Effect of Boiling on Carotenoids content of Pumpkin available in Damascus, Syria

The carotenoids contents of pumpkin were determined in this study by spectrophotometric method. This method was applied on beta-carotene as standard solution as well as samples. The absorption were determined at λmax 454 nm, the analytical method was validated by evaluating linearity, precision, accuracy, LODs, and LOQs of the analyts. Beer’s law is obeyed over the concentration of (0.3-9) µg/ml with a linear regression correlation coefficient of 0.9986, and recovery in range from 98.5-102.2%. After the validation was performed using standard solution, the method then was applied on samples of pumpkin available in local market in Syria after extraction the carotenoids by using Acetone/ Dichloromethane 70/30 .The results ranged between 216.46 - 80.56 µg/g. During boiling for pumpkin, the level of carotenoid reduced between 0.45-13.26 %, 0.18 – 23.07%, 17.3-40.54%, 22.65-51.73%, 14.91_55.82% and 26.16 – 72.56%, for 5, 10, 15, 30, 45 and 60 min respectively. According to the obtained result results, it is concluded that the boiling reduced carotenoid at different rates.

A simple, accurate, precise, reproducible, highly sensitive, an economic spectrophotometric method has been developed for the estimation of finasteride. UV spectrophotometric method is based on the measurement of absorption at a maximum wavelength of 255 nm. The developed method was validated with respect to linearity, accuracy (recovery), Precision (inter and intraday variations), LOD and LOQ. Beer’s law was obeyed in the concentration range of 5 – 25 μg/mL with a correlation coefficient of 0.9933. Results of the analysis were validated statistically and by recovery study. Hence the developed and validated method can be used for estimation of finasteride.

A new simple, rapid, accurate, sensitive and precise spectrophotometric method in ultra violet region has been developed for determination of acipimox (ACX) in bulk and capsule dosage form. Acipimox exhibited maximum absorbance at 231 nm with apparent molar absorptivity of 1.5104 × 104in distilled water. Beer’s law was found to be obeyed in the concentration range 1-10 μg mL-1. Correlation coefficient was found to be 0.9998. The developed method was validated respect to linearity, precision, accuracy. The proposed method is useful for the routine estimation of ACX in bulk and capsule dosage form.

Two simple, sensitive and rapid spectrophotometric methods have been developed for the estimation of Olmesartan medoxomil in bulk and pharmaceutical formulation. Method A is a UV spectrophotometric method in which isopropyl alcohol is used as solvent and exhibited absorption maxima at 285 nm. Beer’s law obeyed in the concentration range of 2 - 12 μg/mL with a correlation coefficient of 0.999. Method B is visible spectrophotometric method based on charge transfer complexation which involves the formation of a yellow colored chromogen when treated with picric acid. The chromogen exhibit absorption maxima at 421 nm and obeyed Beer’s law in concentration range 4-16 μg/mL with a correlation coefficient of 0.999. The results of the methods have been validated statistically and by recovery studies confirmed the accuracy and precision of the methods. The proposed methods are simple, sensitive and economical for quantitative determination of Olmesartan medoxomil in bulk drug and pharmaceutical formulation.

The spectrophotometric method for estimation of CefpodoximeProxetil employed first derivative amplitude UV spectrophotometric method for analysis using methanol as solvent for the drug. CefpodoximeProxetil has absorbance maxima at 235nm and obeys Beer’s law in concentration range 10-50µg/ml with good linearity i.e. r2 about 0.999. The recovery studies established accuracy of the proposed method; result validated according to ICH guideline. Results were found satisfactory and reproducible. The method was successfully for evaluation of CefpodoximeProxetil in tablet dosage form without interference of common excipients.

7-Amino-4-methylcoumarin-3-acetic acid (AMCA) has been found to be a useful fluorophore for immunofluorescence. The present study describes a spectrophotometric method for determining the ratio of moles AMCA to moles protein (or the f/p ratio) in an AMCA-conjugated IgG. The concentration of a substance absorbing light can be determined spectrophotometrically using Beer's Law: Absorbance = Concentration x Extinction coefficient. From Beer's law, one can derive the following formula for determining the f/p ratio of AMCA-IgG conjugates: f/p = (epsilon 280IgG).A350 - (epsilon 350IgG).A280/(epsilon 350AMCA).A280 - (epsilon 280AMCA).A350 where A is the optical density of the conjugate at the given wavelength and epsilon is the extinction coefficient of a substance at the wavelength specified. Using conjugates of model proteins, it was found that the extinction coefficients of the AMCA moiety of AMCA-conjugated protein were 1.90 x 10(4) at 350 nm and 8.29 x 10(3) at 280 nm. Similarly, it was found that the extinction coefficients of swine IgG were 1.56 x 10(3) at 350 nm and 1.26 x 10(5) at 280 nm. Thus, for AMCA-conjugated swine IgG: f/p = (1.26 x 10(5)).A350 - (1.56 x 10(3)).A280/(1.47 x 10(4)).A280 - (6.42 x 10(3)).A350 [corrected]. Based on this formula, the f/p ratios of some AMCA-IgG conjugates useful for immunohistochemistry have been found to range between 6 and 24.

