The aim of this study was to assess the metaphylactic effect of subcutaneous doses of trace minerals (copper, zinc, selenium, and manganese) and vitamins A and E on biochemical (NEFA, BHBA, fructosamine, urea, triglycerides, HDL, and calcium) and hormonal parameters (cortisol, insulin, and IGF-1) of Holstein cows in the transition period. Sixty animals kept in a semi-confinement system, with an average production of 22 liters of milk per day (Farm 1) and 24 liters of milk per day (Farm 2) and a diet according to NRC (2001) with a low metabolic challenge, were divided into two groups: a treatment group, supplemented with trace minerals and vitamins (MTVG) (n=30), and a control group (CG) (n=30). The collection of blood samples was performed at the moments M1 (21 days prepartum), M2 (14 days prepartum), M3 (7 days prepartum), M4 (parturition day), M5 (7 days postpartum), M6 (14 days postpartum), and M7 (21 days postpartum). The experimental design was a randomized block design and the data were submitted to analysis of variance with repeated measures in time within the random variable cow. Differences were considered significant at 5% significance level and trend at 10% level. No difference was observed between treatment and the interaction treatment × day in the assessed parameters. However, some variables showed a significant difference of day. NEFA levels (P < 0.0001) were higher on the parturition day. BHBA concentrations were higher after parturition (P=0.0031), with a peak at the third week of lactation. Fructosamine showed increasing values until the parturition day (P=0.0373), in which higher concentrations were observed. Average values of HDL were higher at 21 days postpartum (P < 0.0001). Triglycerides levels remained high during the prepartum period but showed a sudden drop on the parturition day (P < 0.0001), with lower values postpartum. Regarding serum calcium and IGF-1, lower values were observed on the parturition day (P < 0.001) for both parameters. Cortisol presented a difference regarding day, with higher values on the parturition day (P=0.0151). Thus, the use of trace minerals and vitamins A and E in animals with an adequate body score and low metabolic challenge presents a low response to biochemical and hormonal parameters.

Dry matter and neutral detergent fiber contents act as physical regulators of voluntary intake in ruminants. Therefore, different levels of these components may interfere with animal performance and require careful assessment of the used raw material. This study was conducted with the purpose to assess the effect of soybean hulls and white oat in distinct or associated uses in the concentrate diet on the ingestive behavior of cattle. Thirty-six steers with an initial mean age of 20 months and initial mean weight of 226 kg were used. The diet was composed of 50% sorghum silage and 50% concentrate based on dry matter. The tested diets were soybean hulls (concentrate fraction composed mainly of soybean hulls), white oat grains (concentrate fraction composed mainly of white oat grains), and mixture (concentrate fraction composed of equal parts of soybean hulls and white oat grains). The experimental design was a randomized block design with 12 animals per diet. The data were compared by the PROC MIXED procedure with repeated measures in time, and the means were compared by the least significant difference test at 5% significance level. Diets presented different contents of neutral detergent fiber (66, 56, and 47%) and lignin (3.2, 3.7, and 4.1%) as the percentage of white oat grain increased in the diet. The total rumination time was lower (447 minutes) for animals that received equal parts of soybean hulls and white oat grains in the diet when compared to animals receiving a diet containing soybean hulls (483 minutes) and white oat grains (495 minutes), with no difference from each other. Animals fed a diet containing soybean hulls and white oat grains showed longer feeding time, a higher number of daily meals, shorter rumination time, shorter time spent chewing, and fewer number of chews per bolus and day. This diet results in a better dry matter and neutral detergent fiber rumination efficiencies.

ABSTRACT Introduction: Improving strength levels is important to women with osteoporosis. Resistance and aerobic exercise are effective means of reaching this goal; however, the use of low-load exercises with blood flow restriction is an alternative to traditional methods of exercise to achieve the same strength gains in this population. Objective: To analyze the chronic effects of aerobic and resistance training combined with blood flow restriction on the maximal dynamic strength (MDS) of women with osteoporosis. Methods: Twenty women (61.40±4.63 years of age, 61.82±12.54 kg, 1.51±0.05 m, 27.16±5.55 kg/m²) were randomly assigned to four groups: 1 - high-intensity resistance training (HI); 2 - low-intensity resistance training with blood flow restriction (LI-BFR); 3 - aerobic training with blood flow restriction (ABFR); and 4 - control group (CG). Unilateral knee extension MDS was assessed using the one-repetition maximum (1RM) strength test before and after the 6th and 12th weeks of intervention. The data were analyzed using repeated measures analysis of variance (ANOVA) with a Bonferroni post-hoc test performed using SPSS (version 21.0), considering a significance level of P<0.05 for all tests. Results: Baseline comparisons showed that HI and CG had lower strength levels than LI-BFR and ABFR groups (P<0.05). The ABFR group exhibited a significant increase in MDS between the 1st and the 6th week (9%, P=0.001) and between the 1st and the 12th week (21.6%, P=0.008). The LI-BFR group exhibited increased MDS between the 1st and the 6th week (10.1%, P=0.001), between the 1st and the 12th week (24.2%, P=0.003) and between the 6th and 12th week (12.8%, P=0.030). The HI group exhibited a significant difference between the 1st and the 6th week (38.7%, P<0.001), between the 1st and the 12th week (62%, P<0.001) and between the 6th and 12th weeks (17.4%, P=0.020), whereas the CG had no significant differences between the timepoints (P>0.05). Conclusions: ABFR and LI-BFR effectively increased the MDS of women with osteoporosis.

