of coffee leaves extract showed an antibacterial effect that was better than the fraction of n-hexane, ethanol, and water.

This study aims to determine the antibacterial activity and TLC-bioautography profile of the active fractions of Muntingia calabura L. leaves against Staphylococcus aureus. Muntingia calabura L. leaves were macerated with ethanol 96% then fractionated with n-hexane, ethyl acetate, and methanol solvent, respectively. The antibacterial activity was tested by the Kirby-Bauer method to determine the most active fraction and the lowest concentration that inhibited the growth of Staphylococcus aureus. TLC-bioautography was tested using chloroform: ethyl acetate (2:8) as the mobile phase and silica gel F254 as the stationary phase. Antibacterial activity test of n-hexane and ethyl acetate fractions at a concentration of 10% w/v showed activities with inhibition zone diameter of 0.33±0.288 and 9.66±5.77 mm, respectively. At the same time, the methanol fraction showed no activity. The lowest concentration of ethyl acetate fraction which still showed the inhibition zone was 0.312% w/v. The TLC-bioautography profile showed active spots with an Rf value of 0.82 and had an inhibitory zone diameter of 4.013±0.864 mm. It can be concluded that the ethyl acetate fraction was the most active fraction that inhibited the growth of Staphylococcus aureus and had one active spot on the bioautography test.

Frangipani flowers (Plumeria acuminata L.) Flower is one of the plants that has antibacterial properties. Compounds that act as antibacterial are flavonoids and tannins. The purpose of this study was to prove the antibacterial activity of ethanol, chloroform, and n-hexane fractions of Frangipani flowers (Plumeria acuminata L.) flower as antibacterial against E. coli and S. aureus with Bioautography.The process of extracting plumeria acuminata L. flowers was carried out by maceration method using 70% ethanol. The extract was continued with the fractionation process with ethanol, n-hexane and chloroform solvents. Antibacterial activity testing using the well method with concentrations of 20%, 40%, 60%, 80%, 100%, positive chloramphenicol control, and negative aquadest control. The compound content test used the Thin Layer Chromatography (TLC) method with the mobile phase of chloroform: methanol: water (2:5:3) (v/v).Bioautography test using the contact method, namely the elution TLC plate, was placed on the NA medium containing a bacterial suspension for 3 hours. The biggest antibacterial test results of ethanol fraction of plumeria acuminata L. flowers were at a concentration of 100% with a diameter of inhibition zone of 13.55 mm in E. coli while 14.15 mm S. aureus. The results of TLC showed the presence of flavonoids with a price of Rf 0.70 and tannin at a price of Rf 0.68. Bioautography results showed a inhibition zone with a price of Rf 0.70 in E. coli and S. aureus which are flavonoid compounds. Based on the results of the research that has been done it can be concluded that the ethanol