Fusarium wilt incited by Fusarium oxysporum f. sp. ciceris is an economically damaging disease of chickpea (Cicer arietinum L.). Field experiments on epidemiological studies revealed that sowing during second fortnight of November curtailed the disease severity index (22.50 and 20.83% during 2018-19 and 2019-20, respectively) whereas, sowing during first fortnight of October enhanced the disease severity index (34.86 and 30.41%). The area under disease progress curve was significantly higher in susceptible variety JG 62 and was the least in resistant variety HC 1. The correlation analysis exhibited positive correlation of disease severity index with maximum and minimum temperature while negative correlation with relative humidity morning and evening, irrespective of date of sowing. The principal component analysis depicted resistance index, sowing time and weather parameters as positional factors in determining Fusarium wilt progression.  In susceptible variety, Gompertz model was the best fitted model for simulating the Fusarium wilt epidemic over time.

Abstract Purpose Restless legs syndrome (RLS) is a serious burden for patients which can be measured in economic terms by assessing the money spent on therapy and the willingness to pay. The aim of this study was to assess whether or not patients feel satisfied with the therapy relative to the money they spent on the treatment, and to assess patients’ willingness to pay for therapy that fully eliminates all RLS symptoms. Methods Adult subjects with RLS confirmed by actual international consensus criteria, a positive RLS-Diagnostic Index (RLS-DI) score, and clinical examinations and observations were assessed to generate a disease severity index. An original set of questions was used to collect data on patient satisfaction with therapy and their willingness to pay. Results Among 100 subjects, 27% were not satisfied with therapy; this subgroup was characterized by lower indices of severity of the disease. Patients spent approximately 3% of their income in treating RLS. They are willing to pay up to 8.3% of their income to eliminate symptoms. Conclusions The cost of RLS therapy is a significant part of patient expenditure. Nevertheless, RLS may at times remain so troublesome for patients that they are willing to spend more on therapy to eliminate symptoms.

Bacterial leaf blight (BLB) of rice is an economically important disease caused by Xanthomonas oryzae pv. oryzae (Xoo) throughout the world. To control this disease, bacterial isolate of Bacillus subtilis UiTMB1 was screened for the antagonistic activity against the pathogen in vitro and in vivo studies. A bacterial assay and detached leaf technique were used to evaluate the potential of the bacterium against BLB pathogen in the laboratory. Meanwhile, the glasshouse study was conducted to further examine the aptitudes of the isolate on the disease control and growth-promoting of rice plants. The findings revealed that B. subtilis UiTMB1 is able to control the disease and enhance the growth of rice plants. Rice plants treated with B. subtilis UiTMB1 before being inoculated with BLB pathogen showed less severe disease symptoms with low disease severity index of 3.43 compared to rice plants without B. subtilis UiTMB1 with high disease severity index of 8.4. Besides controlling the disease, B. subtilis UiTMB1 was also promoting plant height, chlorophyll content, number of tillers and biomass of rice plants.