The plasma profile of various hormonal, biochemical and mineral constituents was studied in 32 buffaloes under nutritional management of transition period for 2 months before to 2 months postpartum. The buffaloes that expressed estrus and conceived within 120 days postpartum, irrespective of nutrient treatment or control groups, were classified as fertile (n=15) and rests as infertile (n=17) buffaloes. The variations in blood glucose and plasma profile of progesterone, estradiol, protein, cholesterol, triglycerides, Ca, P, Mg and trace minerals (Zn, Fe, Cu, Co, Mn), if any, on day 60, 30, 15 prepartum, on the day of calving, and on day 15, 30, 45 and 60 postpartum in postpartum conceiving and non-conceiving buffaloes were examined. There was no significant effect of fertile-infertile groups on plasma P4 and E2 profile on any day, except on day 45 postpartum, where E2 was (p less than 0.05) higher in fertile than infertile group (31.47±2.37 vs. 24.29±1.76 pg/ml), indicating early onset of follicular activity with ovulatory estrus in postpartum fertile group. The blood glucose and plasma protein profile also did not vary significantly at any of the intervals between two groups. Further, no significant difference in plasma cholesterol or triglyceride concentrations was found between conceived and non-conceived buffaloes at any of the intervals pre- or postpartum, except that the pooled mean cholesterol level was higher (P less than 0.05) in non-conceived than conceived buffaloes. The plasma concentrations of calcium, inorganic phosphorus and magnesium also did not reveal significant variation between fertile and infertile buffaloes at any of the intervals, though the magnesium value was higher in conceived than non-conceived buffaloes. Similarly, no significant difference was seen in the levels of plasma trace minerals, viz., Zn, Fe, Mn, Cu or Co between fertile and infertile buffaloes at any of the intervals studied. These nonsignificant variations noted between fertile and infertile buffaloes in blood profile could perhaps be due to masking effect of nutritional supplementation as the control group was also optimally managed by the farmers; hence the animals were pooled irrespective of groups for this comparison.

Abstract Background The purpose of this study was to assess the cytotoxicity of various concentrations of ozonated water (OW) on human primary dental pulp cells. Methods Human primary dental pulp cells were isolated from exfoliated primary canine teeth of an 11-year-old patient with good systemic and oral health. Afterwards, cells were divided into 6 experimental groups; four groups of OW in concentrations of 2 mg/L, 4 mg/L, 8 mg/L, and 16 mg/L, untreated control group, and cell culture without cells. Cytotoxicity was evaluated after exposure for 5-min exposure using Mosmann’s Tetrazolium Toxicity (MTT) assay at 0 h and 48 h time points. Data were analyzed using a repeated measures analysis of variance and Post-hoc tests were performed using Bonferroni correction for multiple comparisons. Results All experimental groups showed proliferation at 0 h time point. However, all groups also experienced a decrease in overtime at 48 h time point (p < 0.05). At both time points 2 mg/L OW showed the highest cell viability as well as proliferation. At 0 h time point, the increase in cell viability for all experimental groups was found statistically significant when compared to positive control group (p < 0.05). At 48 h time point, although 8 mg/L and 16 mg/L OW showed statistically significant reduction in compare to 0 h time point, 2 mg/L and 4 mg/L OW groups didn’t experience any statistically significant difference (p < 0.05). Conclusion Considering our findings, due to ozonated water's induced a higher proliferation rate of dental pulp cells, indicating their biocompatibility and a possible adjuvant on irrigating agent in regenerative endodontic procedures.

Robusta coffee plant propagation is recommended to be performed by vegetative methods, one of which is cuttings. Among plant organs that can be used for cuttings is the stem. The research objective was to determine which stem segment from cuttings was able to provide the best root and leaf growth. The layout study used a completely randomized block design (RBD) with an experimental treatment consisting of five stem segments, namely (1) the first stem segment from the upper shoot (A); (2) the second stem segment from the upper shoot (B); (3) the third stem segment from the upper shoot (C); (4) the fourth stem segment from the upper shoot (D); and (5) the fifth stem segment from the upper shoot (E); each treatment had five replications. Data analysis was performed statistically with analysis of variance (ANOVA) and Tukey's HSD (honestly significant difference) test with a significance level of P < 0.05. Robusta coffee cuttings stems from the second stem segment from the upper shoot were able to provide the optimal root growth (root length) and shoot growth (leaf sprout height and leaf sprout growth rate). Leaf sprout number derived from the cuttings was not affected by the stem segment.

ABSTRACT Aim The aim of this study was to evaluate the effect of chlorhexidine and ethanol application on the push-out bond strength and bond durability of fiber posts cemented with an etch-and-rinse adhesive system/resin cement to intraradicular dentin. Materials and methods Fifty-four bovine roots were shaped for the cementation of a fiberglass post and received the application of 37% phosphoric acid. They were then randomly divided into three groups, according to the type of dentin treatment (n = 18) performed: no treatment (control group), 100% ethanol, or 2% chlorhexidine. Next, the adhesive system (Adper Scotch Bond Multipurpose Plus, 3M ESPE) was applied to the dentin, according to the manufacturer's instructions. Glass fiber posts were cemented with dual resin cement (Rely X ARC, 3M ESPE). After 48 hours, the specimens were serially sectioned for push-out test analysis, providing two slices from each root third (cervical, medium and apical), one of which was tested immediately and the other stored in distilled water for 180 days. The data were analyzed with three-way analysis of variance (ANOVA) for repeated measures and Tukey's test at a 5% significance level. Results Intraradicular treatment with chlorhexidine yielded the highest bond strength means, followed by ethanol treatment. The control group presented the lowest bond strength means. Water storage exerted no effect on bond strength values. Conclusion Both chlorhexidine and ethanol improved push-out bond strength to intraradicular dentin, with the former providing the best results, regardless of the storage time. Clinical significance The application of 2% chlorhexidine or 100% ethanol may be an important step that can be taken to enhance bond strength of fiber posts to intraradicular dentin, when dual resin cements are used.
