Phytochemical, antioxidant, anti-stress and cerebroprotective activity of ethanolic extract of stem of Sarcostemma acidum

The current study was designed to assess the phytochemical, antioxidant, anti-stress and cerebroprotective activities of ethanolic extract of stem of Sarcostemma acidum (EESA). The stem of Sarcostemma acidum was collected and extracted with 70% ethanol. The ethanolic extract was subjected to phytochemical screening (Chemical and HPTLC), antioxidant (in-vitro), anti-stress (Mice model) and cerebroprotective activities (Cerebral ischemia model). EESA showed presence of flavonoids as primary phytoconstituents. EESA significantly reduced the immobility period in tail suspension trial and swimming endurance trial. EESA significantly reduced the TBARS levels (21.45±0.56; p<0.01) and augmented tissue antioxidants in cerebral ischemia model. The levels of MOA-A were reduced in the EESA treated animals (54.1±0.2; p<0.001) and cortisol levels also reduced in EESA treated animals (45.1±1.6; p<0.001). Histopathology also supported the biochemical parameters. The EESA effect was compared with reference standard diazepam and Ashwagandha. EESA showed significant antioxidant, anti-stress and cerebroprotective activities and the protective effect might be due to presence of flavonoids as phytoconstituents.

Entada rheedi is claimed to have antistress activity by folklore which is available abundantly in several places of India. The present study was planned to evaluate the phytochemical, anti-oxidant, anti-stress and cerebroprotective activities of ethyl acetate extract of bark of Entada rheedi (EAER). The bark of Entada rheedi was collected and extracted with ethyl acetate. The ethyl acetate extract was subjected to phytochemical screening (chemical and HPTLC), antioxidant (in-vitro), anti-stress (mice model) and cerebroprotective activities (cerebral ischemia model). EAER showed the presence of flavonoids as primary phytoconstituents. EAER significantly reduced the immobility time in swimming endurance and tail suspension test. AbstractLINGO-1 may be involved in the pathogenesis of cerebral ischemia. However, its biological function and underlying molecular mechanism in cerebral ischemia remain to be further defined. In our study, middle cerebral artery occlusion/reperfusion (MACO/R) mice model and HT22 cell oxygen–glucose deprivation/reperfusion (OGD/R) were established to simulate the pathological process of cerebral ischemia in vivo and in vitro and to detect the relevant mechanism. We found that LINGO-1 mRNA and protein were upregulated in mice and cell models. Down-regulation LINGO-1 improved the neurological symptoms and reduced pathological changes and the infarct size of the mice after MACO/R. In addition, LINGO-1 interference alleviated apoptosis and promoted cell proliferation in HT22 of OGD/R. Moreover, down-regulation of LINGO-1 proved to inhibit nuclear translocation of p-NF-κB and reduce the expression level of p-JAK2 and p-STAT3. In conclusion, our data suggest that shLINGO-1 attenuated ischemic injury by negatively regulating NF-KB and JAK2/STAT3 pathways, highlighting a novel therapeutic target for ischemic stroke.

Five traditional herbal preparations were sampled between May-June, 2009 in Kano. The samples were investigated for invitro antibacterial activities against clinical isolates of Staphylococcus aureus. Likewise, phytochemical screening tests were conducted to determine some of the phytochemicals present in the ethanolic and water extracts of the samples. Various concentrations of the extracts were prepared using serial doubling dilutions (5000=l/ml, 2500=g/ml, 1250=g/ml, 625=g/ml and 312.5=g/ml). All the test extracts showed slight antibacterial activity against the test organism, with ethanolic extract of sample E having the highest zone diameter of inhibition, while sample H had the lowest diameter of inhibition. The standard antibiotic disc (Gentamicin) had demonstrated the highest activity on the test organisms. The results of the Phytochemical screening revealed the presence of steroid in all the samples, tannin in samples A, C, D and E, reducing sugars in sample A, D and E respectively. The result of the minimum inhibitory concentration (MIC) was found to be above 312.5=g/ml for samples C, D and E. The aim of present study was to investigate the preliminary phytochemical, physicochemical, TLC, minerals analysis and In-vitro antioxidant activity of leaves of ethanolic extract of Heldigardia populifolia. The preliminary phytochemical screening of ethanolic extract showed the presence of triterpenoids, flavonoids, glycosides, sterols, steroids, phenols, carbohydrates and saponins. The composition of minerals found in the leaf powder was within the permissible limits. TLC analysis of ethanol extract showed the five spots which indicate the presence of five phytoconstituents. The extractive value of ethanol was high than acetone. Ash values were within the limits. The in-vitro antioxidant activity of ethanolic extract increased with increasing the concentration. The ethanolic extract in all the concentration showed the significant antioxidant activity.

