A total of 25 samples of raw cow milk, were collected randomly from street vendors and supermarkets in Zagazig city, Egypt for detection of the probiotic potential of some isolated Lactobacillus strains from raw milk. The obtained results revealed that the Lactobacillus Strains were detected in all the examined samples. The isolated strains were slightly affected by pH 3 and bile condition (0.5%). The obtained results displayed resistance against Erythromycin, Gentamycin and Tetracycline. Meanwhile, Ampicillin, Chloromphnicol, Steptomycin and Vancomycin exhibited an effect against some strains. The isolated strains produced ?-hemolysis, NaCl tolerance was positive for these strains. Also, ? galactosidase was produced.

Background: This study reports an easy method of a veterinary drug investigation in raw milk, based on QuECHERS extraction followed by RP-HPLC-UV analysis. Use of this benchtop system was motivated by its availability and moderate cost relatively to other sophisticated methods such as LC-MS which are more efficient. Methods: This developed method has been optimized and then after validation according to EU legislation, it demonstrated good linearity with R²>0.997, acceptable peak resolution within a short time (<9.5 min) and good recovery of the analyzed drugs (OXY, ALZ and IVR, respectively 87.08, 99.02 and 92.01 %). Additionally, we applied the method to the analysis of cow milk, collected in Nouakchott, capital of Mauritania. Results: The obtained results indicated a mixed level of drug use according to targeted molecules. In 42% of sampled farms, the anti-parasitics IVR and ALZ were detected whereas the antibiotic OXY was detected in 50%. Conclusion: This investigation shows that 17% of the sampled farms exceeded European standards for IVR drug.

In this paper, an investigation on lactic acid bacterial isolates from ethnic goat raw milk samples were examined for their probiotic potential and safety parameters. For this purpose, isolated bacterial cultures were screened based on certain parameters viz., sugar fermentation, tolerance to temperature, salt, low pH, bile salts, and phenol resistance. After that, these bacterial cultures were more estimated in vitro for auto-aggregation, cell surface hydrophobicity, response to simulated stomach duodenum channel, antibiotic resistance, and antimicrobial activity. Besides, probiotic traits show the absence of gelatinase and hemolytic activity supports its safety. The isolate G24 showed good viability at different pH, bile concentration, phenol resistance and response to simulated stomach duodenum passage but it did not show gelatinase and hemolytic activities. Isolate G24 was susceptible to amikacin, carbenicillin, kanamycin, ciprofloxacin, co-trimazine, nitrofurantoin, streptomycin, and tetracycline. Isolate G24 also exhibited antimicrobial action against five common pathogenic bacteria, such as Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Listeria monocytogens, and Salmonella typhimurium. It displayed the maximum auto-aggregation, cell surface hydrophobicity to different hydrocarbons. Following molecular characterization the isolate G24 was identified as Enterococcus hirae with 16S rRNA gene sequencing and phylogeny. E. hirae G24 bears the excellent properties of probiotics.

Objectives: The aim of this study is to evaluate the microbial quality and associated health Methodology: An intensive study was carried out from January¬ to October 2019 at the Tangail district of Bangladesh to assess bacterial contamination of raw milk. A total of 60 samples were examined by following the standard bacteriological methods and the health impact was evaluated with the help of a semi-structured-based questionnaire. Molecular characterization of isolated bacteria was carried out by PCR. Results: Results revealed the mean values of TVC and TCC in raw milk, udder, oil, and utensil samples were 7.6×106, 8.8×106, 4.6×106, and 1.3×106 cfu/ml and 3.7×105, 1.4×106, 4.4×105, and 8.9×104 cfu/ml, respectively. The yeast and mold in raw milk, udder, and oil were 1.5×103, 1.8×103, and 1.3×102cfu/ml, respectively, and the mean values of E. coli in the above-mentioned samples were 1.9×103, 1.8×104, 2.1×103, and 1.6×103cfu/ml, respectively. On the other hand, the mean values of Salmonella spp. in raw milk, udder, oil, and utensil samples were 2.4×102, 7.9×102, 1.5×102, and 1.1×102cfu/ml, respectively. Some selected isolates confirmed by molecular identification were tested for their sensitivity against some common antibiotics used in Bangladesh. Escherichia coli showed 70% resistance to Amoxicillin and 90% sensitivity to Ciprofloxacin whereas Salmonella spp. And Staphylococcus aureus showed 90% resistance to Ampicillin and both were 80% and 90% sensitive to Ciprofloxacin, respectively. Conclusion, Significance, and Impact of Study: Milk producers should be appropriately trained to monitor the overall hygienic conditions surrounding the production and handling of milk until it reaches the consumer. Frequent monitoring of the milk production facility, occasional testing of raw milk, and minimal use of antibiotics will ensure the quality of milk. Consumption of quality milk and avoidance of raw milk consumption will reduce the health risk of the consumers.

A collection of lactic acid bacteria (38 Enterococcus and 41 Lactobacillus strains) was tested for the antilisterial activity against 15 Listeria spp. strains (two L. monocytogenes, one L. ivanovii and 12 L. innocua strains) using agar spot method. Out of all 79 bacteria only six Enterococcus strains (1/3A, 3/3A, 6/4D, 6/1A, 1282 and EN3) exhibited antilisterial activity against almost all used indicator strains, when their live cells were tested. When their cell free neutralised supernatants (CFNS) were tested against four selected indicator strains (L. innocua Ln-03, Ln-06, Ln-10 and L. monocytogenes CCM5576) only two Enterococcus spp. strains were active &ndash; E. faecalis 6/1A strain from raw cow milk of minor interest due to the activity of its CFNS only against L. innocua Ln-06 and thermolability of the compound and E. mundtii 1282 strain from goat raw milk with CFNS active against 13 Listeria spp. strains including L. monocytogenes. E. mundtii 1282 strain produced probably a bacteriocin, because it completely lost the activity after treatment CFNS with proteinase K.

