Nutritional requirements of a Micrococcus species (M. freudenreichii, A.T.C.C. No. 407) were studied. The organism required glutamic acid, thiamine, biotin, magnesium, iron, and potassium for growth. Cells from such a synthetic medium were shown to contain methionine indicating that inorganic sulphur was used. Glutamic acid could not be replaced with glutamine (unheated), aspartic acid, asparagine, nor ammonium salts. The relationship of nutritional requirements of micrococci and staphylococci to classification is discussed.

Background:Systemic sclerosis-related interstitial lung disease (SSc-ILD) involves a combination of inflammation, fibrosis and vascular pathology that is typically assessed on CT imaging as a mixture of ground-glass opacification (GGO) and fibrotic changes. We hypothesized that proteins recovered from bronchoalveolar lavage (BAL) could be used to probe the underlying pathobiology associated with GGO and fibrotic changes.Objectives:(1) To assess the relationship between 68 unique BAL proteins measured in participants of Scleroderma Lung Study (SLS) I1and radiographic and physiologic measures of ILD; (2) To identify inter-correlations among specific proteins to enlighten our understanding of how specific biological pathways contribute to SSc-ILD.Methods:Bronchoscopy was performed on 144 of the 158 participants in SLS I with 103 BAL samples available for analysis. BAL was lyophilized, concentrated 10X and used in a multiplex protein analysis for 68 different cytokines, chemokines and other factors. Kendall tau correlations were performed to assess the relationship between individual proteins and baseline measures of pulmonary function and quantitative CT scores for fibrosis, GGO and total ILD. Those proteins found to correlate significantly with at least 2 clinical measures of ILD were entered into a cluster analysis with inter-correlations expressed as a heatmap.Results:Significant correlations were observed between fibrosis scores and several biologic pathways including pro-fibrotic factors (transforming growth factor beta [TGF-β], platelet-derived growth factor [PDGF]), proteins involved in tissue remodeling (Matrix metallopeptidase [MMP]-1,7,8,9; Hepatocyte growth factor [HGF]), and those involved in monocyte/macrophage migration and activation (Monocyte chemoattractant protein [MCP]-1,3; macrophage colony-stimulating factor [MCSF]). These same pathways correlated with the diffusing capacity for carbon monoxide (DLCO). In contrast, GGO scores correlated primarily with immune and inflammatory mediators (interleukin [IL]-5,8,13,15, IL-1 receptor antagonist and interferon gamma) with only limited overlap to proteins that related to fibrosis. Vascular endothelial growth factor (VEGF) levels were lower in patients with more extensive GGO, fibrosis and diffusion impairment, suggesting that vascular changes are a central feature of SSc-ILD. Specific proteins were highly correlated with one another in a pattern suggesting biologically-related networks (Figure) that might provide additional insight regarding disease pathogenesis.Conclusion:Combining a diverse analysis of BAL proteins with the rich dataset available from SSc-ILD patients participating in SLS I, the study findings suggest the involvement of distinct biologic pathways, inter-related networks, and specific biologic signatures associated with unique radiographic features of ILD. The relationship of these factors to other SSc disease features, patient outcomes and as predictors of treatment responses will be studied in future analyses.References:[1]Tashkin DP, et al. During the first 4 postnatal days, the atrioventricular specialized tissue of the rat contains butyrylcholinesterase alone. The next 7 days are associated with a mixture of both acetyl and butyryl activity, but after the 12th day, acetylcholinesterase is found to predominate largely. It is suggested that this change in activity is related to the growth of adrenergic nerves into the heart on the 4th day. Administration of antinerve growth factor prevents the development of these nerves and is found to delay the onset of the change in cholinesterase activity from butyryl to acetyl from the 5th day until the 21st. Only after the 31st day is acetylcholinesterase the most prominent enzyme in treated animals.

This study aimed to analyze the relationship of insulin-like growth factor 1 with bone mass in obese female. This study used a cross sectional study method using 30 obese women aged 19-23 years, body mass index (BMI) 25-35 kg / m2, normal blood pressure, normal resting heart rate (RHR), normal hemoglobin (Hb). and fasting blood glucose (FBG) <100 mg / dL. Measurement of IGF-1 levels used the Enzym Link Immunosorbent Assay (ELISA) method. Measurement of bone mass using TANITA (Body Composition Analyzer DC3607601 (2) -1604 FA, TANITA Corporation of America, Inc., USA). The data analysis technique used the Pearson product-moment test with Statistical Package for Social Science (SPSS). The results showed that mean levels of IGF-1 (1.17 ± 0.10) ng / mL and bone mass (2.49 ± 0.06) kg (r = 0.712, P ≤ 0.001). Our findings suggest that there was a positive correlation between IGF-1 levels and bone mass

Barsoum and Varshavsky (Proc. Natl. Acad. Sci. U.S.A. 80:5330-5334, 1983) suggest that polypeptide mitogens and the mitogenic tumor promoter 12-O-tetradecanoylphorbol-13-acetate (TPA) stimulate gene amplification by related pathways. I demonstrated that TPA and the polypeptide mitogen fibroblast growth factor (FGF) both increase the frequency of cadmium-resistant variants of Swiss-Webster 3T3 cells. The molecular basis for this phenomenon is the amplification of the metallothionein gene(s). To further characterize the relationship between mitogenesis and gene amplification, I examined the ability of TPA and FGF to increase the frequency of cadmium-resistant colonies in the 3T3 variant cell line 3T3-TNR9. Unlike 3T3 cells, 3T3-TNR9 cells cannot be stimulated by TPA to divide (E. Butler-Gralla and H. R. Herschman, J. Cell. Physiol. 107:59-68, 1981). TPA does not induce an increase in cadmium-resistant colonies of the TPA-nonproliferative 3T3-TNR9, variant, in contrast to its efficacy on 3T3 cells. FGF, a potent mitogen for 3T3-TNR9 cells as well as 3T3 cells, is equally effective for 3T3-TNR9 and 3T3 cells in inducing cadmium-resistant colonies. These data suggest that the pathways of TPA-induced gene amplification and TPA-stimulated mitogenesis share a common step(s). TPA caused transient inhibition of DNA synthesis in both dividing 3T3 and 3T3-TNR9 cells, suggesting that this latter response to TPA is not sufficient to enhance gene amplification.
