Evaluation of Locally Isolated Entomopathogenic Fungi against Multiple Life Stages of Bactrocera zonata and Bactrocera dorsalis (Diptera: Tephritidae): Laboratory and Field Study

Fruit flies including Bactrocera zonata and B. dorsalis (Diptera: Tephritidae) are considered major pests of orchard systems in Pakistan. This study evaluated the laboratory virulence, sub-lethal effects, horizontal transmission, greenhouse, and field-cage efficacy of locally isolated entomopathogenic fungi (EPF) against B. zonata and B. dorsalis. In virulence assays against third instars and adults, all 21 EPF isolates (Beauveria bassiana and Metarhizium anisopliae) tested were pathogenic and caused varying levels of mortality to the fruit flies. Based on the initial screening, four isolates (B. bassiana WG-21 and WG-18 and M. anisopliae WG-07 and WG-02) were selected for further study. The isolate WG-18 was the most virulent against larvae and adults of B. zonata and B. dorsalis followed by WG-21, WG-02, and WG-07. In both species, adults were more susceptible than larvae to all isolates, and pupae were the least susceptible. Isolates WG-18 and WG-21 strongly decreased female fecundity and fertility, the highest adult and larval mortality, and longest developmental time of larvae and pupae. Fungal conidia were disseminated passively from infected to healthy adults and induced significant mortality, particularly from infected males to non-infected females. In greenhouse and field-cage experiments, WG-18 and WG-21 were the most effective isolates in reducing adult emergence when applied to larvae and pupae of both fruit fly species. Our results indicate that B. bassiana isolates WG-18 and WG-21 were the most virulent against multiple life stages of B. zonata and B. dorsalis, and also exerted the strongest sub-lethal effects.

Pollinators and plant nurseries: how irrigation and pesticide treatment of native ornamental plants impact solitary bees

A key conservation goal in agroecosystems is to understand how management practices may affect beneficial species, such as pollinators. Currently, broad gaps exist in our knowledge as to how horticultural management practices, such as irrigation level, might influence bee reproduction, particularly for solitary bees. Despite the extensive use of ornamental plants by bees, especially little is known about how irrigation level may interact with insecticides, like water-soluble neonicotinoids, to influence floral rewards and bee reproduction. We designed a two-factor field cage experiment in which we reared Megachile rotundata (Fabricius) (Hymenoptera: Megachilidae) on containerized ornamental plants grown under two different irrigation levels and imidacloprid treatments (30% label rate dosage of a nursery formulation or an untreated control). Lower irrigation was associated with modest decreases in nectar volume and floral abundance in untreated plants, whereas irrigation did not affect plants treated with imidacloprid. Furthermore, higher irrigation decreased the amount of imidacloprid entering nectar. Imidacloprid application strongly reduced bee foraging activity and reproduction, and higher irrigation did not offset any negative effects on bees. Our study emphasizes the impact of a nursery neonicotinoid formulation on solitary bee foraging and reproduction, while highlighting interactions between irrigation level and neonicotinoid application in containerized plants themselves.

Evaluation of Mass Trapping Devices for Early Seasonal Management of Ceratitis Capitata (Diptera: Tephritidae) Populations

Mass trapping is an environmentally safe alternative to insecticide application for the Mediterranean fruit fly management. The selection of effective trap-attractant combinations for monitoring and mass trapping control remains challenging. The current study explored the attractiveness of trapping devices during spring (early season) and summer (late season) in field cage trials. Five trapping devices were assessed: (a) the commercially available Decis® trap, (b) Tephri trap baited with Biodelear, (c) Tephri trap baited with BioLure, (d) International Pheromone McPhail trap (IPMT) baited with Biodelear, and (e) IPMT baited with BioLure. On a test day, 100 adults (50 males and 50 females) were released in each field cage wherein traps were placed individually. Trap captures were recorded at hourly intervals from 10:30 am to 5:30 pm. Our results showed that Tephri traps baited either with BioLure or Biodelear captured the most adults under low temperatures. Efficacy of Tephri traps baited with BioLure were higher than that of other trap-attractant combinations at high temperatures. Adult captures in Decis® trap were low during both seasons. More males than females were captured at low temperatures. Both efficacy and female selectivity of trapping devices are related to prevailing temperature regimes during spring and summer under semi-field conditions.

Field Cage Assessment of Feeding Damage by Riptortus pedestris on Soybeans in China

The bean bug, Riptortus pedestris, is a major pest of soybeans. In order to assess the critical stages of soybean damage by R. pedestris, we tested the damage to soybeans at different growth stages (R2, R4, and R6) caused by five densities of R. pedestris (1, 2, 3, 4, and 5) through a field cage experiment. The results show that the R4 stage was the most sensitive stage in terms of suffering R. pedestris injury damage, followed by the R6 stage and then the R2 stage. The number of stay green leaves was 7.04 per plant, the abortive pod rate of the soybeans was 56.36%, and the abortive seed rate of the soybeans was 46.69%. The dry weight of the soybeans was 14.20 g at the R4 stage; these values of R4 were significantly higher than at the R2 and R6 stages. However, the dry weight of soybean seed was 4.27 g and the nutrient transfer rate was 27.01% in the R4 stage; these values were significantly lower than in the R2 and R6 stages. The number of stay green leaves, abortive pod rates, and abortive seed rates were all increased significantly with increasing pest density at each stage of soybean growth. However, the nutrient transfer rate was significantly decreased with the increase in the pest density. Soybean nutrition factors changed after they suffered R. pedestris injury; the lipid content of the soybean seed decreased and the lipid content of the soybean plant increased compared to controls, when tested with a density of five R. pedestris in the R4 stage. These results will be beneficial to the future management of R. pedestris in soybean fields.