Fusarium wilt in bananas is one of the most devastating diseases that poses a serious threat to the banana industry globally. With no effective control measures available to date, biological control has been explored to restrict the spread and manage the outbreak. We studied the effective biological control potential of different Trichoderma spp. in the management of Fusarium oxysporum f. sp. cubense tropical race 4 (Foc TR4). Expression of the defense related genes and metabolites in banana plants inoculated with Foc TR4 and treated with effective Trichoderma sp interactions were also studied. The in vitro growth inhibition of Foc TR4 by Trichoderma reesei isolate CSR-T-3 was 85.19% indicating a higher antagonistic potential than other Trichoderma isolates used in the study. Further, in in vivo assays, the banana plants treated with the isolate CSR-T-3 T. reesei had a significant reduction in the disease severity index (0.75) and also had increased phenological indices with respect to Foc TR4 treated plants. Enhanced activity of defense enzymes, such as β-1, 3-glucanase, peroxidase, chitinase, polyphenol oxidase, and phenylalanine ammonia lyase with higher phenol contents were found in the Trichoderma isolate CSR-T-3 treated banana plants challenge-inoculated with Foc TR4. Fusarium toxins, such as fusaristatin A, fusarin C, chlamydosporal, and beauveric acid were identified by LC-MS in Foc TR4-infected banana plants while high intensity production of antifungal compounds, such as ß-caryophyllene, catechin-o-gallate, soyasapogenol rhamnosyl glucoronide, peptaibols, fenigycin, iturin C19, anthocyanin, and gallocatechin-o-gallate were detected in T. reesei isolate CSR-T-3 treated plants previously inoculated with Foc TR4. Gene expression analysis indicated the upregulation of TrCBH1/TrCBH2, TrXYL1, TrEGL1, TrTMK1, TrTGA1, and TrVEL1 genes in CSR-T-3 treatment. LC-MS and gene expression analysis could ascertain the upregulation of genes involved in mycoparasitism and the signal transduction pathway leading to secondary metabolite production under CSR-T-3 treatment. The plants in the field study showed a reduced disease severity index (1.14) with high phenological growth and yield indices when treated with T. reesei isolate CSR-T-3 formulation. We report here an effective biocontrol-based management technological transformation from lab to the field for successful control of Fusarium wilt disease caused by Foc TR4 in bananas.

Mal de Río Cuarto (MRC) is one of the most important viral diseases of maize in Argentina. The disease severity index (DSI) allows to combine the incidence and severity of a disease in a single metric. The genotypic reaction to MRC has been extensively studied in biparental populations. However, this complex trait has not been analyzed by genome-wide association studies (GWAS). The aim of this work is to identify new resistance alleles associated with DSI of MRC in an exotic germplasm from the International Maize and Wheat Improvement Center (CIMMYT). A population of maize lines from CIMMYT was phenotypically evaluated in environments in the area where the disease is endemic. The predictors of genetic effects (BLUP, best linear unbiased predictor) and 78,376 SNP markers (Single Nucleotide Polymorphism) were used to perform the GWAS in 186 maize lines. The values of variance components and mean-basis heritability suggest a wide genotypic variability in the population. The GWAS allowed to identify 11 putative QTL of resistance to MRC. The incorporation of exotic germplasm into local maize breeding programs could contribute favorably to the creation of hybrids with a higher level of resistance to MRC. The predictive ability of associated markers with MRC resistance indicates that marker-assisted selection is an advisable tool for selecting MRC resistant genotypes. Establishment of disease in oil palm seedlings through artificial inoculation of Ganoderma are widely used for studies of various aspects of plant pathology, including epidemiology, etiology, disease resistance, host-parasite interaction and disease control. The estimation of chlorophyll content in the infected seedlings possibly could provide a good indicator for degree of disease or infection, and changes during pathogenesis. Thus, the objective of this study was to evaluate the relationship between disease severity index (DSI) and chlorophyll content in Ganoderma infected oil palm seedlings. Three-month-old oil palm seedlings were infected with Ganoderma inoculum on rubber wood block (RWB), where 44 isolates of Ganoderma were tested. Disease severity index (DSI) and chlorophyll content using a single-photon avalanche diode (SPAD) meter were recorded at 4 weeks interval for a period of 24 weeks after inoculation (WAI). Pearson's correlation analysis and regression analysis were performed to evaluate the relationship between the variables. It was found that the relationship between DSI and SPAD chlorophyll value was inversely proportional (R = -0.92) in a linear trend (R2 = 0.85). Furthermore, the increasing trend of the DSI across the weeks were fitted in a quadratic model (R2 = 0.99). In contrast, the SPAD chlorophyll value declined in a linear trend (R2 = 0.98). The SPAD chlorophyll value could be considered as a better alternative over the DSI as the SPAD chlorophyll value was strongly related to DSI, as well as able to detect physiological changes in the infected oil palm seedlings at the early stages of pathogenesis.
