FDP programme is aimed to demonstrate the ability of various Numerical Weather Prediction (NWP) models to assess the genesis, intensification and movement of cyclones over the north Indian ocean with enhanced observations over the data sparse region and to incorporate modification into the models which could be specific to the Bay of Bengal based on the in-situ measurements. FDP Programme is scheduled in three phases, viz., (i) Pre-pilot phase (15 Oct - 30 Nov 2008, 2009, (ii) Pilot phase (15 Oct - 30 Nov, 2010-2012) and (iii) Final phase (15 Oct - 30 Nov, 2013-14). India is planning to take up aircraft probing of cyclones over the Bay of Bengal during 15 Oct - 30 Nov, 2013-14 with hired aircraft and dropsonde experiments. To accomplish the above objective, the initiative was carried out with priorities on (i) observational upgradation, (ii) modernisation of cyclone analysis and prediction system, (iii) cyclone analysis and forecasting procedure, (iv) warning products generation, presentation & dissemination, (v) confidence building measures and capacity building. Various strategies were adopted for improvement of observation, analysis and prediction of cyclone. Several national institutions participated for joint observational, communicational & NWP activities during the pre-pilot and pilot phases of FDP campaign during 2008-11. The comparison of observational systems before and after FDP indicates a significant improvement in terms of Radar, Automatic Weather Station (AWS), high wind speed recorders over the region. It has resulted in reduction in monitoring and forecasting errors. The performance of NWP models have increased along with the introduction of NWP platforms like IMD GFS, WRF, HWRF and ensemble prediction system (EPS). Salient features of achievements along with the problems and prospects of this project are presented and discussed in this paper. With repeated attempts, the aircraft probing of cyclones could not be possible till now. It is a major challenge for the future campaign during 2013-14.

Abstract The paper is a written summary of an overview oral presentation given at the 1st Spanish Fusion HPC Workshop that took place on the 27th November 2020 as an online event. Given that over the next few years ITER will move to its operation phase and the European-DEMO design will be significantly advanced, the EUROfusion consortium has initiated a coordination effort in theory and advanced simulation to address some of the challenges of the fusion research in Horizon EUROPE (2021-2027), i.e. the next EU Framework Programme for Research and Technological Development. This initiative has been called E-TASC that stands for EUROfusion-Theory and Advanced Simulation Coordination. The general and guiding principles of E-TASC are summarized in the paper. In addition, an overview of the scientific results obtained in a pilot phase (2019-2020) of E-TASC are provided while highlighting the importance of the required progress in computational methods and HPC techniques. In the initial phase, five pilot theory and simulation tasks were initiated: 1. Towards a validated predictive capability of the L-H transition and pedestal physics; 2. Electron runaway in tokamak disruptions in the presence of massive material injection; 3. Fast code for the calculation of neoclassical toroidal viscosity in stellarators and tokamaks; 4. Development of a neutral gas kinetics modular code; 5. European edge and boundary code for reactor-relevant devices. In this paper we report on recent progress made by each of these projects.

BACKGROUND Black men who have sex with men (BMSM) suffer from alarmingly high rates of HIV in the United States. Pre-exposure prophylaxis (PrEP) can reduce risk of HIV infection by 99% among MSM, yet profound racial disparities in the uptake of PrEP persist. Low PrEP uptake in BMSM is driven by poor access to PrEP including inconvenient locations of PrEP-prescribing physicians and distrust of physicians and stigma, which limit communication about PrEP and its side effects. Previous work indicates that offering HIV prevention services in pharmacies located in high poverty, racial minority neighborhoods is feasible and can reduce stigma because pharmacies offer a host of less stigmatized health services (e.g. vaccinations). We present a protocol for a pharmacy PrEP model that seeks to address challenges and barriers to pharmacy-based PrEP specifically for BMSM. OBJECTIVE To develop a sustainable pharmacy PrEP delivery model that can be implemented to increase PrEP access in high poverty, racial minority neighborhoods for BMSM. METHODS This study design is a pilot intervention to test a pharmacy PrEP delivery model among pharmacy staff and BMSM. We will examine the PrEP delivery model’s feasibility, acceptability and safety, and gather early evidence of its impact and cost with respect to PrEP uptake. A mixed methods approach will be performed including three study phases: 1) a completed formative phase with qualitative interviews from key stakeholders; 2) a completed transitional pilot phase to assess customer eligibility and willingness to receive PrEP in pharmacies during COVID-19; and 3) a planned pilot intervention phase which will test the delivery model in two Atlanta pharmacies in high poverty, racial minority neighborhoods. RESULTS Data from the formative phase showed strong support of pharmacy-based PrEP delivery among BMSM, pharmacists and pharmacy staff. Important factors were identified to facilitate implementation of PrEP screening and dissemination in pharmacies. During the transitional pilot phase, we identified 81 individuals who would have been eligible for the pilot phase. CONCLUSIONS Pharmacies have proven to be a feasible source for offering PrEP for White MSM, but have failed to reach the most at-risk, vulnerable population – BMSM. Increasing PrEP access and uptake will reduce HIV incidence and racial inequities in HIV. Translational studies are required to build further evidence and scale pharmacy-based PrEP services specifically for populations that are disconnected from HIV prevention resources.

