In the post-resuscitation period through the I-II-III-IV-V - stages and long - term periods (1 and 3 months) of post – resuscitation disease, the morpho functional activity of β-and d-basophil cells of the adenohypophysis was studied in white male rats with a weight of 180-220 g, who suffered a 10-minute stop of systemic circulation (compression of the vascular bundle of the heart by the method of V. G. Korpachev). False-operated animals served as a control. Morphological, morphometric, histochemical, and cytophotometric studies of the state of both β - and d - basophil cells of the adenohypophysis were performed. We used image analysis using a leys microscope with an electronic micro-nozzle attached to an Intel computer, and the content of the glycoprotein was studied using a cytophotometer. In the postoperative period during the phase I identified in the background of gipergidratace cytoplasmic β - and d - basophilic cells of the anterior pituitary, in the period of II-III stage disease gipergidratace nuclei β - and d - basophilic cells of the anterior pituitary revealed an increase in the excretion of glycoprotein in blood, i.e., the observed compensatory-adaptive reactions. Starting from stage IV and in remote periods of post-resuscitation disease, compensatory and restorative processes were observed against the background of hyperhydration of the nuclei.

THE AIM: to study of the dynamics of structural changes in renal parenchyma of rats exposed to long-term combined effects of thyroxine and propylthiouracilum (PTU). MATHERIAL AND METHODS – studies were performed on mongrel white male rats weighing 250-300g. Hyperthyroidism was caused by daily intragastric administration of thyroxine (T4) in amount of 50g per 100g of body weight over 30 days. On the first day of the experiment animals were divided into 2 groups. Animals of the first group (n = 25) received only T4. The rats of the second group (n = 25) were administrated propylthiouracilum and T4 daily. PTU was administered intragastric in amount of 1 mg per 100g of body weight. Kidney tissue samples were collected on the 10th, 20th and 30th days of the experiment. In addition, there were collected kidney tissue samples of the animals treated with only T4 after 20 days after cessation of hormone. Obtained tissue samples were fixed and treated by the usual method, followed by filling in paraffin. Sections were stained with hematoxylin and eosin. RESULTS – it was established that course of experimental hyperthyroidism leads to significant structural abnormalities of the renal parenchyma. Leading features of kidneys pathology at a hyperthyroidism are rough structural damages of the nephron tubular epithelium. CONCLUSIONS – combined administration in rats of thyroxin and propylthiouracilum has weakly expressed beneficial effect by limiting the development of structural damages to the renal parenchyma and clot formation.

Introduction. The study of the effects of wildfire smoke on the reproductive function is the most important scientific problem at the present stage. The aim of study is to study the behavior and cognitive abilities of sexually mature offspring obtained from white male rats exposed to the smoke of wildfires. Materials and methods. Experiments were carried out on 30 white male rats and 60 adult male of their offspring. The duration of exposure was 4 weeks, 4 hours/day. To obtain progeny exposed males were mated with intact females immediately after the exposure and in long-term period after exposure. Examination of adult progeny was performed using the "open field", Morris water maze, and histological examination of brain tissue. Results. Behavioral and cognitive alterations in offspring of male rats exposed to wildfire smoke were obtained: significant decrease in motor activity and research on higher level of negative emotional state, impaired spatial memory performance. Individuals from offspring obtained in a long-term period after exposure to smoke showed a normalization of the studied parameters to background values. Conclusions. The long-term consequences of exposure to the smoke of wildfires were revealed, manifested in the offspring of the first generation in the form of impaired behavior and cognitive abilities. The recovery period after prolonged smoke intoxication, which is 60 days and is comparable in duration with the duration of spermatogenesis in rats, is important for reducing the risk of developing CNS disorders in offspring.

