Banana (Musa spp.) is mainly grown in the tropical and subtropical countries. Previous study reported that Musa spp. leaves had a potential antioxidant activity, but it was still rarely studied further. In this research, leaves of Musa balbisiana, Musa acuminate, and Musa paradisiaca were extracted using maceration method for 24 hours with three kinds of solvent having different polarities: ethanol (polar), ethyl acetate (semi polar), and hexane (nonpolar). The goal of this research was to compare and determine the stability of the antioxidant activity extracted from different Musa sp. leaves. The highest antioxidant activity is found from Musa balbisiana leaves extract with IC50 value 340.07±22.54 ppm (hexane fraction). Correlation analysis between antioxidant activity, total phenolic content, and total flavonoid of the extracts cannot conclude that the active antioxidant substances in these three banana species leaves were from phenolic or flavonoid groups. This crude extract from Musa balbisiana was then subjected to various pH levels (3.0, 5.0, 7.0, and 9.0) and temperatures (50, 70, and 90°C) to determine the stability of its antioxidant activity. It is found that the best stability condition is at pH 3.0 and temperature of 50°C with an increase of 63.1% in IC50, a decrease of 15.72% in total phenolics, and a decrease of 3.67% in total flavonoids as compared to before treatment.

The aim of this study is to investigate antioxidant activity and total phenolic content of ethyl acetate fraction of Sembung leaves. Sembung (Blumea balsamifera L.) is a plant that has the potential as an antioxidant because it contains secondary metabolites of alkaloids, steroids, flavonoids, saponins, and phenolic compounds. In this study, antioxidant activity was tested by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) for free radical scavenging activity assay. Evaluation of total phenolic content was tested by using the Folin-Ciocalteau method. The results showed that the ethyl acetate fraction of Sembung leaves possess showed medium antioxidant activity with IC50 valueof 221.821 compare with reference standard ascorbic acid with IC50 value 68.25 ppm. The ethyl acetate fraction of Sembung leaves has a total phenolic content of 161.101 mg GAE/g. This study provided that Blumea balsamifera leaves possess antioxidant.

This study aims to determine antioxidant activity and total phenolic content of mangosteen leaves in several different solvents. The antioxidant activity test of the extract was determined by the DPPH method and the total phenolic content was determined by the Folin-Ciocalteu method. The phytochemical test of mangosteen leaves showed the presence of phenolic compounds, saponins, triterpenoids and alkaloids. The extraction results increase with increasing water content in methanol, ethanol and acetone. The highest amount of extract was obtained using 50% acetone solvent (34.98 ± 0.17%). Extraction using 100% ethanol showed the highest total phenolic content (614.25 ± 5.58 mg GAE / g extract) and the lowest IC50 value (5.81 mg / L). The results of this study indicate that the use of 50% acetone solvent can optimize the amount of extract and 100% ethanol can optimize the content of total phenolic compounds and antioxidant activity of mangosteen leaves.

Olax scandens Roxb. (family Olacaceae) available in throughout tropical India. The current study, aerial parts of different concentrates of Olax scandens was evaluated for its in-vitro antioxidant potential by FRAP assay taking ascorbic acid as the standard and estimation of total phenolic content and flavonoids content. The IC50 value was originated that methanolic concentrates of Olax scandens are more efficient in antioxidant activity by FRAP methods compared EA & PE concentrates. The methanolic concentrates of Olax scandens & ascorbate exhibited antioxidant potential possessing IC50 207µg/ml & 50µg/ml by Ferric reducing ability Power assay. The methanolic and EA concentrates of Olax scandens showed the total phenolic content (14.426 ± 0.032, 4.128 ± 0.025) respectively, and ৒lavonoids content (11.526 ± 0.054, 3.682 ± 0.042) respectively. Invitro antioxidant studies show methanolic concentrates of Olax scandens have better antioxidant activity as well as a higher content of total phenolic and flavonoids content. These results indicate that aerial parts of methanolic concentrates Olax scandens could serve as a natural antioxidant, which may be useful in preventing free radical-induced diseases.