fraction of white frangipani flowers has antibacterial activity against E.coli and S. aureus, while the chloroform fraction and n-hexane fraction do not have antibacterial activity. The active compounds that act as antibacterial are flavonoids and tannin, but the ones that provide antibacterial effects are flavonoids with an Rf value of 0.70. Penelitian terhadap ekstrak metanol, fraksi etil asetat, dan fraksi butanol dari rumput laut Eucheuma spinosum menggunakan metode Disc Diffusion Test dilakukan untuk mengetahui peranannya sebagai antibakteri Escherichia coli ATCC 25922, dan Staphylococcus aureus ATCC 6538. Sebagai kontrol digunakan ekstrak (negatif) pelarut metanol, dan Amoxicilin. Konsentrasi sampel uji ditetapkan 10.000 ppm. Hasil pengujian menunjukkan adanya  aktivitas antibakteri yang ditandai dengan terbentuknya zona hambat terhadap bakteri Escherichia coli ATCC 25922.  Besarnya zona hambat secara berurutan untuk ekstrak metanol, fraksi  butanol, dan fraksi etil asetat, ialah : 6,98 mm, 7,85 mm, dan 7,88 mm.  Nilai zona hambat tersebut, relatif sama untuk setiap sampel uji. Sampel uji juga menunjukkan terdapat aktivitas antibakteri, ditandai dengan terbentuknya zona hambat terhadap bakteri Staphylococcus aureus ATCC 6538.  Besarnya zona hambat secara berurutan untuk ekstrak metanol, fraksi  butanol, fraksi etil asetat,  dan kontrol negatif pelarut metanol, ialah : 7,42 mm, 14.40 mm, 7,90 mm, dan 6,75 mm.  Zona hambat fraksi butanol lebih tinggi dibanding yang lain, ini menunjukkan bahwa aktivitas antibakteri Staphylococcus aureus pada fraksi butanol lebih tinggi dibanding fraksi lainnya.Kata kunci:  Antibakteri;  Eucheuma spinosum;  Escherichia coli; Staphylococcus aureus Antibacteria Activities of Extract Sea Algae  Eucheuma spinosum Fresh from Nain Island Waters North Sulawesi  ABSTRACTA research on antibacterial activity of methanol extract, ethyl acetate, and butanol fraction from Eucheuma spinosum seaweed using the disc diffusion method to know the role as  antibacterial activity was tested against Escherichia coli ATCC 25922 and Staphylococcus aureus ATCC 6538. Methanol was used as negative control while amoxicillin was used as positive control. The concentration of the test sample was set at 10,000 ppm. The antibacterial activity was indicated by the formation of an inhibition zone against the tested microorganisms. The inhibition zones against E.coli for methanol extract, butanol fraction, and ethyl acetate fraction were 6.98 mm, 7.85 mm, and 7.88 mm, respectively.  The value of the zone of inhibition is relatively the same for each sample.  In addition, the inhibition zones of methanol extract, butanol and ethyl acetate fraction against Staphylococcus aureus were 7.42 mm, 14.40 mm, 7.90 mm, and 6.75 mm, respectively. The inhibition zone of the butanol fraction is higher than the others, this indicated that the antibacterial activity for butanol fraction is higher than other fractions. Keywords: Antibacterial activity; Eucheuma spinosum; Escherichia coli; Staphylococcus aureus