Cholestasis (disturbance of the liver bile outflow into the duodenum) is a frequent complication of cholelithiasis and other pathologies of the hepatobiliary system. In this case, all metabolism types are disturbances and all body organs and systems, including the nervous system, are affected. The objective of the study was to establish changes in the c-fos immunoreactivity in the cerebellum neurons of rats at different time after modeling subhepatic cholestasis. In this work, we used a material of 60 white male rats 200–250 g in weight. In experimental animals, the ligation of the common bile duct was carried out, in control animals – a false operation while maintaining a physiological bile flow in the duodenum throughout the experiment. Subhepatic cholestasis in rats is accompanied by an increase in the cortex cerebellum on the 2–20th days after the ligation of the common bile duct of the number of neurons with the increased c-fos-immunoreactivity, with the maximum one on the 10th day of the experiment. In surviving animals, after the removal of cholestasis in the long term (45–90 days), the number of neurons with the increased c-fos immunoreactivity, as well as the expression of this protein in neurons is normalized.

Background. Due to the fact that it is not always possible to reproduce all known manifestations of carbohydrate and lipid metabolism disorders, for example, in rodents, it is necessary to verify individual pathogenetic links in animals when modeling the metabolic syndrome in order to select the most optimal natural biotropic factors for studying the effect on them. Aims: To analyze metabolic disorders in the modeling of metabolic syndrome of alimentary genesis in comparison with the combined effect of alimentary factor and chemical agents to select the most appropriate model in the study of natural and weak preformed physical factors. Materials and methods. Metabolic syndrome was modeled in 47 sexually mature outbred white male rats using a hypercaloric diet (HD) of various duration and medications. The first series was the 1st control group (CG1) and the 1st experimental group (EG1). They received HD during 180 days, they were withdrawn from the experiment 30 days after the cancellation of HD. The second series was the 2nd control group (CG2) and three experimental groups. They received HD for 60 days, where the 2nd group (EG2) in association with HD received Mercazolil (10 mg/kg of animal weight) intragastrically for 14 days starting from the 21st day; the 3rd group (EG3) had the same scheme of treatment as in EG2, besides, after taking Mercazolil, they were intramuscularly injected with Nitox 200 at 25 mg/kg once per day during 5 days. Animals of EG2 and EG3 were withdrawn from the experiment 60 days after ending of HD. Animals of the 4th experimental group (EG4) got HD and the same medicine as in EG3. They left the experiment 30 days after ending HD. The content of hormones and biochemical parameters of protein, fat, and carbohydrate in peripheral blood was evaluated using multiple inter-group comparisons and intra-group relationships by Spearmans rank correlation method. Results. There have been detected a large number of biochemical signs of dependence of metabolism regulation on leptin level after long-term HD (180 days), while there were fewer biochemical signs of protein exchange disorder less than with the combined use of HD (60 days) and drugs. Conclusion. It is more appropriate and promising to use a long-term HD as a model of MS close to the natural conditions of its formation, to study the influence of natural and weak preformed physical factors.