AbstractThe stability of tea phenolic compounds is influenced by pH value and digestive processes. However, the complex mixture of constituents in tea may modulate the stability of these compounds during digestion. In this study, tea infusions obtained from green, black, and Oolong tea leaves were exposed to in vitro simulated gastrointestinal digestion, and the stability of ( +)-catechin, caffeine, (−)-epicatechin, epigallocatechin-3-gallate (EGCG), and gallic acid was compared to that of isolated compounds. Changes in antioxidant activity were also evaluated by means of DPPH assay and in a H2O2-induced in vitro oxidative stress model, using Caco-2 cells. The stability of teas antioxidant constituents was different when using teas extract, compared to the reference compound alone, with the total phenolic content being more stable in extracts containing them in higher amount. EGCG degradation correlated well with changes in the DPPH inhibition assay, confirming its pivotal role in the antioxidant activity of tea. Differently, the antioxidant effect in the in vitro cell-based model was much more related to the initial total phenolic content of the extracts, with green tea being more effective than black tea and Oolong tea. Moreover, the antioxidant activity of teas was strongly affected by gastrointestinal digestion. Taken together, these findings suggest a protective role of teas phytocomplex against gastrointestinal digestion of antioxidant constituents. In conclusion, the effect of gastrointestinal digestion on the antioxidant activity of tea should be taken into account, as this may be different from one extract to another and information on the stability of active constituents cannot be extrapolated from data obtained using single compounds.

Four types of traditional Thai rice—polished, black fragrant, glutinous and black glutinous rice—were separately used as raw material for vinegar production. During alcohol fermentation, using enriched baker’s dried yeast (S. cerevisiae) as a starter culture gave the highest ethanol content over 7 days of fermentation. The conversion of ethanol to acetic acid for vinegar production by Acetobacter pasteurianus TISTR 102 was performed for 25 days. The highest amount of acetic acid was detected with glutinous rice fermentation (6.68% w/v). The biological properties of Thai rice vinegars were determined, including the total phenolic content, antioxidant activity, antibacterial activity and cytotoxicity. Black glutinous rice vinegar exhibited the maximum total phenolic content of 133.68 mg GAE/100 mL. This result was related to the antioxidative activity findings, for which black glutinous rice vinegar exhibited the strongest activity against both ABTS•+ and DPPH• radicals. Cytotoxicity against the human colon cancer cell line (HT-29) provided an IC50 value of 74.02 μg/mL and weak activity in a mouse fibroblast normal cell line (L929) with an IC50 value of 171.06 μg/mL. Glutinous rice vinegar was the most effective vinegar for inhibiting pathogenic bacterial growth of both Gram-negative and Gram-positive bacteria. These results suggested that the value of total phenolic content corresponded to the anticancer activity and antioxidant activity results, while antibacterial activity depended on the acidity of rice vinegar.

PENENTUAN TOTAL FENOLIK DAN AKTIVITAS ANTIOKSIDAN FRAKSI ETIL ASETAT DARI TEPUNG PELEPAH AREN (Arenga pinnata) ABSTRAK Tujuan penelitian ini untuk menentukan total fenolik dan aktivitas antioksidan fraksi etil asetat dari tepung pelepah aren (Arenga pinnata). Tepung pelepah aren diekstraksi maserasi menggunakan etanol hasil redestilasi cap tikus kemudian di partisi menggunakan pelarut petroleum eter, etil asetat dan aquades. Penentuan kandungan total fenolik menggunakan metode Folin-Ciocalteu dan aktivitas antioksidan menggunakan metode serapan radikal 1,1-difenil-2-pikrilhidrazil (DPPH). Didapati kandungan total fenolik fraksi etil asetat sebesar 172.857 ± 4.762 mg/kg dan aktivitas penangkal radikal bebas fraksi etil asetat sebesar 90.573 µg/mL. Hasil ini menunjukan bahwa fraksi etil asetat berpotensi sebagai antioksidan alami. Aren’s midrib powder extracted with maceration method with cap tikus redistillated ethanol and then partitioned with solvents such as petroleum ether, ethyl acetate and distillate water. The determination of phenolic content was using Folin-Ciocalteu method and the antioxidant activity was done using diphenylpycrylhydrazyl (DPPH) radical scavenging. Total phenolic value of ethyl acetate fraction is 172.857 ± 4.762 and IC50 value of ethyl acetate fraction is 90.573 µg/mL. This result shows ethyl acetate fraction as natural antioxidant potential. The objective of this research was to identify the recovery of the biological active compound of Musa sp. thru microwave assisted extraction (MAE) system. Antioxidant activity and phenolic compound were investigated on the effect of temperature, microwave power, irradiation time and solid to liquid ratio. The extraction process was carried out using only distilled water as an extracting agent. The result showed that, at 70oC fresh Musa sp. extracted 45% higher antioxidant compared to the dried Musa sp. 500 W was the best power of MAE to extract antioxidant and phenolic compound of ripe Musa sp., while for unripe Musa sp. 800 W was the best. The best irradiation time was 90 s, where it extracted almost 90% antioxidant activity of ripe and unripe Musa sp. Almost 480 mg GA/100 g FW of total phenolic content was extracted on the unripe peel. Solid to liquid ratio of 3 : 60 was the best parameter to extract antioxidant and phenolic content of ripe and unripe Musa sp. Overall, antioxidant and phenolic compound were significantly higher in the unripe peel, followed by ripe peel, unripe pulp and ripe pulp.