Sexual Competitiveness and Induced Egg Sterility by Aedes aegypti and Aedes albopictus Gamma-Irradiated Males: A Laboratory and Field Study in Mexico

The sterile insect technique may prove useful for the suppression of mosquito vectors of medical importance in regions where arboviruses pose a serious public health threat. In the present study, we examined the effects of sterilizing irradiation doses across different ratios of fertile:irradiated males on the mating competitiveness of Ae. aegypti and Ae. albopictus under laboratory and field-cage conditions. For both species, the percentage of females inseminated and the number of eggs laid over two gonotrophic cycles varied significantly in mating treatments involving 1:1, 1:5, and 1:10 fertile:irradiated males compared to controls of entirely fertile or entirely irradiated males but was not generally affected by the irradiation dose. Egg hatching was negatively affected in females exposed to increasing proportions of irradiated males in both laboratory and field cages. Male competitiveness (Fried’s index) values varied from 0.19 to 0.58 in the laboratory and were between 0.09 and 1.0 in field cages, depending on th species. Competitiveness values were negatively affected by th eirradiation dose in both species under field-cage conditions, whereas in the laboratory, Ae. albopictus was sensitive to the dose but Ae. aegypti was not. In general, male competitiveness was similar across all mating regimes. Most importantly, induced egg sterility was positively correlated with the proportion of irradiated males present in the mating treatments, reaching a maximum of 88% under field-cage conditions for both Ae. aegypti and Ae. albopictus males treated with 50 and 40 Gy irradiation, respectively. These results indicate that sterile males produced at our facility are suitable and competitive enough for field pilot SIT projects and provide guidance to decide the optimal sterile:fertile ratios.

Detection of the Nav channel kdr-like mutation and modeling of factors affecting survivorship of Culex quinquefasciatus mosquitoes from six areas of Harris County (Houston), Texas, after permethrin field-cage tests

Culex quinquefasciatus is one of the most important mosquito vectors of arboviruses. Currently, the fastest approach to control disease transmission is the application of synthetic adulticide insecticides. However, in highly populated urban centers the development of insecticide resistance in mosquito populations could impair insecticide efficacy and therefore, disease control. To assess the effect of resistance on vector control, females of Cx. quinquefasciatus collected from six mosquito control operational areas in Harris County, Texas, were treated in field cage tests at three different distances with the pyrethroid Permanone® 31–66 applied at the operational rate. Females were analyzed by sequencing and/or diagnostic PCR using de novo designed primers for detecting the kdr-like mutation in the voltage-gated sodium channel (L982F; TTA to TTT) (house fly kdr canonical mutation L1014F). Females from the Cx. quinquefasciatus susceptible Sebring strain and those from the six operational areas placed at 30.4 m from the treatment source were killed in the tests, while 14% of field-collected mosquitoes survived at 60.8 m, and 35% at 91.2 m from the source. The diagnostic PCR had a with 97.5% accuracy to detect the kdr-like mutation. Pyrethroid resistant mosquitoes carrying the L982F mutation were broadly distributed in Harris County at high frequency. Among mosquitoes analyzed (n = 1,028), the kdr-kdr genotype was prevalent (81.2%), the kdr-s genotype was 18%, and s-s mosquitoes were less than 1% (n = 8). A logistic regression model estimated an equal probability of survival for the genotypes kdr-kdr and kdr-s in all areas analyzed. Altogether, our results point to a high-risk situation for the pyrethroid-based arboviral disease control in Harris County.

Biological Control Potential and Drawbacks of Three Zoophytophagous Mirid Predators against Bemisia tabaci in the United States

Miridae (Hemiptera) of the tribe Dicyphini are important zoophytophagous predators use to control pest arthropods in vegetable crops. However, the risk that their herbivory may cause economic damage could hinder their application as useful biocontrol agents and may limit the likelihood they would meet regulatory requirements for importation. We conducted field cage studies to assess the predation capacity and tomato plant damage of three mirid species established in south USA, a known biocontrol agent (Nesidiocoris tenuis), and two native species (Macrolophus praeclarus and Engytatus modestus). All three species significantly reduced the number of whiteflies (Bemisia tabaci) on tomato plants compared to tomato plants without mirids. More damage, evaluated as the number of necrotic rings, was observed on tomato plants with E. modestus and N. tenuis compared to M. praeclarus. In our experiments that included sesame plants (Sesamum indicum) with tomato plants, mirid numbers increased despite a low number of prey, thus showing a benefit of the plant-feeding habit of these predators. USA’s established mirids may therefore prove to be immediately available biological agents for the management of present and future tomato pests.