The aim of the paper is to study thymus variability in white rats, which were exposed to formaldehyde, and to compare these data with the indicators in control animals. Materials and Methods. The trial enrolled 72 white male rats, initial body weight 40–50 g. The animals were divided into 2 groups (36 rats in each). The first group consisted of control rats. Animals of the second group were exposed to formaldehyde inhalation, 2.766 mg/m3. To characterize the variability of the organ size, centroids were determined. The superposition of landmark configurations was performed using the generalized Procrustes analysis method, MorphoJ 1.06d program. The principal component analysis and canonical analysis of the obtained data were carried out. Results. One-Way ANOVA revealed a high level of intergroup differences in Procrust distance (F=1.34; p<0.0001). The significant effect of the duration of formaldehyde exposure on centroid size was established. The Kruskal-Wallis criterion was 19.778 (p=0.0014). The analysis of the principal components indicated that each of the first 10 components stands for more than 1 % of Procrustes coordinate variance. In this case, the first 7 components compatibly explain 91.398 % of thymus variability. The proportion of the first main component to the total variance of the Procrustes coordinates is 40.236 %. PC1 (-) shows changes in the thymus shape, mostly affecting the tops of its lobes, the middle part of the right boundary and the entire left thymus boundary. The scattering ellipses of the thymus ordinates in rats exposed to formaldehyde, in the first two canonical variables are located higher than those in the control animals. Conclusion. Formaldehyde inhalation leads to thymus changes in white rat. The most significant differences with control data are determined along the second canonical variable. Изучение изменчивости формы тимуса белых крыс, находившихся в условиях влияния формальдегида, и сравнение этих данных с показателями, полученными у контрольных животных. Материалы и методы. Работа выполнена на 72 белых крысах-самцах с начальной массой тела 40–50 г. Животные были разделены на 2 серии (по 36 крыс). Первую серию составляли контрольные крысы. Животные второй серии подвергались ингаляционному воздействию формальдегида (ФА) в концентрации 2,766 мг/м3. Для характеристики изменчивости размеров органов определяли размер их центроидов. Процедуру суперимпозиции конфигураций ландмарок выполняли методом генерализованного прокрустова анализа с использованием программы MorphoJ 1.06d. Проводили анализ главных компонент и канонический анализ полученных данных. Результаты. Однофакторный дисперсионный анализ выявил высокий уровень межгрупповых различий по показателю прокрустовых расстояний (F=1,34; р<0,0001). Установлено значительное влияние продолжительности нахождения животных в условиях воздействия ФА на размер центроида. Критерий Краскела–Уоллиса составил 19,778 (р=0,0014). Анализ главных компонент указывал на то, что каждая из первых 10 компонент объясняет более 1 % дисперсии прокрустовых координат. При этом первые 7 компонент совместно объясняют 91,398 % изменчивости формы тимуса. Вклад первой главной компоненты в общую дисперсию прокрустовых координат составляет 40,236 %. РС1 (–) показывает изменения формы тимуса, в большей степени затрагивающие верхушки его долей, среднюю часть правого контура и весь левый контур тимуса. Эллипсы рассеивания ординат тимусов, принадлежащих крысам, подвергавшимся влиянию ФА, в пространстве первых двух канонических переменных расположены выше по отношению к таковым контрольных животных. Заключение. Ингаляционное воздействие формальдегида приводит к изменению формы тимуса белых крыс. Наибольшие различия с контрольными данными определяются вдоль второй канонической переменной. Abstract Background Testosterone regulates nutrient and energy balance to maintain protein synthesis and metabolism in cardiomyocytes, but supraphysiological concentrations induce cardiac hypertrophy. Previously, we determined that testosterone increased glucose uptake—via AMP-activated protein kinase (AMPK)—after acute treatment in cardiomyocytes. However, whether elevated glucose uptake is involved in long-term changes of glucose metabolism or is required during cardiomyocyte growth remained unknown. In this study, we hypothesized that glucose uptake and glycolysis increase in testosterone-treated cardiomyocytes through AMPK and androgen receptor (AR). Methods Cultured cardiomyocytes were stimulated with 100 nM testosterone for 24 h, and hypertrophy was verified by increased cell size and mRNA levels of β-myosin heavy chain (β-mhc). Glucose uptake was assessed by 2-NBDG. Glycolysis and glycolytic capacity were determined by measuring extracellular acidification rate (ECAR). Results Testosterone induced cardiomyocyte hypertrophy that was accompanied by increased glucose uptake, glycolysis enhancement and upregulated mRNA expression of hexokinase 2. In addition, testosterone increased AMPK phosphorylation (Thr172), while inhibition of both AMPK and AR blocked glycolysis and cardiomyocyte hypertrophy induced by testosterone. Moreover, testosterone supplementation in adult male rats by 5 weeks induced cardiac hypertrophy and upregulated β-mhc, Hk2 and Pfk2 mRNA levels. Conclusion These results indicate that testosterone stimulates glucose metabolism by activation of AMPK and AR signaling which are critical to induce cardiomyocyte hypertrophy.