AbstractType of Solvent Methanol And N-Hexane Against Antioxidant Activity Seaweed Extract Gelidium sp. From Drini Beach Gunungkidul – Yogyakarta             Gelidium sp. the one of red seaweed which has potential as naturalantioxidant. The researh was to know the activity of methanol extract, n-hexane Gelidium sp., and determined totalphenolic compound of pigments (chlorophyll a and carotenoid). Decriptiveexplorative method was used in this research and it was taken from Drini Beach, Gunungkidul, Yogyakarta. Maseration to this sample was done by methanol as solvent, evaporated by rotary evaporator and partitied by n-hexane solvent with separatoryfunnel. Antioxidant activity was determined by transfer electron method using DPPH (1,1-diphenyl-2-picrylhidrazyl) as free radicals. Totalphenolic compound being tested using Folin-Ciocalteu solution with gallic acid as standard and measured at a wavelength of 725 nm, while the chlorophylls were spectrophotometry method and measured at a wavelength of 663 nm and 646 nm as well as carotenoidswere measured at a wavelength of 470 nm. The results showed that IC50 value of methanol extract was 340,10 ppm and n-hexane was 66,25 ppm. IC50 value of methanol extract was categorized as very weak in antioxidant activity, while n-heksan extract was categorized as strong. Total phenolic content in each extract were 46,55 and 135,62 (mg GAE/g extract), chlorophyll a 8,47 and 10,88 (mg/g extract sample) and carotenoids 50,38 and 84,27 (μmol/g extract sample). Keywords: Gelidium sp., Antioxidant, DPPH Abstrak  Gelidium sp. merupakan salah satu rumput laut merah yang berpotensi sebagai sumber antioksidan alami. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan ekstrak metanol dan n-heksan Gelidium sp. (segar), menentukan kadar total fenolat dan pigmen (klorofil a dan karotenoid). Metode deskriptif eksploratif digunakan dalam penelitian ini dan diperoleh diambil dari Pantai Drini, Gunungkidul, Yogyakarta dibersihkan dengan air tawar, dimaserasi dengan pelarut metanol, diuapkan dengan rotary evaporator dan dipartisi dengan pelarut n-heksan menggunakan corong pemisah (separatory funnel). Aktivitas antioksidan ditentukan dengan metode transfer elektron menggunakan DPPH (1,1-diphenyl-2-picrylhidrazyl) sebagai radikal bebas. Kadar total fenolat diuji dengan metode Folin-Ciocalteu dengan asam galat sebagai larutan standar dan diukur pada panjang gelombang 725 nm. Kadar klorofil a diukur dengan metode spektrofotometri pada panjang gelombang 663 nm dan 646 nm sedangkan kadar karotenoid diukur pada panjang gelombang 470 nm. Hasil penelitian menunjukkan nilai IC50 ekstrak metanol sebesar 340,10 ppm dan ekstrak n-heksan 66,25 ppm. Nilai IC50ekstrak metanol termasuk kategori aktivitas antioksidan sangat lemah sedangkan n-heksana termasuk kategori kuat. Latar Belakang: Antioksidan adalah senyawa yang mampu menangkal atau meredam efek negatif oksidan dalam tubuh, bekerja dengan cara mendonorkan satu elektronnya kepada senyawa yang bersifat oksidan sehingga aktivitas senyawa oksidan tersebut dapat dihambat. Herba poguntano (Picria fel-terrae Lour.) digunakan sebagai obat antidiabetes dan telah dilaporkan mengandung senyawa kimia yang terdiri dari alkaloid, saponin, flavonoid, dan polifenol yang merupakan metabolit sekunder yang bersifat antioksidan. Tujuan Penelitian: Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan serta mengetahui kandungan fenolat dan flavonoid total dari ekstrak etanol herba poguntano. Metode Penelitian: Ekstrak diperoleh secara maserasi dengan pelarut etanol 96%, lalu dipekatkan dengan alat rotary evaporator hingga didapatkan ekstrak kental. Ekstrak diuji dengan metode pemerangkapan radikal bebas DPPH (1,1-diphenyl-2-picrylhydrazyl) serta mengukur kandungan fenolat total dan flavonoid total dengan mengukur absorbansi menggunakan spektrofotometri uv-vis yang diinkubasi berturut-turut selama 60, 90 dan 40 menit pada panjang gelombang 516, 775 dan 432 nm. Hasil: Hasil pengukuran aktivitas antioksidan dengan metode pemerangkapan radikal bebas DPPH dari ekstrak etanol herba poguntano menunjukkan kekuatan antioksidan dalam kategori sangan kuat dengan nilai IC50 21.79 ppm dan kuersetin menunjukkan kekuatan antioksidan dalam kategori sangat kuat dengan nilai IC50 4.9 ppm. Hasil pengukuran kandungan fenolat total dengan metode Folin-Ciocalteau menunjukkan angka 66.29 mg GAE/g ekstrak serta hasil pengukuran kandungan flavonoid total menunjukkan angka 19.26 mg QE/g ekstrak. Kesimpulan: Ekstrak etanol herba poguntano mempunyai aktivitas antioksidan dengan kategori sangat kuat serta memiliki kandungan fenolat total yang tinggi dibandingkan dengan kandungan flavonoid total. Background: Antioxidants are compounds capable of counteracting or reducing the negative effects of oxidants in the body. Antioxidants work by donating an electron to an oxidant compound so that the activity of the oxidant compound can be inhibited. Poguntano (Picriafel-terrae Lour) is used as an antidiabetic drug and has been reported to contain chemical compounds, , as secondary metabolites that have antioxidant properties. Objective: This study aimed to determine antioxidant activity and total phenolic and flavonoid content of the ethanol extract of poguntano. Research Method: . The dried materials were macerated using 96% ethanol and then it was concentrated using rotary evaporator to obtain crude extract. The extract was tested by DPPH free radical scavenging method (1,1-diphenyl-2-picrylhydrazyl) and the total phenolic and flavonoid content was determinedby measuring absorbance using uv-vis spectrophotometry which was incubated successively for 60, 90 and 40 minutes at wavelength 516, 775 and 432 nm. Results: The evaluationof antioxidant activity using DPPH free radical scavenging method ofethanol extract of poguntano showed the antioxidant activitywas in very strong category with IC50 value of 21.79 ppm and quercetin showed the antioxidant activitywas in very strong category with IC50 value of 4.9 ppm. The result of the measurement of total phenolic content by the Folin-Ciocalteu method was66.29 mg GAE/g extract and the result of measurements of total flavonoid content was19.26 mg QE/g extract. Conclusion: Ethanol extract of poguntanohas antioxidant activity in a very strong category and it has higher total phenolic content compared with total flavonoid content.

Leaves and drupes of the olive varieties "Koroneiki", "Lianolia Kerkyras", "Mastoidis", "Arbequina", "Adramytini", "Megaritiki", "Gaidourelia", "Kalamata", "Konservolia" and "Chalkidiki", were collected at different seasons during the years 2008 & 2009 and investigated for their total phenolic content and antioxidant activity. Total phenolic content and antioxidant activity were determined according to Folin-Ciocalteau and DPPH assays, respectively. Different varieties presented differences in the total phenolic content and total antioxidant activity per tissue. For both years, most varieties showed higher total phenolic content in new season leaves than in green and black drupes. New season leaves on April showed the highest total phenolic content than the leaves of September and December of the same year, with a few exceptions. There was a positive correlation between the antioxidant activity and the total phenolic content. Highest phenolic content was accompanied by a lower IC50 value and as a result higher antioxidant activity.