ABSTRACT: Pseudomonas, the main genus of gram-negative microorganisms isolated from milk, is psychrotrophic, biofilm-forming, and thermo-resistant deteriorating enzyme producers. The aim of this study was to quantify Pseudomonas spp. in goat’s and cow’s milk produced in the Paraná state, Brazil, to evaluate the deteriorating activity of the isolates at mesophilic and psychrotrophic conditions and to identify, at the species level, the isolates with alkaline metalloprotease (aprX gene) production potential. Microbiological, biochemical and molecular methods were used for isolating, confirming and identifying of isolates. The mean counts were 1.6 (±6.3)x104 and 0.89(±3)x102 CFU/mL for goat and bovine milk samples, respectively, immediately after milking. Of the Pseudomonas colonies isolated from goat milk (n=60), 91.7% showed proteolytic potential when incubated at 35°C/48 h and 80% at 7°C/10 days, and lipolytic potential was observed in 95% of the isolates incubated in mesophilic and 78.3% at refrigeration conditions. From the isolates of bovine milk (n=20), 35% showed proteolytic activity only when incubated at 35°C/48 h, and lipolytic potential was observed in 25% of the isolates incubated at 7°C/10d and 35°C/48h. It was observed that 83.3% and 25% of the isolates genetically confirmed as Pseudomonas spp. of goat and bovine milk showed the potential for alkaline metalloprotease production, with the species P. azotoformans, P. koreensis, P. gessardii, P. monteilii and P. lurida being the most frequent in goat milk and P. aeruginosa the only species identified in cow milk.

This study aimed to isolate Staphylococcus aureus in refrigerated raw cow milk, and identify the presence of enterotoxin-expression genes, enterotoxin production and adherence ability, and antimicrobial resistance potential of the isolated strains. Fifty raw milk samples obtained in different dairy farms were analyzed for S. aureus and evaluated in the isolates the presence of genes associated with the production of major staphylococcal enterotoxins and biofilm formation. In vitro assays were also performed to evaluate the production of enterotoxins and adherence ability, and the antimicrobial resistance. One half (25/50) of raw milk samples presented coagulase-positive staphylococci and 95.2% of the isolates were confirmed to be S. aureus. Among them, 42.4% were carrying genes for enterotoxins production; however, only one isolate was able to produce enterotoxins. All S. aureus isolates were carrying at least two genes associated with biofilm formation and 95.2% isolates was able to adhere upon the in vitro assay. All isolates demonstrated antimicrobial resistance potential to one or more of the tested antibiotics.

The environment, including animals and animal products, is colonized by bacterial species that are typical and specific of every different ecological niche. Natural and human-related ecological pressure promotes the selection and expression of genes related to antimicrobial resistance (AMR). These genes might be present in a bacterial consortium but might not necessarily be expressed. Their expression could be induced by the presence of antimicrobial compounds that could originate from a given ecological niche or from human activity. In this work, we applied (meta)proteomics analysis of bacterial compartment of raw milk in order to obtain a method that provides a measurement of circulating AMR involved proteins and gathers information about the whole bacterial composition. Results from milk analysis revealed the presence of 29 proteins/proteoforms linked to AMR. The detection of mainly β-lactamases suggests the possibility of using the milk microbiome as a bioindicator for the investigation of AMR. Moreover, it was possible to achieve a culture-free qualitative and functional analysis of raw milk bacterial consortia.

Camel milk is an important protein source for the nomadic communities living in the arid lands of the world, In recent years there has been an increase in consumption of non-bovine milk as an alternative protein source for humans. Camel milk seems to be containing larger amount of total proteins, such as lactoferrin and immunoglobulins as compared to the cow milk, which may be responsible for the better antimicrobial properties. The casein fraction of milk proteins consists of four caseins, namely as1-casein, as2-casein, b-casein, and k-casein. Casein genetic polymorphisms are important due to their effects on quantitative traits and technological properties of milk. This work was designed to study occurrence of polymorphism of a-casein in native Bikaneri camel (Camelus dromedarius) raw milk sample and to characterize these variants on molecular level.

In this paper the results of the analysis of the milk somatic cell count are presented, as well as correlation between the somatic cell count and content of certain chemical parameters in milk (milk proteins, fat, lactose and dry matter without fat) determined in collective samples of milk obtained from cows reared in intensive rearing system, during two production years. The research was carried out by control of collective milk samples from cows reared on family holdings. Somatic cell count, as well as the chemical quality of milk, were controlled daily in the laboratory for raw milk in dairy plant AD ?Mlekara? - Subotica using the apparatus CombiFoss 6200 FC. In this investigation, statistically significant correlation (P<0,001) between all observed milk parameters was determined. Positive, weak and statistically highly significant correlation between the content of milk fat and proteins in milk and somatic cell count was established. It was also established that the variability of chemical parameters of milk and somatic cell count is also under the influence of different factors, such as: month of control, year of the research and farm.