Epidendrum coryophorum belongs to the Orchidaceae family. Traditional uses of some species for this genus include infusions of the leaves used for kidney problems, treat influenza, conjunctivitis, liver pain, relieve kidney symptoms, and hypoglycemic effect. This work's objective was to determine the phytochemical profile of the ethanolic extract of Epidendrum coryophorum leaves and to evaluate the potential anti-inflammatory activity in vitro of the extract employing the erythrocyte membrane stabilization method. The phytochemical screening carried out in this work suggested phenols, coumarins, flavonoids, tannins, steroids, and sterols in the ethanolic extract of Epidendrum coryophorum leaves. Cardiotonic glycosides and carbohydrates were also found. The ethanolic extract's UV-Vis spectrum showed absorption maxima at 268 nm and 332 nm, which could correspond to flavonoids of the flavonoid classes, 3-OH substituted flavonols, or isoflavones. The quantitative determination of total phenols of the ethanolic extract was carried out using the Folin-Ciocalteu method. The total phenolic content expressed as mg Gallic acid equivalent (G.A.E.) per gram of extract was found to be 19,96 mgGAE/g of Epidendrum coryophorum. The ethanolic extract of Epidendrum coryophorum leaves showed hemolysis inhibition values ​​of 18,19% at 1,0 mg/mL, 38,98% at 1,5 mg/mL and 40,94% at 2,5 mg/mL compared with aspirin (positive control) giving values ​​of 65,33% at 1,0 mg/mL, 72,26% at 1,5 mg/mL and 73,75% at 2,5 mg/mL. The values ​​obtained for inhibition of hemolysis with ethanolic extract, compared with the values ​​obtained with a pure anti-inflammatory, are significant and demonstrate anti-inflammatory activity in Epidendrum coryophorum. A comparative study was designed to evaluate the in vitro antidiabetic activity from the aqueous extract of dried pericarp of Prunus amygdalus Bail and ethanolic extract of dried flower of Hibiscus rosa-sinensis L. against the standard Acarbose.The aqueous extract of P. amygdalus and ethanolic extract of H. rosa-sinensis were extracted by the continuous hot extraction process.The qualitative phytochemical screening of aqueous extract and ethanolic extract reveals the presence of phytosterols, phenols, carbohydrates, flavanoids, saponins, amino acids and alkaloids, phenol, tannins, cardiac glycoside, saponin and flavonoids, respectively. In vitro pharmacological activity of P. amygdalus and H.rosa-sinensis were performed and compared by α-amylase inhibition assay method. The percentage inhibition of α-amylase by P. amygdalus, H. rosa-sinensis and Acarbose (50 &100 μg/mL) at dose of 50mg/mL and 100mg/mL were found to be 60.11±2.74%,74.86±0.31*%, 71.36± 1.86%, 81.49±1.72%, 82.33±1.10% and 95.37±0.56*%, respectively. The IC50 values were found to be 40.26μg/mL for acarbose, 49.63mg/mL for ethanolic extract of H. rosa-sinensis and 56.42mg/mL for aqueous extract of P. amygdalus.

Pentavalent antimonials are the first choice for the treatment of human leishmaniasis. However in rural areas the traditional plants may be preferred for the treatment of lesions. In recent years a number of papers are published related to the natural products especially plant derivative with infectious diseases. The present work was undertaken to evaluate the antileishmanial activity of Pterodon pubescens which is a native tree widely distributed over the central region of Brazil and used in folk medicine as wine infusions to treat inflammatory disease. The phytochemical screening and the biological essay of ethanolic extract of Pterodon pudescens (PPE) leaves at the concentrations of 150, 300, 450, 600 µg/ml were tested in vitro in Leishmania amazonensis-infected macrophages to support its traditional medicinal use as a leishmaniasis remedy. Phytochemical screening of PPE has shown the presence of catechemical tannins, steroids, triterpenoids and flavonoids. The biological test suggests that PPE were found to control parasite burden of cell cultures in dose-dependent manner. These findings highlight the fact that the apparent potency of Pterodon pudescens compounds, together with their widely distribution over Latin America and Brazil, may represent a promising antileishmanial agent.

BACKGROUND: The popular commercially cultivated Pleurotus ostreatus mushroom contains very high nutrients and bioactive compounds with high antioxidant activity. The ethanolic extract seems to be the most active in preparation. AIM: This study has an aim to compare the phytochemical analysis of a fresh, dry and ethanolic extract of Pleurotus ostreatus, to measure the total phenolic content and antioxidant activities in vitro of ethanolic extracts of Pleurotus ostreatus. METHODS: The fresh plant's materials (FPM), dry plants materials (DPM), ethanolic extracts were macerated with 70% (EE70) and 96% ethanol (EE96) of Pleurotus ostreatus which were used for phytochemical analysis, and EE96 was used for antioxidant activity in vitro. The phytochemical analysis was conducted using the Dragendorf and Meyer, FeCl3 test, Salkowsky method, Lieberman method, amyl alcohol, foam test and the NaOH reagent. The total phenol test was carried out using the Follin-Ciocalteu method. The antioxidant activity was tested using the ABTS and H2O2 essay. RESULTS: The phytochemical screening showed that the flavonoid, phenolic compounds, tannin, saponin, alkaloids, and steroids were detected in the FPM, DPM, EE70 and also the EE96. The alkaloid, however, was not identified by the Meyer Reagent in the FPM and DPM. The DPM and EE70 seemed to have the highest amount of saponin based on the foam that was formed. Meanwhile, steroids and flavonoids were detected at a higher level in the EE96, based on the strength of visible colour. However, triterpenoid and quinones could not be identified. In the total phenol test, there was an amount of 6.67 μg phenol in a 1 mg extract sample which was equivalent to 1 mg of Gallic Acid. The EE96 has an IC50 of 108.07 μg/mL for ABTS and an IC50 reduction of 229.17 μg/mL. The process of Pleurotus ostreatus drying did not reduce the content of active substances. The polar active substances seem to be more soluble in the EE70 than the EE96. CONCLUSION: The higher the bioactive substances in the preparation, the more significant the bio-therapeutic effects. Ethanolic extract of Pleurotus ostreatus has a phenol content and a good antioxidant action.