Abstract Background Laparoscopic Heller myotomy (HM) has gained acceptance as the gold standard of treatment for achalasia. However, 10–20% of the patients will experience symptom recurrence, thus requiring further treatment including pneumodilations (PD) or revisional surgery. The aim of our study was to assess the long-term outcome of laparoscopic redo HM. Methods Patients who underwent redo HM at our center between 2000 and 2019 were enrolled. Postoperative outcomes of redo HM patients (redo group) were compared with that of patients who underwent primary laparoscopic HM in the same time span (control group). For the control group, we randomly selected patients matched for age, sex, FU time, Eckardt score (ES), previous PD, and radiological stage. Failure was defined as an Eckardt score > 3 or the need for re-treatment. Results Forty-nine patients underwent laparoscopic redo HM after failed primary HM. A new myotomy on the right lateral wall of the EGJ was the procedure of choice in the majority of patients (83.7%). In 36 patients (73.5%) an anti-reflux procedure was deemed necessary. Postoperative outcomes were somewhat less satisfactory, albeit comparable to the control group; the incidence of postoperative GERD was higher in the redo group (p < 0.01). At a median 5-year FU time, a good outcome was obtained in 71.4% of patients in the redo group; further 5 patients (10.2%) obtained a long-term symptom control after complementary PD, thus bringing the overall success rate to 81.6%. Stage IV disease at presentation was independently associated with a poor outcome of revisional LHD (p = 0.003). Conclusions This study reports the largest case series of laparoscopic redo HM to date. The procedure, albeit difficult, is safe and effective in relieving symptoms in this group of patients with a highly refractory disease. The failure rate, albeit not significantly, and the post-operative reflux are higher than after primary HM. Patients with stage IV disease are at high risk of esophagectomy.

Lung hepatoid adenocarcinoma (HAC) is a rare primary lung carcinoma pathologically characterized by hepatocellular carcinoma-like tumor cells, the majority of which produce alpha-fetoprotein (AFP). The clinical prognosis of lung HAC is generally poor, and effective therapeutic regimens for inoperable or recurrent cases have not been established. Here, we report a case of AFP-producing lung HAC with brain metastasis with long-term disease control, treated with the 5-fluorouracil-derived regimen S-1. The patient was a 66-year-old male admitted to the hospital with alexia. Chest X-ray revealed a massive tumor in the left upper lobe, and a head CT scan revealed a metastasis in the left parietal lobe. The laboratory data showed a remarkably elevated AFP level (97,561 ng/mL). Pathological assessment of the resected brain tumor revealed HAC, which was compatible with the lung biopsies. Together with the absence of other metastatic lesions, a final diagnosis of primary lung HAC, stage IV T4N3M1b, was given. The patient first underwent non-small cell lung cancer chemotherapy regimens (carboplatin and paclitaxel as the first line, and pemetrexed as the second line), but had clinical progression. After third-line oral S-1 (tegafur/gimeracil/oteracil) administration, the serum AFP level significantly dropped and the patient achieved long-term disease control without relapse, surviving more than 19 months after disease presentation. The autopsy result was consistent with the diagnosis of primary lung HAC, and immunohistochemical staining was AFP+, glypican 3+, and spalt-like transcription factor 4+. Here, we report the case of a rare primary lung HAC with apparent disease control on S-1 therapy, together with a literature review.

CONTEXT AND OBJECTIVE: Long-term totally implantable catheters (e.g. Port-a-Cath®) are frequently used for long-term venous access in children with cancer. The use of this type of catheter is associated with complications such as infection, extrusion, extravasation and thrombosis. Embolism of catheter fragments is a rare complication, but has potential for morbidity. The aim here was to report on two cases in which embolism of fragments of a long-term totally implantable catheter occurred. DESIGN AND SETTING: Case series study at Hospital do Servidor Público Estadual, São Paulo. METHODS: Retrospective review of catheter embolism in oncological pediatric patients with long-term totally implantable catheters. RESULTS: The first patient was a 3-year-old girl diagnosed with stage IV Wilms' tumor. Treatment was started with the introduction of a totally implantable catheter through the subclavian vein. At the time of removal, it was realized that the catheter had fractured inside the heart. An endovascular procedure was necessary to remove the fragment. The second case was a boy diagnosed with stage II Wilms' tumor at the age of two years. At the time of removal, it was noticed that the catheter had disconnected from the reservoir and an endovascular procedure was also necessary to remove the embolized catheter. CONCLUSION: Embolism of fragments of totally implantable catheters is a rare complication that needs to be recognized even in asymptomatic patients.
