Determination of Trigonelline, Nicotinic Acid, and Caffeine in Yunnan Arabica Coffee by Microwave-Assisted Extraction and HPLC with Two Columns in Series

Abstract A simple, rapid method was developed for simultaneous extraction of trigonelline, nicotinic acid, and caffeine from coffee, and separation by two chromatographic columns in series. The trigonelline, nicotinic acid, and caffeine were extracted with microwave-assisted extraction (MAE). The optimal conditions selected were 3 min, 200 psi, and 120°C. The chromatographic separation was performed with two columns in series, polyaromatic hydrocarbon C18 (250 × 4.6 mm id, 5 μm particle size) and Bondapak NH2 (300 × 3.9 mm id, 5 μm particle size). Isocratic elution was with 0.02 M phosphoric acid–methanol (70 + 30, v/v) mobile phase at a flow rate of 0.8 mL/min. Good recoveries and RSD values were found for all analytes in the matrix. The LOD of the three compounds was 0.02 mg/L, and the LOQ was 0.005% in the matrix. The concentrations of trigonelline, nicotinic acid, and caffeine in instant coffee, roasted coffee, and raw coffee (Yunnan Arabica coffee) were assessed by MAE and hot water extraction; the correlation coefficients between concentrations of the three compounds obtained were close to 1.

An efficient microwave-assisted extraction (MAE) technique was developed to extract polysaccharides from Enteromorpha prolifera (PEP). The operating parameters, such as microwave power, liquid/material ratio, temperature and extraction time, were optimized using orthogonal array design coupled with single factor method. PEP yield was determined by the phenol-sulfuric acid method. The optimum extraction conditions were determined as follows: microwave power, 700 W; liquid/material ratio, 40:1 (mL/g); temperature, 70 °C; and extraction time, 25 min. Under such conditions, PEP yield reached to 10.79 %. In comparison with conventional hot water extraction and ultrasonic-assisted extraction, MAE showed obvious advantages in terms of high extraction efficiency, saving energy, rapidity, solvent consumption, and so on. The data demonstrated that MAE could be a fast and reliable method for quantitative analysis of PEP. The scavenging capability of PEP to DPPH/hydroxyl radical reached to 65.2 % and 41.2 % at the concentration of 0.5 mg/mL. The reducing power of PEP was 0.354. Compared with butylated hydroxytoluene (BHT) and gallic acid (GA), the experimental results showed that DPPH radical scavenging activity of PEP was higher than that of BHT. Thus, PEP had good potential as a natural antioxidant used in functional food or medicine industries.

Evodia lepta (E. lepta) is a traditional Chinese herbal medicine with various biological activities. One of the active components of this widely used medicinal plant is believed to be an oligosaccharide. The extraction yields, structural characteristics, antioxidant, and antitumor activities of four oligosaccharide extracts obtained by hot water extraction (HEO), ultrasound-assisted extraction (UEO), enzyme-assisted (EEO), and microwave-assisted extraction (MEO) were investigated. Matrix-assisted laser desorption/ionization-time of flight-mass spectrometry (MALDI-TOF-MS), X-ray diffraction (XRD), and Scanning electron microscopy (SEM) results indicated that the extraction methods had a difference on the molecular mass distribution, structure, and morphology of the EOs. In addition, HEO and MEO showed strong antioxidant activities, which might be related to their uronic acid and protein contents. More interestingly, MEO was more active toward MDA-MB-231 cells compared to other cells, and cell growth inhibition was proposed to occur through apoptosis. Overall, microwave-assisted extraction is a promising technique for the extraction of high quality EO.

Acanthopanax Senticosus is a traditional medicinal plant in China. Flavonoids are one of the main active ingredients of this plant. Four extraction technologies of flavonoids from Acanthopanax Senticosus were investigated and compared, including microwave-assisted extraction, ultrasonic extraction, reflux extraction and Soxhlet extraction. The results showed that microwave-assisted extraction was most suitable for the extraction of flavonoids from Acanthopanax Senticosus because of its high effect and short extraction time. The found optimum extraction conditions were that the ethanol concentration was 50% (v/v), solid–liquid ratio was 1:50, extraction temperature was 50°C and irradiation time was 9 min. Quantification was performed by means of UV–Vis spectrophotometry with chromogenic system of NaNO2–Al (NO3)3–NaOH. Under the optimum conditions, the calibration curve for the analyte was linear with the correlation coefficients greater than 0.9999. The average recovery was 101.8%, and its RSD was 0.87%(n=5). The total content of flavonoids was18.72 mg g−1.

Polysaccharides are a main active substance in Panax ginseng; however, microwave-assisted extraction used to prepare P. ginseng polysaccharides (MPPG) has rarely been reported, and knowledge of the bactericidal activity of P. ginseng polysaccharides remains low. Thus, this study was designed to investigate the extraction of P. ginseng polysaccharides by using two methods—hot water extraction and microwave-assisted extraction—and compare their chemical composition and structure. In addition, their antibacterial and antioxidant activities were also determined. The data implied that P. ginseng polysaccharides extracted by microwave-assisted extraction possessed a higher extraction yield than hot water extraction (WPPG) under optimized conditions, and the actual yields were 41.6% ± 0.09% and 28.5% ± 1.62%, respectively. Moreover, the preliminary characterization of polysaccharides was identified after purification. The WPPG with the molecular weight (Mw) of 2.07 × 105 Da was composed of Man, Rib, Rha, GalA, Glu, Gal, and Arab, and the typical characteristics of polysaccharides were determined by IR spectra. Compared with WPPG, MPPG had a higher Mw, uronic acid content, and Glu content. More importantly, the antioxidant activity of MPPG was higher than WPPG, which was probably ascribed to its highly Mw and abundant uronic acid content. Besides, both of them exhibited high bactericidal activity. These results demonstrate that microwave-assisted extraction is an effective method for obtaining P. ginseng polysaccharides, and MPPG could be applied as an antioxidant and antibacterial agent.

Objective: The purpose of this study was to formulate a phytosome lotion from Mangkokan leaf using green solvent extraction. Methods: Ionic liquid, 1-butyl-3-methylimidazolium tetrafluoroborate, combined with microwave-assisted extraction, was used to obtain flavonoid compounds. Mangkokan leaf extract was formulated into a phytosome with three different ratios of phospholipon (1:0.5), (1:1), and (1:2). Three phytosome formulas were analyzed to identify the best formula. The analysis included morphology, particle size, and zeta potential, entrapment efficiency, and spectrum analysis using an FTIR spectrophotometer. The formula which chosen was formulated into three lotion formulas with different concentration (20%; 25%; 30%) then, evaluated for physical stability, and hair growth activity test included hair length, weight, and diameter. Minoxidil and non-phytosome lotion were used as comparison. Data were analyzed using two-way ANOVA. Results: The FTIR of phytosome was confirmed a complex formation of extract and phospholipon. The phytosome morphology was displayed irregular spherical vesicles. Phytosome with the ratio of phospholipon (1:0.5) was formulated into lotion because has a smaller particle size (289 nm), and higher entrapment efficiency (99.76%±0.24) than others. Phytosome lotions displayed higher activity than 2% minoxidil (p = 0.0001). Phytosome lotion (30%) thus showed the highest efficacy for hair length, weight and diameter. Conclusion: Mangkokan leaf that extracted using 1-butyl-3-methylimidazolium tetrafluoroborate solvent combined with microwave-assisted extraction then formulated into phytosome lotion has higher activity than 2% minoxidil. Lotion that contains 30% mangkokan leaf phytosome was showed the highest efficacy.