This study was conducted to determine the activity of Jatropha gossypifolia L (jarak merah’s) leaf extract as an antioxidant and antibactery against acne causing bacteria (Propionibacterium acnes). To obtain an J. gossypifolia L jarak merah’s leaf extract by maceration using 96% ethanol, and then fractionated with n-hexane and ethyl acetate. Testing of antioxidant activity by DPPH method and antibactery activity test by the paper disc method. Antioxidant activity test has done for ethanol fraction, ethyl acetate fraction and n-hexane fraction, while testing for antibacterial activity performed on fractions active as antioxidant. The test results demonstrate the antioxidant activity of ethyl acetate fraction and the ethanol fraction as a fraction very strong with IC50 value of 22.33 ppm for ethyl acetate fraction and 41.04 ppm for ethanol fraction. Then the fraction of ethyl acetate and ethanol fraction antibactery activity test, the result is known that both factions have the ability to inhibit the growth of bacteria Propionibacterium acnes demonstrated by the inhibition zone is formed. In 1000 ppm is formed diameter of the greatest inhibition zone that is equal to 6 mm for ethyl acetate fraction and 5 mm for ethanol fraction.

The aims of the study are to evaluate bioactive compounds, antibacterial and cytotoxic potential of D. lasianthera. This orchid grows well all over New Guinea Island as an ornamental plant because of their beautiful flowers. Orchids also known rich of its phytochemical compounds which already used as a traditional medicine in many countries around the world. However, research in pharmacological fields is still limited. In this study, leaves and stem of D. lasianthera were powdered and extracted with ethanol followed by fractionated using n-hexane, ethyl acetate, and ethanol solvent. Extract as well as fraction were tested for phytochemical screening and determined antibacterial activity against Escherichia coli and Staphylococcus aureus using Disc Diffusion Method. Brine Shrimp Lethality Test (BSLT) was used to observe cytotoxic potential of leaves and stem extract and fraction at 10, 50, 100, 250, 500, 750, and 1000 ppm. The results showed that the leaves and stem extract contained tannin and alkaloids, separately. The ethanol extract of D. lasianthera showed 7.35 mm (leaves) and 7.43 mm (stem) inhibition zone against S. aureus Furthermore, the maximum inhibition zone of ethanol fraction of leaves and n-hexane fraction of stem were 8.42 mm and 8.10 mm separately. The LC50 of stem extract and fractions in these study were 699.3 ppm (ethanol extract), 602.1 ppm (ethyl acetate), 329.6 ppm (n-hexane fraction) and 676 ppm (ethanol fraction), whereas for leaves, only ethyl acetate fraction has toxict activity with an average LC50 833.2 ppm. Key words: D. lasianthera; phytochemical screening; antibacterial; cytotoxicity; Papua.

Generally, Gluta renghas L. is known because of its very toxic latex which can cause hard irritations to skin. Although, rengas latex has efficacy as an antibacterial agent. Related to previous research, it has been reported that there are urisol, rengol, glutarengol, laccol, and thitsiol in rengas latex. The woody trunk was reported to contain flavonoid, benzenoid, lipid, and steroid compounds. The leaves, were reported shows that the leaves of G. renghas contain an anticholinesterase substance and can be reduced the rate of hydrolysis of acetylcholine. Extraction of rengas leaves has been carried out. It has been shown phenols, steroids, and coumarins compounds in methanol extract, flavonoids, phenols, saponins and alkaloids compounds in ethyl acetate extract, steroids and alkaloids compounds in hexane extract. In the present, cytotoxicity and antibacterial activity have been tested. The results show that the great cytotoxicity activity by ethyl acetate extracts which have an LC50 value is 123,718 µg/mL (R2 0.9822), while the great antibacterial activity shown by methanol extract 1,000 µg/mL with a diameter of inhibition zone 19.02 mm (S. aureus) and 16.06 mm (E. coli).

Aims: This study was aimed at evaluating the antimicrobial activities of fractions of Rauwolfia vomitoria against some isolates including; Salmonella typhi, Escherichia coli, Candida albicans, Aspergillus niger, Microsporon canis, and Trichophyton rubrum. Methodology: Primary extraction was done on the dried and pulverized plant samples (leaves, barks, and root) using methanol, after which the crude extracts was fractionated using butanol, ethyl acetate, and n-hexane respectively. The samples were duly labelled according to the plant parts and solvents used. Agar diffusion and dilution methods on Mueller-Hinton agar (MHA) and Sabouraud dextrose agar (SDA) where used to determine the minimum inhibitory concentrations (MIC) and minimum bactericidal and fungicidal concentrations (MBC/MFC) respectively. The percentage inhibition diameter growth (PIDG) of the extracts and fractions were also determined. Results: From the results, eleven (11) samples showed inhibitory activity on one or more of the test isolates. Extended spectrum activity was observed with five (5) of the samples, including; RVL ethyl acetate, RVL n-hexane, RVB crude, RVR crude, and RVR ethyl acetate against S. typhi, E. coli, and M. canis. Antifungal activity was observed only against M. canis. The ethyl acetate fraction of the leaves (RVLE) and crude methanol extract of the bark (RVBC) gave the least MIC both against M. canis at a concentration of 25 mg/ml, while the ethyl acetate fraction of the leaves (RVLE) at 12.5 mg/ml also against M. canis gave the least MBC/MFC. Using the inhibition zone diameters (IZD), the obtained PIDG showed apparently that the ethyl acetate fraction of the leaves (RVL ethyl acetate) was the most active against S. typhi (28.57%) and E. coli (20%). Conclusion: The study lends support to the traditional use of the plant especially in treating gastro intestinal tract and skin infections.