Abstract Wellbore stability problems, such as stuck pipe and tight spots, are one of the most critical risks that impact drilling operations. Over several years, Oil and Gas Operator in Middle East has been facing problems associated with stuck pipe and tight spot events, which have a major impact on drilling efficiency, well cost, and the carbon footprint of drilling operations. On average, the operator loses 200 days a year (Non-Productive Time) on stuck pipe and associated fishing operations. Wellbore stability problems are hard to predict due to the varying conditions of drilling operations: different lithology, drilling parameters, pressures, equipment, shifting crews, and multiple well designs. All these factors make the occurrence of a stuck pipe quite hard to mitigate only through human intervention. For this reason, The operator decided to develop an artificial intelligence tool that leverages the whole breadth and depth of operator data (reports, sensor data, well engineering data, lithology data, etc.) in order to predict and prevent wellbore stability problems. The tool informs well engineers and rig crews about possible risks both during the well planning and well execution phase, suggesting possible mitigation actions to avoid getting stuck. Since the alarms are given ahead of the bit, several hours before the possible occurrence of the event, the well engineers and rig crews have ample time to react to the alarms and prevent its occurrence. So far, the tool has been deployed in a pilot phase on 38 wells giving 44 true alarms with a recall of 94%. Since mid-2021 operator has been rolling out the tool scaling to the whole drilling operations (over 40 rigs).

Summer monsoon season of the year 2009 resulted in a major drought on the scale of India with rainfall deficiency of 23% from the normal. This was the monsoon season when a pilot phase of the programme Continental Tropical Convergence Zone (CTCZ), a planned multiyear programme to understand the complex interactions among the land, ocean, atmosphere, biosphere components of the regional monsoon climate system, was undertaken. The paper attempts to document the major features in the evolution of monsoon 2009 and provides a preliminary diagnosis of the causes for monsoon drought.

The spread of COVID-19 has created numerous challenges in the field of archive management. Limited in-house office space, furloughs of personnel, and inconsistency, has highlighted the potential for the Carl Albert Congressional Research and Studies Center Archives (Center) to develop and implement improved accessibility measures to thousands of linear feet of material. Addition ally, the Center has found unique opportunities to collaborate with multiple academic institutions to propose large-scale digitization program exhibitions using the Center’s remote workflow model. One of the largest, most complex collections the Center has worked with during this time is the Political Commercial Collection (the Collection), which holds 119,000 film, audio, and videotape recordings of commercials aired between 1936 and present. It is the largest collection of political commercials in the world. The Center has developed a working pilot digitization project that has currently resulted in access to 16,000 digital videos for public researchers and over 10,000 available for on-line streaming during the pilot phase between April 16, 2020, and December 1, 2020. This paper presents the practical application of the Center’s simplified “Linear Reciprocity Workflow Model” to provide a systematic solution for digital and long-term preservation of complex collections. The Center has proven that limited personnel and reduced resources need not interrupt continued access to archival repositories.

