Abstract Experiencing stress during sensitive periods of brain development has a major impact on how individuals cope with later stress. Although many become more prone to develop anxiety or depression, some appear resilient. The mechanisms underlying these differences are unknown. Key answers may lie in how genetic and environmental stressors interact to shape the circuits controlling emotions. Here we studied the role of the habenulo-interpeducuncular system (HIPS), a critical node of reward circuits, in early stress-induced anxiety. We found that a subcircuit of this system, characterized by Otx2 expression, is particularly responsive to chronic stress during puberty, which induces HIPS hypersensitivity to later stress and susceptibility to develop anxiety. We further show that Otx2 deletion restricted to the HIPS counteracts these effects of stress. Together, these results demonstrate that Otx2 and stress interact, around puberty, to shape the HIPS stress-response, revealed here as a key modulator of susceptibility/resilience to develop anxiety.

The work is devoted to the improvement of career guidance work in the specialty "Architecture and Urban Planning". Research objectives: 1. To study and generalize knowledge and experience on the organization of career guidance work. 2. Investigate the patterns of formation and development of thinking at different ages. 3. Summarize the data on career guidance work in the specialty "Architecture and Urban Planning" at the Dnieper State Academy of Civil Engineering and Architecture. 4. To substantiate the peculiarities of the organization of early career guidance work taking into account the laws of formation and development of different ages children thinking. The study was conducted by summarizing a number of theoretical provisions of various scientific concepts and practical experience, as an information base used official data published in the scientific literature. Conclusions. The particularity of the specialty "Architecture and Urban Planning" requires early specialization of future potential applicants for higher education, as well as the acquisition and improvement of certain skills from an early age. Early career guidance in the specialty "Architecture and Urban Planning" is the key to successful entry into the Free Economic Zone, as well as obtaining a specialty that meets the individual characteristics of the child and the maximum professional realization in future work. As a means of early career guidance, in full, should be used as a means of informing potential applicants, as well as training and selection measures for innate (features of thinking, channel perception activity) and acquired skills. Taking into account the sensitive periods of the child's development, during the organization of cognitive activity, significantly reduces the burden on the child and reorients educational activities to meet the relevant needs of certain functional systems during their formation and development. In order to save costs in the system of "learning − acquisition of knowledge" career guidance work associated with the development of certain abilities, it is desirable to conduct mainly in the relevant sensitive periods, taking into account the stages of formation and development of thinking.

Abstract Greater childhood adversity predicts a higher likelihood of later self-injurious thoughts and behaviors (SITB). There is little research focused on whether the timing of childhood adversity predicts SITB. The current research examined whether the timing of childhood adversity predicted parent- and youth-reported SITB at age 12 and 16 years in the Longitudinal Studies of Child Abuse and Neglect (LONGSCAN) cohort (n = 970). We found that greater adversity at age 11–12 years consistently predicted SITB at age 12 years, while greater adversity at age 13–14 years consistently predicted SITB at age 16 years. These findings suggest there may be sensitive periods during which adversity may be more likely to lead to adolescent SITB, which can inform prevention and treatment.

AbstractEarly childhood and pregnancy are two sensitive periods of heightened immune plasticity, when exposure to adversity may disproportionately increase health risks. However, we need deeper phenotyping to disentangle the impact of adversity during sensitive periods from that across the total lifespan. This study examined whether retrospective reports of adversity during childhood or pregnancy were associated with inflammatory imbalance, in an ethnically diverse cohort of 53 low-income women seeking family-based trauma treatment following exposure to interpersonal violence. Structured interviews assessed early life adversity (trauma exposure ≤ age 5), pregnancy adversity, and total lifetime adversity. Blood serum was assayed for pro-inflammatory (TNF-a, IL-1ß, IL-6, and CRP) and anti-inflammatory (IL-1RA, IL-4, and IL-10) cytokines. CD14+ monocytes were isolated in a subsample (n = 42) and gene expression assayed by RNA sequencing (Illumina HiSeq 4000; TruSeq cDNA library). The primary outcome was a macrophage-associated M1/M2 gene expression phenotype. To evaluate sensitivity and specificity, we contrasted M1/M2 gene expression with a second, clinically-validated macrophage-associated immunosuppressive phenotype (endotoxin tolerance) and with pro-inflammatory and anti-inflammatory cytokine levels. Adjusting for demographics, socioeconomic status, and psychopathology, higher adversity in early life (ß = .337, p = 0.029) and pregnancy (ß = .332, p = 0.032) were each associated with higher M1/M2 gene expression, whereas higher lifetime adversity (ß = −.341, p = 0.031) was associated with lower immunosuppressive gene expression. Adversity during sensitive periods was uniquely associated with M1/M2 imbalance, among low-income women with interpersonal violence exposure. Given that M1/M2 imbalance is found in sepsis, severe COVID-19 and myriad chronic diseases, these findings implicate novel immune mechanisms underlying the impact of adversity on health.

In recent years, the area dedicated to modern irrigated almond plantations has increased significantly in Spain. However, the legal irrigation allocations are lower than the maximum water requirements of the crop in most cases. Therefore, almond growers are forced to implement regulated deficit irrigation strategies on their farms, applying water stress in certain resistant phenological periods and avoiding it in sensitive periods. Given the need to monitor the water status of the crop, especially in the most sensitive periods to water stress, the objective of this work was to evaluate the sensitivity of two UAV-based crop water status indicators to detect early water stress conditions in four almond cultivars. The field trial was conducted during 2020 in an experimental almond orchard, where two irrigation strategies were established: full irrigation (FI), which received 100% of irrigation requirements (IR), and regulated deficit irrigation (RDI), which received 70% of IR during the whole irrigation period except during the kernel-filling stage when received 40% IR. The UAV flights were performed on four selected dates of the irrigation season. The Crop Water Status Index (CWSI) and the Normalized Difference Vegetation Index (NDVI) were derived from thermal and multispectral images, respectively, and compared to classical water status indicators, i.e., stem water potential (Ψstem), stomatal conductance (gs), and photosynthetic rate (AN). Of the four flights performed, three corresponded to mild water stress conditions and a single flight was performed under moderate water stress conditions. Under mild water stress, CWSI was not able to capture the differences between FI and RDI trees that were observed with Ψstem. Under moderate stress conditions, CWSI was sensitive to the water deficit reached in the trees and showed significant differences among both irrigation treatments. No differences were observed in the CWSI and NVDI response to water stress among cultivars. Although NDVI and CWSI were sensitive to water stress, the low signal intensity observed in NDVI makes this index less robust than CWSI to monitor crop water stress. It can be concluded that UAV-based CWSI measurements are reliable to monitor almond water status, although for early (mild) levels of water stress, Ψstem seems to be the preferred option.