Abstract A new, simple, and rapid spectrophotometric method for the microdetermination of dithianon, on the basis of its reaction with a dithiocarbamate, is described. The red color, which develops instantaneously when mixing the fungicide with the reagent in acetonitrile, is stable for at least 1 h and is measured at 520 nm. Beer's law is applicable up to 12 μg/mL dithianon concentration. The method has been successfully adapted to the analysis of the fungicide in commercial formulations and its residues on grains and apple (fruit and leaves). A photometric titration method for formulation analysis of the fungicide has also been developed.

Abstract A new UV spectrophotometric method was developed for quantitative evaluation of ceftazidime preparations. The UV detector was set at 255 nm. Beer's law is obeyed in the concentration range of 7.014.0 g/mL. The method was found to be selective, linear, accurate, and precise in the specified ranges. Intra- and interday variability for the method were <2 relative standard deviation. Common excipients used as additives in pharmaceutical preparations do not interfere with the proposed method. This method was successfully used for quantification of ceftazidime in pure form and in pharmaceutical preparations.

Determination of lanthanum in the raffinate by spectrophotometry was studied, using tribromoarsenazo as chromogenic reagent. The addition of ascorbic acid can mask the interference of other metal cations in the solution. Under the conditions of ascorbic acid (10g·L-1) 5 mL, hydrochloric acid 0.04 mol·L-1 and tribromoarsenazo (0.5g·L-1) 2 mL, Beer’s law is obeyed in the range of 0-10 μg·(25mL)-1 for lanthanum, with correlation coefficient higher than 0.99. Beer’s law can be applicable for the wavelength range of 580-660 nm. There is no linear relation at too high or too low acidic environment.

In this study, new spectrophotometric methods were developed for estimation of cefdinir (CFD); the research consists of two methods. The first method depends on the conversion of function group amine in cefdinir to diazonium salt and then combined with the 2, 5-Dimethylphenol (2, 5-DMP) reagent in the alkaline medium. The formed azo dye has a purple colour with absorption intensity at λmax 510 nm. Concentration range was obeyed Beer's law at (1-50 μg / ml), correlation coefficient was 0.9998, molar absorptivity was 1.54 × 104 L.mol-1.cm-1 and the detection limit was 0.04 μg.ml-1. The second method is including cloud point extraction (CPE) of a trace amount of the formed azo dye in the first method followed by measuring with a UV-visible spectrophotometer. Concentration range that obeyed the Beer's law was (0.2-6 μg / ml), correlation coefficient was 0.9998, molar absorptivity was1.48×105 L.mol-1.cm-1, detection limit was 0.004 μg.ml-1, Pre-concentration factor was 25 and Distribution coefficient was 125.The proposed methods were applied and proved their compatibility for estimating of ingredient compound in pure samples and pharmaceuticals by comparing them with previous studies. This research aims to partially and collectively analyze the influence of the academic atmosphere, the provision of facilities on the work spirit of permanent lecturers and to analyze the most dominant variable in influencing the work spirit of permanent lecturers at STIE Trisna Negara OKU Timur. The regression analysis results and the correlation between academic atmosphere and employee work spirit show the regression model Ŷ = 21,908 + 0.551X1 + e with a correlation coefficient of 0.589. The results of the regression analysis and the correlation between the provision of facilities on employee work spirit show the regression model Ŷ = 46.446 + 0.027X2 + e with a correlation coefficient of 0.033. The results of multiple regression analysis and the correlation between the academic atmosphere and the provision of facilities together on work spirit show the regression model Ŷ = 19.792 + 0.553X1 + 0.043X2 + e with a correlation coefficient of 0.592 at the 95% confidence level, it is found that the academic atmosphere and provision facilities can simultaneously predict employee work spirit. From the multiple linear regression equation above, it shows that the academic atmosphere variable (X1) has a more dominant influence on employee work spirit than the provision of facilities.