Abstract Background: IL-21 expanded NK cells have high expression of CD16 and have demonstrated antibody-dependent cell-mediated cytotoxicity (ADCC) activity in combination with monoclonal antitumor antibodies (mAb). Mogamulizumab (moga) is a mAb targeting CCR4 that is defucosylated to enhance its binding to CD16, thereby enhancing ADCC of NK cells against targets expressing CCR4. We designed a pilot phase I clinical trial studying this combination in patients with relapsed/refractory (r/r) CTCL and ATLL. The study is soon opening to accrual at the OSU James Cancer Center (NCT04848064). Study is conduced under IND 26888. Preclinical data: Allogeneic NK cells obtained from buffy coat (Red Cross Blood Bank), were expanded for 14 days on CSTX002 feeder cells, cryopreserved, and then thawed and recovered for 48 hours prior to testing. Malignant T-cells were incubated with moga (at 10ng/µl) for 30 minutes prior to co-culture with NK cells and cytotoxicity was determined by the calcein release assay (Somanchi et al, J Vis Exp 2011). Malignant T-cells were obtained from peripheral blood from 3 patients with multiply relapsed CTCL and all have circulating Sezary cells and from CCRF-CEM cell line (T-ALL cell line that expresses CCR4). No significant cytotoxicity was observed with moga alone and significant synergy in cytotoxicity was observed between and moga and NK cells in all 3 patient samples and also CCRF-CEM cell line (figure 1: A and B). Two-fold increase in ADCC was observed with addition of moga to NK cells (p=0.0272; figure 1C) Design: Patients will receive lymphodepleting chemotherapy (Fludarabine/Cyclophosphamide) on days -5 to -3 prior to cell infusion, moga weekly for 4 doses starting on day -7 (prior to the first NK cell infusion) and then every 2 weeks until toxicity or progression. Patients will receive third-party ideal-donor mbIL-21 expanded NK cells once every 2 weeks for 6 total doses (Figure 1D). Donors meeting ideal-donor characteristics from National Marrow Donor Program were identified in collaboration with Be The Match Biotherapies. PBMNC were collected by apheresis, CD3-depleted, expanded for 14 days as previously described, and cryopreserved in ready-to-infuse aliquots. NK cells will be thawed and infused in 2 dosing cohorts; 3x10 7 and 1x10 8 cells/kg in a standard dose-escalation design. Primary endpoint is the maximum tolerated dose of NK cells given in combination with standard-dose moga. Dose-limiting toxicity (DLT) is defined as any steroid refractory graft vs host disease (GVHD), severe NK cell-related toxicities, or other unusual events occurring from D-7 until D+84 post last dose of NK cells. Dose-escalation will proceed in the standard 3+3 fashion. Secondary endpoints include overall response rate (ORR) per ISCL/USCLC/EORTC consensus panel, for CTCL, and international consensus panel, for ATLL and progression-free and overall survival. Correlative endpoints include quality of life impact as captured by Skindex-16 score, serum cytokine levels in blood, persistence of NK cells by chimerism studies, correlation between CCR4 staining of tumor cells in skin and trafficking of NK cells to skin by immunohistochemistry or immunofluorescence in serial skin biopsies and ORR (Figure 1D). Abbreviated eligibility: Eligible patients will be 18 years, or older, with biopsy-proven, measurable, stage IB-IVB relapsed or refractory CTCL or ATLL, progressing on at least one standard chemotherapy. Other eligibility criteria include: ECOG performance status of ≤ 1, no systemic anti-neoplastic therapy within a week or 3 half-lives, adequate laboratory parameters including: absolute neutrophil count ≥1000/mm³, platelet count ≥50,000/mm³, total bilirubin ≤ 2 x upper limit of normal (ULN), AST/ALT ≤ 3 x ULN or ≤ 5 x ULN in patients with documented hepatic involvement by lymphoma, and calculated creatinine clearance ≥ 50 ml/min., disease free of prior malignancies for ≥ 2 years with exception of treated basal cell, squamous cell carcinoma of the skin, or carcinoma in situ of the cervix or breast and life expectancy ≥ 90 days. Patients who were pre-treated with moga, pregnant, HIV positive, with active hepatitis B and C, active CNS involvement, active grade II-IV acute or extensive chronic GVHD or other serious medical comorbid conditions are excluded. BACKGROUND VITALISE is a H2020 project that aims to harmonize Living Lab procedures and facilitate the access to European Health and Wellbeing research infrastructures. In this context, this study presents a joint research activity (JRA) that will be conducted within VITALISE, in transitional care domain, in order to test and validate the harmonized Living Lab procedures and infrastructures. The collection of data from various sources (ICT, clinical and patient reported outcome measures) demonstrated capacity to assess risk and support decision during care transitions but there is no harmonized way of combining this information. OBJECTIVE This study primarily aims to evaluate the feasibility and benefit of collecting multichannel data across Living Labs on the topic of transitional care and to harmonize the data processes and collection. Secondly, we aim to investigate the collection and use of digital biomarkers and explore initial patterns in the data that demonstrate the potential to predict transition outcomes such as readmissions and adverse events. METHODS The current research protocol presents a multi-center, prospective, observational cohort study that will consist of three phases, running consecutively in multiple sites: a co-creation phase, a testing and simulation phase and a transnational pilot phase. The co-creation phase aims to build a common understanding among different sites, investigate the differences of hospitalization discharge management among countries and the willingness of different stakeholders to use technological solutions in the transitional care process. The testing and simulation phase aims to explore ways of integrating observation of a patient’s clinical condition, patient involvement and discharge education in transitional care. The objective of the simulation phase is to evaluate the feasibility and the barriers that are faced by a healthcare professional in assessing transition readiness. The transnational pilot phase takes input from co-creation and testing and stimulation phase. The aim is to pilot the already designed activities from previous phases and collect data to conduct a first predictive analysis. RESULTS The co-creation phase will be completed by April 2022. The testing and simulation phase will begin in September 2022 and will partially overlap with the deployment of the transnational pilot phase that will start the same month. The data collection of the transnational pilots will be finalized by the end of June 2023. Data processing is expected to be completed by March 2024. The results will consist of guidelines and implementation pathway for large scale study and the analysis for identifying initial patterns in the acquired data. CONCLUSIONS The knowledge acquired though this research will lead to harmonized procedures and data collection for Living Labs that support transitions in care. In addition, this research contributes to the increase in capacity to perform Big Data analytics while accounting for each local context and across Living Labs.
